수확 전후 칼슘-키토산 및 고농도 CO2 단기처리에 의한 '설향' 딸기 과실의 저장성 증진 (Increase of Strawberry Fruit Shelf-life through Preharvest Spray of Calcium-chitosan and Post-harvest Treatment with High Pressure CO2)

  • 수확 전후 키토산 및 $CO_2$ 단기처리가 '설향' 딸기 과실의 저장성과 품질에 미치는 영향을 조사하기 위하여 본 실험을 수행하였다. 과실 경도는 수확 시기가 늦어질수록 감소하였고 수확 전 칼슘-키토산 누적 처리는 수확 당시의 과실 경도를 유의하게 증가시키지 못하였다. 그러나 수확 전 키토산 처리와 수확 후 100% $CO_2$ 단기처리를 병행하였을 때 저장 중 경도 유지와 과피 무름 및 부패를 억제하였다. 수확 전 키토산 살포는 과실의 무름 장해 및 부패 발생 억제에 효과적이나 수확 시기가 늦어질수록 칼슘-키토산의 수확 전 처리 효과가 감소하였다. 결론적으로 수확 전 칼슘-키토산과 수확 후 $CO_2$의 병행 처리는 딸기 과실의 경도 증가는 물론 과피 색의 변화 및 부패를 억제시키므로 딸기의 저장성 증진을 위한 수확 후 처리기술로 유용하였다.

수입 과일에 살포된 수확 후 처리농약(Post-harvest pesticide)의 경시적 변화 (Residue Levels of Pesticides in Post-Harvest Treated Import Fruits During Storage)

  • 수입과일을 대상으로 captan, chlorpyrifos, methidathion 및 kresoxim-methyl 을 각각 1000, 250, 400 및 157 $mgkg^{-1}$ 농도로 수확 후 처리 하여 저장 기간에 따른 경시적 변화를 관찰한 결과 captan은 처리 직후 과일 전체에서 0.9-12.5 $mgkg^{-1}$, 과육에서 ND-0.23 $mgkg^{-1}$ 으로 조사 되었으며, 4주 후에는 과일 전체에서 0.7-3.2 $mgkg^{-1}$, 과육에서 ND-0.67 $mgkg^{-1}$으로 조사되었다. chlorpyrifos은 처리 직후 과일 전체에서 0.4-2.2 $mgkg^{-1}$, 과육에서 ND-0.32 $mgkg^{-1}$으로 조사 되었고, 4주 후에는 과일전체에서 0.3-0.9 $mgkg^{-1}$, 과육에서 ND-0.02 $mgkg^{-1}$으로 조사 되었다. methidathion은 처리 직후 과일 전체에서 0.7-3.1 $mgkg^{-1}$, 과육에서 ND-0.05 $mgkg^{-1}$으로 조사되었고, 4주 후에는 과일 전체에서 0.4-2.0 $mgkg^{-1}$, 과육에서 ND-0.05 $mgkg^{-1}$으로 조사 되었다. kresoxim-methyl은 처리 직후 과일 전체에서 1.3-2.1 $mgkg^{-1}$, 과육에서 ND-0.16 $mgkg^{-1}$으로 조사 되었고, 4주 후에는 과일 전체에서 1.3-1.8 $mgkg^{-1}$, 과육에서 ND-0.15 $mgkg^{-1}$으로 조사 되었다. 농약처리 4주 후 농도의 감소율은 captan이 52%로 가장 높았으며 다음으로 chlorpyrifos, methidathion 및 kresoximmethyl이 각각 47, 41 및 11% 순으로 조사 되었다.

Cloning of Phospholipase D from Grape Berry and Its Expression under Heat Acclimation

  • To investigate whether phospholipase D (PLD, EC 3.1.4.4) plays a role in adaptive response of post-harvest fruit to environment, a PLD gene was firstly cloned from grape berry (Vitis Vinifera L. cv. Chardonnay) using RT-PCR and 3'- and 5'-RACE. The deduced amino acid sequence (809 residues) showed 84.7% identity with that of PLD from Ricinus communis. The secondary structures of this protein showed the characteristic C2 domain and two active sites of a phospholipid-metabolizing enzyme. The PLD activity and its expression in response to heat acclimation were then assayed. The results indicated PLD was significantly activated at enzyme activity, as well as accumulation of PLD mRNA and synthesis of new PLD protein during the early of heat acclimation, primary suggesting that the grape berry PLD may be involved in the heat response in post-harvest grape berry. This work offers an important basis for further investigating the mechanism of post-harvest fruit adaptation to environmental stresses.

온주밀감 부패 곰팡이의 분리 및 동정 (Collection and Identification of Molds from Citrus Oranges during Post-Harvest Storage)

  • 수확 후 감귤의 장기저장은 주로 곰팡이에 의한 부패로 어려움을 겪어왔다. 본 연구에서는 감귤부패 곰팡이를 수집, 동정함으로써 수확후 저장성을 증대하는데 기여하고자 하였다. 1994년 가을부터 1995년 봄에 걸쳐 대기중에 노출된 채로 상온에 저장 중 부패된 온주밀감으로부터 14개의 상이한 균주를 분리하였다. 저장 중 각 곰팡이에의한 병해의 발생빈도는 Penicillium italicum 25.8%, Monila candida 19.8%, Alternaria citri 18.1%, Mucor hiemalis 11.0%, Phomopsis citri 6.6%, Botrytis cinerea 5.5%, Phoma citricarpa 3.8%, Glomerella cingulata 3.8% 그리고 P. digitatum 1.1% 순이었고, 기타 곰팡이가 4.5%를 차지 하였다. 한편 생리적 특성상 동정된 모든 균주가 펙틴 분해효소를 생산하였다. 이러한 연구결과는 감귤의 부패와 곰팡이의 펙틴분해능간에는 연관성이 있음을 시사한다. 수집된 곰팡이 균주는 감귤의 부패를 방지하는 연구에 이용될 것이다.

자외선 유도 형광의 사과 성숙도 평가 적용 (UV/blue Light-induced Fluorescence for Assessing Apple Quality)

  • Chlorophyll fluorescence has been researched for assessing fruit post-harvest quality and condition. The objective of this preliminary research was to investigate the potential of fluorescence spectroscopy for measuring apple fruit quality. Ultraviolet (UV) and blue light was used as an excitation source for inducing fluorescence in apples. Fluorescence spectra were measured from 'Golden Delicious' (GD) and 'Red Delicious' (RD) apples using a visible/near-infrared spectrometer after one, three, and five minutes of continuous UV/blue light illumination. Standard destructive tests were performed to measure fruit firmness, skin and flesh color, soluble solids and acid content from the apples. Calibration models for each of the three illumination time periods were developed to predict fruit quality indexes. The results showed that fluorescence emission decreased steadily during the first three minutes of UV/blue light illumination and was stable within five minutes. The differences were minimal in the model prediction results based on fluorescence data at one, three or five minutes of illumination. Overall, better predictions were obtained for apple skin chroma and hue and flesh hue with values for the correlation coefficient of validation between 0.80 and 0.90 for both GD and RD. Relatively poor predictions were obtained for fruit firmness, soluble solids content, titrational acid, and flesh chroma. This research has demonstrated that fluorescence spectroscopy is potentially useful for assessing selected quality attributes of apple fruit and further research is needed to improve fluorescence measurements so that better predictions of fruit quality can be achieved.

A NONDESTRUCTIVE NIR SPECTROMETER : DEVELOPMENT OF A PORTABLE FRUIT QUALITY METER

  • The quality of agricultural products is very important factor for consumers. In Japan, quality is sometimes more important than cost. Usually, the quality of fresh food products is determined in terms of shape, color, size, etc. However, these indices are not always associated with taste, leaving consumers to complain. Recently, two types of the fruit quality meter (a tabletop type - K-FS200 and a portable type - K-BA100, Kubota Corp.) using NIR technology were introduced in Japan. A tabletop instrument is for post harvest use and a portable one is for precision agriculture use. The both meters use the NIR region from 600nm to 1000nm in the interactance mode to determine quality factors related to taste. The instruments can measure sugar content and acidity of such fruit as apples, tomatoes, tangerines and other fruits. The measurement is timely, nondestructive and precise. For example, the coefficient of variation (CV) is less than 6% for sugar in most fruits. The K-FS200 has been evaluated in supermarkets, grading facilities, and wholesalers in Japan. The introduction of the K-FS200) has drawn attention to taste quality and its use is becoming more popular. In addition, researchers or farmers are becoming interested in measuring product ingredient not only after harvest but also during growing in the field so that they can make intelligent judgements concerning soil amendments, such as fertilizers and water, employs the fiber probe for flexible measurement and is battery powered for field use. Design of the fruit quality meters will be discussed. Applications to fruit quality will be presented.

하이퍼 스펙트랄 반사광 및 형광 산란을 이용한 사과 품질 측정 (Apple Quality Measurement Using Hyperspectral Reflectance and Fluorescence Scattering)

  • Hyperspectral reflectance and fluorescence scattering have been researched recently for measuring fruit post-harvest quality and condition. And they are promising for nondestructive detection of fruit quality. The objective of this research was to develop a model, which measure the quality of apple by using hyperspectral reflectance and fluorescence. A violet laser (408 nm) and a quartz tungsten halogen light were used as light sources for generating laser induced fluorescence and reflectance scattering in apples, respectively. The laser induced fluorescence and reflectance of 'Golden Delicious' apples were measured by using a hyperspectral imaging system. Fruit firmness, soluble solids and acid content were measured using standard destructive methods. Principal component analyses were performed to extract critical information from both hyperspectral reflectance and fluorescence data and this information was then related to fruit quality indexes. The fluorescence models had poorer predictions of the three quality indexes than the reflectance models. However, the prediction models of integrating fluorescence and reflectance performed consistently better than the individual models of either reflectance or fluorescence. The correlation coefficient for fruit firmness, soluble solid content, and tillable acidity from the integrated model was 0.86, 0.75, and 0.66 respectively. Also the standard errors were 6.97 N, 1.05%, and 0.07% respectively.

과수육종에 있어 생명공학의 이용 전망 (The Prospectss and Utilization of Biotechnology for the Improvement of Fruit Breeding)

  • The major objectives of fruit breeding lie in improvement of cultivar, easy to be cultivated and of high quality, in order to produce unexpensive, delicious fruit both for fresh fruit market and processing. Recently, fruit breeding in Korea has contributed to breeding of several superior cultivars in major fruit crops, resulting in appreciable improvement in qualities such as skin color, taste and fruit-bearing habit concerned with productivity. In spite of accomplishments mentioned above, the need for both highly disease-resistant cultivars and long-keeping, physiological disorder-resistant cultivars to meet long distance transsportation in the temperate fruit crops of apples, oriental pears, stone fruits such as peaches, and grapes grown in Korea is rapidly pressing more than ever, as cultivars of today susceptible to pests and diseases and vulnerable to physiological disorders are very expensive and time-consuming in post-harvest handling and management. Thus, imporvements made in the above problems through breeding level will lead to the really enhanced productivity in fruit industry. The major impediments of tree size, the long length of juvenile period and the highly heterogeneous genetic composition to the improvement of fruit crops are responsible for the lower amount and rate of improvements of fruit crops as compared to annuals. Considering the expected limitations of the above problems to be solved through conventional breeding methods and strategy, a turning point of breeding a near perfect cultivar would be laid down if innovative breakthroughs in biological technology will be realized in applying some of the techniques of genetic manipulation at the molecular level to the cultivar improvement of fruit crops, such as the selective insertion of DNA carrying genes that govern desirable characteristics. More than anything else, those traits such as fruiting habit deciding productivity, elements of fruit qualities conditioned by monogene, and disease-and pest-resistance of vital importance for successful fruit growing are urgently desired to be improved by advancement of biotechnology for they are more than difficult and need long period to be attained through conventional breeding method.

배 신품종 '창조'의 성숙 중 품질 요인 변화 및 수송온도 환경에 따른 반응성 (Evaluation of the Fruit Quality Indices during Maturation and Ripening and the Influence of Short-term Temperature Management on Shelf-life during Simulated Exportation in 'Changjo' Pears (Pyrus pyrifolia Nakai))

  • 국내에서 육성된 중생종 동양배 품종인 '창조'는 성숙기간 중 지속적으로 과실이 비대하여 600g대의 편원형 과실로 발달하였다. 성숙기간 중 경도는 경시적으로 저하되어 만개 후 153일인 9월 13일에 32.58N으로 조사되었고 만개 후 160일에는 26.44N으로 급격히 경도가 저하되었다. 식감과 관련된 품질요인 중 과실의 전분함량은 만개 후 153일에 0.737, 만개 후 160일에 0.451로 전분함량이 급속히 감소하였고, 과육의 세포벽함량을 조사하였던 결과, 과실비대가 증가하고 과실성숙의 진행과 더불어 알코올불용성물질 함량은 낮아지는 경향을 보였다. 과피색차 중 적색도($a^*$)는 만개 후 139일 이후 양의 값을 보여 외관상의 과피녹색 발현이 소실되었는데 과피의 SPAD 값을 조사한 결과에서도 동일하게 엽록소의 감소를 확인할 수 있었다. Hue angle 역시 성숙과 연화기간 중 유의하게 감소하는 경향을 보여 수확시기의 지표로 활용 가능하다고 판단되었다. 성숙기간 중 과육 내에 축적되는 당 성분을 분석한 결과, 만개 후 153일에 총량이 최대치를 보였는데 전 생육시기에 걸쳐 전체 당성분 중 과당이 우점하고 있었고, 과실성숙도가 높아질수록 자당의 비율이 유의하게 증가하여 만개 후 160일에 수확한 과실에서는 자당이 과당을 넘어 우점하여 과숙단계로 생각되었다. 따라서, 배 신품종 '창조'의 적정 수확시기는 만개 후 153일인 9월 13일 이전으로 평가되었고 수확시기를 놓친 경우에는 경도 등 과실의 품질이 급격하게 하락하는 것으로 사료되었다. 대만 지역 수출을 모의하여 단기수송온도 설정을 위한 수확 후 저온순화 실험을 실시한 결과, 만개 후 146일에 수확한 과실이 만개 후 153일에 수확한 과실에 비하여 $25^{\circ}C$ 유통 후 높은 경도를 유지하는 경향을 보였고 최종 수송 목표온도인 $5^{\circ}C$로 저하되는 속도가 느릴수록 과육장해의 발생심도 및 에틸렌발생량이 높은 경향을 보였으므로 '창조' 품종에 있어 동남아 지역 수출 시 생리장해 및 과실품질 유지를 위해서는 수확시기를 만개 후 146일 이전에 설정하는 것이 바람직할 것으로 판단되었다. 본 연구결과를 바탕으로 '창조' 배의 목적별 수확시기 결정 및 과실의 선별 기술에 일조하고 수출 시장에서의 한국산 배의 신인도 확보에 기여할 것으로 기대된다.

Feasibility Study on Styrofoam Layer Cushioning for Banana Bulk Transport in a Local Distribution System

  • Purpose: This study evaluates a new banana bulk packaging method under the real transport conditions of Sri Lanka. Methods: A field evaluation of optimized 8-mm thick Styrofoam sheets used as the cushioning material was applied. A trial transport was conducted from Thambuttegama to Colombo using a medium-sized open truck, with banana leaves as the control material. Data were recorded at the farmer, transporter, retailer, and consumer stages of the supply chain. Mechanical damage, physiological loss in weight, fruit firmness, total soluble solids, ripeness index, visual quality ratings, and the physical damage index of the bananas were measured at each stage. A cost-benefit analysis was also conducted for both packaging methods. Results: The 8-mm styrofoam sheets significantly reduced (p < 0.05) the mechanical damage from 26.3% to 12.9% compared to the conventional method for long-distance transport, and the physiological loss in weight showed a decrease of 2.88%. The loss of firmness of the fruits followed a simmilar pattern for both methods until reaching the retailer, but at the consumer was significantly higher (p < 0.05) for the control. However, the physical damage index at the retail stage for the control showed symptoms of physical injury, whereas the bananas transported using the cushioning materials exhibited only minor symptoms. Further, the visual quality of the fruits after transport from the farmer to the consumer was preserved, which is one of the main factors affecting consumer preference and retail price. The proposed method increases the profit margin by 51.2% for Embul bananas owing to the reduced postharvest losses. Conclusion: The 8-mm thick Styrofoam sheets reduced the physical damage to the bananas, with the quality parameters maintained at the prefered level. Moreover, profits may be increased.
