Immunohistochemical observations of proliferating cells in distal epiphyseal tissue of chicken femurs (닭의 대퇴부 골단조직의 세포증식에 대한 면역조직화학적 관찰)

  • The present study was focussed to assess the proliferating cells in the distal epiphyseal tissue of the chicken femur by immunohistochemical staining methods. Four chickens were administrated intraperitoneally by twice consecutive injections, 1 day interval with bromodeoxyuridine(Brdur, 0.05 mg/gm BW/time), and then were killed by exsanguination of jugular vein at 2 hours after last injection. Samples were taken from femur distal epiphyseas of chicken. Labeling indexes(LI) were calculated as the ratio of the number of anti-Brdur monoclonal antibody-labeled cells in the each tissue layers from basal layer of the integument to bone marrow. The overall LI were found to be $13.90{\pm}3.44%$, $30.03{\pm}7.52%$, $16.00{\pm}9.41%$, $0.00{\pm}0.00%$ and $60.03{\pm}13.39%$ at basal layer of integument, perichordrium, reseving zone in cartilage, hypertrophic zone in cartilage and bone marrow respectively. LI in proliferating zone of cartilage were found to be $36.99{\pm}7.59%$, $32.83{\pm}5.38%$ and $22.02{\pm}6.27%$ at reserving zone side region, middle region, and hypertrophic zone side region respectively. The tissue layers with higher LI were odered as bone marrow, reserving zone side region in proliferating zone, middle region in proliferating zone, perichondrium, hypertrophic zone side region in proliferating zone. reserving zone of cartilage and basal layer of integument. These data indicate that the overall LI in the each tissue layer of distal epiphyseas of the chicken femur were concluded to be higher than that in another tissue of adult birds but hypertrophic zone of cartlage were appeared to be not proliferating cells.

  • This study was desinged to investigate the effect of estrogen(Est) on the proliferating of progesterone(Prog) target cells. The spayed 13 rats(Wistar, approximately 300gm) were randomly alloted into 3 groups. One group was the control group and another Prog-treated group was injected with 1mg of Prog/rat/day for 2 consecutive days, and Estand Prog-treated group was injected intramuscularly with $17{\beta}$-estradiol $20{\mu}g/rat/day$ for 3 consecutive days and then with Prog for 2 days as above from 4th day. Rats were administrated intraperitoneally with bromodeoxyuridinc(Brdur,0.2mg/BW once) befero 2 hours of exanguination. In gross finding, the groups with more level of dimension and weight on the uterus were ordered as Est- and Prog-treated group, Prog-treated group and control group. The investigation by immunohistochemical methods using paraffin sections of the uteri was performed by using anti-Brdur antibody for labeling proliferating cells of Prog target cells. The groups with higher labeling index(LI) were ordered as Prog-treated grop, Est- and Prog- treated group and control group. The number of proliferating cells from Prog target cells in the rats were rather deceased by Prog injection following Est injection than prog injection only. The cell types with higher LI in the wall layers of all 3 groups were ordered as endometrial stromal cells, glandular epithelial cells, luminal epithelial cells, myometrial muscle cells and serosa methodelial cells, and the region with highest LI was functional zone of the endometrium and the region with lower LI was muscular layer and then those with lowest LI was serosa and also the considerable different LI from individual rat were observed.

Ginsenosides Promote Proliferation of Cultured Ovarian Germ Cells Involving Protein Kinase C-mediated System in Embryonic Chickens

  • The effect of ginsenosides (GS) on germ cell proliferation was evaluated with a chicken ovarian germ-somatic cell coculture model and the mechanism involving protein kinase C (PKC) pathway was investigated. Ovarian cells were cultured in serum-free McCoy's 5A medium and challenged with GS alone or in combinations with PKC activator (phorbol 12-myristate 13-acetate, PMA) or inhibitor ($H_7$) for 48 h. The number of germ cells was counted and the proliferating cells were identified by immunocytochemistry of proliferating cell nuclear antigen (PCNA). Results showed that GS significantly increased germ cell proliferation and this stimulating effect was further increased by PMA, but inhibited by H7, in a dose-dependent manner. Moreover, GS-elevated PCNA expression and the PCNA -labeling index of germ cells displayed similar changes with the increased numbers of germ cells. These results indicated that GS stimulated proliferation of ovarian germ cells with involvement of the PKC-mediated system.

Observations of a endometrial stromal sarcoma in the rat uterus wall (Rat의 자궁벽에 발생된 stromal sarcoma의 관찰)

  • The present observation was focussed mainly on the histopathological and immunohistochemical finding of the tumor developing from the outside of rat uterus wall. In gross finding, this tumor were a oval form mass and 0.75cm and 1cm in greatest demension. The sectioned surface showed uniform, solid, gray-white. In histopathological finding, the tumor showed the form of lymphoid nodule-like structure and was composed of the cells with karryorrhexis, karyolysis and necrosis in the central zone, mature cells in the paracentral zone, and concentric layers of compressed, flattened and atrophied smooth muscle fibers by the proliferating of the cells closely resembling endometrial stromal ells in the peripheral zone and a few smaller nodule-like structure in certain areas of the peripheral zone. In immunohistochemical observations, the proliferating cells presents only among the concentric layers of the smooth muscle fibers in the peripheral zone. Labeling index of proliferating cells in the peripheral zone was $4.34{\pm}1.28%$. These tumor cells did not metastasized in another orgns and labeling index was low, So it was concluded that this tumor was benign stomal sarcoma.

  • The cytologic finding of proliferating trichilemmal tumor is not well documented and have difficulties in the cytological analysis of the fine needle aspirates. This rare dermatopathologic entity may be encountered during aspiration biopsy of subcutaneous masses, and is, occasionally, confused with other neoplasm, particulary squamous carcinoma owing to its close cytological resemblance. We report a case of proliferating trichilemmal tumor in the left breast. The patient is a woman and had a lump in the breast for 20 years. Fine needle aspiration revealed a few small clusters of squamoid cells in the background of amorphous and calcified material. After excisional biopsy, the mass was confirmed as proliferating trichilemmal tumor. The main cytologic features discriminating squamous cell carcinoma were bland nature of epithelial cells, absence of atypical dyskeratotic cells, and rich amount of amorphous material.

Immunohistochemical analysis of the effect of low power GaAlAs laser treatment on the expression of proliferating cell nuclear antigen (PCNA) in full-thickness excisional wound of rat skin (CaAlAs 저출력 레이저 자극이 흰쥐의 피부 전층결손 절제 창상의 치유시 proliferating cell nuclear antigen(PCNA)발현에 대한 면역조직화학법적 분석)

  • We evaluated the effect of low power GaAsAl laser on re-epithelization in full-thickness excisional wound of rat skin. Two full-thickness excisions were made on the back of the experimental animals. Low power laser applications with 10mW intensity were treated experimental animals twice a day for 7 days. On the seventh postoperative day the quantitative analysis of re-epithelization was performed using immunohistochemical staining for proliferating cell nuclear antigen (PCNA). The majority of PCNA immunoreactive cells was observed at epithelial cells in the margin of full thickness excisional wound. The low power laser treatments significantly increased the number of PCNA immunoreactive cell as compared to that of non treated animal group (p<0.01). The shape of PCNA immunoreactive cell appeared as small dark, round to ovoid structures. Most PCNA immunoreactive cells exhibited a high intensity of staining that contrasted sharply with the surrounding background. In conclusion, these findings suggest that GaAlAs laser treatments effectively enhance the epithelial wound healing by the stimulating cell proliferation. Furthermore, the majority of cell proliferation occurred in the margin of full thickness excisional wound.

Effects of nerve cells and adhesion molecules on nerve conduit for peripheral nerve regeneration

  • Background: For peripheral nerve regeneration, recent attentions have been paid to the nerve conduits made by tissue-engineering technique. Three major elements of tissue-engineering are cells, molecules, and scaffolds. Method: In this study, the attachments of nerve cells, including Schwann cells, on the nerve conduit and the effects of both growth factor and adhesion molecule on these attachments were investigated. Results: The attachment of rapidly-proliferating cells, C6 cells and HS683 cells, on nerve conduit was better than that of slowly-proliferating cells, PC12 cells and Schwann cells, however, the treatment of nerve growth factor improved the attachment of slowly-proliferating cells. In addition, the attachment of Schwann cells on nerve conduit coated with fibronectin was as good as that of Schwann cells treated with glial cell line-derived neurotrophic factor (GDNF). Conclusion: Growth factor changes nerve cell morphology and affects cell cycle time. And nerve growth factor or fibronectin treatment is indispensable for Schwann cell to be used for implantation in artificial nerve conduits.

Immunohistochemical application of anti-proliferating cell nuclear antigen antibody on the development of uterus following sex hormone administration in rats (성 hormone이 rat 자궁 발달에 미치는 영향에 대한 proliferating cell nuclear antigen 항체의 면역조직학적 응용)

  • The study was designed to investigate the effects of progesterone and estrogen on the uterus of rats by immunohistochemical methods using Proliferating Cell Nuclear Antigen (PCNA) antibody. Eighteen female rats(Wistar), weighing initially about 300g, were ovariectomized. These rats were divided into four groups, progesterone-treated group, estrogen-treated group, estrogen+progesterone-treated group, and control group, progesterone-treated group was injected with 1mg of progesterone per rat per day for 2 days and estrogen-treated group with $20{\mu}g$ of $17{\beta}-estradiol$ for 3 days and estrogen+progesterone-treated group with $17{\beta}-estrdiol$ for 3 days and then with progesterone for 2 days as above. In gross findings, the uteri were markedly hypertrophied by estrogen treatment but were not affect in size by progesterone treatment. Immunohistochemical investigation was performed on the cell types with higher appearance of PCNA positive reaction cells in four groups. The groups with higher appearance of the stromal cells were ordered as estrogen-treated group, progesterone-treated group, estrogen+progesterone-treated group, and control group. The muscle cells were ordered as progesterone-treated group, estrogen-treated group, estrogen+progesterone-treated group, and control group. Positive reaction cells of the stromal cells were total 4.6 times higher than those of muscle cells. Therefore, the affect of the hypertrophy on the uterus by estrogen was larger than those of progesterone and affect on the uterus by stromal cells were larger than those of muscle cells. The group with more PCNA positive reaction cells of luminal epithelial cells were ordered as control group, progesterone-treated group, estrogen+progesterone-treated group, and estrogen-treated group, and glandular epithelial cells were ordered as estrogen+progesterone-treated group, progesterone-treated group, control group, and estrogen-treated group. It was suggested that estrogen and progesterone did not affect on the proliferating cells of luminal epithelial cells and affection of progesterone on the development of glandular epithelial cell was larger than that of estrogen.

Correlation Analysis of Apoptosis and Cell Proliferating Marker (Ki-67) in Thyroid Tumors (갑상선 종양에서 아포프토시스(Apoptosis)와 세포증식 표지자(Ki-67)의 상관관계 분석)

  • To clarify the growth mechanisms of thyroid tumors, we investigated apoptotic cells in 88 thyroid tumors, consisting of 24 adenomas, 58 papillary thyroid carcinomas, and 6 undifferentiated carcinoma, using terminal deoxynucleotidyl transferase mediated deoxyuridine triphosphate digoxigenin-nick end labeling (TUNEL). The cell proliferating marker was also evaluated immunohistochemically using the monoclonal antibody to Ki-67 antigen (MIB-1) in the same tumors. The apoptosis was expressed as a percentage of the TUNEL-positive cells in the tumor cells, and a proliferating marker, being the percentage of Ki-67 positive cells, was counted up each tumor. The statistical analysis were used analysis of variance (ANOVA) and student's t-test that were analyze the differences in the rate of each histological types of the thyroid tumors. The overall level of apoptosis was extremely low in all histological types of the thyroid tumors analyzed, the mean apoptosis being $0.31{\pm}0.40$ in adenoma, $0.55{\pm}0.48 $in papillary thyroid carcinoma, and $4.60{\pm}3.27$ in undifferentiated carcinoma. The Ki-67 protein in the thyroid tumor subtypes was significantly lower in adenoma and papillary carcinoma, at $2.45{\pm}2.99$ and $6.27{\pm}4.42$, respectively, than that in undifferentiated carcinoma at $26.47{\pm}23.88$ (p<0.0001). There was no correlation between clinicopathological factors and apoptosis or Ki-67 in papillary thyroid carcinoma. In conclusion, our findings suggest that apoptosis occurs infrequently in thyroid tumor, and that cell proliferating maker Ki-67 markedly differs according to the thyroid tumor subtypes. Moreover, the ratio between proliferating cells and apoptotic cells may reflect thyroid tumor progression.

Convergence of Cancer Metabolism and Immunity: an Overview

  • Cancer metabolism as a field of research was founded almost 100 years ago by Otto Warburg, who described the propensity for cancers to convert glucose to lactate despite the presence of oxygen, which in yeast diminishes glycolytic metabolism known as the Pasteur effect. In the past 20 years, the resurgence of interest in cancer metabolism provided significant insights into processes involved in maintenance metabolism of non-proliferating cells and proliferative metabolism, which is regulated by proto-oncogenes and tumor suppressors in normal proliferating cells. In cancer cells, depending on the driving oncogenic event, metabolism is re-wired for nutrient import, redox homeostasis, protein quality control, and biosynthesis to support cell growth and division. In general, resting cells rely on oxidative metabolism, while proliferating cells rewire metabolism toward glycolysis, which favors many biosynthetic pathways for proliferation. Oncogenes such as MYC, BRAF, KRAS, and PI3K have been documented to rewire metabolism in favor of proliferation. These cell intrinsic mechanisms, however, are insufficient to drive tumorigenesis because immune surveillance continuously seeks to destroy neo-antigenic tumor cells. In this regard, evasion of cancer cells from immunity involves checkpoints that blunt cytotoxic T cells, which are also attenuated by the metabolic tumor microenvironment, which is rich in immuno-modulating metabolites such as lactate, 2-hydroxyglutarate, kynurenine, and the proton (low pH). As such, a full understanding of tumor metabolism requires an appreciation of the convergence of cancer cell intrinsic metabolism and that of the tumor microenvironment including stromal and immune cells.
