Wheat (Triticum aestivum L.) grain texture is an important determinant of milling properties and end product use. Two linked genes, puroindoline a (PINA) and puroindoline b (PINB), control most of the genetic variation in wheat grain texture. Wheat seed proteins were examined to identify PINA and PINB gene using two pre-harvest sprouting wheat cultivars; Jinpum (resistant) and Keumgang (susceptible).Wheat seed proteins were separated by two-dimensional electrophoresis with IEF gels over pH ranges: pH 3-10. A total of 73 spots were digested with trypsin resulting peptide fragmentation were analyzed by matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF/MS). Mass spectra were automatically processed and searched through NCBInr, SWISS-PORT and MSDB database with mono isotopic masses and complete gene sequence were found by UniProt database. Puroindoline a and puroindoline b that is responsible for grain texture related with baking performance and roughness. Two spots were found Pin b (16.7 kDa) and Pin a (16.3 kDa) in Jinpum compare to seven spots were identified Pin a (16.1 kDa, 16.3 kDa) and Pin b (16.7 kDa, 9.5 kDa and 14.4 kDa) in Keumgang. Some selected spots were identified puroindoline like grain softness protein (16.9 kDa, 17 kDa and 18.1 kDa) in Keumgang. Moreover, to gain a better inferring the identification of puroindoline related proteins using proteomics, we accomplished a complete gene sequence of PINA and PINB gene in pre-harvesting sprouting wheat seeds between resistant (Jinpum) and susceptible (Keumgang).
This study was conducted in order to figure out the energy consumption pattern in rice production system of Korea, with literature investigation focused on energy requirement in rice production system in worldwide. The investigation reveals that 24,994 MJ/ha was needed to produce 4,500kg of rice production or 5.55 MJ/kg was consumed. The major enegry consumptions are resulted from the application of nitrogen fertilizer, fuel for farm machinery, and farm machinery embodied energy, which showed somewhat different energy consumption pattern than that of the developed country, like, U.S.A. Based on the machinery chosen in this investigation, it was found that 32.7% of the fuel energy, 2,431.8MJ, was consumed in drying operation, 32.2%, 2.402MJ, in tillage and land preparation. 25.8%, 1.923.6MJ in harvest. A linear relationship was found in pre harvest energy input and output of rice production.
The purpose of this study was to establish postharvest management techniques including a temperature pre-conditioning protocol for maintaining fruit quality in newly developed early-season Korean pear cultivar 'Hanareum' (Pyrus pyrifolia Nakai). The fruits were treated with three different pre-conditioning temperatures (21, 25, or $29^{\circ}C$) for 4 days according to the harvest time (103 or 110 days after full bloom, DAFB). The percent weight loss was relatively low in the fruits subjected to low pre-conditioning temperature regardless of harvest time. The firmness of the fruits treated with pre-conditioning at $21^{\circ}C$ remained high during 20 days of simulated marketing at $25^{\circ}C$, although all treated fruits showed a general decline of firmness with extended time of simulated marketing. These fruits also showed higher appearance and a lower incidence of mealiness disorder symptoms. During the experimental periods, the production of ethylene was lower in the fruits pre-conditioned at $21^{\circ}C$ in comparison with those of treated at 25 and $29^{\circ}C$. High respiration rates were obvious in the fruits pre-conditioned at high temperature ($29^{\circ}C$), especially in the optimum-harvested fruits, where respiration was approximately two times higher than that of fruits exposed to $21^{\circ}C$ during pre-conditioning. However, the respiration rate was similar during simulated marketing at $25^{\circ}C$ regardless of harvest time. These results demonstrated that temperature pre-conditioning at $21^{\circ}C$ is a simple and effective postharvest technique for summer harvested Korean pear cultivars including 'Hanareum'.

BACKGROUND: This study was conducted to evaluate the residual characteristics and safety assessment of αcypermethrin and deltamethrin in minor crops, chives and spring onion cultivated in greenhouse. METHODS AND RESULTS: The insecticides α-cypermethrin 2% EC and deltamethrin 1% EC used in the experiment were diluted 1,000 times and then sprayed on chives and spring onion twice with 1-week intervals at 0, 7, 14, and 21 days before harvest. The residual insecticides were extracted from the minor crops using QuEchERS method and analyzed by GC-MS/MS. The average initial residues of α-cypermethrin and deltamethrin in chives after 21 days decreased from 2.74 to 0.82 mg/kg and 1.12 to 0.16 mg/kg, respectively. Similarly, in spring onion the residues after the same periods decreased from 0.26 to <0.01 mg/kg for α-cypermethrin and from 0.07 to <0.01 mg/kg for deltamethrin. CONCLUSION(S): The PHIs (pre-harvest intervals) for α-cypermethrin and deltamethrin in chives are recommended as 14 days before harvest with twice applications of the pesticides, whereas for α-cypermethrin in spring onion PHI of 7 days before harvest is recommended with 3 times of applications and PHI of 21 days for deltamethrin. The theoretical maximum daily intakes of cypermethrin and deltamethrin were 68.8% and 64.2%, respectively, indicating that residues of both compounds did not pose considerable health risks to consumers.
BACKGROUND: This study was performed to investigate pre-harvest residue limit (PHRL) in Chwinamul, to estimate biological half-life for residue of each pesticide. Chwinamul was sprayed with pesticides of standard and double application rate. Chlorfenapyr and fenarimol were sprayed once on Chwinamul at 10 days before harvest, and it was sampled 7 times and analysed the residual change of two pesticides. METHODS AND RESULTS: Chwinamul sample was extracted with acetonitrile and partitioned with dichloromethane, and pesticide residues were determined with GCECD. Method quantitation limit (MQL) of chlorfenapyr was 0.10 mg/kg and that of fenarimol was 0.02 mg/kg. Recoveries of chlorfenapyr at two fortification levels of 1.0 and 5.0 mg/kg were $94.2{\pm}$1.70 and $99.0{\pm}1.61%$, respectively. Recoveries of fenarimol at two fortification levels of 0.2 and 1.0 mg/kg were $92.1{\pm}2.14$ and $83.1{\pm}1.98%$, respectively. CONCLUSION(s): The biological half-lives of chlorfenapyr were about 3.5 days at standard application rate, and 3.4 days at double application rate. The biological half-lives of fenarimol were about 6.0 days at standard application rate, and 5.9 days at double application rate. The PHRLs of chlorfenapyr were recommended as 13.02 and 6.25 mg/kg for 10 and 5 days before harvest, respectively. And the PHRLs of fenarimol were recommended as 2.80 and 1.67 mg/kg for 10 and 5 days before harvest, respectively.
This study evaluated the hydration, gelatinization, and saccharification properties of rice processing for beverage development. The properties of rice were studied on 10 rice cultivars (Samkwang, Ilpum, Seolgaeng, Anda, Dasan-1, Goami-4, Danmi, American rice, Chinese rice, and Thai rice) and employing four kinds of pre-treatment methods (dry grain, wet grain, dry flour, and wet flour). The results showed that moisture content of rice was between 11.88~15.26%. Increase in soaking time along with highest water absorption was noted in American rice cultivar (46.81%). The water binding capacity of Thai rice was higher when compared to that of other rice flours. In addition, solubility and swelling power of rice were 4.52~26.65% and 0.19~2.05%, respectively. The amylose content of Goami-4 was higher in rice processing. Using a rapid visco analyzer (RVA), the initial pasting temperature of Danmi cultivar was found to be the highest; the peak viscosities of Anda cultivar and Dasan-1 cultivar, and Chinese rice were higher than of those of other rice flours. After saccharification, the pH, soluble solids content, and reducing sugar content of rice processed through different pre-treatment methods were in the range of 6.22~7.08, $4.67{\sim}16.07^{\circ}Brix$, and 0.35~11.67% (w/w), respectively. In terms of color values, the L-value of dry grain, a-value of wet (grain, flour), and b-value of dry sample (grain, flour) were found to be the highest. Assessment of various factors and cultivars characteristics of the raw grains are of importance in the development of rice beverage.
BACKGROUND: This study was performed to evaluate the residue dissipation of boscalid and spinetoram on crown daisy and sweet pepper affected by the morphology of the crop. The half-lives and dissipation rate constants for boscalid and spinetoram on crown daisy and sweet pepper were calculated. And then lower limit of 95% confidence interval for dissipation rate constant could be used to propose the pre-harvest residue limit. METHODS AND RESULTS: The pesticide products diluted according to the pesticide label were applied one time on crown daisy and sweet pepper at 2 field sites, respectively. Initial concentration of boscalid and spinetoram on crown daisy after application were in the range of 72.80~117.15 mg/kg and 2.82~4.67 mg/kg, respectively. And Initial concentration of boscalid and spinetoram on sweet pepper were in the range of 1.58~1.62 mg/kg and 0.10~0.21 mg/kg, respectively. Boscalid and spinetoram for crown daisy dissipted below the maximum residue limit(MRL) at 10 and 2 days after application, respectively. All residues concentration of boscalid and spinetoram for sweet pepper below the MRL at 0 day after application. The half-lives based on dissipation rate constant for boscalid and spinetoram on crown daisy were 4.2~4.9 days and 3.0~2.4 days respectively. And the half-lives for boscalid and spinetoram on sweet pepper were 6.7~7.0 days and 2.8~4.0 days respectively. CONCLUSION: The difference in initial concentration of boscalid and spinetoram among crop commodities were due to different crop morphology with larger surface areas. This study was suggested that pre-harvest residue limit would be calculated from lower limit of 95% confidence interval for dissipation rate constant and would be useful to protect consumers by controlling the pesticide residues in crop.
This study aimed to investigate the effects of pre-harvest sprouting (PHS)-damaged grains of rice on seed viability, germination percentage and seedling emergence rate. As comparing the seed lots of same cultivar 'Hopum' with different PHS percentages, seed lot with 8% PHS (high PHS percentage) showed 30% lower germination than that with 1% PHS (low PHS percentage). The difference of seedling emergence rate and seed viability percentages between the two seed lots was consistent with that of germination percentage. PHS-damaged brown rice was observed protruded embryo, discolored endosperm and fungi infected grains. PHS grains were classified into two groups, PHS-I showing endosperm discoloration in one third of a grain and PHS-II in more than half of a grain. The seed lot with high PHS percentage had much more PHS-II grains than that of low PHS percentage. PHS-II grains showed remarkably lower germination percentage than PHS-I grains and got severely moldy during germination test. In laboratory experiment, morphological changes and re-germination ability of seeds which dried after germination by soaking were tested. Sprouted seeds of 3mm shoot length showed half discolored endosperm and had a strong resemblance with PHS-II grains. Re-germination percentage of 3mm sprouted seeds sharply decreased than 2 mm sprouted seeds. Sprouted seeds which were treated for accelerated-aging (AA) for 24hr at $45^{\circ}C$ were significantly reduced in re-germination percentage compared to that of non-AA-treated sprouted seeds. These results indicate that PHS with over 3mm shoot length can severely damage not only grain appearance but also seed viability, germination percentage and seedling emergence rate. Therefore, we conclude that larger difference of germination percentage or seedling emergence rate between seed lots having smaller difference of PHS percentage might be caused by accelerated seed deterioration in PHS-damaged rice grains.

Seed dormancy is an adaptive trait in which seeds do not germinate under unfavorable environmental conditions. Low dormancy seeds are easily germinated under optimal environmental conditions, and these characteristics greatly reduce the yield and quality of crops. In the present study, we compared the pre-harvest sprouting (PHS) rate of two cultivars, Joun and Jopyeong, using the Winkler scale after heading day and temperature of the test. The PHS rate increased as the Winkler scale after heading day increased from 700℃ to 1100℃ and the temperature of the test increased. In all conditions, the PHS rate of Jopyeong was higher than that of Joun. RNA-sequencing was used to analyze the cause of the high PHS rate. We analyzed the biological metabolic processes related to the abscisic acid (ABA) metabolite pathway using the KEGG mapper with selected differentially expressed genes in PHS seeds. We found that the expression of ABA biosynthesis genes (OsNCEDs) was down-regulated and that ABA catabolic genes (OsCYP707As) was up-regulated in PHS seeds. However, the quantitative real-time PCR results showed that Joun had a higher expression of OsNCEDs than that of Jopyeong, but OsCYP707As did not yield a significant result. Joun displayed higher ABA content than that of Jopyeong not only during ripeness time but also during PHS treatment. Taken together, we provided evidence that the ABA content remaining in the seed is important to the PHS rate, which is determined by the expression level of the ABA biosynthesis gene OsNCEDs.
