Aurora A kinase is a mitotic serine/threonine kinase whose proposed functions include the maturation of centrosomes, G2/M transition, alignment of chromosomes at metaphase, and cytokinesis. In this study, we investigated the effect of MLN8237, an aurora A kinase inhibitor, on the postovulatory aging of oocytes based on the frequency of oocyte fragmentation, cdk1 kinase activity, and cyclin B degradation. The fragmentation of ovulated oocytes during prolonged culture was inhibited by treatment with MLN8237 in a concentration-dependent manner. The frequency of fragmented oocytes was significantly lower in oocytes treated with 2 ${\mu}M$ MLN8237 (13%) than in control oocytes (64%) after two days of culture. Most of the control (non-fragmented) oocytes (91%) were activated after two days of culture. In comparison, only 22% of the MLN8237-treated oocytes were activated; the rest of the oocytes (78%) were still in metaphase with an abnormal spindle and dispersed chromosomes. Next, cdk1 activity and the level of cyclin B were examined. The level of cyclin B and cdk1 activity in MLN8237-treated oocytes were nearly equal to those in control oocytes. Our results indicate that MLN8237 inhibited the fragmentation of ovulated oocytes during prolonged culture, although it blocked the spontaneous decrease in activity of cdk1 and degradation of cyclin B. This mechanism of inhibition is different from that in oocytes treated with nocodazole, which have high levels of cdk1 activity and cyclin B.
This study was carried out to obtain the basic information about the large scale propagation of ginseng (Panax ginseng C.A. Meyer). Therefore, the stem cuttings of 1-year old ginseng, treated with various concentrations of plant growth regulators for 5 seconds (quick dipping) and 24 hours (prolonged soaking), were cultured. The root formation of stem cuttings was varied with the concentrations, kinds, and treatment methods of plant growth regulators. Besides normal-looking roots various malformed roots were observed. In the prolonged soaking method, the culture of stem cuttings, treated with 10 ppm of IBA or NAA, resulted in profuse root regeneration. And stem cuttings, in quick dipping method, treated with 2000 ppm of IBA or NAA resulted in more excellent root regeneration. In general, IBA was more vigorous for the root formation than NAA, The treatment with 50 ppm kinetin or 100 ppm BA brought good result for the retardation of senescence of stem cuttings and BA treatment was more effective than kinetin. As for the saponin content of roots derived from stem cutting culture, the roots, formed by non-treatment of growth regulators, were higher in saponin content than those formed by treatment of growth regulators.

The ratio of indirubin/indigo was highest under conditions of prolonged culture and reduced culture oxygenation.

The aim of this study was to evaluate different methods for Trichomonas vaginalis diagnosis during pregnancy in order to prevent maternal and perinatal complications. A total of 386 vaginal exudates from pregnant women were analyzed. T. vaginalis was investigated by 3 types of microscopic examinations direct wet mount with physiologic saline solution, prolonged May-Grunwald Giemsa (MGG) staining, and wet mount with sodium-acetate-formalin (SAF)/methylene blue method. PCR for 18S rRNA gene as well as culture in liquid medium were performed. The sensitivity and specificity of the microscopic examinations were evaluated considering the culture media positivity or the PCR techniques as gold standard. The frequency of T. vaginalis infection was 6.2% by culture and/or PCR, 5.2% by PCR, 4.7% by culture, 3.1% by SAF/methylene blue method and 2.8% by direct wet smear and prolonged MGG staining. The sensitivities were 83.3%, 75.0%, 50.0%, and 45.8% for PCR, culture, SAF/methylene blue method, and direct wet smear-prolonged MGG staining, respectively. The specificity was 100% for all the assessed methods. Microscopic examinations showed low sensitivity, mainly in asymptomatic pregnant patients. It is necessary to improve the detection of T. vaginalis using combined methods providing higher sensitivity, such as culture and PCR, mainly in asymptomatic pregnant patients, in order to prevent maternal and perinatal complications.
The color difference value of fabrics dyed with browning extracts from mushroom were as follows. 1. Two hours of dyeing depending on pH at 95℃ exhibited colors of YR and Y. The color of wool and nylon was darkest brown at pH 4, but light brown or yellow as pH increased. The color of silk was darkest brown at pH 5 and pH changes resulted in the same tendency as in wool. 2. In wool the color was changed to darker brown as time prolonged, however, there was no further color change after 9hr and value and chroma of wool declined. 3. At pH 4, the increase in temperature turned colors of sil, wool and nylon into dark brown from light yellow. 4. As the result of repetitive dyeing, colors of silk, wool and nylon gradually turned into darker brown. The repetitive dyeing was more effective in dark color and high colorfastness compared to a prolonged dyeing. 5. The amount of mordant resulted in color changes between light yellow and dark brown without various color changed. 6. In color change according to methods of mordant, post-mordant with CuSO₄·5H₂O and with SnCl·2H₂O showed colors of green and light yellow, respectively.
In this study, the changes in fashion consumption culture in the post-COVID-19 era were examined through big data analysis. Considering that the Chinese market plays a pivotal role in the global fashion industry, big data was collected in the most famous and professional fashion network in China, LADYMAX.cn. As a result of text mining and social network analysis, three major changes were identified as the emerging fashion consumption culture in the post-COVID-19 era. First, as a trend in new media consumption, COVID-19 disease and the development of digital technology tended to encourage consumers to put more importance on the relationship between bloggers and fans than previously. Second, as a trend in reward consumption, consumers tended to be rewarded for their hard work to relieve and comfort their high stress caused by spending a long time worrying about the prolonged COVID-19 situation. Third, as a trend in home-economy consumption, consumers tended to prefer homewear and sportswear more because they were spending longer times at home as the social distancing period was prolonged.

Antithrombotic and fibrinolytic activity of natto was evaluated on platelet aggregation in vitro and in vivo. Natto showed inhibitory effects on platelet aggregation induced by adenosine 5’diphosphate (ADP) and collagen. Orally administered natto also showed fibrinolytic activity in hypercholesterolemia rats. Normal levels of natto, when administered for four weeks, shortened euglobulin clot lysis time (ECLT) and prolonged partial thromboplastin time (PATT) significantly compared to non-treated group. In addition, the natto treatment decreased total cholesterol in serum. These results showed that intake of normal levels of natto can elicit antithrombotic and fibrinolytic effects, suggesting its consumption may improve blood circulation.
Seventy four microorganisms, which have antagonistic activity against to Fusarium culmorum, were tested for their inhibitory effect on colony development of obligate biotroph Erysiphe graminis f. sp. hordei Marchal, the causal agent of powdery mildew on barley plants. Of these, 13 actinomycetes isolates were shown to reduce the colony development of mildew completely by application of their 10% cell-free culture filtrates on barley leaves. An Isolate, A252, was the most powerful antagonist and its antifungal activity was further assessed. The colony development of mildew was significantly reduced by application of the 1% cell-free culture filtrate of isolate A252. In comparison to the control, the protective and curative application of 10% cell-free culture filtrate from A252 showed 88.5% and 96.1% reduction of colony numbers respectively. By the protective application, 68.3% of the inhibition was observed after 9 days of treatment, thus showed prolonged inhibitory effect. In vitro test, complete inhibition of the mycelial growth of Microdochium nivale was achieved by the treatment of 1% A252 culture filtrate and 80.2% of inhibition was observed by the 0.1% treatment.
Biodegradation of an explosive compound, glyceryl trinitrate (GTN), was studied with a denitrifying microbial culture grown in a sequencing batch reactor and a GTN acclimated denitrifying culture. The GTN acclimated culture, which were fed on GTN for 1 month, degraded GTN regioselectively via denitration on C1 position as compared to C2 position denitration by denitrifying culture that has never been exposed to GTN. Accumulation of two isomeric glyceryl dinitrates (GDNs) in both culture medium suggests that GDN denitration is the rate-limiting step in GTN biodegradation. The first order GTN degradation rate normalized to cell concentration of the acclimated culture was calculated to be 0.045 (${\pm}0.002$) L/g-hr. Increasing concentration of electron acceptor(nitrate) resulted in discouraged GTN degradation. According to microbial community analysis, prolonged GTN exposure resulted in 25% increase in the genus level of the GTN acclimated culture with the disappearance of two dominating denitrifying microbial species of Methyloversatilis universalis and Hyphomicrobium zavarzinii in the denitrifying culture.
