Primary aldosteronism with an aldosterone-producing adenoma and contralateral adrenal hyperplasia: A case report (양측성 부신비대와 알도스테론 생성 선종이 동시에 발현된 증례)

  • Primary aldosteronism has been found more often among patients with hypertension. Primary aldosteronism can be caused by an aldosterone-producing adenoma, bilateral adrenal hyperplasia, or rarely by an adrenal carcinoma. An initial diagnostic test for aldosteronism is a measurement of the plasma renin activity and aldosterone concentration. For example, up to 20% of patients with hypertension showed increased plasma aldosterone concentration/renin activity ratio. If surgery is planned, an adrenal vein sampling is necessary for exact localization. Spironolactone, an aldosterone antagonist, is the drug of choice for patients with an aldosterone-producing adenoma or hyperplasia. It can control elevated blood pressure in most primary aldosteronism patients. However, unilateral laparoscopic adrenalectomy is the best treatment for aldosterone-producing adenoma or asymmetrical aldosterone production in patients with uncontrolled hypertension. Here we report a patient with primary aldosteronism caused by unilateral adrenal hyperplasia and a contralateral adrenal adenoma who required as many as five different kinds of antihypertensive medications for controlling elevated blood pressure. The adrenal adenoma was successfully removed by unilateral adrenalectomy and the blood pressure had been controlled well after the surgery.

Isolation of lysozyme producing bacteria capable of solubilizing microbial cells (미생물 용해가 가능한 Iysozyme 분비 균주의 분리 및 특성)

  • Lysozyme-producing microorganisms were isolated to obtain bacteria which can efficiently solubilize microbial cells. Cells of normal and chloroform-treated Escherichia coli and Micrococcus Iysodeikticus were used as model substrates to isolate lysozyme-producing microorganisms and investigate the efficiency of cell lysis. The culture supernatant of the isolate New1 (98% similarity of 16S rDNA sequence with Thermomonas haemolytica) showed different lytic characteristics for different substrates. Thermal treatment (autoclave) of substrate cells showed a significant effect on cell solubilization by culture supernatant of the New1. For autoclaved substrate cells, E. coli, M. Iysodeikticus and chloroform-treated E. coli were solubilized by 58.7%, 49.4% and 79.1%, respectively, in the culture supernatant of New1. The lytic activity of New1 was mainly caused by lysozyme produced by the isolate. It was also showed that New1 exhibited high protease activity and a little cellulase activity.

  • Industrial strain improvement is concerned with developing or modifying microorganisms used in production of commercially important fermentation products. The aim is to reduce the production cost by improving productivity of a strain and manipulating specific characteristics such as the ability to utilize cheaper raw materials or resist bacteriophages. The traditional empirical approach to strain improvement is mutation combined with selection and breeding techniques. It is still used by us to improve the productivity of organisms in amino acids, organic acids and enzymes production. The breeding of high L-lysine-producing strain Au112 is one of the outstanding examples of this approach. It is a homoserine auxotroph with AEC, TA double metabolic analogue resistant markers. The yield reaches 100 g/l. Besides, the citric acid-producing organism Aspergillus niger, Co827, its productivity reaches the advanced level in the world, is also the result of a series mutations especially with $^60Co{\gamma}$-radiation. The thermostable .alpha.-amylase producing strain A 4041 is the third example. By combining physical and chemical mutations, the strain A 4041 becomes an asporogenous, catabolite derepressed mutant with rifamycin resistant and methionine, arginine auxotroph markers. The .alpha.-amylase activity reaches 200 units/ml. The fourth successful example of mutation in strain improvement is the glucoamylase-producing strain Aspergillus niger SP56, its enzyme activity is 20,000 units/ml, 4 times of that of the parental strain UV-11. Recently, recombinant DNA approach provides a worthwhile alternative strategy to industrial strain improvement. This technique had been used by us to increase the thermostable .alpha.-amylase production and on some genetic researches.

Studies on the Pseudomonas aeruginosa Isolated from Infected Patient (감염환자에서 분리한 녹농균의 특성에 관한 연구)

  • Seventy-two strains of Pseudomonas aeruginosa isolated from the patients were tested for pigment production, exoenzyme production and antimicrobial susceptibility. In the pigment production test, 23.6% of total 72 strains produced both pyocyanine and pyoverdin. Pyoverdin and pyomelanin producing strains were in 9.7%, and 5.5% produced pyoverdin and pyorubin. Strains producing of all of exoenzyme, protease, elastase and lecithinase were in 5.6%. The most common type of exoenzyme production was both protease and elastase producing. Protease producing strain were 23.6%, Among the 72 strains, 50% produced protease. As the result of antimicrobial susceptibility in the isolated 20 strain, most strain were resistant to sulfamethoxazole(90%), but sensitive to other tested antibiotics more than 60%. The MIC50 and MIC90 level of tested antibiotics to 70 strains were 128 $\mu\textrm{g}$/$m\ell$, 512 $\mu\textrm{g}$/$m\ell$ for KM, 8,256 $\mu\textrm{g}$/$m\ell$ for GM, 8, 128 $\mu\textrm{g}$/$m\ell$ for CPZ, and 8, 64 $\mu\textrm{g}$/$m\ell$ for PIPC respectively.

  • The Purpose of this study Gerber company AM-300 system of the automatic system of Producing the original form the automatic system of Producing the original form of "Jogori(a Korean Jacket)' and Grading by the usage of computers and find out its efficiency. In the result, the auther has found out the following facts and became confident on the facts; The AM-300 program of the automatic system enabled to produce the original form of 'Jogori' and Grading fitting in a short time and definately, and which indicated that the automatically producing system of the original form of 'Jorgori' and Granding is efficient. Even in the aspect of education, it has been acknowledged that there is necessity of using computers, the accumulation of techincs and technology based on traditions by cultivating professional designers, and computerization so that the composition of 'Hanbok' (Korean clothes) should be rational and scientific. In addition, advertisement and education on the traditionalism and superiority of 'Hanblk' are indispensable and absolutely necessary. Also, to succeed folk costumes rightly, the usage of computers is thought to be a way to effectiveness. So far in the study, only the automatic system of producing the original form of 'Jogori' and Grading through computers is emphasized on, however in the future, such an automatic system should be continuously supplemented, studied on and developed even in other various fields such as in pattern making, design, products planning, etc..

  • Matrix producing carcinoma of the breast is a variant of heterologous metaplastic carcinoma which is defined as "overt carcinoma with direct transition to a cartilaoenous and/or osseous stromal matrix without an intervening spindle cell zone or osteoclastic cells". This tumor is very rare, occuring in less than 0.2% of total breast carcinoma, but the prognosis is better than other metaplastic carcinoma. We experienced a case of fine needle aspiration(FNA) cytologic finding of matrix producing carcinoma of the breast. A 75-year old woman, who presented a right huge breast mass$(9{\times}8cm)$ during 10months, was examined. Mammography reveals right lateral mass with even density without calcification. Breast ultrasonography shows multifocal hypoechogenic cystic change in the huge mass, suggesting resolving hematoma or carcinoma or sarcoma with necrosis. On cytologic finding of FNA, myxoid matrix was the dominant feature and the rest of the material was composed of scanty isolated atypical cells with large irregular nuclei. The histologic finding was moderately differentiated adenocarcinoma with abundant cartilagenous matrix and focal squamous metaplasia.

USING LOCAL FEED MATERIALS FOR FEEDING EGG PRODUCING BIRDS IN THE KINGDOM OF TONGA

  • A trial involving 200 day-old 288 Shaver chicks in a randomized experimental design tested the effects of using diets based on local feed materials (LF). Commercial imported counterpart feeds (CF) served as control diets. Birds were raised from 0-6 w on Chick Starter feed, from 7-17 w on Pullet Developer feed and from 18 to 72 w on Layer feed. The results obtained indicated that there were no significant differences in the performance of birds fed LF and CF diets during the Chick Starter and Pullet Developer phases. During the Laying phase, there were also no differences in laying percentage, (66 vs 65) and in egg size (62 vs 63 g) between LF and CF diets. Feed cost was lower on LF diet had a better egg yolk colour score, (4 vs 1) than those fed on CF diet. Feed cost was lower on LF than CF diets and the feed cost for producing eggs was approximately 50 percent lower on LF compared to CF (P$ 0.06 vs P$ 0.12). It was concluded that the use of locally available feed resources produces comparable performance to that obtained using commercial imported feeds. Secondly, using local materials markedly reduces feed cost and cost of producing eggs.

Selection of High Tobramycin-Producing Mutants (Tobramycin 고생산성 변이주의 분리)

  • An improved method for the selection of high tobramycin-producing mutants of Streptomyces tenebrarius ATCC 17920 was investigated. By the use of apramycin-containing media, low nebramycin-producing mutants were eliminated. Strain No. 23, resistant to apramycin and kanamycin B and sensitive to tobramycin, was isolated from soils, identified as Pseudomonas paucimobilis and used as a test organism for overlaying the mutants on agar plate. While inhibition zones were not shown when the parent strains were overlaid with soft agar containing the strain No. 23, clear zones were shown when high tobramycin-producing mutants were overlaid. Using this screening strategy, 58 mutants showing clear zones had been isolated. antibiotic activities of which were incresed to 3~8 fold compared to that of parent strain.

  • This experiment was carried out to investigate on the flora and cardinal wild mushroom in Tricholoma matsutake producing sites at the Namweon area. The results were as follows: Flora of Tricholoma matsutake producing sites were prevailed over at tree layer (Pinus densiflora S. et. Z.), shrub layer (Rhododendora mucronulatum TURCZ., Quercus serrata THUNB.) and herb layer (Carex humilis LEYSS.). Piedominant wild mushrooms were Boletaceae (Suillusplacidus Bon.) and Ramariaceae (Ramaria botrytis Fr.)

Characterization of Mouse Hybridoma Producing Monoclonal Antibody Paragonimus westermani (폐디스토마에 대한 Mmonoclonal Antibody생산 세포주개발에 대한 연구)

  • In this study, hybridoma techniques were applied to produce monoclonal antobodies to Paragonimus westermani, commonly known as lung distoma. Balb/c mice were immunized intraperitoneally every week with increasing doses of purfied protein of Paragonimus westermani adult worms begining with 0.3/mg/mouse/7 days and ending with 0.5mg at 28th day. Spleen cells from these immunized mice were hybridized with myeloma cells (NS-1) and the hybridized cells were selected in HAT media. The antibody secreting cells among the hybrid cells were initially selected by ELISA. Those initially selected cells were further screened by the criteria of antibody producing activity, and seneral cell lines among them were further tested with immunodiffusion method. One hybridoma clone produced IgM and another clone produced IgG1. The supernatant of the hybridoma clone producing IgM had titer 1:64 and the hybridoma clone producing IgG1 had titer 1:256 measured by immunofluorescence technique.
