Mutagenic Activity of Organic Pollutans in Drinking Water in Seoul (수도권 상수중 유기오염물질의 돌연변이원성)

  • To measure the mutagenic activity of micro-organic pollutants in drinking water, mutagenicity test was conducted using Salmnella typhimurium TA 98 strain on the water sample taken from three water supply stations and six tap water in Seoul in July and November 1987. The results were as follows : 1. The average amounts of organic matters in raw, treated, and tap water sampled in July were 0.38mg/l, 0.28mg/l, and 0.45mg/l. respectively, and sampled in November were 0.34mg/l, 0.24mg/l, and 0.22mg/l, respectively. The amount of organic matters of tap water sampled in November did not increase while that of tap water sampled in July increased compare to those of raw or treated water. 2. The amount of organic matters is the highest in neutral fraction compare to acidic and basic fractions. 3. In the five out of six tap water and raw water of Paldang and Kuui station sampled in July, the mutagenicity ratios were greater then two (both direct and indirect mutagenicity). 4. In the three out of six tap water and raw and treated water of Kuui station sampled in November, the mutagenicity ratios were greater than two. 5. While mutagenic activities were low in acidic and basic fraction, they were high in neutral fraction. The samples which had high mutagenic acitivity in the total amount also showed high mutagenic activity in neutral fraction. 6. While mutagenic acitivity was decreased after the treatement of water, it was increased in tap water as the distance from the water supply station increases.

Pharmacological Studies on Butanol Fraction of Puerariae Radix (갈근 Butanol Fraction의 약리 작용에 관한 연구)

  • Effective BuOH fraction was extracted from Puerariae radix (Pueraria thunbergiana), and several pharmacological activities were examined. From several pharmacological examinations, it was found that BuOH fraction has an antipyretic activity against typhoid vaccine pyrexia in rabbit, a papaverine-like antispasmodic activity on isolated ileum and jejunum of rat, an analgesic effects on mice treated by 0.7% HAc, and an anti-inflammatory activity of carrageenin edema in rat. It was found that BuOH fraction has detoxication effects on acute and subacute toxic rat induced by alcohol in consequence of biochemical tests of serum. Therefore it was considered that BuOH fraction has preventive and therapeutic effects in alcohol intoxication.

  • This Study was carried out develop antitumor effect of the n-butanol soluble of fraction of Perilla frutescens on human skin melanoma cells. The antitumor activity of various fractions obtained form n-butanol soluble fraction of Perilla frutescens was evaluated in human skin melanoma cells. The antitumor activity of the n-butanol soluble fraction in human skin melanoma cells was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, neutral red (NR) assay and sulforhordamine B protein (SRB) assay of colorimetic assay methods. The light microscopic study was carried out to observe morphological changes of cultured human skin melanoma cells. These results were obtained follows; The fractions 5 and 6 of the n-butanol soluble fraction of P frutescens were shown significant antitumor activities. The number of human skin melanoma cells were decreased and tend to form cell cluster by treatment with actions 5 and 7 of the n-butanol soluble fraction of P. frutescens. The fraction 6 of the the n-butanol soluble fraction showed the highest antitumor activity on P. frutescens. These results suggest that the fraction 6 of the n-butanol soluble fraction of P. frutescens may be a valuable choice for the studies on the treatment of human skin tumors.

Effects of the water soluble fraction of the musk on the activities of murine peritoneal macrophages (사향(麝香)의 수용성분(水容性分)이 생쥐 복강내(腹腔內) 거식세포(巨食細胞)의 활성(活性)에 미치는 영향(影響))

  • The musk has been reported to have significant anti-inflammatory activities in clinical use and several animal models and we examined the effects of water soluble fraction(WSF) of the musk on murine peritoneal macrophages. WSF decreased the production of nitric oxide from the lipopolysaccharide(LPS)-treated murine peritoneal macrophages and also reduced the phagocytic activity of macrophages on the opsonized sheep red blood cells(SRBC). Transcriptional expression level of the inducible nitric oxide synthase(iNOS) was also decreased and the viability of the treated macrophages was not affected by WSF, suggesting that the effects could be partly explained by transcriptional regulation. Contrary to down-regulating iNOS expression, WSF slightly increased the release of tumor necrosis factor-alpha$(TNF-{\alpha})$, which implied its selective action on cellular pathways activated by LPS. Our results showed that anti-inflammatory activities of the musk could be partly explained by the inhibitory effects of the water soluble fraction on the macrophageal activation.

  • The objective of this study was to investigate the preventive effect of ginseng on acetaldehyde-induced acute toxicity in mice . Compared to the control group, treatment with acetaldehyde inhibited the hepatic cytosolic xanthine oxidise activity with increase in dose. The inhibition of enzyme activity was not changed after dialysis. Pretreatment with ginseng butanol fraction prevented the inhibition of enzyme activity by acetaldehyde. In conjunction with the our previous results (Yakhak Hoeji, 29, 18 (1985)), these results suggest that the most likely mechanism for the observed preventive effects of ginseng against the acetaldehyde-induced acute toxicity may be the decrease hepatic acetaldehyde level.

Effect of School-Based Fluoride Mouth Rinsing Program in a Socially Deprived Community

  • Purpose: The objective of this study was to evaluate the caries prevention effect of school-based fluoride mouth rinsing (FMR) program implemented in the Hanuul district of Mongolia, which has a very low socioeconomic status and extremely poor infrastructure for oral health. Materials and Methods: One hundred and seventy children aged from 6 to 8 years of the FMR school and 187 children aged from 6 to 8 years of the control school completed the baseline survey. Children from the FMR school rinsed with 0.05% sodium fluoride everyday under supervision, while those from the control school did not. Adjusted caries preventive fraction (CPF) for 2 years were calculated to evaluate the effect of the FMR program. Result: After 2 years, 288 schoolchildren remained in the study. Decayed, missing or filled permanent teeth (DMFT) and index of the FMR and the control schools at baseline were 0.11 and 0.12, respectively, and the average DMFT increment of the FMR and the control schools after 2 years were 0.35 and 0.65, respectively. The adjusted CPF of DMFT was 48.5%. Conclusion: These findings show that a school-based FMR is an effective caries preventive program in a socially deprived community with poor infrastructure for oral health.

A Comparison of Green, Delta, and Monte Carlo Methods to Select an Optimal Approach for Calculating the 95% Confidence Interval of the Population-attributable Fraction: Guidance for Epidemiological Research

  • Objectives: This study aimed to compare the Delta, Greenland, and Monte Carlo methods for estimating 95% confidence intervals (CIs) of the population-attributable fraction (PAF). The objectives were to identify the optimal method and to determine the influence of primary parameters on PAF calculations. Methods: A dataset was simulated using hypothetical values for primary parameters (population, relative risk [RR], prevalence, and variance of the beta estimator ) involved in PAF calculations. Three methods (Delta, Greenland, and Monte Carlo) were used to estimate the 95% CIs of the PAFs. Perturbation analysis was performed to assess the sensitivity of the PAF to changes in these parameters. An R Shiny application, the "GDM-PAF CI Explorer," was developed to facilitate the analysis and visualization of these computations. Results: No significant differences were observed among the 3 methods when both the RR and p-value were low. The Delta method performed well under conditions of low prevalence or minimal RR, while Greenland's method was effective in scenarios with high prevalence. Meanwhile, the Monte Carlo method calculated 95% CIs of PAFs that were stable overall, though it required intensive computational resources. In a novel approach that utilized perturbation for sensitivity analysis, was identified as the most influential parameter in the estimation of CIs. Conclusions: This study emphasizes the necessity of a careful approach for comparing 95% CI estimation methods for PAFs and selecting the method that best suits the context. It provides practical guidelines to researchers to increase the reliability and accuracy of epidemiological studies.

Antimutagenic Compounds Indentified from Chloroform Fraction of Persimmon Leaves

  • Methanol extract of dried persimmon leaves was refractionated using hexane, chloroform, ethylacetate, n-butanol aqueous fractions. Among these chloroform fraction showed the highest inhibition rate on the mutagenicities of aflatoxin B₁(AFB₁) and 3,2' -dimethyl-4-amino-bipheny1(DMAB) in Salmonella typhimurium TA 100. Chloroform fraction was further fractionated into eight fractions by silica gel column chromatography and thin layer chromatography(TLC). The fraction 5 on TLC exhibited the highest antimutagenic activities in AFB₁and DMAB. 2,4-Decadienal, dihydro-4-methyl-2(3H)-furanone, hexanoic acid 1,4-bis(1-methy1 ethy1)benzene, heptanoic acid, phenol, octanoic acid, nonanoic acid and benzoic acid were tentatively identified from this antimutagenic fraction by GC-MS.

Verification of Cytotoxicity Against Cancer Cell Line and Estrogen-like Activity of Cheongkukjang (청국장의 암세포주에 대한 세포독성 및 에스트로겐 유사활성 검증)

  • In order to evaluate the cancer preventive and estrogenic compounds in soybean and Cheongkukjang, MTT assay and in vitro test system for the evaluation of the estrogenic activity were applied. The fractions from the ethanol extract of soybean and Cheongkukjang were prepared by the systematic extraction procedure with the solvents such as hexane, ethyl ether, butanol, methanol and H$_2$O. Ethyl ether fractions of soybean and Cheongkukjang showed the highest cytotoxicity against U937 cell line in dose dependent manner, and ethyl ether fraction of Cheongkukjang showed two times higher cytotoxicity than that of soybean. Aqueous fraction of soybean and ethyl ether fraction of Cheongkukjang revealed the highest estrogenic activity and activity was higher in the fractions of Cheongkukjang than soybean. Mixture of Spirulina and Cheongkukjang showed synergistic activity. These observations concerning cancer preventive and estrogen effects of soybean and Cheongkukjang suggest that these materials possess useful ingredients for the prevention of cancer and/or postmenopausal disorder.

Effects of Ephedra Herba ethyl-acetate fraction on high fat diet induced hyperlipidemic mice (마황 에틸아세테이트 분획물이 고지방 식이로 유발된 생쥐의 지질대사에 미치는 영향)

  • Objective : Ephedra Herba has been widely used for patients with common cold, asthma in eastern countries, especially china japan and korea. Recently it has been also used for obesity in clinic with high frequency. This study was designed to investigate the effects of Ephedra Herba ethyl-acetate fraction (EEAF) on hyperlipidemic mice. Method : Effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured in this experiment, and in addition, histopathological and gene expression changes in liver tissue was also observed. Results : In our study, EEAF did not affect weight gain in hyperlipidemic mice. Oral administration of EEAF lowered levels of total cholesterol which were elevated by induction of hyperlipidemia. And administration of EEAF lowered fasting blood glucose significantly. By carrying out ontological analysis, large numbers of genes were identified in up or down regulated genes. The expression of the genes that were altered in response to high-fat diet was restored to normal levels in EEAF treated mice, with a recovery rate of 49%. And it was considered that fatty acid metabolism was one of important key pathway of the recovery. Conclusion : Results in our study suggest that EEAF can prevent obese and through regulation of dyslipidemia and hyperglycaemia.
