We investigated the effects of a high-protein diet and resveratrol supplementation on immune cells changes induced by abdominal irradiation in rats. Female Wistar rats were divided into 5 groups: 1) control diet, 2) control diet with irradiation 3) 30% high-protein diet with irradiation, 4) normal diet with resveratrol supplementation and irradiation, and 5) 30% high-protein diet with resveratrol supplementation and irradiation. We measured blood protein and albumin concentrations, lipid profiles, white blood cell (WBC) counts, proinflammatory cytokine production, and splenocyte proliferation in rats that had been treated with a 17.5 Gy dose of radiation 30 days prior. A high-protein diet affected plasma total cholesterol and very low density lipoprotein-cholesterol levels, which were increased by the radiation treatment. In addition, the lymphocyte percentage and immunoglobulin M (IgM) concentration were increased, and the neutrophil percentage was decreased in rats fed a high-protein diet. Resveratrol supplementation decreased the triglyceride (TG) level, but increased the IgM concentration and splenocyte proliferation. Proinflammatory cytokine production was lower in rats fed a high-protein diet supplemented with resveratrol than in rats fed a control diet. The results of the present study indicate that high-protein diets, with or without resveratrol supplementation, might assist with recovery from radiation-induced inflammation by modulating immune cell percentages and cytokine production.
Twenty cows, by order of calving, were used in a completely randomized $2{\times}2$ factorial experiment. Variables were tow protein levels (14 and 18% crude protein) and concentration of fat (2 and 6% ether extract) in diets. Fat addition, via unprocessed whole sunflower seed, insured forage utilization in diets to meet energy requirement of cows. A total of 36 wks of lactation was subdivided into three 12-wk stages of lactation. Net energy lactation was set at 1.72, 1.57 and 1.42 Mcal/kg for each stage. Higher protein diets improved the efficiency of energy (FCM/net energy intake) which was particularly noted for diets containing high fat (85.7%). However, diets with low protein-high fat resulted in the lowest efficiency (67.7%). No difference in milk yield and butterfat was due to different levels and combinations of protein and lipid in diets. High protein diets depressed blood cholesterol and glucose compared to low-protein counterparts. Relative decline in milk production was slower for lower fat diets than for higher fat groups, especially mid to later stage of lactation. Results of this experiment tend to support our thesis on the synergistic effect of dietary protein and energy (lipid) upon efficiency of lactation.
The experiment was carried out to estimate the energy and protein requirements of Khaki Campbell $\times$ Thai Native laying ducks. To estimate the energy requirement, 5 experimental diets were formulated at levels of 2.70, 2.75, 2.80, 2.85 and 2.90 Mcal ME/kg. These diets were equal in ME/CP ratio (170/l). A total of 150 18-week old laying ducks were assigned to 5 energy level treatments, each comprising 3 replicates of 10 birds each, and they were fed the diets for 18 weeks. To estimate the protein requirement, 90 18-week old laying ducks were divided into 9 groups of 10 birds each, and they were assigned to 3 protein level treatments, each comprising 3 replicates. The levels of protein in the diets were 13.5%, 15.0% and 16.5%, and all diets were isocaloric (2.8 Mcal ME/kg). As a result, the 16.5% protein diet gave significantly better egg production than the 13.5% and 15.0% protein diets, however, no significant difference in egg production was found among the energy levels. Feed cost to produce 1k eggs was lower in the 2.70 Mcal and 16.5%protein diets than in the higher ME and lower protein diets.
This study was performed to investigate effect of dietary protein and calcium levels on cad-mium intoxication in rats. Adult Sprague-Dawley male rate(245$\pm$21g) were blocked into 18 groups of 7 animals according to body weight Nine experimental diets different with protein(40%, 15%, 7%) and calcium (1.3%, 0.6%, 0.1%) levels were prepared. Nine groups of animals were fed each diet with 50ppm cadmium in drinking water and the other 9 groups without cadmium for 30days. Results were summarized as follows: 1) Body weight gain F. E. R(Food Efficiency Ratio) and weights of liver kidney and femur were higher in high protein groups among cadmium exposed groups. 2) Cadmium contents in liver and intestine were higher in rats fed high protein diet or low calcium diet among cadmium exposed groups. Fecal cadmium excretion was highest in high protein-high calcium diet group among cadmium exposed animals. Metallothionein contents in liver kidney and intestine were higher in animals exposed to cadmium and fed high protein diets. 3) Gel filtration chromatography of cytosolic solution showed that the higher dietary protein and calcium levels were the more cadmium was found in metallothionein fractions. 4) No gross histopathological change was seen in liver kidney and intestine of cadmium exposed rats. However a significant increase of smooth endoplasmic reticulum which was alleveated by high protein-high calcium diet was observed. Results obtained indicated that not only high protein diet but also high calcium diet showed preventive effect on cadmium intoxication by increasing the induction of metallothionein syn-thesis and decreasing the cadmium absorption.
Effects of different operating factors including superficial air velocity (SAV), hydraulic residence time (HRT), protein concentration, and foam overflow height on protein removal by a foam fractionator in simulated seawater aquaculture system were investigated. This experiment was conducted on batch and consecutive modes at different combinations of the affecting factors. The foam fractionator had a diameter of 20cm and a height of 120cm and the experiment was conducted with synthetic wastewater. In 5 consecutive trials, protein concentrations in culture tank water decreased faster when the foam fractionator was operated at higher SAVs and lower HRTs. In batch trials, protein removal rates increased with an increase in SAV but decreased with an increase in URT. Higher protein concentrations in the bulk solution resulted in higher protein removal rates. Protein concentrations in the collected foam condensates increased but the foam overflow rates decreased with the increase of foam overflow heights. The results of this experiment indicate that foam fractionation would be an effective way for protein removal in seawater aquaculture systems and the performance of the foam fractionator depends largely on the operating parameters, especially SAV.
To evaluate the quality of fish extracts with spicy vegetables (garlic, onion and ginger) in suppressing fishy oder, fish extracts of crucian carp, loach, bastard halibut and jacopever were processed at 100 $^{\circ}C$ for 6 hours, and their in vitro and in vivo protein qualities were determined . Protein and total lipid contents were closely related to the degree of discarding floated lipid on fish extracts and the kinds of added apicy vegetables . Boiling (10$0^{\circ}C$) , appeared to improve in vitro protein qualities slightly more than hydrocooking (11$0^{\circ}C$), but those with mild processing tended to result in better protein qualities than high temperature cooking (136-14$0^{\circ}C$). Spicy vegetables did not have remarkable effects on improving in vitro protein quality parameters. Fish extracts with 10% ginger were generally higher in in vitro protein quality than with the other vegetables . In spite of higher in vivo protein digestibility of fish extracts containing spicy vegetables processed under mild conditions(10$0^{\circ}C$), PERs of those extracts were not higher htan those of extranct processed at high temperature.
This experiment was conducted to study the effects of blood-mixed and heat-treated protein feeds on protein degradation in the rumen, flow of protein to the abomasums and availability of undegraded protein in the intestine of sheep in a $4{\times}4$ Latin square design. Soybean oil meal, rapeseed meal, and whole soybean were mixed with fresh swine blood and dried at $140^{\circ}C$ for 2 h. Proportionate disappearance of apparently digested OM in the postrumen for the blood and heat treated protein group was ranged from 43.2 to 50.5% as compared with 28.0% for the unheated soybean oil meal diet. The treated protein supplements were resulted in greater total N and NAN flow passing at the abomasums than untreated soybean oil meal diet was fed. The quantities of undegraded feed N passing at the abomasums for the treated protein diets was approximately twice as high as that of the untreated soybean oil meal diet and the estimated amount of undegraded N of the protein supplement itself was 79.1 to 84.2% as compared with 15% of soybean oil meal.
In preparation of textured soybean protein, drying process of the isolated protein affected its gelling property and other related characteristics such as water holding capacity and viscosity. In model systems, denaturation of the protein, as determined in terms of nitrogen solubility index (NSI), was appeared to be a parameter of the gel strength of soybean protein isolate. The gel strength was maximum when the protein was denatured properly during drying process of which the NSI was 43 in this experiment and decreased at either the higher or the lower NSI. It indicated that proper denaturation of the protein during drying operation is advantagous for the preparation of textured soybean protein but not neccesary to make highly undenatured one.
The investigation assessed the influence of Compositae plants consumption on the protein profile in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats by injection of STZ (45 mg/kg body weight) into tail vein. The rats were randomly assigned to five groups: normal and STZ-control fed an AIN-93 diet, and groups whose diets were supplemented with 10% Compositae powder containing Artemisia iwayomogi (A. iwayomogi), Atractylodes lancea (A. lancea) or Taraxacum mongolicum (T. mongolicum). To observe the effects of Compositae plants in the animal model, the levels of protein in liver, kidney, lung, pancreas, and brain were determined after 4 weeks. The level of protein in kidney increased significantly in rats receiving the A. iwayomogi- and T. mongolicum-supplemented diet compared to the STZ-control group. The level of protein in lung was increased significantly in the A. iwayomogi-supplemented group. Blood glucose level correlated well with brain protein level but did not correlate with other protein levels. Also, blood glucose correlated inversely with kidney, lung and brain protein levels. It is suggested that supplementation with A. iwayomogi in diabetic rats leads elevates protein in kidney and lung.
An inhibitor of farnesy-protein transferae is known to be a fgood candidate for antitumor agent that block the oncogenic activity of Ras protein . We recently isolated and characterized dihydrotanshinone I from Salvia miltiorrhiza Bunge (Danshen), an oriental herb, which has an inhibitory activity of toposimerase I to some cancer cell lines. In order to examine the molecular mechanism of dihydrotanshinone I, we studied the farmesy-Protein Transferase activity by dihydrotanshinone I. As a result, we found that result, we found that dihydrotanshinone I showed inhibitory effect on farnesyl-protein transferase with $IC_{50}$ value of 15 ug/ml. This result suggest that dihydrotanshinone I may be an useful anticancer agent with the inhibitory activity of farnesyl-protein transferase.
