Endophytic fungi are little known for their role in gibberellins (GAs) synthesis and abiotic stress resistance in crop plants. We isolated 10 endophytes from the roots of field-grown soybean and screened their culture filtrates (CF) on the GAs biosynthesis mutant rice line - Waito-C. CF bioassay showed that endophyte GMH-1B significantly promoted the growth of Waito-C compared with controls. GMH-1B was identified as Penicillium minioluteum LHL09 on the basis of ITS regions rDNA sequence homology and phylogenetic analyses. GC/MS-SIM analysis of CF of P. minioluteum revealed the presence of bioactive $GA_4$ and $GA_7$. In endophyte-soybean plant interaction, P. minioluteum association significantly promoted growth characteristics (shoot length, shoot fresh and dry biomasses, chlorophyll content, and leaf area) and nitrogen assimilation, with and without sodium chloride (NaCl)-induced salinity (70 and 140 mM) stress, as compared with control. Field-emission scanning electron microcopy showed active colonization of endophyte with host plants before and after stress treatments. In response to salinity stress, low endogenous abscisic acid and high salicylic acid accumulation in endophyte-associated plants elucidated the stress mitigation by P. minioluteum. The endophytic fungal symbiosis of P. minioluteum also increased the daidzein and genistein contents in the soybean as compared with control plants, under salt stress. Thus, P. minioluteum ameliorated the adverse effects of abiotic salinity stress and rescued soybean plant growth by influencing biosynthesis of the plant's hormones and flavonoids.

This is the first report that paromomycin, an antibiotic derived from Streptomyces sp. AG-P 1441 (AG-P 1441), controlled Phytophthora blight and soft rot diseases caused by Phytophthora capsici and Pectobacterium carotovorum, respectively, in chili pepper (Capsicum annum L.). Chili pepper plants treated with paromomycin by foliar spray or soil drenching 7 days prior to inoculation with P. capsici zoospores showed significant (p < 0.05) reduction in disease severity (%) when compared with untreated control plants. The disease severity of Phytophthora blight was recorded as 8% and 50% for foliar spray and soil drench, respectively, at 1.0 ppm of paromomycin, compared with untreated control, where disease severity was 83% and 100% by foliar spray and soil drench, respectively. A greater reduction of soft rot lesion areas per leaf disk was observed in treated plants using paromomycin (1.0 μg/ml) by infiltration or soil drench in comparison with untreated control plants. Paromomycin treatment did not negatively affect the growth of chili pepper. Furthermore, the treatment slightly promoted growth; this growth was supported by increased chlorophyll content in paromomycin-treated chili pepper plants. Additionally, paromomycin likely induced resistance as confirmed by the expression of pathogenesis-related (PR) genes: PR-1, β-1,3-glucanase, chitinase, PR-4, peroxidase, and PR-10, which enhanced plant defense against P. capsici in chili pepper. This finding indicates that AG-P 1441 plays a role in pathogen resistance upon the activation of defense genes, by secretion of the plant resistance elicitor, paromomycin.
Inhibition of flowering in Lemna perpusilla 6746 by 30 mM sucrose was reversed by the addition of acetylcholine (>$10^{-4}M) supplemented with 10^{-4}M$ ascorbic acid to 1/10-strength Hunter's growth medium. The reversible effect of acetylcholine was found to be greater at early stages of flowering than in the later period. Promotive effects of both acetylcholine ($10^{-3}M) and eserine(10^{-5}M$) on flowering in the short-day plant under various photoperiodic conditions were studied. It was indicated that the application decreased length of the critical dark period for the floral induction, and it was also shown that the endogenous status of acetylcholine was involved in the floral response which had a correlation with phytochrome. Interruption of inductive dark periods by red irradiation (1min) immediately followed by far-red light (1 min) completely inhibited flowering, while the addition of acetylcholine and eserine to the medium under the same condition slightly promoted flowering, indicating possible involvement of phytochrome system in acetylcholine activity for photoperiodic sensitivity of floral response in Lemna perpusilla 6746.
This experiment was carried out to establish a year-round production system of pathogen-free stock through micropropagation in Fritillaria thunbergii as medicinal bulbous plant. The effect of different types of culture method and plant growth regulators, activated charcoal and mannitol on bulblet formation and subsequent growth were investigated. The MS solid medium containing 1. 0 mg/L kinetin and 0. 3 mg/L NAA was effective on bulblet formation and propagation rate compared to liquid and suspension culture. Addition of activated charcoal at 0. 01% to 0. 1% in the medium promoted bulbing of cultured bulblets and shoot formation. Addition of 1% to 2% mainnitol in MS medium was effective on the formation of bulblet and subsequent growth of bulblets compared to control. In addition of inhibitors, $10{\sim}100\;mg/L$ B-9 and Chloromequat had effective to stimulate bulblet growth but their effects were not so much as mannitol treatment. ABA treatment had detrimental effect on survival rate of explant and bulblet formation.
BACKGROUND: Light-emitting diodes (LEDs) with lower electric cost and the specific wavelength have been considering as a novel light source for plant production in greenhouse conditions as well as in a closed culture system. Supplementary lighting for day-length extension was considered as light intensity, light quality, and/or photoperiod control on plant growth and development. Effects of supplementary blue or red LED radiation with lower light intensity on growth of Ageratum (Ageratum houstonianum Mill., cv. Blue Field), African marigold (Tagetes erecta L., cv. Orange Boy), and Salvia (Salvia splendens F. Sello ex Ruem & Schult., cv. Red Vista) were discussed during sunrise and sunset twilight in the experiment. METHODS AND RESULTS: Supplementary lighting by blue and red LEDs for 30 (Treatment B30; R30) or 60 (Treatment B60; R60) min. per day were established in greenhouse conditions. Photosynthetic photon flux for supplementary radiation was kept at $15{\mu}mol\;m^{-2}\;s^{-1}$ on the culture bed. Natural condition without supplementary light was considered as a control. The highest shoot and root dry weights were shown in African marigold exposed by red light for 60 min. per day. Supplementary blue and red lighting regardless of the radiation time significantly stimulated development of lateral branches in African marigold. Stem growth in Ageratum and Salvia seedlings was significantly promoted by red radiation as well as natural light. CONCLUSIONS: Extending of the radiation time at sunrise and sunset twilight using LEDs stimulated reproductive growth of flowering plant species. Different characteristics on growth under supplementary blue or red lighting conditions were also observed in the seedlings during supplementary radiation.
For determining the effect of extracts exudated from ten industrial plant resources on germination and early growth of shoot and root of radish the allelopathic compounds screened. Germination rate and seedling growth of radish differed depending on plant species and extract concentration. Lower levels of 1% extracts exudated from industrial plant resources promoted the rate of germination of radish while higher levels of 10% extract inhibited the rate of germination. Germination rate, shoot growth and root growth of radish in higher concentration of extract also differed depending on the plant species exudated and extract concentraton of industrial plant resources. Higher concentration of extracts from Xanthium strutnarium L. Angellica gigas Nakai, and Solanum nigrum L. were completely inhibited the germination while those from Cichorium intybus L., and Polygonatum officinale Allioni were not inhibited. Extracts exudated from Xanthium strutnarium L, Angellica gigas Nakai, and Solanum nigrum L. showed the strong inhibition of shoot and root growth of radish while those from Cichorium intybus L., and Polygonatum officinale Allioni did not show the bioactivity in shoot and root growth of radish..
To establish the mass propagation methods of Weigela subsessilis,, a promising native plant species for horticultural use, several factors influencing rooting from stem cuttings were evaluated. Softwood cuttings showed best rooting rate, 70~77%, in all the cutting media tested, 2.8 times more than hardwood cuttings in which perlite was best medium. The rooting from hardwood cuttings was promoted by higher concentration of all growth regulators used,500 mg.L$^{-1}$ NAA being the best with 80%. More than 80% of softwood cuttings treated by growth regulators produced roots, especially 100, 500, and 1000mg.L$^{-1}$ NAA producing 97%. Roofings from semi-wood cuttings was enhanced by all the growth regulators, except 1000 mg.L$^{-1}$ concentration in which rooting was reduce. Higher rooting rate was obtained by higher concentration and longer soaking duration of NAA in case of hardwood cutting. Softwood cutting showed 100 % rooting by soaking treatment with 500 mg.L$^{-1}$ NAA for 3 min. Rooting rate of semi-hardwood cuttings was promoted by higher concentration and longer soaking with growth regulators, However, the degree of improvement was lower than soft and hardwood cutting.
The influences of light generated by LEDs on shoot growth and photosynthesis of Tea plant(Camellia sinensis L.) were evaluated. The growth characteristics were investigated after 45 days of culture under four different light qualities: fluorescent lamp, red LED, blue LED, red+blue+white LED. Shoot growth was promoted by red light, especially root length and area were further promoted under the red+blue+white LED. Also, T/R ratio and Chlorophyll content were highest in red+blue+white. Fluor Cam was used to measure the fluorescence images of the plants, inhibition of photochemical efficiency(Fv/Fm) were not changed in all treatment. However, non-photochemical quenching(NPQ) were found rapidly increasing in blue LED, these results were that blue LED were inhibit photosynthetic efficiency and must be considered for efficiently in vitro cultivation of the tea plant. The above results suggest that light qualities could be an important factor to foster in vitro growth of the species. Also, In order to produce healthy plants, it is effective to using light qualities of red+blue+white LED on in vitro culture of the tea plant. These results could be used to mass propagating shoot and produce of healthy seedling.
This study was conducted to evaluate the effect of mulching material and planting density on growth and bulb development of shallot. The transparent PE film was better than black PE film as a mulching material for the overwintering shallot crop. Transparent PE film mulching promoted plant growth and increased marketable yield by 21% as compared with that of the black PE film-mulched crop. However, the bulb size was not significantly affected by the type of mulching film. The height and width of ridge and planting density significantly affected the growth and bulb yield of the moisture sensitive shallot. Bulb yield of the shallot planted in five rows in 120 cm wide ridges (20,833 plants per 10 a) was 1,332 kg per 10 a, which was 1.7 times as high as that by the crop grown in three rows in 120 cm wide ridges (12,500 plants per 10 a). In conclusion, the shallot crop is recommended to be cultivated in five rows in 120 cm ridges mulched with the transparent PE film.
