X-선 분석법을 이용한 내화도료의 화재안전성 평가 방법에 관한 기초연구 (A Preliminary Study on the Fire Safety Testing Method for Fire-resistance Paints Using an X-ray Analysis Method)

  • 내화도료는 화재 시 발포를 통해 단열 성능을 향상시켜 철골 구조의 열전달을 저감하기 위해 사용되고 있는데, 만일 표준규격을 만족시키지 못하는 내화도료가 사용된다면 철골 구조의 내화 성능에 문제가 발생할 우려가 있다. 내화도료의 내화 성능은 육안으로는 평가가 어려워 현장을 대상으로 하는 간편하면서도 정확한 평가방법의 개발이 요구되고 있다. 본 연구에서는 X-선 분석법(XRF, XRD)을 이용한 내화도료의 화재안전성 평가 방법에 대한 기초연구를 수행하였다. X-선 분석법을 적용한 결과 실제 화재시험을 통한 내화 성능에 따라 특정 성분이 검출되는 것을 확인하였다. X-선 형광분석(XRF)을 통해 인(P)과 염소(Cl) 원소 성분이 일반도료에 비해 내화도료에 훨씬 많이 함유되어 있는 것을 알 수 있었다. X-선 회절분석(XRD)을 통해 내화도료의 주된 결정성 물질로 확인된 폴리인산 암모늄(ammonium polyphosphate)이 일반도료에는 포함되어 있지 않는 것으로 나타났다. X-선 분석법은 휴대용 XRF와 XRD 분석 장비를 통해 머지않아 현장평가방법으로의 적용이 가능할 것으로 예상된다.

Effects of Ionizing Radiation on Postharvest Fungal Pathogens

  • Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

  • A helminth glutathione S-transfease, 26 kDa isozyme from Clonorchis sinensis was cocrystallized with inhibitors by the hanging-drop method with monomethylether 550 as a precipitant. The crystals, grown in the presence of S-methylglutathione or trans-4-phenyl-3-buten-2-one, suitable for X-ray analysis, belong to the hexagonal space group P6₂(or P6₄), with unit cell parameters a= b= 97.6Å, c=117.4Å. X-ray diffraction data were collected with 2.5Å resolution.

Preliminary Study on the Visualization and Quantification of Elemental Compositions in Individual Microdroplets using Solidification and Synchrotron Radiation Techniques

  • Quantifying the solute composition of a cloud droplet (or a whole droplet) is an important task for understanding formation processes and heating/cooling rates. In this study, a combination of droplet fixation and SR-XRF microprobe analysis was used to visualize and quantify elements in a micro-scale droplet. In this study, we report the preliminary outcome of this experiment. A spherical micro-scale droplet was successfully solidified through exposure to ${\alpha}$-cyano-acrylate vapor without affecting its size or shape. An X-ray microprobe system equipped at the beam line 37XU of Super Photon ring 8 GeV (SPring-8) was applied to visualize and quantify the elemental composition in an individual micro-scale droplet. It was possible to reconstruct 2D elemental maps for the K and Cl contained in a microdroplet that was dispensed from the 10-ppm KCl standard solution. Multi-elemental peaks corresponding to X-ray energy were also successfully resolved. Further experiments to determine quantitative measures of elemental mass in individual droplets and high-resolution X-ray microtomography (i.e., 3D elemental distribution) are planned for the future.

Crystallization and preliminary X-ray analysis of API5-FGF2 complex

  • API5 is a unique oncogenic, non-BIR type IAP nuclear protein and is up-regulated in several cancers. It exerts several functions, such as apoptosis inhibition, cell cycle progression, cancer immune escape, and anticancer drug resistance. Although structural studies of API have revealed that API5 mediates protein-protein interactions, its detailed molecular functions remain unknown. Since FGF2 is one of API5's major interacting proteins, structural studies of the API5-FGF2 complex will provide insight into both proteins' molecular function. We overexpressed and purified API5 and FGF2 in Escherichia coli and crystallized the API-FGF2 complex using polyethylene glycol (PEG) 6000 as a precipitant. Diffraction data were collected to a $2.7{\AA}$ resolution using synchrotron X-rays. Preliminary diffraction analysis revealed that the API5-FGF2 complex crystal belongs to the space group $P2_12_12_1$ with the following unit cell parameters: a = 46.862, b = 76.523, $c=208.161{\AA}$. One asymmetric unit with 49.9% solvent contains one API5-FGF2 complex. Molecular replacement calculation, using API5 and FGF2 coordinates, provided a clear electron density map for an API5-FGF2 complex.

Overexpression, Crystallization, and Preliminary X-Ray Crystallographic Analysis of the Alanine Racemase from Enterococcus faecalis v583

  • Alanine racemase, a bacterial enzyme belonging to the fold-type III group of pyridoxal 5'-phosphate (PLP)-dependent enzymes, has been shown to catalyze the interconversion between L- and D-alanine. The alanine racemase from the pathogenic bacterium Enterococcus faecalis v583 has been overexpressed in E. coli and was shown to crystallize an enzyme at 295 K, using polyethylene glycol (PEG) 8000 as a precipitant. X-ray diffraction data to $2.5{\AA}$ has been collected using synchrotron radiation. The crystal is a member of the orthorhombic space group, $C222_1$ with unit cell parameter of a=94.634, b=156.516, $c=147.878{\AA},\;and\;{\alpha}={\beta}={\gamma}=90{\AA}$. Two or three monomers are likely to be present in the asymmetric unit, with a corresponding $V_m\; of\;3.38{\AA}^3\;Da^{-1}\;and\;2.26{\AA}^3\;Da^{-1}$ and a solvent content of 63.7% and 45.5%, respectively.

Subaru Weak-lensing Analysis of the Merging Cluster ZwCL 1447.2+2619 at z=0.37

  • ZwCL 1447.2+2619 is a merging galaxy cluster at z=0.37 with clear substructures in X-ray emission and galaxy distribution. In addition, the system possesses distinct radio relics. In order to constrain the merger scenario, it is necessary to measure both the distribution and mass of the cluster dark matter. We perform weak lensing analysis of ZwCL 1447.2+2619 using Subaru imaging data. We detect clear lensing signal from the cluster after carefully addressing instrumental systematics. In this poster, we present our preliminary results on our mass reconstruction and discuss the comparison with X-ray and radio results.

Discovery of the prominent radio relics in the cluster merger ZwCL J1447+2619

  • Diffuse radio emissions at the outskirt of merging galaxy clusters called radio relics provide a unique channel to understand the merger history. We present a recent discovery of double radio relics in the cluster merger ZwCL1447+2619 from our recent Giant Metrewave Radio Telescope observations. Both Band 3 (300-500 MHz) and Band 4 (550-850 MHz) data reveal a large (~1Mpc) and thin (~40kpc) radio relic ~1Mpc from the cluster X-ray center and a small radio relic (~0.3 Mpc) on the opposite side. These remarkable radio data together with Subaru weak-lensing analysis and Chandra X-ray observations enable us to reconstruct the merger scenario. Our preliminary analysis suggests that the cluster ZwCL J1447+2619 is a post-merger near its returning phase. In addition, using Keck DEIMOS spectroscopy, we find many "green" and "blue" member galaxies are located between the radio relics, a possible indication of merger shock-driven star formation activities.

THE ORBITAL EPHEMERIS OF THE PARTIAL ECLIPSING X-ray BINARY X1822-371

  • X1822-371 is a low mass X-ray binary with an accretion disk corona exhibiting partial eclipses and pulsations in the X-ray band. We update its orbital ephemeris by combining new RXTE observations and historical records, with a total time span of 34 years. There were 11 RXTE observations in 2011 but the eclipsing profile can be seen in only 4 of them. The eclipsing center times were obtained by fitting the profile with the same model as previous studies. Combined with the eclipsing center times reported by Iaria et al. (2011), the O-C analysis was processed. A quadratic model was applied to fit the O-C results and produced a mean orbital period derivative of $\dot{P}_{orb}=1.339(25){\times}10^{-10}s/s$, which is slightly smaller than previous records. In addition to the orbital modulation from the orbital profile, we also present our preliminary results for measuring the orbital parameters using the orbital Doppler effect from the pulsation of the neutron star in X1822-371. The updated orbital parameters from eclipsing profiles will be further compared with the ones from pulsar timing.

  • Plasmid Achromobacter secretion (PAS) factor is a putative secretion factor that induces the secretion of periplasmic proteins. PAS factor from Vibrio vulnificus was crystallized at 294 K by the hanging drop vapor-diffusion method. It was isolated as a monomer during the purification procedures. The native crystal belongs to the F222 space group with unit cell parameters a = 56.1, b = 74.4, $c=80.0{\AA}$, ${\alpha}={\beta}={\gamma}=90^{\circ}$. The crystal was soaked in cryoprotectant containing 1 M NaBr for 1 h for MAD phasing. The diffraction limit of the Br-MAD data set was $1.9{\AA}$ using synchrotron X-ray irradiation at beam line BL-18B at the Photon Factory, Japan.
