사용자 중심설계를 이용한 임산모 건강관리 교육프로그램 개발 연구 (Development of a Health Educational Program for Maternal Health)

  • Purpose: The purpose of this paper is to describe the development of health educational program for maternity using principle of user centered design. Method: Research process includes five distinct phases: needs assessment, analysis, design, development/testing/revision, and application release. Results: This program includes an introduction, pregnancy test and fetal assessment, maternal and fetal change during pregnancy, self-care methods during pregnancy, complication of postpartum period, process of labor, complication of pregnancy, self care during the postpartum period, information for parenting skill, father page, FAQ, helping resources and institutions. Conclusion: The program will be distributed to health centers for maternal health education. The second phase of this project was evaluation this program for further development of the program. The end result of this program will be pregnant women with a high degree of usability. Author believe this program have true potential in helping maternal health promotion and successful parenting.

Age- and Area-Dependent Distinct Effects of Ethanol on Bax and Bcl-2 Expression in Prenatal Rat Brain

  • Cell proliferation and differentiation are critical processes in a developing fetal rat brain, during which programmed cell death (PCD) also plays an important role. One of the decisive factors for PCD is Bcl-2 family proteins, where Bax induces cell death, whereas Bcl-2 acts as an inhibitor of PCD. As maternal drinking is known to cause fetal alcohol syndrome (FAS) or malformation of the fetal brain during pregnancy, the objective of the present study was to investigate whether maternal ethanol exposure alters the PCD-related Bax and Bcl-2 protein expression during fetal brain development. Pregnant female rats were orally treated with 10% ethanol and the subsequent expressions of the Bax and Bcl-2 proteins examined in the fetal brain, including the forebrain, midbrain, and hindbrain, from gestational day (GD) 15.5 to GD 19.5, using Western blots, in situ hybridization, and immunohistochemistry. With regard to the ratio of Bcl-2 to Bax proteins (Bcl-2/Bax), the Bax protein was dominant in the forebrain and midbrain of the control GD 15.5 fetuses, except for the hindbrain, when compared with the respective ethanol-treated groups. Moreover, Bcl-2 became dominant in the midbrain of the control GD 17.5 fetuses when compared with the ethanol-treated group, representing an alternation of the natural PCD process by ethanol. Furthermore, a differential expression of the Bcl-2 and Bax proteins was found in the differentiating and migrating zones of the cortex, hippocampus, thalamus, and cerebellum. Thus, when taken together, the present results suggest that ethanol affects PCD in the cell differentiation and migration zones of the prenatal rat brain by modulating Bax and Bcl-2 expression in an age- and area-dependent manner. Therefore, this is the first evidence that ethanol may alter FAS-associated embryonic brain development through the alteration of Bax and Bc1-2 expression.

  • Objectives : The purpose of this study was to examine the prenatal, perinatal and developmental risk factors of attention-deficit hyperactivity disorder (ADHD), compared to unaffected siblings (SIB), and typically developing children (TC). Methods : Subjects with ADHD, their SIB, and TC were recruited from the child psychiatry outpatient clinic of the Asan Medical Center Children's Hospital. The parents of the children completed questionnaires on perinatal and developmental risk factors. Results : Fifty-eight subjects with ADHD (41 boys, $7.7{\pm}1.3years$), 21 SIB (8 boys, $8.2{\pm}1.8years$), and 22 TC (8 boys, $8.5{\pm}2.1years$) were included. The ADHD group showed higher rates of maternal stress during pregnancy than the SIB group (p=.002), and the ADHD group showed higher rates of familial psychiatric history than the TC (odds ratio, 8.76 ; 95% confidence interval, 1.69 to 45.45). Conclusion : These findings suggest that among perinatal and developmental factors, maternal stress during pregnancy contribute to the development of ADHD. Future prospective studies will be needed in order to determine the causal relationship between perinatal risk factors and development of ADHD.

  • The present study was designed to investigate the morphological developments of the stomach in the prenatal and postnatal rats. The gastric fundus of 16- to 22-day-old fetuses, neonates, 7-day-old, 14-day-old, 21-day-old, and adult rats were observed by transmission electron microscopy. 1. In the 19-day-old fetuses, the gastric pits and the parietal cells were the primitive state, but the parietal cells contained numerous microvilli and mitochondria. 2. In the 20-day-old fetuses, the intracellular canaliculus appeared in the parietal cells, and glycogen granules were localized mainly in the basal part of the epithelial cells and parietal cells. 3. In the 20-day-old fetuses, the mucous granules and zonular occludens appeared in the gastric glandular cells adjacent to the parietal cells. 4. In the 21-day-old fetuses, the chief cells were identified in the lamina propria and these cells contained granular endoplasmic reticulum and zymogen granules. 5. In the 20- to 22-day-old fetuses, the definitive parietal cells were firstly appeared in pairs or groups within the lamina propria. These cells tended to be placed near the basement membrane and blood capillary, and frequently tended to be extruded into lumen of gland. 6. The endocrine cells were appeared at the basal areas of the gastric gland after the 14-day-old.

단일 태아세포에서의 PEP-PCR을 이용한 성의 결정과 Dystrophin 유전자 분석 (Analyses of Dystrophin Gene and Sex Determination using PEP-PCR in Single Fetal Cells)

  • Recently, through the development of the primer extension preamplification(PEP) method which amplifies the whole genome, simultaneous multiple DNA analysis has become possible. Whole genome from each single cell can be amplified using 15 base oligonucleotide random primer. The greatest advantage of PEP-PCR is the ability to investigate several loci simultaneously and confirm results by analysing multiple aliquots for each locus. This technique led to the development of preimplantation genetic disease diagnosis using blastomere from early embryo, sperm, polar body and oocyte. In this study, we applied PEP-PCR in 20 cases of single amniocyte and 20 cases of single chorionic villus cell for the clinical application of the prenatal and preimplantational genetic diagnosis. We analysed 7 gene loci simultaneously which are 46, 47 exons related to dystrophin gene, two VNTR (variable number tandem repeat) markers using 5'dysIII, 3'CA related to dystrophin gene and DYZ1, DYZ3, DYS14 regions on chromosome Y. In all the tests, 97.5% of PEP-PCR amplifications with single cells were successful. We obtained 38/40 (95%) accuracy in gender determination through chromosome analysis comparison. Therefore, these results have significant implications for a sperm or oocyte analysis and prenatal or preimplantational genetic diagnosis.

  • The immunocytochemical localization of tyrosine hydroxylase(TH) is examined in the developmental forebrain of mongolian gerbil in order to determine changes in the distribution and cytology of neurons. At each of the different developmental stage, including prenatal(E)14, E16, E18, E20, postnatal(P)0, P2, P4 days, mongolian gerbils were sacrificed. In E14, TH-IR cells were predominantly round or oval in shape and their processes were very short. In olfactory blub, TH-IR cells were begun to appear from E20. In the striatum, we observed only fibers of TH-IR at all ages. From E16, TH-IR perikarya and fiber were present in periventricular nucleus and paraventricular nucleus of hypothalamus. The changes from the early to the late prenatal stages of development appear to be the result of an increase in number of TH-IR perikarya and processes. These results were shown slight differences to other rodents.

  • Pentose phosphate shunt의 첫 작용때 나타나는 효소 glucose-6-phosphate dehydorgenase의 활성을 출생전 및 출생후 연령에 따라서 쥐(white rat)의 간세포를 암수 성별로 spectrophotometer를 써서 측정하였다. 이 활성이 16일 된 배에서 가장 높았고 (54 I.U.), 그때부터 출생후 13일 까지 조금씩 저하되지만 (17 I.U.), 다시 서서히 증가해서 19일에는 53 I.U.까지 높아졌다. 성별로 본 결과는 일반적으로 수컷보다 암컷에서 G-6-PD의 활성이 약간 높게 나타났다.

Immunoreactivity of PCNA in the Cerebellum of Developing Guinea Pig

  • The investigation of the embryonic development of the cerebellum has a long history. The postnatal normal development of the cerebellum in rodents and other animals became a popular topic for morphological investigations nearly a century ago. However, surprisingly, only a few studies are available regarding the prenatal normal development of the rodent cerebellum, especially in guinea pigs. Cell proliferation is essential for the development of the nervous system. The assessment of cell proliferation can be achieved by using various methods. In this study, we investigated the cell proliferation of the cerebellar cortex in guinea pigs at different stages of pregnancy and in postnatal life. Fetuses were obtained by cesarean section at 50 or 60 days of gestation (dg). Immunohistochemistry was performed with proliferating cell nuclear antigen (PCNA) antibody in the cerebellum. Strong PCNA immunoreactivity was observed in the external granular layer (EGL), which is a neurogenic zone in the cerebellum. The proportion of PCNA-IR cells was greater at 1 week than at 60 dg in lobule I, but not lobule VIII. After 50 dg, the width of the EGL continued to decline until 1 week, due to the maturation of the EGL cells. These results demonstrate the pattern of PCNA immunoreactivity in the developing cerebellum of guinea pigs. This serves as a guideline to study abnormal cerebellum development.

  • This documentary study explored the stages of human development and education in the Choson Dynasty with the use of "Non Eu, Ye Gi, So Hak, Tae Gyo Sin Gi, and Dong Yi Bo Gam. The findings were compared with Western writings of a comparable period. It was found that education in the Choson Dynasty was largely concerned with mental attitude and conduct in ordinary life (education for manners and living). Education claimed to stand for unity of knowledge and behavior. It was founded on a Confucian view of education with a saint as the ideal model to be emulated. In the early years, home influence was emphasized. This was later replaced by institutional education. Additional education frequently extended to ten years and even forty or more years for the development of leaders. Based on the above findings. It was concluded that (1) the Western stages of human development exemplified by the Puritans and by Rousseau were concerned only with progressive changes from the postnatal stage to adolescence but the stages or the Choson Dynasty applied to the entire life span, from prenatal life through old age: (2) the developmental stages of males and females were defined differently as a result of differences in educational contents and sex-role distinctions: (3) life span educational objectives were Confucian and the preparatory stages were long coming to fruition in late senescence: and (4) although education was centered on manners and based on Confucianism, much of human development in the Choson Dynasty corresponds to that of modern times.

TRAF6 Distinctly Regulates Hematopoietic Stem and Progenitors at Different Periods of Development in Mice

  • Tumor necrosis factor receptor-associated factor 6 (TRAF6) is identified as a signaling adaptor protein that regulates bone metabolism, immunity, and the development of several tissues. Therefore, its functions are closely associated with multiple diseases. TRAF6 is also involved in the regulation of hematopoiesis under steady-state conditions, but the role of TRAF6 in modulating hematopoietic stem and progenitor cells (HSPCs) during the developmental stages remains unknown. Here, we report that the deletion of TRAF6 in hematopoietic lineage cells resulted in the upregulation of HSPCs in the fetal liver at the prenatal period. However, in the early postnatal period, deletion of TRAF6 drastically diminished HSPCs in the bone marrow (BM), with severe defects in BM development and extramedullary hematopoiesis in the spleen being identified. In the analysis of adult HSPCs in a BM reconstitution setting, TRAF6 played no significant role in HSPC homeostasis, albeit it affected the development of T cells. Taken together, our results suggest that the role of TRAF6 in regulating HSPCs is altered in a spatial and temporal manner during the developmental course of mice.
