Bacillus licheniformis KJ-9를 이용한 항균발효퇴비의 생산 (Production of Antifungal Compost by Using Bacillus licheniformis KJ-9)

  • 환경친화적인 항균발효퇴비를 생산할 목적으로 식물병원성 균주에 대하여 항균활성이 우수한 B. licheniformis KJ-9를 톱밥우분에 접종하여 고품질의 항균발효퇴비(B. licheniformis KJ 9 fermented compost, BLC)의 개발을 행하였다. B. licheniformis KJ-9가 생산한 물질의 온도 안정성은 $100^{\circ}C$, 10분간 열처리하여도 약 60% 이상의 항균활성을 유지하였으며, pH 안정성은 산성보다는 pH 7.0 이상의 중성 및 알칼리성 영역에서 높은 항균활성을 유지하였다. 퇴비를 제조하는 과정에서 BLC의 경우는 우분 냄새가 확실히 줄어들었으며 또한 퇴비의 발효시간도 3일 정도 단축되었다. 고추의 pot실험 결과 BLC를 첨가한 시험구 이외의 다른 시험구에서는 고추 잎이 마르고 약간의 황색반점이 발생하였다. 그리고 BLC를 이용한 시험구는 밭 흙만을 사용한 시험구 및 밭 흙에 기존의 판매퇴비(commercial available compost, CC)를 혼합한 시험구보다 잎의 수가 많고(1.5~2배), 줄기 및 뿌리의 성장이 빠르며, 줄기가 굵은(1.5~3배) 것으로 나타났다. 마늘 및 들깨를 재배한 현장적용 실험결과 두 시험구 모두 자연발생적인 병충해의 피해는 없었지만 BLC를 사용한 밭에서 마늘 및 들깨의 생육이 현저하게 빠른 것을 관찰할 수 있었다.

하수오니 시용이 옥수수 및 배추 생육과 토양의 화학성 변화에 미치는 영향 (Effect of Sewage Sludge Application on Growth of Corn and Chinese Cabbage and Chemical Properties of Soil)

  • 본 연구는 하수오니의 농경지 이용 가능성을 밝히고자 중랑, 가좌, 나주 및 용인 하수 종말 처리장에서 채취한 하수오니를 처리한 토양에서 옥수수와 배추를 재배하여 생육 및 토양의 화학성 변화에 미치는 영향을 검토하였다. 옥수수의 생육조사결과 오니 처리율이 증가할수록 초기 생육이 저조하였다. 배추의 생육 조사 결과 초기 3주간은 12.5 및 $25Mg\;ha^{-1}$ 처리구가 좋은 생육을 보였고 후기에는 50및 $100Mg\;ha^{-1}$처리구의 생육이 더 좋은 결과를 나타내었다. 그러나 최대 생장을 보인 처리구도 화학비료 시용구에서의 생육에는 미치지 못했다. 양이온치환용량, 유기물 및 질소의 경우는 처리간의 차이는 뚜렷하였지만 재배 전후에는 큰 차이가 없었다. Na, K, Ca, Mg 등의 염기류들도 비슷한 경향을 보였다. 토양포화침출액의 EC는 재배 전과 비교하여 재배후에 감소하였고 오니처리량이 많을수록 감소폭이 커서 재배전보다 오니 처리량 간의 차이가 줄었으나 유료인산 함량은 재배중 뚜렷한 증가 경향이 나타났다. 따라서 오니시용기준은 질소가 아닌 인산을 기준으로 하여 처리량을 결정하며 부족분은 화학비료로 보충하도록 하는 것이 바람직한 것으로 사료된다.

Effect of Azospirillum brasilense and Methylobacterium oryzae Inoculation on Growth of Red Pepper (Capsicum annuum L.)

  • Plant growth-promoting effects of rhizobacterial inoculation obtained in pot experiments cannot always be dependably reproduced in fields. In this study, we investigated the effect of inoculation with Azospirillum brasilense and Methylobacterium oryzae, which have displayed growth promoting effects in several pot experiments, on growth and fruit yield of red pepper under field condition in a plastic-film house. Four rows spaced 90 cm apart were prepared after application of compost ($10Mg\;ha^{-1}$), and red pepper seedlings (Capsicum annum L., Nocgwang) were transplanted in each row with 40-cm space. Experimental treatments were consisted of A. brasilense CW903 inoculation, M. oryzae CBMB20 inoculation, and uninoculated control. Twelve plots, 10 plants per plot, were allotted to the three treatments with four replicates in a completely randomized design. At the time of transplanting, 50 mL of each inoculum ($1{\times}10^8cells\;mL^{-1}$) was introduced into root zone soil of each plant, and re-inoculated at 7 and 14 days after transplant. Plant growth and fruit yield were measured during the experiment. Both A. brasilense CW903 and M. oryzae CBMB20 could not promote growth of red pepper plants. All growth parameters measured were not significantly different among treatments. There were large variations in fruit yield recorded on plot basis, and no statistically significant differences were found among treatments. The failure to demonstrate the expected plant growth promoting effect of the inoculants is possibly due to various environmental factors, including weather and soil characteristics, reducing the possibility to express the potential of the inoculated bacterial strains.

  • Bromoxynil은 쌍떡잎 특이적 제초제로써 폭 넓게 이용되고 있으며 반감기가 매우 짧다. Bromoxynil을 3,5-dibromo-4-hydroxybenzoic acid로 분해하는 nitrilase를 암호화한 bxn 유전자를 식물 벡터인 pGA482에 도입하고 Agrobacterium과의 동시배양을 통해 담배와 상추에 형질전환하였다. Kanamycin을 이용해 형질전환 식물체를 선별하고 완전한 식물체로 분화시켰다. Northern hybridization을 통해 bxn 유전자의 발현정도를 검정하고 liaf-disc와 pot assay를 통해 형질전환 식물체가 고농도의 bromoxynil에 저항성을 보임을 확인하였다.

  • This study was carried out to research microorganisms having the antifungal activity against ginseng Alternaria blight pathogen Alternaria panax and ginseng anthracnose pathogen Colletotrichum gloeosporioides. Eleven Bacillus strains. were isolated from Korean traditional soybean paste and Kimchi. Among the 11 isolates, DJ5, KC1, KC2 and KC4 showing antagonistic activity on the mycelial growth of A. panax and C. gloeosporioides in pairing culture were finally selected as the antagonistic microorganisms. Based on 16s rRNA sequence and phylogenetic tree analysis, they were identified as Bacillus spp.. The selected microorganisms were investigated antagonistic activity by measured leaf-segment colonization in pot test. When Bacillus sp. were injected after A. panax treatment, KC1, KC2 and KC4 showed similar effect to chemical pesticides treated control. To measure preventive effect of Bacillus sp, antagonistic microorganisms were injected and C. gloeosporioides was treated in pot. When measuring the effectiveness for the prevention of Anthracnose, All Bacillus spp. showed approximately 83~90 % degree of superior preventive effect. In general, The four Bacillus spp. isolated from Korean traditional fermented foods showed therapeutic effect of Alternaria blight and preventive effect of Anthracnose.

한국에서의 벼밤나방(Sesamia inferens Walker)의 생태와 약제방제에 관한 연구 (Studies on ecolgy and control for the purplish stem borer (Sesamia inferens Walker) in Korea)

  • 1. 한국에서는 벼 밤나방 (Sesamia inferens Walker)이 중남부지방에서 발생하여 이화명충과 유사한 가해상을 나타내고 있으며 유충태로 주로 지하부 도경에서 월동하고 년 2-3회 발생하였다. 2, 각 충태별 코기는 난이 $0.645\pm0,037\times0.399\pm0.757mm$, 유충이 $18.32\pm3.72mm$, 용이 $15.8\pm2.0mm$ 성충자아가 $11.56\pm1.35mm$, 웅아는 $10.36\pm0.87mm$였다. 3. 각충태별 기간은 각 화기에 따라 차가 컸으나 7-9월경인 2화기가 가장 짧았으며 그 기간은 란기$6.37\pm0.49$일, 유충기 $32.13\pm4.16$일, 용기 $13.38\pm0.92$ 성충기 $4.76\pm2.09$일였다. 4. 방제약제로는 Pot 시험결과 무처리 피해경율 $62.6\%$에 대하여 m.Parathion G $12.0\%$, EPNec$22.9\%$, Diazinon G $24.7\%$, Labaycid G $27.3\%$의 순으로 양호한 방제효과를 나타내었다.

세균성 시들음병에 대한 식물성 유산균(Lactobacillus sp.)의 저해효과 (Antagonistic Effect of Lactobacillus sp. Strain KLF01 Against Plant Pathogenic Bacteria Ralstonia solanacearum)

  • KLF01으로 명명된 세균이 토마토의 뿌리에서 분리 되었고 생화학적, 유전학적 검증을 통해 Lactobacillus sp.로 동종 되었다. In vitro 실험에서 식물성 세균인 Ralstonia solanacearum를 비롯하여, Xanthomonas axonopodis pv. citri, Xanthomonas campestris pv. vesicatoria, Eriwinia pyrifoliae, Eriwinia carotovora subsp. carotovora group에게도 저해 효과를 나타내는 것으로 확인되었다. 고추와 토마토를 이용한 in vivo 실험에서는 특히 R. solanacearum에 대해 대조구와 비교 시들음병의 진행을 저해하는 것으로 관찰되었다.

Propagation by In Vitro Zygotic Embryos Cultures of the Quercus myrsinifolia

  • Zygotic embryo culture was performed to propagate evergreen oak, Quercus myrsinifolia, which has recalcitrant seeds and is difficult to propagate by cuttings. Zygotic embryos appeared in WPM medium after 14 days, and after 56 days, they developed into complete plants with cotyledons and roots. The medium suitable for zygotic embryo culture was 1/4 WPM medium, showing a shoot growth of 2.43 cm and root growth of 8.7 cm after 8 weeks of culture. As a result of investigating the effect of GA3 on the growth of plants germinated from zygotic embryos through GA3 treatment, the best growth was shown in 0.5 mg/l GA3 treatment. The in vitro rooting and growth of IBA-treated zygotic embryo-derived plants were good in the 0.5 mg/l IBA treatment and rooting and shoot growth were not observed at higher concentrations. And the callus induction rate also increased as the concentration of IBA increased. Plants grown in vitro were transferred to a plastic pot containing artificial soil and acclimatized in a greenhouse for about 4 weeks, resulting in more than 90% survival. As a result of this study, the zygotic embryo culture method was confirmed to be effective for mass propagation of Q. myrsinifolia. The results of this study are expected to contribute significantly to the mass propagation of elite Q. myrsinifolia.

"고사십이집(攷事十二集)"의 조리가공에 관한 분석적 연구(2) (A Study on the Cooking in 'The Kosa-sibi Jip' (2))

  • In this paper, twenty-five kinds of food presented in Sooljip(戌集) 5 and 6 of Food collections of 'Kosa-sibi Jip(攷事十二集)' have been classified into four : Staple food, subsidiary food, Tuck(rice cake) and Han-gwa(Korean confectionery), and Tang-jng and tea. Cooking processes have been examined and scientifically analyzed in terms of cooking, Fourteen kinds of Jook (thick gruel with cereal) as well as Urak-Jook were presented among the methods of making Jook, one of staple foods. Milk and ground rice were boiled together into Urak-Jook, which was nutritious because of carbohydrate, added to milk. Hong-sa Myun was mode of ground shrimps, ground bean, ground rice and flour which were kneaded together. It was a nutritiously balanced food. Nineteen kinds of Kimchi presented in this book were classified by the recipes. The five of Jook-soon Ja, U-so Ja, Tam-bok Ja and Jo-gang were made by adding red malt and cereals(boiled rice or candies). Jo-gang, Jo-ga and Jo-gwa-chae were made by adding salt and rice wine. With salt and fermenters added, eight were made. Chim-jup-jeo-ga was made by adding Jang(soy-bean sauce) and the inner chaff of wheat instead of salt. The four of Ka-za-san, Hwang-gwa-san, Tong-gwa-san and Jo-gang were made by adding salt and vinegar. Jo-gang was made by adding salt, rice wine, residue of rice wine and candies. The four of Kae-mal-ga, Ku-cho-chim-chae, Un-gu-hwa and Suk-hwa-chim-chim-chae were made by adding salt and spices. San-got-Kimchi was made without salt. San-got-Kimchi and Suk-hwa-chim-chae were made originally in Korea. Suk-hwa-chim-chae, in particular, was first classified as a kind of Kimchi in this book and oysters were added, which is notable. Pork could be preserved longer when smoked oven the weak fire of thatch ten days and nights. Dog meat was sauced and placed on the bones in a pot. A porcelain was put on the top of the pot. Flour paste sealed the gap between the porcelain and the pot. Some water was poured into the porcelain, and the meat was steamed, with two or three thatched sacks burned, which was a distilled dry steaming. This process has been in use up to now. Various cooking methods of chicken were presented from in Umsik-dimi-bang to in Chosun Musang Sinsik Yori Jebup. These methods were ever present regardless of ages. Such measuring units as Guin(斤) and Nyang(兩) were most frequently used in cooking processes of this book, except in case of Jang(soy bean sauce), vinegar and liquor. Twenty eight kinds of kitchenware and cookers were used, of which porcelains wee most used and pans and sieves followed. The scientific eight cooking methods were as follows. First, salt was refined through saturated solution. Next, it was recommended Hong-sa Myun containing shrimps should not be taken along with pork, which is thought to be a proper diet in terms of cholesterol contained by shrimps and pork. Third, meat was coated with thin gruel and quickly roasted and cleared of the dried gruel membrane, which prevented nutrients from exuding and helped to make the meat well-done. Fourth, The fruit of paper mulberry trees has the protease which can soften meat. Therefore when meat was boiled with th fruit of paper mulberry trees, it can be softened easily. Fifth, pork was smoked over the weak fire of thatch. Sixth, in cooking dog meat, distilled dry steaming raised the boiling point and made it possible to preserve meat longer. Seventh, in boiling the sole of a bear, lime was added, which made meat tender by making the pH lower or higher than that of raw meat. Finally, in boiling down rice gluten, a porcelain in the pot prevented boiling over the brim, which is applied to pots in which to boil medical herbs.

자생식물로부터 내건성 식물의 최적인자 선발과 생육특성 (Selection Indices to Identify Drought-tolerance and Growth Characteristics of the Selected Korean Native Plants)

  • 본 연구에서는 내건성 식물 선발을 위한 최적 인자를 구명하고, 이들 내건성 식물들의 생장특성을 조사하였다. 건조저항인자에 대한 나머지 건조저항일(RD), 엽면적(LD), 단위증산량(UTR), 상대함수량(RWC), 상대수분손실량(RWL), 엽면적(LA), 기공수(SN) 및 기공면적(SA) 등 6개 인자들을 비교한 결과 상대수분손실량과 단위증산량이 건조저항일에 많은 영향을 주는 것으로 나타났다. PCA분석 결과 SA, LA, RD는 한그룹으로 RWC와 SN은 다른 한 그룹으로 구분되었고, UTR은 SA, LA와, RWL은 RWC와 SN과 음의 상관을 보였다. 상기 결과를 종합하여 느릅나무, 도깨비바늘, 뚝갈, 매듭풀, 새, 더위지기, 맑은대쑥, 독말풀, 긴담배풀, 소리쟁이, 비수리, 장구채, 개기장, 도깨비가지를 내건성 식물 종으로 선발하였고, 이들에 대한 생장시험을 수행하였다. 절토사면지에서 내건성 식물의 줄기생장은 식물 종간에 약간의 차이를 보였는데, 줄기생장과 잎의 수는 느릅나무를 제외하고 폿트에서는 차이를 보이지 않았다. 그러나 절토사면지에서 내건성 식물 뿌리 생장과 T/R율은 폿트생장과 큰 차이를 보였고, 특히 T/R율은 폿트묘보다 월등히 낮았다. 이 결과로 보아 선발된 내건성 식물은 절토사면지와 같이 척박한 토양에서도 잘 적응할 것으로 나타나 내건성 식물의 육종 등에 이용될 수 있을 것으로 판단된다.
