구아노신일인산의 결정화에 대한 수용성 고분자의 영향 (Influence of Water Soluble Polymers on Crystallization of 5-Guanosine Monophosphate)

  • 고분자를 저분자 유기물질의 결정화 과정에 사용하면 결정형성과정을 중간 단계, 즉 메조크리스탈 단계에서 멈출 수 있다. 메조크리스탈은 수백 나노미터-수 마이크로미터의 결정들이 스스로 배열되어 결정성을 갖는, 결정과 고분자의 복합초구조로 정의할 수 있다. 본 연구에서는 구아노신일인산의 메조크리스탈 형성에서 고분자의 영향과 그와 관련된 변수들에 대해 알아 보았다. OM과 SEM 분석을 통해 고분자유도 결정화에 의한 GMP 결정은 전형적인 GMP입자와는 다른 모폴로지를 보임을 확인하였고, XRD 분석을 통해 새로운 polymorph를 가짐을 확인하였다. 이러한 결과는 고분자에 의해 GMP의 결정구조가 달라졌음을 의미하며, 결정 안에 함유되는 물의 함량이 달라져 다른 수화물 구조를 보임을 TGA 분석을 통해 확인하였다.

Phylogenetic Analysis of Mitochondrial DNA Control Region in the Swimming Crab, Portunus trituberculatus

  • The control region of mitochondrial DNA (13516-14619) is located between srRNA and $tRNA^{lle}$ gene in swimming crab, Portunus trituberculatus. The present study was investigated the genetic polymorph isms of the control region in samples of P. trituberculatus collected at coastal waters of the Yellow Sea in Korea. A total of 300 substitution and indel polymorphic sites were identified. In addition to SNPs and indel variation, a hypervariable microsatellite motif was also identified at position from 14358 to 14391, which exhibited 10 alleles including 53 different suballeles. When the hypervariable microsatellite motif was removed from the alignment, 95 haplotypes were identified (93 unique haplotypes). The nucleotide and haplotype diversities were ranged from 0.024 to 0.028 and from 0.952 to 1.000, respectively. The statistically significant evidence for geographical structure was not detected from the analyses of neighbor-joining tree and minimum-spanning network, neither. This result suggest that population of P. trituberculatus are capable of extensive gene flow among populations. We believed that the polymorph isms of the control region will be used for informative markers to study phylogenetic relationships of P. trituberculatus.

Analyses of Single Nucleotide Polymorphisms and Haplotype Linkage of the Human ABCB1 (MDR1) Gene in Korean

  • Single nucleotide polymorph isms (SNPs) in the MDR1 gene that are responsible for drug efflux can cause toxicity. Therefore, this study determined the SNPs of the Korean MDR1 gene, and analyzed the haplotypes and a linkage disequilibrium (LD) of the SNPs determined. The frequency of 9 SNPs from the MDR1 gene was determined by PCR-RFLP analyses of 100 to 500 healthy individuals. The frequcies of the SNPs were C3435T (47.7%), G2677T (37.6%), G2677A (4.4%), T1236C (21.7%), T129C (8%), A2956G (2.5%), T307C (1.5%), A41aG (9.2%), C145G (0%), and G4030C (0%). Analyses of the haplotype structure and an estimation of the LD of the combined polymorph isms demonstrated that the frequency of the 1236T-2677G-3435T haplotype is much higher in Koreans (14.1%) than in Chinese and western black Africans and the C3435T SNP in Koreans appears to have LD with T129C in Koreans for the first time. These results provide insight into the genetic variation of MDR1 in Koreans, and demonstrated the possibility of a new LD in this gene.

  • We have analyzed the structures of $Alq_3$ in different polymorphs by solid-state NMR. On the basis of the results, OLEDs were fabricated from different polymorphs of $Alq_3$. The current efficiency of the device fabricated from the mixture of $\alpha$-, $\gamma$-, and $\delta-Alq_3$ powders was higher than that from $\alpha-Alq_3$.

Biomineralization of Calcium Carbonate Polymorphs by the Bacterial Strains Isolated from Calcareous Sites

  • Microbially induced calcium carbonate precipitation (MICCP) is a naturally occurring biological process that has various applications in remediation and restoration of a range of building materials. In the present investigation, five ureolytic bacterial isolates capable of inducing calcium carbonate precipitation were isolated from calcareous soils on the basis of production of urease, carbonic anhydrase, extrapolymeric substances, and biofilm. Bacterial isolates were identified as Bacillus megaterium, B. cereus, B. thuringiensis, B. subtilis, and Lysinibacillus fusiformis based on 16S rRNA analysis. The calcium carbonate polymorphs produced by various bacterial isolates were analyzed by scanning electron microscopy, confocal laser scanning microscopy, X ray diffraction, and Fourier transmission infra red spectroscopy. A strain-specific precipitation of calcium carbonate forms was observed from different bacterial isolates. Based on the type of polymorph precipitated, the technology of MICCP can be applied for remediation of various building materials.

다수회 투여용 주사용액의 오염 -증류수와 푸로카인을 중심으로- (Contamination of Multiple Dobe Solutions for Injection distilled water and procaine)

  • This study was performed from July to December 1974 to investigate the rates of contamination of distilled water for injection and local anesthetics. 222 vials were sampled from 7 different places of St. Mary's Hospital The samples were cultured for isolation of microorganisms. The results were as follows; 1. Of 222 vials, 11 were Contaminated by 6 types: Staphylococcus epidermis, Bacillus subtilis, Mima polymorph a, Alcaligenes faecalis, Herellea vaginicala and pepto- streptococcus. 2. 5(9.1 %) of 55 vials from surgical wards, 2 (4.2 %) of 48 vials from surgical OPD, and 4 (12,l %) of 33 vials from pediatric wards were contaminated 3. Between rates of contamination of distilled water for injection and local anesthetics and the number of punctures, there was no significant relationship. (CR=1.42 P〉0.1) 4. There was no significant relationship (CR=1.02 P〉0.1) between the rates of contamination of vials and the duration of the first and the last punctures. 5. The ratios of contamination between 75% alcohol and 2% phenol used for disinfection of rubber lids of vials did not show significant difference.

Precipitated Calcium Carbonate Synthesis by Simultaneous Injection to Produce Nano Whisker Aragonite

  • The synthesis of pure calcium carbonate nanocrystals was achieved using a simultaneous injection method to produce nano particles of uniform size. These were characterized using scanning electron microscopy and powder X-ray diffraction. The nano particles were needle-shaped aragonite polymorphs, approximately 100-200 nm in length. The aragonite polymorph of calcium carbonate was prepared using aqueous solutions of $CaCl_2$ and $Na_2CO_3$, which were injected simultaneously into double distilled water at $50^{\circ}C$ and then allowed to react for 1.5 h. The resulting whisker-type nano aragonite with high aspect ratio (30) is biocompatible and potentially suitable for applications in light weight plastics, as well as in the medical, pharmaceutical, cosmetic and paint industries.

Genetic Analysis of Kallikrein-Kinin System in the Korean Hypertensives

  • The kallikrein-kinin system affects regulation of blood pressure, and genes encoding for the components of this system have been considered as good candidates for hypertension. To evaluate the relationship between genetic polymorphisms of candidate genes involved in this system and hypertension, we performed case-control studies using genetic markers in Korean normotensives and hypertensives, respectively. By association study, there was a marginal association with hypertension in AA genotype distribution of A1789G polymorphism in the hKLK1 gene (P=0.0754). Thus, this genetic polymorphism may weakly contribute to the susceptibility to hypertension in Koreans. We also observed that significant linkage disequilibrium exists among three polymorphic sites in the hKLK1 gene studied, suggesting that the three genetic polymorph isms can be useful as genetic markers in clinical association studies. Further studies using larger sample sizes and more genetic markers will be needed to clarify genetic influence of kallikrein-kinin system for hypertension.

Crystal Forms of Ketorolac

  • Four crystal forms of ketorolac have been obtained by recrystallization in organic solvents under variable conditions. Different ketorolac polymorphs and pseudo polymorph were characterized by X-ray powder diffraction crystallography (XRD), Differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). In the dissolution studies in water at $37{\pm}0.5^{\circ}C$ four crystal forms showed different patterns. The solubility of Form I were the highest. The solubility decreased in rank order: Form I> Form II > Form III > Form IV. Form land Form III were shown to have a good physical stability at room temperature for 60 days. However, Form II is converted to Form III and Form IV is converted to Form I after 60 days storage. Therefore, these observations indicate that crystalline polymorphism for ketorolac is readily inter-convertible and the relationship may have to taken into consideration in the formulation of the drug.
