Advancing Risk Assessment through the Application of Systems Toxicology

  • Risk assessment is the process of quantifying the probability of a harmful effect to individuals or populations from human activities. Mechanistic approaches to risk assessment have been generally referred to as systems toxicology. Systems toxicology makes use of advanced analytical and computational tools to integrate classical toxicology and quantitative analysis of large networks of molecular and functional changes occurring across multiple levels of biological organization. Three presentations including two case studies involving both in vitro and in vivo approaches described the current state of systems toxicology and the potential for its future application in chemical risk assessment.

  • With increasing offshore oil exploration, drilling, and production activities, and with a huge amount of petroleum being transported by tankers and pipelines through the ocean and coastal environment and resoures has been realized among government decision makers, oil industry personnel, and the general public. Assessment must be made of the potential risk of damage resulting from the exploration, development and transportation activities, based on predictive impact evaluations of the fate of hypothetical or real oil spills.(omitted)

인공신경망 기반 VRF 시스템 제어 (ANN-Based VRF (variable refrigerant flow) system control)

  • This study aimed at developing control algorithms for operating a variable refrigerant flow (VRF) heating and cooling system with optimal system parameter set-points. Two artificial neural network (ANN) models, which were respectively designed to predict the heating energy cost and cooling energy amount for upcoming next control cycle, was developed and embedded into the control algorithms. Performance of the algorithms were tested using the computer simulation programs - EnergyPlus, BCVTB, MATLAB in an incorporative manner. The results revealed that the proposed control algorithms remarkably saved the heating energy cost by as much as 7.93% and cooling energy consumption by as much as 28.44%, compared to a conventional control strategy. These findings support that the ANN-based predictive control algorithms showed potential for cost- and energy-effectiveness of VRF heating and cooling systems.

  • In this article, We propose research propositions about correlates of job interest, job difficulty, job effort, and job performance as well as job performance and emotions while working. The purpose of this study is to review existing research on emotions and job performance, particularly emotions at work. The purpose of this article is to review existing research on job performance and emotions, particularly at the job relevant cognitions level. In this study, We propose research propositions about correlates of job performance and emotions while working. The purpose of this article is to develop and propose research propositions about potential correlates of perceived performance at the job relevant level. Correlates include those that should function mainly as antecedents of performance, such as job difficulty, job interest, and job effort in the study are emotions experienced concurrently with job performance. Therefore, We suggest the predictive direction and bivariate relationship of the set of among variables in explaining job performance and emotions.

  • For advanced civil engineering structures a service life of hundred up to hundred fifty and even two hundred years is sometimes required. The prediction of the performance of concrete structures over such a long period requires accurate and reliable predictive models. Most of the presently used, mostly experience based models don't have the quality and reliability that is required for reliable long-term predictions. The models designers are searching for should be based on an accurate description of the relevant degradation mechanisms. The starting point of such models is a realistic description of the microstructure of the concrete. In this presentation the need and the role of fundamental microstructural models for predicting the performance of concrete structures will be presented. An example will be given of a microstructural model with a proven potential for long-term predictions. Besides this also the role of models in general, i.e. in the whole design and execution process of concrete structures, will be dealt with. Finally recent trends in concrete research will be presented, like the research on self-healing cement-bases systems.

Immunotherapy in Head and Neck Squamous Cell Cancer

  • Prognosis in relapsed metastatic head and neck squamous cell cancer (RM-HNSCC) is dismal. Platinum based chemotherapy in combination with Cetuximab is used in first-line setting, while no further validated options are available at progression. Immunotherapy has produced durable clinical benefit in some patients with RM-HNSCC although the premises are several patients are nonresponders. Studies are ongoing to determine predictive factors and the ideal setting/combination of novel immunotherapies. In this paper, we discuss the past and present of immunotherapy in head and neck cancer and provide an up-to-date information regarding the potential ways to improve immunotherapy outcomes in HNSCC.

  • The increasing popularity of autonomous unmanned aerial vehicles (UAVs) can be attributed to their wide range of applications. 3D path planning is one of the crucial components enabling autonomous flight. In this paper, we present a novel 3D path planning algorithm that generates and utilizes curvature-based trajectories. Our approach leverages circular properties, offering notable advantages. First, circular trajectories make collision detection easier. Second, the planning procedure is streamlined by eliminating the need for the spline process to generate dynamically feasible trajectories. To validate our proposed algorithm, we conducted simulations in Gazebo Simulator. Within the simulation, we placed various obstacles such as pillars, nets, trees, and walls. The results demonstrate the efficacy and potential of our proposed algorithm in facilitating efficient and reliable 3D path planning for UAVs.

Spatiotemporal Impact Assessments of Highway Construction: Autonomous SWAT Modeling

  • In the United States, the completion of Construction Work Zone (CWZ) impact assessments for all federally-funded highway infrastructure improvement projects is mandated, yet it is regarded as a daunting task for state transportation agencies, due to a lack of standardized analytical methods for developing sounder Transportation Management Plans (TMPs). To circumvent these issues, this study aims to create a spatiotemporal modeling framework, dubbed "SWAT" (Spatiotemporal Work zone Assessment for TMPs). This study drew a total of 43,795 traffic sensor reading data collected from heavily trafficked highways in U.S. metropolitan areas. A multilevel-cluster-driven analysis characterized traffic patterns, while being verified using a measurement system analysis. An artificial neural networks model was created to predict potential 24/7 traffic demand automatically, and its predictive power was statistically validated. It is proposed that the predicted traffic patterns will be then incorporated into a what-if scenario analysis that evaluates the impact of numerous alternative construction plans. This study will yield a breakthrough in automating CWZ impact assessments with the first view of a systematic estimation method.

  • Cardiac organoids have emerged as invaluable tools for assessing the impact of diverse substances on heart function. This report introduces guidelines for general requirements for manufacturing cardiac organoids and conducting cardiac organoid-based assays, encompassing protocols, analytical methodologies, and ethical considerations. In the quest to employ recently developed three-dimensional cardiac organoid models as substitutes for animal testing, it becomes imperative to establish robust criteria for evaluating organoid quality and conducting toxicity assessments. This guideline addresses this need, catering to regulatory requirements, and describes common standards for organoid quality and toxicity assessment methodologies, commensurate with current technological capabilities. While acknowledging the dynamic nature of technological progress and the potential for future comparative studies, this guideline serves as a foundational framework. It offers a comprehensive approach to standardized cardiac organoid testing, ensuring scientific rigor, reproducibility, and ethical integrity in investigations of cardiotoxicity, particularly through the utilization of human pluripotent stem cell-derived cardiac organoids.

Predictive Value of Sensory Nerve Conduction in Carpal Tunnel Syndrome

  • Objective : Electrodiagnostic test has shown diagnostic sensitivity and specificity in carpal tunnel syndrome[CTS]. This study was to evaluate the correlation between clinical outcome of endoscopic carpal tunnel ligament release[ECTR] and the predictive value of sensory nerve conduction. Methods : From January 1998 to December 2004, 87 patients [44 right hand, 37 left hand, 6 bilateral hands] with CTS who underwent ECTR were followed up in our hospital for an average of 24 months. We retrospectively analyzed the results with previous medical records. All patients underwent electrodiagnostic test and ECTR. The patients were divided into three groups according to the electrodiagnostic test results. Group [A] was normal sensory nerve response, Group [B] was slowing sensory response and Group [C] was no sensory response. Improvement of the symptom after ECTR was assessed using a visual analogue scale[VAS] score. Results : Differences between the three groups on the correlation of severity of sensory potential and duration of preoperative symptoms were significant. The mean value of improved VAS scores for the three groups were $6.0{\pm}0.96$ in the Group A, $6.11{\pm}0.48$ in the Group B and $6.14{\pm}0.53$ in the Group C. There was no statistically significant difference between the severity of sensory nerve response and improvement in VAS score after ECTR. Complications included a wound infection, a case of skin necrosis, and two patients with persistent symptoms without any improvement. Conclusion : Although electrodiagnostic test has been known to be useful, sensory nerve response is considered not to be a good prognostic value for carpal tunnel syndrome after ECTR.
