  • The amino acids in the mucilage and its hydrolyzed products from Ulmus coreana, Nakai root were detected by thin layer chromatography. The mucilage contained 10 kinds of amino acids; alanine, aspartic acid, glutamine, glutamic acid, hydroxyproline, isoleucine, leucine, proline, phenylalanine and tyrosine. In the hydrolyzed mucilage 14 kinds of amino acids were detected; alanine, aspargine, arpartic acid, glutamine, glutamic acid, hydroxyproline, isoleucine, leucine, methionine, proline, phenylalanine, serine, tyrosine and valine.

Change in Photosynthesis, Proline Content, and Osmotic Potential of Corn Seedling under High-Saline Condition

  • To identify salt-tolerance characteristics of corn seedling was treated in solution of 0, 50 and 100 mM NaCl of hydroponic cultivation. In photosynthesis of corn seedling, there was no large difference between 50mM and 0 mM NaCl solution, however, in 100 mM NaCl solution, the tolerance gradually decreased to $76\%,\;49\%,\;and\;31\%$ after one day, four days, and seven days, respectively, in comparison to 0 mM NaCl solution. Osmotic potential of corn in seedling period was significantly decreased with increasing saline level, however, free proline content in the plant on the ground was significantly increased with increasing saline level and with the lapse of time. In terms of correlation among major characteristics, there was a highly significant positive difference between osmotic pressure potential and photosynthesis, However, highly negative correlation was found between osmotic pressure potential and free proline content. In addition, it was expected that young seedling of corn with saline tolerance may be utilized in the transplantation in salt-accumulated land. Based on above-shown result, in terms of saline tolerance of Chalok-2 variety, growth suppression was serious with 100mM NaCl solution. However, growth was expected that seedling growth would be favorable under 50 mM NaCl solution.

  • The distribution of water soluble free amino acids in various parts of sheep muscles were observed. amino acids of cystine, histidine, aspartic acid, arginine, hydroxy proline, threonine, glutamic acid alanine, proline, methionine, tryptophan, leucine, iso-leucine, glycine, creatine, and two or four different kinds of amino acid content in each tissues, M. longissimus. Overall, the distribution of content of amino acids are seemed to be different for each tissues, while for hydroxyproline the result was negative.

  • We prepared a simple, economic and versatile preparative system for an enantiomeric separation. Hydrophobic amino acid enantiomers were resolved in the preparative scale by a centrifugal liquid chromatography on the N-hexadecyl-L-proline coated reversed phase. The factors controlling the retention and resolution of racemic amino acids such as the concentration of Cu(Ⅱ), pH of the eluent, the type and content of organic modifier, and rpm of CLC were examined. Several mg of hydrophobic amino acid enantiomers were separated preparatively. To separate all of different amino acid enantiomers, various coating material will be investigated.

  • Oligopeptidase B (OpdB) is a serine peptidase widespread among bacteria and protozoa that has emerged as a virulence factor despite its function has not yet been precisely established. By using an OpdB-overexpressing Escherichia coli strain, we found that the overexpressed peptidase makes the bacterial cells specifically less susceptible to several proline-rich antimicrobial peptides known to penetrate into the bacterial cytosol, and that its level of activity directly correlates with the degree of resistance. We established that E. coli OpdB can efficiently hydrolyze in vitro cationic antimicrobial peptides up to 30 residues in length, even though they contained several prolines, shortening them to inactive fragments. Two consecutive basic residues are a preferred cleavage site for the peptidase. In the case of a single basic residue, there is no cleavage if proline residues are present in the $P_1$ and $P_2$ positions. These results also indicate that cytosolic peptidases may cause resistance to antimicrobial peptides that have an intracellular mechanism of action, such as the proline-rich peptides, and may contribute to define the substrate specificity of the E. coli OpdB.

Proline and Ammonia Accumulation in the Zoysiagrass Infected with Large Patch (라지 팻치에 감염된 잔디에서 프롤린과 암모니아의 축적)

  • To investigate the response of proline and ammonia to pathogen infection, plant growth and relevant chemical component were examined in large patch-infected or healthy (control) zoysiagrass during 6 days after treatment. Pathogen-infection increased root mortality by 30% compared to control. Soluble protein was not significantly affected by pathogen-infection except in the leaf at day 6. Ammonia concentration also increased significantly in both leaves and roots of pathogen-infected plants. Proline concentration in leaves and roots increased to 3.4- and 4.5-fold, respectively, compared to those of control at day 6. These results suggest that proline accumulation may be a sensitive biochemical indicator representing the stress intensity caused by pathogen infection in zoysiagrass.

  • The Rhizobium and Azospirillum spp. were isolated from the root nodules of several leguminous plants and rhizosphere of various paddy rice varieties. The growth of the nitrogen-fixing strains isolated was largely inhibited in yeast extract-mannitol medium (AMA) containing 0.6 M NaCl. In response to osmotic stress, the nitrogen-fixing strains accumulate intracellular free glutamate. The growth and nitrogenase activity of Rhizobium and Azospirillum were increased by addition of osmoprotectants such as proline, glycine betaine, and glutamate during salt stress. Glycine betaine was the most effective among exogenous osmoprotectants tested. In the absence of sodium chloride, nitrogenase activity seem to be slightly decreased by the presence of the proline or glycine betaine. These results revealed that nitrogenase activity was repressed by fixed nitrogens such as proline or glycine betaine.

Evaluation of biochemical and free radical scavengers of Digitaria exilis L. under osmotic stress

  • Digitaria exilis L. is an under-utilized crop with high nutritional and medicinal values. It thrives in and is well-adapted to arid areas with low soil nutrients. Using biochemical markers, this study investigates the mechanisms by which D. exilis responds to osmotic stress. Three accessions Dinat Iburua (DIN), Jakah Iburua (JAK) and Jiw Iburua (JIW) were collected from National Cereal Research Institute, Niger State. Two accessions, NG/11/JD/061 and NG/11/JD/062 were also collected from National Centre for Genetic Resources and Biotechnology, Ibadan. Murashige and Skoog medium of approximately 1.2 L was supplemented with polyethylene glycol 6000 to create osmotic pressures of -9.29, -13.93, -20.13, -26.32, -32.51, and 0 MPa (control). Sterilized seeds were inoculated in the medium and placed in the growth room for 4 weeks. Proline accumulation was significantly high in all JAK plants under osmotic stress. Proline and ascorbate peroxidase (p<0.05) activities were directly correlated, thus reinforcing the survivability of JAK during stress. Catalase (CAT) activity was also significantly induced in JAK under osmotic stress, which synergistically improved its tolerability. As a result, >50% of OH-, H2O2, and NO radicals were scavenged. However, other accessions including DIN, NG061, NG062, and JIW showed variations in their responses to different levels of osmotic stress, although not significant. Therefore, JAK possesses a well-equipped free radical quenching system that is protected by the accumulation of the osmolyte proline; therefore, accession JAK is considered osmotolerant. CAT and superoxide dismutase activities were osmostabilized against oxidative stress by proline.

Influence of Aluminum Chloride on Growth and Chemical Components of Rice Seedling (염화알루미늄 처리가 벼 유묘생육 및 체내 성분함량에 미치는 영향)

  • The experiment was carried out to determine Al tolerance ranges of two rice cultivars(Ilpumbyeo and Hyangmibyeo) during germination and early growth. The amount of 0, 300, 600 and 900ppm Al were treated in the germination media. Plant height and root length of rice cultivars were decreased significantly with Al concentration. The reduction of growth was more remarkable in root than in shoot. Germination rates in all Al concentrations were decreased comparing with untreated control. Two rice cultivars were not different in Al concentrations. As Al concentration was increased, chlorophyll content were decreased. Ca and Mg contents in 300ppm of Al treatment were sharply increased. Free proline contents of both rice cultivars grown at the 300, 600, and 900ppm of Al treatments were increased only in the 3 days after Al treatments.

Effect of NaCl Stress on Inorganic Ion, L-Proline, Sugar and Starch Content of Soybean Seedlings

  • This study conducted to elucidate the change of the cation content (Na$^{+}$, $K^{+}$, and $Ca^{2+}$), the L-proline content, and the sugar and starch content in the stems, roots, and leaves of three cultivars of the 30 days old seedling soybeans (Glycine max L. cv. Danwonkong, Hwang-keumkong, and Kwangankong) after 100 mM NaCl stress containing 1/2 Hoaglands nutrient solution in the sand culture. The reduction of the dry matter weight after 100 mM NaCl treatment among cultivars was higher in the order of Kwangankong, Danwonkong, and Hwang-keumkong. The highest reduction of the dry matter weight was occurred at the roots among three parts of plant. The Na$^{+}$ content increased with NaCl treatment in overall and specially greatly increased in roots and stems. The $K^{+}$ and $Ca^{2+}$ content decreased with NaCl treatment at the roots and stems. The $K^{+}$ content, however, at the leaves increased in all three cultivars with the NaCl treatment. The L-proline content with NaCl stress increased greatly. The increment of the L-proline content at the stems and roots of Hwangkeumkong was lower than that of other two cultivars, K wangankong and Danwonkong. The sugar content decreased with NaCl treatment at the stems and leaves. The starch content also decreased at the stems and leaves with NaCl treatment.aCl treatment.
