Optimal Conditions of Reaction Flavor for Synthesis of Crab-like Flavorant from Snow Crab Cooker Effluent (홍게 자숙액으로부터 게향 제조를 위한 반응향의 최적화)

  • To develop a crab-like flavorant from snow crab cooker effluent (SCCE, $20^{\circ}Brix$), optimal reaction conditions were determined using response surface methodology (RSM) combined with reaction flavoring technology (RFT). Using five variables (proline, glycine, arginine, methionine, fructose), RSM based on a five-level central composite design was applied to evaluate sensory acceptance (odor, taste, and overall acceptance) as dependent variables. A model equation obtained from RSM showed 0.88 of R-square for odor, 0.90 for taste, and 0.95 for overall acceptance with 0.07 lack of fit in overall acceptance (P<0.05). Odor score (predicted value) was 7.21 in the saddle point. Optimal flavoring conditions for making a crab-like flavorant were as follows: addition of 0.29 g of proline, 0.63 g of glycine, 0.61 g of arginine, 0.02 g of methionine, and 1.07 g% (w/v) of fructose into SCCE with RFT (90 min at $130^{\circ}C$). Odor score obtained under optimal conditions was 7.56, which was higher than the predicted value.

Angiotensin I Converting Enzyme Inhibitory Activity of Hot-Water Extract and Enzymatic Hydrolysate of Fresh Water Fish (담수어 열수추출물 및 효소가수분해물의 Angiotensin I 전환효소 저해작용)

  • Hot-water extract and enzymatic hydrolysate prepared from fresh water fish such as carp, snakehead, eel and israeli cart were tested for inhibitory activity against Angiotensin I converting enzyme(ACE). ACE inhibitory activity of enzymatic hydrolysate was higher than that of hot-water extract, and was the highest in enzymatic hydrolysate of carp among the tested samples. ACE inhibitory activity of 70% ethanol soluble fraction was higher than that of precipitated fraction, the highest in enzymatic hydrolysate of carp. Molecular weight of active fraction was about 1,400 in hot-water extract and slightly above in enzymatic hydrolysate. Amino acid of active fraction of hot-water extract was abundant in glycine, alanine, leucine and proline, whereas amino acids of aspartic acid, glutamic acid, glycine, alanine, valine, leucine and proline were abundant in enzymatic hydrolysate. $IC_{50}$/(amounts of inhibitors need for 50% inhibition) of hot-water extract was the range of 50.3~56.9$\mu\textrm{g}$, those of enzymatic hydrolysate 42.6~57.7$\mu\textrm{g}$.

In Vitro Test on Allelopathic Effects of Leaf Extracts from Phytolacca americana and Armoracia rusticana (미국자리공 및 겨자무 잎 추출물의 Allelopathy 효과 기내 검정)

  • Allelopathic effects on some crops(rice, barley, wheat, Chinese cabbage, leaf mustard, onion, welsh onion, tobacco, red pepper) for extracts of Phytolacca americana and Armoracia rusticana were investigated in MS solid medium. Germination percentage, radicle length, hypocotyl length, plant height, fresh weight, dry weight, and seedling vigor were rapidly inhibited by methanol extract from Phytolacca americana according to increase of the concentration in test plants. Autotoxicity of Phytolacca americana began to appear at concentrations greater than 50$\mu$l methanol extracts per 20ml medium. The fresh weight and dry weight of Phytolacca americana were also decreased by its own methanol extract according to increase of the concentration. Initial amounts of growth of all crops were also inhibited by methanol extracts from Armoracia rusticana, but the inhibitory effects were lowered than those of methanol extracts of Phytolacca americana. Of four fractions, ethyl acetate, petroleum ether, $1^{st}H_2O, 2^{nd}H_2O$, reextracted from methanol extracts of Phytolacca americana, the ethyl acetate fraction showed the highest allelopathic effects on germination percentage and initial amounts of growth. Chlorophyll contents of rice, barley and Chinese cabbage were more inhibited in the ethyl acetate fraction than in the other fractions. Free proline content of Chinese cabbage was increased 31.2 times in the 100$\mu l$ ethyl acetate fraction, and the contents of rice and barley were also increased according to concentration levels in the ethyl acetate fraction.

Effects of Copper and Zinc on Germination, Chlorophyll and Organic Compounds in Seedling of Rice (Oryza sativa L.) (구리와 아연 처리가 벼의 발아, 유묘의 엽록소와 유기산 함량에 미치는 영향)

  • The experiment was conducted to elucidate the growth response of rice seedling to heavy metals, copper and zinc. The copper and zinc concentration in rice seedling was 50mg/l and 100mg/l, respectively. Germination. chlorophyll and organic compounds were investigated in seedling of rice with or without heavy metals. Root growth and germination were inhibited at 50mg/l Cu and at 100mg/l compared to control and such reduction in growth was more remarkable at 100mg/l Zn than at 50mg/l Cu. Chlorophyll content was slightly reduced at both 50mg/l Cu and 100mg/l Zn. Malic acid was remarkably increased at 50mg/l Cu compared with that at Zn 100mg/l regardless of rice cultivars. The content of organic acids such as succinic, citric, and malic acid and free proline was increased with treatment of copper and zinc.

Growth and Physiological Characteristics in a Halophyte Suaeda glauca under Different NaCl Concentrations (염생식물 나문재의 염농도에 따른 생장 및 생리적 특성)

  • This research was carried out to investigate the plant growth, inorganic ion and amino acid content characteristics in a halophyte, Suaeda glauca, under different NaCl concentrations for cultivating in the reclaimed land. S. glauca was hydroponically cultivated under 0, 50, 100, 200, 300 and 400 mM NaCl concentrations with Hogland's nutrient solution. To evaluate growth response under different NaCl concentrations, plant height and number of branches, dry weight, Fv/Fm value, and photosynthetic efficiency were investigated. To find out physiological characteristic, inorganic ion contents and amino acids in the plant were evaluated. The optimum concentration of NaCl for plant growth were 50 mM. The plant growth were gradually decreased in the concentration ranged from 100 to 400 mM. As increasing of NaCl concentration, Na ion was increased, but K, Ca, Mg ions were decreased in the plant. The amino acid contents were varied due to NaCl concentrations, but most of amino acids content in total plant was the lowest at 50 mM. Conversely, proline was exceptionally high at 50 mM of NaCl concentration. The Fv/Fm value was the highest at 50 mM of NaCl concentration. From these results, the optimum salt concentration for the growth of S. glauca was 50 mM, but the plant seems to adapt in a variety of salt environments in view of the change of ions and amino acids depending on salt concentration and the maintenance of photosynthetic efficiency even under high salt condition.

Grain Yield and Physiological Responses of Water Stress at Reproductive Stage in Barley (보리 생식생장기의 수분부족이 수량 및 몇 가지 생리적 반응에 미치는 영향)

  • To cope with increasing importance of water stress in food crop production, some physiological characteristics, their cultivar-differences and grain yield of winter barley cultivars in response to water stress during reproductive stages were studied employing three covered-barley cultivars, Milyang 12, Durubori, and Olbori, one naked-barley cultivar, Baegdong, and one two-row malting-barley cultivar, Hyangmaeg. The barley grown in pot-soil was conditioned for 10 days under water stress, varying the time of water stress : 20 days before heading, 10 days before heading and the time of heading. The decrease in growth due to water stress varied greatly with the cultivars and time of water stress. The greatest injury occurred when water stress was imposed for 10 days from 10 days before heading : the culm length of water-stressed plants have shown reduced by 85∼98% of the non-stressed; the number of spikes per plant by 52∼83%; the number of grains per spike by 71∼86%; 1,000-grain weight by 80∼84%; yield per pot by 60∼94%. The number of spikes per plant as one of yield components was most sensitively affected. As a whole, the drought resistance of cultivars was high in the order of Olbori> Milyang 12 and Durubori> Hyangmaeg>Baegdong. On rewatering the plants after termination of the water stress treatment the recovery rate of free proline content and relative turgidity of flag leaf were higher in 3 covered-barley cultivars, and lower in cultivars Baegdong and Hyangmaeg.
