The author has observed the tissue reaction of the absolute alcohol infection of rat oral mucosa. 0.5ml absolute alcohol was injected subcutaneously on the mucobuccal fold of rat. And the rats were sacrifieced at intervals of one day, 3rd, 1 week, 2 week and 4 week after alcohol injection. The microscopic tissue sections were made and stained with hematoxylin and eosin. The results were are as follows; 1. Degeneration and shrinkage of fibroblasts and coagulative necrosis were observed one day to and three day after alcohol injection. 2. Although coagulative necrosis and tissue degeneration occurred, the inflammatory infiltration was not prominent especially there were scarcely any polymorphonuclear leukocytes in that field. 3. Granulation tissue with moderate small round cell infiltration were replaced the necrotic area at one week after injection and the fibroblast proliferate into the granulation tissue at two week group. 4. At four week after injection, the damaged area recovered by fibroblastic proliferation and collage formation, but there were
Bacterial lipopolysaccharide (LPS) plays a major role in stimulating the synthesis and release of the principal osteoclast-activating cytokines, namely, interleukin 1 and tumor necrosis factor-$\alpha$ from immune cells. Although rnonocytes/macrophages are the main producers of these cytokines, recent evidence has indicated that polymorphonuclear leukocytes (PMN) have the ability to release IL-1 and TNF-$\alpha$. Calcium hydroxide has been shown to be an effective medicament in root canal infections, reducing the microbial titre within the canal. It has been proposed that the therapeutic effect of Ca(OH)$_2$ may also be the result of direct inactivation of LPS. The purpose of this study was to investigate whether treatment of Porphyromonas endodontalis LPS with calcium hydroxide alters its biological action as measured by human PMN secretion of IL-1 and TNF-$\alpha$, and it was compared with Escherichia coli LPS. P. endodontalis ATCC 35406 was cultured in anaerobic condition, and LPS was extracted using the hot-phenol water extraction method and purified. Purchased E. coli LPS was also purified. 100 $\mu\textrm{g}$/ml of each LPS in pyrogen free water were incubated with 25mg/ml Ca(OH)$_2$ at 37$^{\circ}C$ for 7 days. The supernatants were subjected to ultrafiltration, and the isolates were lyophilized and weighed. PMNs were obtained from peripheral blood by centrifugation layered over Lymphoprep. The cells were resuspended (4$\times$10$^6$ cells/ml) in RPMI 1640 followed by treatment with various concentrations of LPS (0, 0.1, 1, 10$\mu\textrm{g}$/ml) for 24 hours at 37$^{\circ}C$ in 5% $CO_2$ incubator. The supernatants of cells were collected and the levels of IL-1$\alpha$, IL=1$\beta$ and TNF-$\alpha$ were measured by enzyme-linked immunosorbent assay. The results were as follows ; 1. The levels of IL-1$\alpha$, IL-1$\beta$, TNF-$\alpha$ from PMN treated with each LPS were significantly higher than those released from unstimulated PMN of the control group (p<0.05). 2. The levels of all three cytokines released from PMN stimulated with each calcium hydroxide treated LPS were significantly lower than those released from PMN stimulated with each untreated LPS (p<0.05), while they were not significantly different from those released from unstimulated PMN of the control group (p>0.05) 3. The levels of secretion for all three cytokines were affected in a dose-dependent manner in PMN stimulated with each LPS (p<0.05), but not in PMN stimulated with each calcium hydroxide treated LPS (p>0.05). 4. The levels of all three cytokines released from PMN stimulated with p. endodontalis LPS were significantly lower than those released from PMN stimulated with E coli LPS (p<0.05).

The objectives of this study were to compare the effectiveness of different indicators of mammary inflammation in buffalo and to evaluate the association of the indicators with buffalo milk yield, composition, and rennet coagulation properties. This study was carried out at four buffalo farms in central Italy using a total of 50 lactating buffalo. Milk from each buffalo was tested at the beginning, middle, and end of lactation. To evaluate the relationship between mastitis markers and milk components, three classes were defined for each of the following markers: total somatic cell count (TSCC), differential somatic cell count (DSCC), and bacteriological results The regression coefficient for the reference method and the alternative method of determining TSCC was 0.81, indicating that the method routinely used to analyze buffalo milk consistently underestimated actual TSCC. The milk samples positive for udder-specific bacteria also had higher TSCC values than the samples that were negative for bacteria ($872{\times}10^3$/ml vs. $191{\times}10^3$/ml). In samples that were positive for udder-specific bacteria, polymorphonuclear leukocytes (PMN) made up greater than 50% of the cells. Moreover, only 1% of the samples in the lowest TSCC class were positive for bacteria. The correlation between TSCC and PMN was stronger (0.70), and PMN values in buffalo milk increased significantly when the TSCC class changed from low (38%) to medium and high (56% and 64%). Milk yield was negatively related to TSCC. Significant changes in lactose (4.87%, 4.80% and 4.64%) and chloride content (0.650 mg/ml, 0.862 mg/ml and 0.882 mg/ml) were also observed with increasing TSCC values. Higher TSCC was associated with impaired rennet coagulation properties: the clotting time increased, while the curd firming time ($p{\leq}0.05$) and firmness decreased. We concluded that in buffalo as in dairy cows, TSCC is a valid indicator of udder inflammation; we also confirmed that a value of $ 200{\times}10^3 cells/ml should be used as the threshold value for early identification of an animal affected by subclinical mastitis. In addition to its association with significantly decreased milk yield, a TSCC value above this threshold value was associated with changes in milk composition and coagulating properties.
Immunomodulatory and therapeutic potential of Enrofloxacin was studied in bovine sub clinical mastitis (SCM). The therapeutic efficacy was adjudged by Somatic cell count and Total bacterial count of the milk, whereas, the immuno modulatory potential of the drug was assessed by measuring myeloperoxidase (MPO) and acid phosphates (ACP) enzyme level in the milk leukocytes. Forty-five cows were divided into three equal groups. Gr I consisting 15 cows served as healthy control, whereas, 30 cows (SCM), Gr II and Gr III, selected on the basis of California Mastitis Test (CMT) positive reaction. Gr II cows received 150 mg of Enrofloxacin, once a day for three days and Gr.III received sterile 5 ml PBS (pH 7.4) for 7days, both the treatment were given by intramammary route. The observation was made up to 30 days post-treatment (PT). The CMT of the healthy milk was negative (0), whereas, it ranged between 1 point score and 2 point score in SCM. The Somatic cell count (SCC) and Total bacterial count (TBC) decreased significantly (p<0.05) on day 3 PT in GrII cows in Enrofloxacin treated group, however, such changes were insignificant in PBS treated group. Traces of MPO and ACP enzyme were found in the healthy milk. The mean ACP level enhanced by 70% on day 3 PT in GrII and only 18.7% in Gr. III cows. The mean MPO level enhanced to 32% in Gr. II and 18 % in Gr. III cows on day 3 PT. Concomitant use of Enrofloxacin in SCM at sub optimal dose was found to reduce the bacterial load by increasing the bactericidal enzyme level in the milk polymorphonuclear cells (PMNs) in bovine SCM, which indicates its immunomodulatory potential in mastitis.
Ischemia followed by reperfusion in the presence of polymorphonuclear leukocytes (PMNs) results in a marked cardiac contractile dysfunction. Amrinone, a specific inhibitor of phosphodiesterase 3, has an antioxidant activity against PMNs. Therefore, we hypothesized that amrinone could attenuate PMNs-Induced cardiac dysfunction by suppression of reactive oxygen species (ROS) produced fby PMNs. In the present study, we examined the effects of amrinone on isolated ischemic (20 min) and reperfused (45 min) rat hearts perfused with PMNs. Amrinone at $25\;{\mu}M$, given to hearts during the first 5 min of reperfusion, significantly improved coronary flow, left ventricular developed pressure (P<0.001), and the maximal rate of development of left ventricular developed pressure (P<0.001), compared with ischemic/reperfused hearts perfused with PMNs in the absence of amrinone. In addition, amrinone significantly reduced myeloperoxidase activity by 50.8%, indicating decreased PMNs infiltration (p< 0.001). Superoxide radical and hydrogen peroxide production were also significantly reduced in fMLP- and PMA-stimulated PMNs pretreated with amrinone. Hydroxyl radical was scavenged by amrinone. fMLP-induced elevation of $[Ca^{2+}]_i$ was also inhibited by amrinone. These results provide evidence that amrinone can significantly attenuate PMN-induced cardiac contractile dysfunction in the ischemic/reperfused rat heart via attenuation of PMNs infiltration into the myocardium and suppression of ROS release by PMNs.
This investigation is to determine whether the surface complexation of iron influence acute pulmonary responses induced by silica. For this study, three varieties of cation complexed silica were used: $silica-H^+,\;-Zn^{2+},\;and\;-Fe^{3+},$ since the first two are not active in the transport of electrons and generate little free radicals relative to the dust with the surface iron. Rats (270 to 280 g) were intratracheally (IT) instilled with saline, $silica-H^+,\;-Zn^{2+},\;or\;-Fe^{3+}$(5 mg in 0.5 ml saline). After 4 h, cell number, type, and differentiation were analysed in the bronchoalveolar lavage cells, and the levels of lactate dehydrogenase (LDH) and total protein were determined in the lavage fluid. In addition, bronchoalveolar lavage cells were cultured, and nitric oxide production was measured using nitrate assay. Inducible nitric oxide synthase (iNOS) mRNA in the bronchoalveolar lavage cells was also determined by northern blot analysis. Differential counts of the lavage cells showed that red blood cells were increased by 9-, 8-, and 13-fold and total leukocytes (lymphocytes plus polymorphonuclear neutrophils) by 48-, 36-, and 33-fold, following IT $silica-H^+,\;-Zn^{2+},\;and\;-Fe^{3+},$ respectively compared with the saline group. Meanwhile, there were no significant differences in red blood cells and total leukocytes among any of the cation complexed silica groups. The levels of LDH and total protein in the lavage fluid were significantly increased by 3- to 4-fold. However, compared among these silica groups, $Fe^{3+}$? complexation did not significantly change the LDH activity and total protein. NO production in cultured bronchoalveolar lavage cells was elevated by 2-fold, following IT any of the silica treatments compared with the saline group. Furthermore, the steady-state levels of iNOS mRNA in the lavage cells were greatly increased. There were any differences in iNOS mRNA expression among the silica-treated groups as with NO production. These findings suggest that surface complexed iron may not influence the acute pulmonary responses resulted from 4h exposure to silica.
Objective : Reactive oxygen metabolites and polymorphonuclear leukocytes have been implicated in the pathophysiology of reperfusion injury. The mechanisms involved in superoxide-mediated leukocyte adherence remain unclear, however, nitric oxide[NO] may contribute to this response. The present study is undertaken to elucidate mechamisms controlling NO based mechanisms that regulated leukocyte-endothelial interactions in the cerebral vasculature after global cerebral ischemia and reperfusion. Methods : Pial venular leukocyte adherence of anesthetized newborn piglets was quantified by in situ fluorescence videomicroscopy through closed cranial windows during basal conditions and during 2hours of reperfusion after global ischemia induced by 9minutes of asphyxia. Nitric oxide synthase[NOS] was inhibited by local window superfusion of L-nitroarginine[NA]; superfusion of sodium nitroprusside[SNP] was used to donate NO. Results : The mean number of adherent leukocytes to cerebral venules in the 9minutes asphyxia and 2hours reperfusion group were $161{\pm}19$ compared with $13{\pm}4$ in the nonasphyxial group. Superfusion of L-NA through the cranial window for 2hours resulted in leukocyte adherence similar to that observed during the initial 2hours of reperfusion after asphyxia. Leukocyte adherence was not additionally increased in asphyxic animal treated with L-NA. SNP inhibited asphyxia induced leukocyte adherence back to control levels. Conclusions : Nitric oxide inhibits leukocyte adherence to cerebral venules during the initial hours of reperfusion after asphyxia, and that NO supplementation inhibit asphyxia induced leukocyte adherence back to control levels. These results indicate that NO is an important factor in ischemia-reperfusion induced leukocyte adherence.
FSH는 미성숙 설치류의 난포성장을 촉진하며, 강소형성 난포의 퇴화비율을 감소시킨다. 본 연구는 미성숙 생쥐에 난포성숙호르몬을 투여한 후 유발되는 난포의 조직학적인 변화를 규명하기 위해 시행되었다. 3주령의 ICR생쥐에 10 i.u.의 재조합 난포자극호르몬을 복강주사한 후 1일, 2일, 3일에 좌측 난소의 조직학적 변화를 관찰하였다. 강소형성전 난포의경우 FSH처리 후 시간에 따라 퇴화난포의 비율이 증가하였으나 강소형성 난포의 경우에는 유의한 변화를 보이지 않았다. 퇴화되는 양상은 난포 내 세포자연사하는 과립세포의 증가, 대식세포 및 다형다핵백혈구의 증가 등이 관찰되었다. 이상의 결과로 보아, 과량의 FSH처리 후에 유발되는 난포의 퇴화는 과립세포의 세포자연사뿐 아니라 급성 염증반응을 수반하는 것으로 생각된다.
4,4'-diaminodiphenyl sulfone (dapsone) is a sulfone drug that has antibacterial effects on a variety of bacteria, especially Mycobacterium leprae; thus, it has been used to treat leprosy. Previous studies demonstrated that dapsone inhibits integrin-mediated adherence of neutrophils and production of prostaglandin $E_2$ by polymorphonuclear leukocytes. Hence, dapsone may act in immune cells and regulate cell-mediated inflammation processes. However, its anti-inflammatory effects remain unclear. The present study demonstrated that dapsone modulates the production of inflammation-related cytokines in immune cells. We employed the spleen cells of mice, which are major immune cells, and lipopolysaccharide (LPS) as a causative agent of inflammation for experiments. Dapsone induced a proportional change in splenocyte subsets and the apoptosis of spleen cells. Interestingly, dapsone decreased the production of tumor necrosis factor-alpha and interleukin (IL)-10, but not IL-6, in LPS-treated spleen cells. In other assays, we measured the dapsone-induced production of nitric oxide (NO) and the expression of activation markers of spleen cells. Dapsone decreased NO production in LPS-treated spleen cells. Taken together, our results demonstrate that dapsone has anti-inflammatory effects in immune cells and provide new insight into the potential uses of this agent.
The state of pulp and dental hard tissue during the process of resorption and shedding of 104 non-carious primary teeth was assessed by histomorphologic study. The teeth were extracted from the Korean school children aged 5 to 15 and classified into preshedding, shedding, delayed shedding groups according to the age of the child at the time of extraction, The results were as follows: 1. The inflammatory cell infiltration in primary pulp tissues occurred in 61.5% of observed teeth and were increased in the order of preshedding, shedding and delayed shedding stages. 2. The odontoclasts were appeared in 43.3% of observed teeth and were increased in the order of preshedding, shedding and delayed shedding stages. 3. The polymorphonuclear leukocytes and odontoclasts couldn't be observed in the exfoliated primary pulp tissues before the process of resorption has involved the root surface about 1.8mm subjacent to the cemento-enamel junction. 4. The intact root surface was the longest and largest in the preshedding stage. 5. The resorption process of primary teeth roots occurred mainly in the stage of preshedding and shedding stages. 6. There was a tendency of repair of resorbing root surfaces in the delayed shedding stage.
