Do neonicotinoid insecticides impaired olfactory learning behavior in Apis mellifera?

  • Bee's population is declining and disappearing at alarming rate. There are many factors responsible for declining the population of bees including diseases, natural enemies, environmental conditions and pesticides. Insecticides play its role dramatically for their population decline and neonicotinoid insecticides are critically important due to their wide application for pest control. Keeping in view of above problem, effect of neonicotinoid insecticides on olfactory learning behavior in Apis mellifera was observed using Proboscis Extension Reflex (PER) method. In this method, bees were harnessed in centrifuges tubes and feed on insecticides mixed sugar solution after three hours hunger. Bees were checked by feeding on non-treated sugar solution to observe PER response. Minimum proboscis extension was observed for acetamiprid and imidacloprid with 26% and 20% respectively at their recommend field doses while it was maximum for dinotefuran and thiamethoxam with 73% and 60% respectively. Only 40% bees showed response when exposed at 1/10 concentration of field dose for imidacloprid and the least at 1/100 of field dose. At control (Sugar solution) about 90% bees showed PER response. Among these neonicotinoid insecticides tested, imidacloprid and acetamiprid were the most damaging which impaired the olfactory learning performance in Apis mellifera.

Measurement and Comparison of Morphology of Developmental Stages of Chestnut Weevil, Curculio sikkimensis(Coleoptera: Curculionidae) (밤바구미 (Curculio sikkimensis)의 발육단계별 형태측정 및 비교)

  • Measurements were made on morphology of each developmental stages of the chestnut weevil, Curculio sikkimensis, reared in the laboratory and field from 2003 to 2006. The size of egg was 0.8${\pm}$0.03 mm. The escaping larvae were measured, in average, as 98 mg in body weight, 10.65 and 3.99 mm in body length and width, and 1.70 mm in head width. Pupal size of female and male was 7.01 and 6.53 mm, respectively. The fresh weight (0.343 g), body length (7.76 mm) and width (3.38 mm), and head width (1.60 mm) of female adults were significantly bigger than those (0.268 g, 7.14 mm, 3.01 mm and 1.37 mm, respectively) of male adults. Proboscis length (6.53 mm) and antennal length (5.47 mm) of female was also significantly longer than those (3.56 and 4.63 mm, respectively) of male. The larvae of C. sikkimensis overwintered for 1~3 years and their body weight, body length, and body width were decreased. The ratio between proboscis length and body length, the basipodite position attached to the proboscis, and shape of the sex organ on the abdominal end could be used to discriminate sexes.

Characteristics of Feeding Behaviors of Myzus persicae (Hemiptera: Aphididae) Depending on Inflow Concentrations of Dinotefuran (작물체내로의 dinotefuran 유입농도에 따른 복숭아흑진딧물 (Myzus persicae) (Hemiptera: Aphididae)의 섭식행동)

  • The changes of feeding pattern and the amount of insecticide penetrated into the leaf were monitored for 420 min after treatment of three concentrations of dinotefuran. At about 30 min after treatment of insecticide, the overall feeding pattern in EPG (Electrical Penetration Graph) monitoring was started to change, and there was significant difference in proportions of NP (non-penetration), PA (pathway activity), and phloem-feeding patterns among the different concentrations of dinotefuran treatment. Especially, as the amount of insecticide penetrated into a leaf increased, the reactive behaviors against this insecticide such as withdrawal of proboscis and movements of stylet of aphid were more quickly exhibited. And also, total time at which the proboscis of the aphids did not penetrate the plant was getting longer. The amount of dinotefuran penetrated into a leaf was monitored with HPLC. When the recommended concentration (100 mg/L) of the insecticide was treated, 5.24${\sim}$7.24 mg/L of the insecticide was detected from the leaf, and the proboscis of the aphids was apparently withdrawn from the leaf at approximately 60 minutes after treatment of this insecticide.

A New Record of Glycerid Polychaete, Glycera fallax (Polychaeta: Glyceridae) from Korea

  • A newly recorded glycerid polychaete collected from mud of the tidal flat in the southwestern coast of Korea is identified as Glycera fallax Quatrefages, 1850 based on the morphologies of papaillae and ailerons on proboscis, parapodial postsetal lobes, and branchiae. In this paper, we provide a detailed description and illustration of Korean materials of this species and a key to the species of the genus Glycera from Korean waters.

  • As a carrier of malaria and sneak of blood, mosquitoes are regarded as an unpleasant insect. However, there are novel phenomena that happen inside a mosquito. Among them, we focused on the blood sucking function of a female mosquito. The main objective of this study was to investigate the mosquito's pumping mechanism in order to resolve the problem encountered when we inject or transport biologic fluids into a micro-chip. To analyze the pumping mechanism, we visualized the blood sucking process inside a female mosquito. Flow characteristics of blood flow in a proboscis were investigated experimentally using a micro-PIV velocity field measurement technique. The anatomical variation of head, thorax, abdomen which work as pumps and valves, was visualized using the syncrotron X-ray micro-imaging technique.

Gustatory Receptors Required for Avoiding the Toxic Compound Coumarin in Drosophila melanogaster

  • Coumarin is a phenolic compound that mainly affects the liver due to its metabolization into a toxic compound. The deterrent and ovicidal activities of coumarin in insect models such as Drosophila melanogaster have been reported. Here we explore the molecular mechanisms by which these insects protect themselves and their eggs from this toxic plant metabolite. Coumarin was fatal to the flies in a dosage-dependent manner. However, coumarin feeding could be inhibited through activation of the aversive gustatory receptor neurons (GRNs), but not the olfactory receptor neurons. Furthermore, three gustatory receptors, GR33a, GR66a, and GR93a, functioned together in coumarin detection by the proboscis. However, GR33a, but not GR66a and GR93a, was required to avoid coumarin during oviposition, with a choice of the same substrates provided as in binary food choice assay. Taken together, these findings suggest that anti-feeding activity and oviposition to avoid coumarin occur via separate mechanisms.

First Record of the Genus and Species, Thetys vagina (Thaliacea: Salpida: Salpidae) in Korea

  • Pelagic tunicate, Thetys vagina Tilesius, 1802, is newly reported from Korean waters. The genus Thetys Tilesius, 1802 is also first recorded as Korean fauna. Thetys vagina is the only valid species in the genus Thetys. It is distinct from other genera by having at least 16 body muscles widely interrupted, no anterior proboscis, bent alimentary canal in the solitary oozooid, and five narrow body muscles, no ventral peduncle, an almost compact loop alimentary canal in the aggregate blastozooids. The specimens of T. vagina examined in this study were collected at the subtidal zone of Ulleung-do Island by scuba diving. In this paper, the detailed descriptions and photographs of both the solitary and aggregate living forms of T. vagina are provided.

  • On the inner side of each operculum of the crucian carp, Carassius auratus (n=10), the leech, Limnotrachelobdella sinensis of 1-4 individuals were parasitic. The leeches had approximately 41.0 mm in total length and 11 mm in width. These body was composed with anterior sucker, neck, trunk and posterior sucker and average length was 2.3 mm, 7.2 mm, 23.3 mm and 8.7 mm respectively. To both sides of the trunk lateral vesicle of 11 pair existed. When observed by SEM, anterior sucker was hemisphere shape and the mouth where proboscis comes out existed with the its center. Proboscis was connected the esophagus directly. Under light microscopy, bloodsucking gill of C. auratus showed lamella fusion, hypertrophy the epithelial cell of the filament and lamella, increased mucocytes and congested capillaries. On the other hand, necrotic and hydropic degeneration epithelial cell of the lamella, and infiltration of the macrophages from some individuals were suggested the secondary infection with the bacteria or virus after bloodsucking activity of the leech.

Extraintestinal Migration of Centrorhynchus sp. (Acanthocephala: Centrorhynchidae) in Experimentally Infected Rats

  • Reptiles were known to serve as paratenic hosts for Centrorhynchus (Acanthocephala: Centrorhynchidae) in Korea, but the infection course in experimental animals was not elucidated yet. In this study, the tiger keelback snakes (Rhabdophis tigrinus) were collected and digested with artificial pepsin solution, and the larvae of Centrorhynchus were recovered from them. Then, the collected larvae were orally infected to rats for developmental observations. In rats, all the larvae were observed outside the intestine on day 3 post-infection (PI), including the mesentery and abdominal muscles. As for the development in rats, the ovary of Centrorhynchus sp. was observed at day 15 PI, and the cement glands were 3 in number. Based on the morphological characteristics, including the arrangement of proboscis hooks, these larvae proved to be a species of Centrorhynchus, and more studies were needed for species identification.
