Recently molybdenum alloys have been introduced as accident tolerating materials for cladding of fuel rods. Molybdenum element has seven stable isotopes with different neutron absorption cross section used in various fields, including nuclear physics and radioisotope production. This study presents separation approaches for all intermediate isotopes of molybdenum element by squared-off cascades using a newly developed numerical code with Salp Swarm Algorithm (SSA) optimization algorithm. The parameters of cascade including feed flow rate, feed entry stage, cascade cut, input feed flow rate to gas centrifuges (GCs), and cut of the first stage are optimized to maximize both isotope recovery and cascade capacity. The squared off and squared cascades are studied, and the efficiencies are compared. The results obtained from the optimization showed that for the selected squared off cascade, Mo94 in four separation steps, Mo95 in five steps, Mo96 in six steps, Mo97 in seven steps, and Mo98 in two steps are separated to the desired concentrations. The highest recovery factor is obtained 63% for Mo94 separation and lowest recovery factor is found 45% for Mo95.

These issues raise animal welfare concerns that currently limit the acceptability and applicability of the technology. It is expected that improved reprogramming of the donor genome will increase cloning efficiencies realising a wide range of new agricultural and medical opportunities. Efficient cloning potentially enables rapid dissemination of elite genotypes from nucleus herds to commercial producers. Initial commercialization will, however, focus on producing small numbers of high value animals for natural breeding especially clones of progeny-tested sires, The continual advances in animal genomics towards the identification of genes that influence livestock production traits and human health increase the ability to genetically modify animals to enhance agricultural efficiency and produce superior quality food and biomedical products for niche markets. The potential opportunities in animal agriculture are more challenging than those in biomedicine as they require greater biological efficiency at reduced cost to be economically viable and because of the more difficult consumer acceptance issues. Nevertheless, cloning and transgenesis are being used together to increase the genetic merit of livestock; however, the integration of this technology into farming systems remains some distance in the future.

본 실험은 유휴 논토양에 톨 페스큐를 재배하였을 때, 액상우분뇨를 물로 희석하거나 희석하지 않고 시기를 다르게 시용하여 계절별 및 연간 건물수량과 사료가치를 조사하고 화학비료 시비에 따른 건물수량과 사료가치도 비교하여 액상우분뇨의 적정 시용 시기와 희석수준을 결정하고자 실시되었으며, 그 결과를 요약하면 다음과 같다. 1) 액상우분뇨를 물로 희석하지 않거나 희석하여 시용하였을 때 연간 건물수량이 ha당 각각 $10.57{\sim}12.51$ 톤(평균 11.50 ton DM/ha)과 $11.31{\sim}14.81$ 톤 (평균 13.13 ton DM/ha)으로 무시비구의 ha당 9.21톤 보다 높은 건물수량을 나타내었는데(Table 2), 액상우분뇨를 이른봄과 여름철 시용한 구(SA구)와 이른 봄, 늦봄 및 여름철 분할 시용한 구(SUA구) 그리고 액상우분뇨를 물로 희석하여 봄철에 전량 시용한 구(DS구), 이른봄과 여름철에 시용한 구(DSA 구) 및 이른 봄, 늦봄 및 여름철에 분할 시용한 구(DSUA구) 등에서 무시비구 보다 유의하게 높은 건물수량을 나타내었다(p<0.05). 2) 질소와 인산 및 칼리를 시비한 구에서는 ha당 15.38 톤의 연간 건물수량을 나타내어 무시비구와 인산과 칼리 시비한 구(10.68 톤/ha) 보다도 유의하게 높은 건물수량을 나타내었다(p<0.05). 또한 인산과 칼리 시용구의 연간 건물수량은 액상우분뇨 시용구의 평균 연간 건물 수량보다도 낮았다. 3) 화학비료구에서 무기태 질소의 건물생산 효율은 연 평균 31.3kg DM/kg N이었으며, 예취 시기별로 보면 2번초>1번초>3번초 순으로 낮아졌지만 액상우분뇨 질소의 연간 건물생산 효율은 무 희석구와 희석구가 각각 15.3과 26.1kg DM/kg N을 나타내었고 2번초에서 가장 높았다. 한편 무기태 질소 대비 액상우분뇨 질소의 건물생산효율은 무희석구와 희석구가 각각 48.9와 83.4%에 도달하였다. 4) 톨 페스큐의 연간 평균 조단백질 함량은 액상우분뇨 시용구에서 $9.9{\sim}11.6%$로 무시비구의 9.5%와 화학비료 구의 $9.0{\sim}9.8%$ 보다 유의하게 높았지만(p<0.05), 연간 평균 NDF와 ADF 함량은 무시비구가 가장 낮았고 무시비구의 상대 사료가치(RFV)와 TDN 함량은 모든 처리구보다 유의하게 높았다(p<0.05). 5) 액상우분뇨 시용과 희석시용으로 톨 페스큐의 연간 조단백질 수량과 가소화 양분수량은 무시비구 및 인산과 칼리를 시용한 구 보다 유의하게 높았는데(p<0.05), 이러한 경향은 액상우분뇨를 희석하여 이른 봄, 늦봄 및 여름철에 분할 시용한 구(DSUA구)에서 더욱 뚜렷하였다.

Transgenic animals have become important tools for the production of therapeutic proteins in the domestic animal. Production efficiencies of transgenic animals by conventional methods as microinjection and retrovirus vector methods are low, and the foreign gene expression levels are also low because of their random integration in the host genome. In this study, we investigated the homologous recombination on the porcine ${\beta}$-casein gene locus using a knock-in vector for the ${\beta}$-casein gene locus. We developed the knock-in vector on the porcine ${\beta}$-casein gene locus and isolated knock-in fibroblast for nuclear transfer. The knock-in vector consisted of the neomycin resistance gene (neo) as a positive selectable marker gene, diphtheria toxin-A gene as negative selection marker, and 5' arm and 3' arm from the porcine ${\beta}$-casein gene. The secretion of enhanced green fluorescent protein (EGFP) was more easily detected in the cell culture media than it was by western blot analysis of cell extract of the HC11 mouse mammary epithelial cells transfected with EGFP knock-in vector. These results indicated that a knock-in system using ${\beta}$-casein gene induced high expression of transgene by the gene regulatory sequence of endogenous ${\beta}$-casein gene. These fibroblasts may be used to produce transgenic pigs for the production of therapeutic proteins via the mammary glands.

To treat wastewater efficiently by a one-step process of nitrogen removal, a new bacterial strain producing $N_2$ gas from ${NH_4}^+$ under an aerobic condition was isolated and identified. The cell was motile and a Gram-negative rod, and usually occurred in pairs. By 16S-rDNA analysis, the isolated strain was identified as Enterobacter asburiae with 96% similarity. The isolate showed that the capacity of $N_2$ production under an oxic condition was approximately three times higher than that under an anoxic condition. Thus, the consumption of ${NH_4}^+$ by the isolate was significantly different in the metabolism of $N_2$ production under the two different environmental conditions. The optimal conditions of the immobilized isolate for $N_2$ production were found to be pH 7.0, $30^{\circ}C$ and C/N ratio 5, respectively. Under all the optimum reaction conditions, $N_2$ production by the immobilized isolate resulted in reduction of ORP with both the consumption of DO and the drop of pH. The removal efficiencies of $COD_{Cr}$, and TN were 56.1 and 60.9%, respectively. The removal rates of $COD_{Cr}$, and TN were the highest for the first 2.5 hrs with the removal $COD_{Cr}/TN$ ratios of 32.1, and afterwards the rates decreased as reaction proceeded. For application of the immobilized isolate to a practical process of ammonium removal, a continuous operation was executed with a synthetic medium of a low C/N ratio. The continuous bioreactor system exhibited a satisfactory performance at 12.1 hrs of HRT, in which the effluent concentrations of ${NH_4}^+$-N was measured to be 15.4 mg/L with its removal efficiency of 56.0%. The maximum removal rate of ${NH_4}^+$-N reached 1.6 mg ${NH_4}^+$-N/L/hr at 12.1 hrs of HRT(with N loading rate of $0.08\;Kg-N/m^3$-carrier/d). As a result, the application of the immobilized isolate appears a viable alternative to the nitrification-denitrification processes.
The demand for the production of gene-defective mice from embryonic stem (ES) cells is increasing to clarify decisive gene function in vivo. Although blastocyst injection is widely used to generate ES cell-mediated knockout mice, coculture method has been alternatively used because of several advantages, such as low cost and simple procedure. Thus, this experiment was designed to demonstrate the feasibility of the coculture method using J1 ES cells, which are known to be efficient for blastocyst injection. Eight-cell embryos were harvested from 2.5 days post-coitum (dpc), denuded with acid tyrode's solution, and transferred onto trypsinized J1 ES cells. Aggregation was carried out following two typical methods, which are simple coculture method and aggregation in groove prepared by aggregation needle. Successfully aggregated-embryos were developed to blastocysts for 24 h and transferred into uterus of pseudo-pregnant foster mother. Chimeric offspring was judged by coat pigmentation. In this study, we could obtain chimeric mice from all the two aggregation methods, but the chimera production efficiencies in coculture using groove were three times higher at least than those in the other group. In conclusion, these observations suggest that coculture method should be available for production of knockout mice from J1 ES cells. Presently, the germ-line transmission rates of the chimeras produced from the two methods are under investigation.
본 연구는 실제 아파트 단지의 재생이 필요한 환경 인프라를 경제적이고 효율적인 녹색 인프라 기술로 재생하기 위한 계획을 수립하였으며, 단지내 물과 폐기물의 자원 순환 및 재이용을 위해 단지형 도시 중수 생산 기술과 단지형 유기성 폐기물 에너지화 기술, 무막힘 탄성포장 우수 재이용 기술, 도시농장 기술을 현장에 적용한 결과를 보여주고 있다. 도심 중수 생산 기술은 하수, 우수, 지하수를 대상으로 중수를 생산하여 단지내 공공용수로 이용하기 위한 기반 기술이며, 무막힘 탄성포장 우수배수구 기술은 단지 내에서 우수의 재이용율 극대화시키기 위한 기반 기술이다. 유기성 폐기물 에너지화 기술은 단지에서 발생하는 음식폐기물 및 분뇨 등의 유기성 폐기물을 처리하는 과정에서 바이오가스와 비료를 만들어 에너지원으로 활용할 수 있으며, 도시농장은 재생용수, 재생에너지, 재생비료를 활용하여 주거민들의 건강 및 여가공간으로 활용할 수 있는 기반기술이다. 이를 통해 단지 내에서 자원 순환 및 재이용이 가능한 하나의 에코시스템을 구축하였으며, 재생 후 녹색지표결과에서 에너지 부문은 4.23, 물부문은 0.32, 폐기물 부문은 0, 토지이용 부문은 0, 환경부문은 2.12가 상승하는 결과를 가져올 수 있었다.
에너지 소비의 증가와 화석 연료의 감소로 인해 바이오디젤과 같은 재생 가능한 대체 에너지 자원이 관심을 받고 있다. 미세조류를 이용한 바이오디젤은 기존의 농작물과 경쟁하지 않는 것과 더불어 많은 장점을 갖고 있다. 본 연구에서는 미세조류 배양의 생산 비용 절감과 축산 폐수 처리라는 두 가지 목표를 충족시키지 위해 돈분 액체 비료를 사용하였다. 옥외 배양 시스템(Small Scale Raceway Pond; SSRP)과 희석된 돈분 액체 비료를 이용하여 단일 미세조류 Chlorella sp. JK2, Scenedesmus sp. JK10 과 혼합 토착 미세조류 CSS를 20일 동안 각각 배양하였다. 미세조류 혼합균주인 CSS의 바이오매스 생산과 지질 생산성은 각각 $1.19{\pm}0.09gL^{-1}$, $12.44{\pm}0.38mgL^{-1}day^{-1}$로 단일 종에 비해 2배 이상 높았다. 돈분 액체 비료의 TN, TP의 제거율 역시 혼합 토착 미세조류 CSS에서 93.6%, 98.5%로 단일 종의 이용에 비해 30%이상 높은 제거 효율을 보여주었다. 이를 통해 돈분 액체 비료는 미세조류 배양에 필요한 N과 P를 제공하며, 미세조류를 이용한 SSRP를 통하여 영양염류를 제거할 수 있는 가능성을 확인하였다. 또한 미세조류 배양을 위한 생산 비용의 감소로 경제성 있는 바이오디젤의 생산 가능성을 확인하였다.
미생물연료전지(MFC)는 하 폐수내의 유기물로부터 전기를 생산할 수 있는 획기적인 기술이지만, 실용화를 위해서는 하 폐수 내의 질소를 제거할 수 있어야 한다. 본 연구에서는 두 개의 대면적 SEA (separator electrode assembly)로 구성된 평판형 외기환원전극 미생물연료전지(FA-MFC)를 이용하여 질산화 전배양의 유무와 식종원에 따른 총질소제거율을 평가하였다. 질산화 전배양 단계에서 FA-MFC의 질산화율은 식종원과는 무관하게 99% 이상을 나타냈다. 질산화 및 탈질 단계에서 300 mg-COD/L 이하의 낮은 유기물 농도에서는 전배양을 하지 않은 조건의 총질소제거율이 가장 높았다. 유기물 농도가 증가할수록 더 높은 총질소제거율을 나타냈으며, 유기물제거율은 모든 조건에서 95% 이상을 나타냈지만, 종속영양탈질에만 이용되지는 않은 것으로 판단된다. FA-MFC의 전기 발생량은 매우 낮았지만, 유기물과 질소를 동시에 제거할 수 있다는 장점이 있기 때문에 획기적인 하 폐수처리공법으로 발전시킬 수 있으리라 기대된다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
