Physicochemical Characteristics and Antioxidant Activity of Heated Radish (Raphanus sativus L.) Extracts (열처리 무 추출물의 이화학적 특성과 항산화 활성)

  • This study investigated the changes of physicochemical characteristics and antioxidant activity of heated radish (Raphanus sativus L.). Raw radish was heated at various temperatures ($110{\sim}150^{\circ}C$) for 2 hr. The heated radish was extracted with 70% ethanol and then fractionated with hexane, ethyl acetate, and aqueous. Total polyphenol contents, $IC_{50}$ value of electron donating ability (EDA), and total antioxidant activity (AEAC) increased with increasing heating temperature. The maximum total polyphenol content was $256.26{\pm}9.61$ mg/100 g at $150^{\circ}C$ (control: $27.90{\pm}1.28$ mg/100 g), $IC_{50}$ value was $1.34{\pm}0.004$ mg/mL at $150^{\circ}C$ (control: $34.93{\pm}0.039$ mg/mL), and AEAC was $53.10{\pm}1.155$ mg AA eq/g at $150^{\circ}C$ (control: $6.721{\pm}0.122$ mg AA eq/g). The ethyl acetate fraction showed higher total polyphenol contents and stronger antioxidant activities than hexane and aqueous fractions. Total polyphenol content was 133.62 mg/100 g (at $140^{\circ}C$), $IC_{50}$ values of EDA and AEAC content were 0.39 mg/mL (at $150^{\circ}C$) and 183.72 mg AA eq/g (at $140^{\circ}C$), respectively.

Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening (왕겨초액의 대량생산과 항노화 및 미백 효과)

  • Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Dioscorea batatas Decne (마(Dioscorea batatas Decne)의 항균, 항산화 및 항혈전 활성 평가)

  • Yam (Dioscorea batatas Decne) has been recognized as healthy food due to its various biological activities, such as anti-obesity, anti-constipation, anti-proliferation, anti-mutagenic activity, and decrease of blood glucose and cholesterol level. In this study, the methanol extract and its organic solvent fractions were prepared from dried Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activity were evaluated, respectively. The 66.7% of methanol extract was fractioned into water residue, and most of total-polyphenol/total-flavonoids were found in ethylacetate fraction. This ethylacetate fraction at $500\;{\mu}g/disc$ concentration showed strong antibacterial activity against gram-negative and gram-positive bacteria, except Staphylococcus aureus. Antifungal activity was not observed in methanol extract and its fractions. But, strong antithrombin activity was found in methanol extract. At 4.8 mg/mL concentration, thrombin time was 99.5 sec, which is coincides with 30% activity of aspirin, a commercial available antithrombosis agent. The ethylacetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $80.5\;{\mu}g/mL$), compared to vitamin C ($IC_{50}$ of $15.2\;{\mu}g/mL$). Also, the ethylacetate fraction showed strong SOD-like activity and reducing power, which are coincide with 43% of vitamin C and 82.7% of butylated hydroxytoluene activity, respectively.

Biological Activities of Magnolia denudata Desr. Flower Extracts (목련(Magnolia denudata Desr.) 꽃 추출물의 생리활성)

  • The antioxidant, antiproliferation, and nitrate synthesis inhibitory effects of Magnolia denudata extracts (ME) were evaluated. The ME was extracted with 70% (v/v) ethanol and fractionated with solvents of hexane, chloroform, ethyl acetate, n-buthanol and aqueous. The ethyl acetate fraction contained the highest phenolic and flavonoid contents of 427.10 mg garlic acid eq/g and 356.05 mg catechin eq/g, respectively. The ethyl acetate fraction showed strong 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity with a 50% inhibition concentration ($IC_{50}$) of 0.20 mg/mL and total antioxidant activity was 0.90 mg AA eq/100 mg. From the results of cytotoxic effects of HCT116, NCL-H460, and HepG2 human cancer cells by MTT assay on the ME and its solvent fraction, chloroform fraction showed the highest cytotoxic effect ($IC_{50}$ value: 0.14, 0.37, and 0.41 mg/mL, respectively). Nitrate synthesis inhibitory effect of ME and its solvent fractions on nitric oxide synthase activity in LPS stimulated RAW 264.7 cells were decreased in dose-dependent manners, and $IC_{50}$ value of hexane and chloroform fractions were 0.39 and 0.49 mg/mL, respectively.

Antioxidant Properties and Protective Effects of Inula britannica var. chinensis Regel on Oxidative Stress-induced Neuronal Cell Damage (금불초 추출물의 항산화 효과 및 산화 스트레스에 대한 신경세포 보호작용)

  • The antioxidant properties and protective effects of Inula britannica on ${H_2}{O_2}$-induced SH-SY5Y neuroblastoma cell damage were investigated. A series of solvent fractions, including hexane(Fr.H), petroleum ether, chloroform, ethyl acetate(Fr.EA), and water fraction(Fr.W), were prepared from the 70% methanol extracts of Inula britannica. Fr.W had the highest total contents of phenolics and flavonoids, followed by Fr.EA. The antioxidant properties of the fractions were also evaluated by analyzing their scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radicals, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals, and nitric oxide. Fr.W showed the strongest activities in all assays. The concentrations of Fr.W that resulted in 50% reductions of the DPPH and ABTS radicals were 20.7 ${\mu}g$/mL and 39.4 ${\mu}g$/mL, respectively. Fr.W showed the weakest cytotoxic activities on the SH-SY5Y cells, whereas it effectively protected ${H_2}{O_2}$-induced cell death, increasing cell survival by 35.0-77.0% at a concentration range of 62.5-250 ${\mu}g$/mL. In this range, Fr.W also significantly decreased intracellular ROS levels by 34-39%. Overall, the antioxidant properties of Inula britannica can contribute to rescuring neuronal cells from oxidative stress-induced cell injury.

Comparative Analysis of the Amino Acids and Effective Components from Viola mandshurica (자화지정(Viola mandshurica)의 아미노산과 유용성분 비교 분석)

  • This study compared amino acids and effective components of Viola mandshurica produced in Korea with those of Chinese plants, as part of a study on nutritional andfunctional materials, to aid in the development of a valuable food. The contents of reducing sugar, free sugar, polyphenols, and soluble protein in the flowers of V. mandshurica were higher than in other fractions, being 10,485.11 mg%, 118.85 mg%, 1,259.26 mg%, and 23.67 mg%, respectively. Total flavonoid content of Chinese leaves was the highest at 2,370.75 mg%, followed by 1,844.43 mg% in Korean leaves. Free amino acids were highest in Korean leaves (5,345.21 mg%), followed by roots (4,692.24 mg%), flowers (3,807.87 mg%), and leaves of China plants (3,613.81 mg%). Arginine accounted for 55% of free amino acids in the roots, and all plant fractions contained more valine, aspartic acid, glutamic acid, alanine, and arginine, than other amino acids. The total content of amino acid derivatives was the highest (9,058.80 mg%) in Korean leaves. Especially, $\alpha$-aminoadipic acid constituted over 50% of total amino acid derivatives in all plant parts of V. mandshurica. When mineral contents were analyzed, leaves grown in China showed the highest content of 5,917.57 mg%, and the contents of K, Ca, and Mg were much greater than those of other minerals, together comprising over 95% of total mineral content in leaves and flowers from Korea.

Antioxidant and growth inhibitory activities of Mesembryanthemum crystallinum L. in HCT116 human colon cancer cells (아이스플랜트의 항산화 및 HCT116 인체 유래 대장암세포 성장억제 활성)

  • Purpose: This study examined the antioxidant and cancer cell growth inhibitory activities of an ethanol extract and different solvent fractions of Mesembryanthemum crystallinum L. (ice plant). Methods: The ice plant was freeze-dried, extracted with 99.9% ethanol, and then fractionated with hexane, ethyl acetate, butanol, and water. The total polyphenol content (TPC), total carotenoid content (TCC), 2,2-diphenyl-1-picrylhydrazyl radical-scavenging activity (RSA), and ferric reducing antioxidant power (FRAP) were measured. Assays using 2',7'-dichlorofluorescin-diacetate and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide were performed to measure the intracellular reactive oxygen species (ROS) and cell growth, respectively. Annexin V/propidium iodide staining and cell cycle analysis were performed for the detection of apoptosis and cell cycle arrest. Results: TPC, TCC, RSA, and FRAP of the ethanol extract (EE) were 3.7 mg gallic acid equivalent/g, $13.2{\mu}g/g$, 21.0% (at a concentration of 5 mg/mL), and 21.0% (at a concentration of 5 mg/mL), respectively. Among the different solvent fractions, the butanol fraction (BF) showed the highest TPC (5.4 mg gallic acid equivalent/g), TCC ($86.6{\mu}g/g$), RSA (34.9% at 5 mg/mL), and FRAP (80.8% at 5 mg/mL). Treatment of HCT116 human colon cancer cells with EE and BF at concentrations of 250 and $500{\mu}g/mL$ reduced the levels of intracellular ROS. Concomitantly, EE and BF resulted in the dose-dependent inhibition of cell growth (at the concentrations of 125, 250, and $500{\mu}g/mL$ for 24 ~ 48 h) and the induction of apoptosis (at the concentrations of 250 and $500{\mu}g/mL$ for 48 h) in HCT116 cells. An increased G2/M cell population was also found in the BF-treated cells. Conclusion: These results suggest that ice plant possesses antioxidant and growth inhibitory activities in colon cancer cells.

Antimicrobial Activity of Aqueous Ethanol Extracts of Perilla frutescens var. acuta Leaf (차조기의 에탄올과 물 혼합 추출물의 항균활성)

  • This study was conducted to examine the antimicrobial activity of Perilla frutescens var. acuta leaf fractions extracted with a mixture of ethanol and water. The Ca and Mg contents of the leaf were 595.75 mg% and 467.0 mg%, respectively, and they were the highest among all of the test minerals. The extract yield increased w e content of water in e extraction solvent. Antimicrobial activity against Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa was found in the 50,70 and 95% ethanol extracts. Of the various fractions extracted from the mixture of ethanol and water, the ethyl acetate fraction showed antibacterial activity against all microorganisms tested in this experiment, and the antibacterial activity of ethyl acetate fraction from the water extract was the strongest. The phenol and flavonoid content in the ethyl acetate fraction showed no correlation with the concentration of ethanol in the extract solvent; however, their contents were higher in the 30% ethanol and water extraction which the antimicrobial activity of the extract was the strongest.

Comparison of Ingredients and Antioxidant Activity of Roasted Aerial Parts of Elaeocarpus sylvestris Extracts and Fractions (로스팅 담팔수 지상부 추출물 및 분획물의 성분 분석 및 항산화 활성)

  • Elaeocarpus sylvestris var. ellipticus is an evergreen tree of the family Elaeocarpaceae, which is a plant that grows naturally only in subtropical regions due to its vulnerability to cold. E. sylvestris has been reported to have flavonoids, coumarins, and polyphenols, and it is reported that these components have antibacterial and antioxidant effects. In this study, a roasting technique was introduced to increase the physiological activity of E. sylvestris, and antioxidant, total phenol, and total flavonoid content were measured after confirming changes in the ingredients of roasted E. sylvestris extracts and fractions. We analyzed the extracts before and after roasting and found an increase in the content of four components (brevifolin, ellagic acid, quercetin, and kaempferol), with the roasted extracts showing better antioxidant activity. The optimal roasting condition was confirmed to have the best antioxidant effect when roasting at 200 ℃ for 30 min, and the total phenol and total flavonoid content were also confirmed to be the best. E. sylvestris extract produced under optimal roasting conditions has been confirmed to exhibit improved antioxidant effects, and it is believed that it can be used as a cosmetic and food material in the future.

Antioxidant, Antimicrobial, and Antitumor Activities of Partially Purified Substance(s) from Green Tea Seed

  • The aim of this study is to evaluate the antioxidant, antimicrobial, and antitumor activities of various concentrations of partially purified substance(s) from green tea seed (Camellia sinensis L.). The total polyphenol contents of each fraction (non-adsorption fraction: F-1, fraction eluted with 40% methanol: F-2, and fraction eluted with 100% methanol: F-3) purified by Diaion HP-20 column chromatography were, in the increasing order: F-1 (3.7 mg tannic acid equivalents, TAB/g) < F-3 (23.2 mg TAB/g) < seed extracts (26.2 mg TAB/g) < F-2 (42.7 mg TAB/g). The scavenging activities toward the 1,1-diphenyl-2-picyrylhydrazyl (DPPH) radical were, in decreasing order: F-2 (93.3%) > butylated hydroxytoluene (BHT; 89.8%) > ascorbic acid (89.3%) > leaf extracts (70.3%) > F-3 (15.9%) > seed extracts (15.8%) > F-1 (14.8%) at a 0.1% concentration. In studies on antimicrobial activities, the results indicate that the growth of yeast (Candida albicans KCCM 11282 and Cryptococcus neoformans KCCM 50544) was inhibited more so than that of other fungi (Alternaria alternate KCTC 6005 and Rhizoctonia solani). In addition, it appears that the antitumor activities of the F-1, F-2, and F-3 fractions at a concentration of $50\;{\mu}g/mL$ showed 6, 7, and 23% growth inhibition of the HEC-1B cell line, 14, 11, 82% inhibition of the HEP-2 cell line, and 8, 16, and 81% inhibition of the SK-OV-3 cell line, respectively. Overall these results indicate that the antioxidant activity is greatest in the F-2 fraction, and the antimicrobial and antitumor activities are greatest in the F-3 fraction.
