Choline-Lipid Release from Normal and Transformed Cells

  • The effect of albumin on phosphatidylcholine (PC) metabolism in Hep-G2, 3T3-H.ras, and 3T3 cells pre-labelled with [Me-$^3H$]choline was studied. The [$^3H$]choline was more efficiently taken up and incorporated into cellular phospholipids in 3T3-H.ras cells than in Hep-G2 and 3T3 cells. In each of the three cell lines, most of the [$^3H$]choline metabolized into the phospholipids was incorporated into PC and only minor was incorporated into lysophosphatidylcholine (LPC). Bovine serum albumin stimulated the release of [$^3H$]LPC and [$^3H$]PC from each of the three cell lines pre-labelled with [$^3H$]choline. [$^3H$]PC was also released in the absence of albumin but [$^3H$]LPC was not. The efficiency of LPC secretion represented as the proportion of medium [$^3H$]LPC to cellular [$^3H$]choline lipid during a chase period is approximately 9 to 14 times greater in 3T3 cells compared with the transformed 3T3-H.ras and Hep-G2 cells. A similar comparison of published data for rat hepatocytes with Hep-G2 shows secretion to be 35~75 times greater from the rat hepatocytes than from Hep-G2. Also, PC secretion from 3T3 cells was 1.6 times more effective than from 3T3-H.ras, whereas rat hepatocytes secrete PC 2.8~3.8 times more effectively than does Hep-G2. The measurement of specific radioactivity of cellular PC in pre-labelled 3T3 cells showed it to be similar to that of the secreted PC. However, the specific radioactivity of secreted LPC was markedly lower than that of the cellular PC, which suggests that LPC is being secreted from a PC pool distinct from that used for PC secretion.

Effects of Active Mandibular Exercise for Mouth Opening Limitation Patients after Maxillomandibular Fixation Release: A Non-Randomized Controlled Trial (악간고정 제거후 개구장애 환자에게 적용한 능동적 하악운동의 효과: 비무작위 대조군 설계)

  • Purpose: The aim of this study was to evaluate the effects of active mandibular exercise (AME) in patients with limited mouth opening after maxillomandibular fixation (MMF) release. Methods: The study used a quasi-experimental, nonequivalent control group and a pre test-post test design. Sixty-two patients with Maxillomandibular Fixation Release were assigned to the experimental (n=31) or control group (n=31). The AME was performed in the experimental group for 4 weeks. The exercise AME consisted of maximal mouth opening, lateral excursion and protrusive movement. These movements were repeated ten times a day. After the final exercise of the day, the number of tongue blades used for mouth opening was noted. The effect of AME was evaluated after MMF release at different time intervals: a) immediately, b) after 1 week, c) after 2 weeks, d) after 4 weeks, and e) after 12 weeks. The exercise was assessed using the following criteria: a) mandibular movements, b) pain scores associated with maximal mouth opening, c) discomfort scores associated with range of movement, and d) daily life activities that involve opening the mouth. Results: The experimental group showed significant improvement regarding the range of mandibular movements (maximal mouth opening (F=23.60, p<.001), lateral excursion to the right side (F=5.25, p=.002), lateral excursion to the left side (F=5.97, p=.001), protrusive movement (F=5.51, p=.001)), pain score (F=39.59, p<.001), discomfort score (F=9.38, p<.001). Daily life activities that involve opening the mouth were more favorable compared to those in the control group. Conclusion: The AME in patients after MMF release is helpful for increasing mandibular movement range, decreasing pain and discomfort, and improving day life activities that involve opening the mouth. Therefore, AME is highly recommended as an effective nursing intervention.

Potential Water Retention Capacity as a Factor in Silage Effluent Control: Experiments with High Moisture By-product Feedstuffs

  • The role of moisture absorptive capacity of pre-silage material and its relationship with silage effluent in high moisture by-product feedstuffs (HMBF) is assessed. The term water retention capacity which is sometimes used in explaining the rate of effluent control in ensilage may be inadequate, since it accounts exclusively for the capacity of an absorbent incorporated into a pre-silage material prior to ensiling, without consideration to how much the pre-silage material can release. A new terminology, 'potential water retention capacity' (PWRC), which attempts to address this shortcoming, is proposed. Data were pooled from a series of experiments conducted separately over a period of five years using laboratory silos with four categories of agro by-products (n = 27) with differing moisture contents (highest 96.9%, lowest 78.1% in fresh matter, respectively), and their silages (n = 81). These were from a vegetable source (Daikon, Raphanus sativus), a root tuber source (potato pulp), a fruit source (apple pomace) and a cereal source (brewer's grain), respectively. The pre-silage materials were adjusted with dry in-silo absorbents consisting wheat straw, wheat or rice bran, beet pulp and bean stalks. The pooled mean for the moisture contents of all pre-silage materials was 78.3% (${\pm}10.3$). Silage effluent decreased (p<0.01), with increase in PWRC of pre-silage material. The theoretical moisture content and PWRC of pre-silage material necessary to stem effluent flow completely in HMBF silage was 69.1% and 82.9 g/100 g in fresh matter, respectively. The high correlation (r = 0.76) between PWRC of ensiled material and silage effluent indicated that the latter is an important factor in silage-effluent relationship.

Effects of ultrasound coupled with potassium permanganate pre-treatment of sludge on aerobic digestion

  • The biodegradability and decomposition efficiencies increase with the pre-treatment of sludge in a digestion process. In this study, the feasibility of ultrasound coupled with potassium permanganate oxidation as a disintegration method and digestibility of aerobic reactor fed with disintegrated sludge with ultrasound coupled potassium permanganate were investigated. The first stage of the study focused on determining the optimum condition for ultrasonic pre-treatment for achieving better destruction efficiency of sludge. The second part of the study, the aerobic digestibility of sludge disintegrated with ultrasound and potassium permanganate oxidation alone and combined were examined comparatively. The results showed that when 20 min of ultrasonic pre-treatment applied, the specific energy output was 49384 kJ/kgTS with disintegration degree of 58.84%. During the operation of aerobic digester, VS/TS ratios of digesters fed with disintegrated sludge decreased indicating that disintegration methods could obviously enhance aerobic digestion performance. The highest reduction in volatile solids was 75% in the digester fed with ultrasound+potassium permanganate disintegrated sludge at the end of the operation compared to digester fed with raw sludge. Total Nitrogen (TN) and Total Phosphorus (TP) levels in sludge supernatant increased with this combined method significantly. Besides, it promoted the production of ${\bullet}OH$, thus enhancing the release of Carbon (C), Nitrogen (N) and Phosphorus (P) from the sludge. Disintegration with all methods used in this study could not improve Capillary Suction Time (CST) reduction in disintegrated digesters during the operation. The results demonstrated that the combined ultrasound treatment and potassium permanganate oxidation method improves the biodegradability compared to control reactor or their single application.

  • In case of segmental injury of trachea, the primary repair is very difficult, because the trachea is fixed in place without movability from the surrounding tissue. In addition to special anatomical situation of trachea, any ideal artificial prosthesis for tracheal replacement is not still appeared. Many authors proposed several procedures for the tracheal repair, but satisfactory results were few. Among the proposed procedures end to end anastomosis of trachea was noted superior when both ends of trachea could be approximated by mobilization of upper and lower injured trachea. The author's experiment was designed to determine the effect about decreased tension on trachea when the hyoid bone was released downward from the surrounding muscular structures. The experimental dogs were divided into two groups, suprahyoid releasing group (SH R) and control group of intact hyoid. SH R group was subdivided into two groups accord ing to he degree of tension. The experimental results were as follow; 1. SH R group: In view of X-ray, the distance between the angle of mandible and the displaced hyoid bone was lengthened downward. And it's range was from 1.3cm (38%) minimally to 2.7cm(108%) maximally. 2. Control group: The distance between the angle of mandible and hyoid was same in both pre and postoperation. As the result of this experiment study, the suprahyoid release technique seems to be the efficient method that enable of release the trachea maximally. And it should be expected that the SH R technique is applicable clinically.

The effect of Myofascial Release and Infrared on the Range of Motion and Pain in Persons with Neck Pain (적외선치료와 근막이완술이 경부통증환자의 경추가동범위와 통증에 미치는 영향)

  • Purpose : The purpose of this study was to evaluate the Influence of Infra red(IR) and myofascial release(MRF) on the range of motion and pain in persons with neck pain. Methods : 24 subjects with neck pain participated in the experiment. All subject randomly assigned to the IR group and MFR group. Both groups receive 10minutes, 3 times per week during 3 weeks period. laser exercise(LEX) used to measure range of motion of neck and visual analog scale(VAS) used to measure pain. All measurements of each subject were measured at pre-treatment and post-treatment. Results : 1. The neck flexion, extension, right-sidebending, left-rotation, right-rotation range of motion of MFR group was significantly increased.(P<0.05). 2. The neck left-sidebending, left-rotation, right-rotation range of motion of IR group was significantly increased.(P<0.05). 3. As to compare two group, MFR group increases ROM more than IR group. Conclusion : These data suggests that MFR is more beneficial than IR.

  • The effect of equivalence ratio and velocity on the stability of flame in dump combustor was studied in an atmospheric pressure, laboratory scathe dump combustor operating natural gas. Traditionally, peak-to-peak pressure, fluctuation of the heat release rate and Rayleigh index were used to find and control the combustion instability. Cross correlation coefficients, Ci,j which is defined as the normalized value of the integration of the product of two of the mixer pressure, dump plane pressure and heat release rate, are introduced to see whether the flame is stable or not. Ci,j shows more sensitive to combustion status than Rayleigh index in steadily burning flame. Also, the result indicates that the amplitude of Ci,j between heat release and mixer pressure goes up before the flame at the rich de-stabilizing equivalence ratio near $\psi$=0.85. t means Ci:j at this case has a potential to detect the de-stablizing moment in prior to becoming unstable in dump combustor.

Effect of Myofacial Release and Cryotherapy in Acute Whiplash Injury

  • The purpose of this study is to provide the case of influence of myofacial release(MFR) and cryotherapy in acute whiplash injury. The subjects were consisted of 3 adults(2 males, 1 female) who had been diagnosed with whiplash injury. The performance period was 1 weeks, 6 times and 1 time is 40min(MFR: 20min, cryotherapy: 20min). We measured neck pain(Visual analogue scale, VAS) and cervical alignment(cervical curvature & line of gravity). All measurements of each subject were measured at pre-treatment and post-treatment. The results are listed below. The neck pain(VAS) index decreased from 7.54cm to 4.11cm. The cervical curvature increased from $23.62^{\circ}$ to $29.76^{\circ}$. The line of gravity decreased from 8.03mm to 3.66mm. Based on the results, it can be suggested that both MFR and cryotherapy can be used to neck pain and alignment in acute whiplash injury.

Sterilization Test of Microorganisms of Slow-released ClO2 Gas Gel-Pack (서방출형 이산화염소 가스 젤팩의 미생물 살균 시험)

  • Even though chlorine dioxide ($ClO_2$) is utilized in a pre-treatment due to its effective sterilizing activity for microorganisms and its safety for food, it has a limitation in maintaining freshness of the food product. In this study, a low-concentration $ClO_2$ gas was produced in a packaging form of air-permeable gel pack so that it could be released continuously over several days. The amount of $ClO_2$ gas emission and microbial inactivation effect against foodborne pathogens were measured during the release of $ClO_2$ gas. As a result of measuring the change of color in order to confirm whether the chlorine dioxide gas was eluted in the form of a sustained release, the yellowness was significantly higher at higher gel pack concentration and higher value during storage periods. The slow-released $ClO_2$ gel-pack showed clear inactivation effect against Escherichia coli and Staphylococcus aureus with 99.9% inactivation efficiency. As a result of measuring the sterilization effect of Listeria monocytogenes by the concentration of chlorine dioxide gas, the sterilization effect was increased as the concentration was increased. Therefore, the slow-released $ClO_2$ gel-pack is feasible to apply for industry usages.

  • Objective: MMP-8 is a neutrophil enzyme and its level increases in some inflammatory diseases, including periodontal disease. We knew that the lipopolysaccharide of E.coli(E-LPS) induced MMP-8 release from human neutrophils. E-LPS is known to induce the production and release of inflammatory cytokines through CD14, Toll-like receptor(TLR). In the present study, we investigated whether MMP-8 release by E-LPS is induced via CD14-TLR pathway and the cellular mechanism of MMP-8 release in human neutrophils. Material and methods: Human neutrophils were isolated from the peripheral blood of healthy donors and pre-incubated in medium containing antibodies against CD14, anti-TLR2 and anti-TLR4 or several inhibitors of microtubules and microfilaments and then incubated with E-LPS. The cells were treated TPCK and E-LPS simultaneously. The MMP-8amount in the culture medium was determined using ELISA. Results: E-LPS increased MMP-8release from neutrophils and its induction was inhibited by anti-CD14 and anti-TLR4 but not by anti-TLR2 antibodies. The inhibitors of microtubule and microfilament polymerization significantly decreased E-LPS-induced MMP-8release. TPCK inhibited E-LPS-induced MMP-8 release. Conclusion: These results suggest that MMP-8 release is induced by E-LPS via the CD14-TLR4 signal pathway in human neutrophils and may be depedent on microtubule and microfilament systems and $NF-{\kappa}B$ pathway.
