생리활성물질 처리가 priming 고추종자의 발아에 미치는 영향을 분석한 결과, ASM 0.1 mM 처리에서는 치상후 7일째 98%의 발아율을 보였으며 T50은 0.96일이였다. 그러나 ASM 0.5 mM 처리에서는 17%의 낮은 발아율을 보여 발아가 억제되었다. INA 0.01 mM 처리에서는 치상 후 2일째에 90% 이상의 발아율을 보였지만, 0.1 mM 처리에서는 치상 후 5일째에 90%의 발아율을 보였다. T50은 INA 0.01과 0.1 mM 처리에서 각각 0.65와 6.03일로 나타났다. BABA와 JA 처리는 priming에 의한 발아촉진 효과에 영향을 미치지 않았다.
Lupus-like syndrome is characterized by multiple organ injuries including lungs and kidneys. Endotoxin induces a transiently intent systemic inflammatory response and indirectly transient acute lung injury in normal condition. However, whether endotoxin may trigger the persistent development of lung injury in chronic, inflammatory lupus-like syndrome compared with normal condition remains unclear. We examined the pulmonary vascular permeability and production of proinflammatory cytokines, such as TNF-${\alpha}$, IL-6, IL-10 and IFN-${\gamma}$, which play prominent roles in the pathogenesis of lupus-like tissue injury, 6 hand 72 h after i.p. lipopolysaccharide (LPS; endotoxin) injection in pristane-primed chronic inflammation ICR mice characterized by a lupus-like syndrome. These results demonstrated that levels of serum IL-6, IL-10 and IFN-${\gamma}$ and bronchoalveolar lavage (BAL) IL-6 and IFN-${\gamma}$ were remarkably increased 6 h in LPS-exposed pristane-primed mice compared with pristane-primed controls, while pulmonary vascular permeability and levels of serum and BAL TNF-${\alpha}$ were not. And levels of BAL TNF-${\alpha}$, IL-6 and IL-10 were significantly enhanced 72 h in LPS-exposed pristane-primed mice compared with pristane-primed controls. Also, LPS significantly induced the increased in vitro production of TNF-${\alpha}$, IL-6 and IL-10 by lung cells obtained from LPS-exposed pristane-primed mice compared with LPS-exposed normal mice. Our findings indicate that LPS may trigger persistent progression of lung injury through late overproduction of BAL TNF-${\alpha}$, IL-6, and IL-10 in lupuslike chronic inflammation syndrome compared with normal condition.
Tobacco seeds (Nicotiana tabacum L. cv KF109) were primed in the polyethylene glycol 6000(PEG) solution and then stored at 5 and $25^{\circ}C$ under 40, 60 and 80% relative humidity (RH) conditions for six months. The effect of storage temperature and humidity on mean germination time ($T_{50}$), longevity and germination of the primed tobacco seeds were compared. Untreated seeds (control) stored at $5^{\circ}C$ showed high germinability throughout the entire storage period and humidity, and a decline in germinability showed after 6 months at 60% RH and after 3 months at 80% RH when stored at $25^{\circ}C$, Primed seeds retained high germinability until 6 months at 60% RH and 3 months at 80% RH when stored at $5^{\circ}C$ but showed a significant decline in germinability after 3 months at 40% RH, and 1 months at 60% and 80% RH, respectively when stored at $25^{\circ}C$, Primed seeds were completely lost viability when stored at $25^{\circ}C$ under 60% RH for 6 months and under 80% RH for 3 months.
Pluripotent stem cells could self-renew and differentiate into various cells. In particular, porcine pluripotent stem cells are useful for preclinical therapy, transgenic animals, and agricultural usage. These stem cells have naïve and primed pluripotent states. Naïve pluripotent stem cells represented by mouse embryonic stem cells form chimeras after blastocyst injection. Primed pluripotent stem cells represented by mouse epiblast stem cells and human embryonic stem cells. They could not produce chimeras after blastocyst injection. Populations of embryonic stem cells are not homogenous; therefore, reporter systems are used to clarify the status of stem cells and to isolate the cells. For this reason, studies of the OCT4 reporter system have been conducted for decades. This review will discuss the naïve and primed pluripotent states and recent progress in the development of porcine OCT4 reporter systems.
Bone marrow is a hematological and immunological organ that provides multiple immune cells, including B lymphocytes, and thus plays a critical role in the efficacy of vaccine. We previously demonstrated that Bordetella (B.) bronchiseptica antigen has high immunogenicity in spleen cells, a peripheral immune organ. In this study, we investigated the immunogenicity of B. bronchiseptica antigen in bone marrow cells, a central immune organ. B. bronchiseptica antigen increased the cellular activity of bone marrow cells and significantly enhanced the production of nitric oxide, IL-6, and TNF-${\alpha}$. Bone marrow cells primed with B. bronchiseptica antigen in vivo were harvested and stimulated with the same antigen in vitro. The stimulation of B. bronchiseptica antigen significantly increased the cellular activity and proliferation rate of the primed cells. B. bronchiseptica antigen also greatly induced the production of antigen-specific antibody in the primed cells. Taken together, the present study demonstrated that B. bronchiseptica antigen can stimulate bone marrow cells, a central immune organ, and recall the immune response of the primed bone marrow cells.
This study was carried out to investigate the changes in composition of the non-volatile organic acid, fatty acid and polyphenolic compounds during air-curing in burley tobacco leaves, and the effect of curing methods on the contents in air-cured leaves. The air-cured variety, (Nicotiana tabacum cv KB108) was normally grown at the Chonju tobacco experiment station in 1998. Plants designated for the each curing methods were harvested on the same date, and the ripe leaves for primed curing were harvested at the tenth leaf position from the top on the stalk. The major compounds of non-volatile organic acid and fatty acid were malic, citric, oxalic, palmitic, and linolenic acid. The concentrations of malic acid, unsaturated fatty acids, chlorogenic acid and rutin in cured leaves were remarkably decreased during curing, while citric acid was increased. The changes of these compounds showed the similar pattern during both primed and stalk curing. In connection with curing methods, the contents of malic, linoleic and linolenic acid were higher in excessive cured leaves than those in primed cured or stalk cured leaves, while the content of citric acid was lower in excessive cured leaves than that in primed cured or stalk cured leaves.
Background: The usefulness of DNA vaccine at priming step of heterologous prime-boost vaccination led to DNA vaccine closer to practical reality. DNA vaccine priming followed by recombinant viral vector boosting via systemic route induces optimal systemic immunity but no mucosal immunity. Mucosal vaccination of the reversed protocol (recombinant viral vector priming-DNA vaccine boosting), however, can induce both maximal mucosal and systemic immunity. Here, we tried to address the reason why the mucosal protocol of prime-boost vaccination differs from that of systemic vaccination. Methods: To address the importance of primary immunity induced at priming step, mice were primed with different doses of DNA vaccine or coadministration of DNA vaccine plus mucosal adjuvant, and immunity including serum IgG and mucosal IgA was then determined following boosting with recombinant viral vector. Next, to assess influence of humoral pre-existing immunity on boosting $CD8^+$ T cell-mediated immunity, $CD8^+$ T cell-mediated immunity in B cell-deficient (${\mu}K/O$) mice immunized with prime-boost regimens was evaluated by CTL assay and $IFN-{\gamma}$-producing cells. Results: Immunity primed with recombinant viral vector was effectively boosted with DNA vaccine even 60 days later. In particular, animals primed by increasing doses of DNA vaccine or incorporating an adjuvant at priming step and boosted by recombinant viral vector elicited comparable responses to recombinant viral vector primed-DNA vaccine boosted group. Humoral pre-existing immunity was also unlikely to interfere the boosting effect of $CD8^+$ T cell-mediated immunity by recombinant viral vector. Conclusion: This report provides the important point that optimally primed responses should be considered in mucosal immunization of heterologous prime-boost regimens for inducing the effective boosting at both mucosal and systemic sites.
본 연구는 당근과 양파에서 발아력 증진에 미치는 osmotic priming과 SMP효과를 비교하기 위해 수행되었다. Osmotic priming과 SMP 처리과정중 작물별 수분흡수율은 처리 1시간 후 급속한 수분흡수가 이루어졌고, 두 작물 모두 osmotic priming이 SMP보다 수분흡수 속도가 빨랐다. 이러한 경향은 처리 4시간까지 유지되었다. 처리최종일의 수분흡수율은 당근과 양파에서 osmotic priming이 SMP보다 2%높았다. Osmotic priming과 SMP 처리는 두 작물 모두 발아율을 향상시키지 못했지만 $T_{50}$ 및 MDG는 단축되어 조기발아 하였으며, 이러한 경향은 저온인 $15^{\circ}C$에서 더욱 현저하였다. Osmotic priming과 SMP 처리 후 초기함수율로 재건조하면 당근에서는 발아속도에 큰 차이가 없었으나, 양파에서는 표면건조에 비해 발아속도가 지연되었다. Osmotic priming과 SMP 상호처리간 발아력을 비교한 결과 두 작물 모두 SMP 처리가 osmotic priming보다 발아촉진에 효과적이었다. 당근과 양파종자를 osmotic priming과 SMP 처리하면 묘출현율도 향상되었고, 묘출현에 소요되는 일수를 단축시켜 신속한 묘출현을 유도하였다. 그러나 묘출현에 이은 유묘의 초기생육에는 통계적인 유의성은 인정되지는 않았으나 osmotic priming과 SMP 처리된 종자는 무처리에 비해 향상되었다.
양파 종자의 초기발아 촉진을 위한 priming 처리시 그 적정 조건을 구명하기 위하여 priming 처리 약제의 종류와 농도, priming 처리 기간과 온도, priming 처리후 온도조건에 따른 발아율에 대한 실험 결과를 요약하면 다음과 같다. 1. 양파종자의 초기 발아 촉진 및 발아율 향상을 위한 priming 처리결과 최종 발아율 향상 및 발아 소요일수 단축에 효과적 이었으며, priming 처리에 가장 효과적인 약제는 150 mM 농도의 KCl이였으며, priming 처리기간 및 온도는 $10^{\circ}C$에서 6 일간 처리하는 것이 가장 효과적이었는데 발아율은 96%로 일반종자의 82% 에 비해 14%가 높았다. 2. Priming 처리한 종자는 발아온도 $20^{\circ}C$ 이상보다는 $10^{\circ}C$ 및 $15^{\circ}C$의 저온에서 발아율 향상 및 발아 소요일수 단축 효과가 커서 적온보다는 저온에서 priming 처리 효과가 극대화 될 것으로 판단된다. Priming 처리한 후 종자 건조 조건은 $20^{\circ}C$에서 5 시간 건조시키는 것이 가장 좋았다.
fMLP에 의하여 자극된 중성호성 백혈구에서의 superoxide 생성, myeloperoxidase 유리, 칼슘 동원과 백혈구 부착에 나타내는 adenosine과 $N^6-cyclopentyladenosine$의 효과를 관찰하였다. 또한 이들의 효과를 C5a와 PMA의 자극효과에 대하여 그리고 lipopolysaccharide-primed 중성호성 백혈구의 반응에 대하여 관찰하였다. 이와 함께 adenosine의 억제작용에 있어 cAMP의 관여 여부를 조사하였다. 연구 결과로 부터 fMLP에 의해 자극된 중성호성 백혈구에서의 superoxide 생성, 탈과립과 세포내 칼슘 동원과 백혈구 부착은 adenosine 수용체에 의하여 조절된다고 추정된다. Adenosine은 protein kinase C의 활성화에 따른 백혈구 반응의 자극에 영향을 나타내지 않을 것으로 시사된다. Nonprimed 세포에 비하여, lipopolysaccharide-primed 중성호성 백혈구에서 fMLP에 의한 superoxide 생성은 adenosine의 영향을 적게 받을 것으로 여겨진다. Adenosine 존재하에서 백혈구 반응에 나타내는 theophylline의 억제효과는 세포내 cAMP 축적에 기인할 것으로 추정된다.
