Purification and Characteristics of New Biopolymer Produced by Alkaline-Tolerant Bacillus sp. (알칼리 내성 Bacillus sp.가 생산하는 생물 고분자의 정제 및 특성)

  • Biopolymer from alkaline-tolerant Bacillus so. was purified, and its physico-chemical and structural properties were investigated. Crude biopolymer, precipitated by acetone from culture broth was fractionated into two fractions by gel chromatography on Sephadex G-200. Among two fractions, one fraction(PS I), which an acidic biopolymer precipitated by the CPC(cetylpyridinium chloride) treatment was studied further. PS I fraction had carboxyl groups and was positive at color reaction of sugar. PS I fraction also showed UV absorbance at 190-225nm. The purified acidic biopolymer was composed of 4% glucose, 8% glucosamine and 88% glutamic acid. Sugar components of the purified acidic biopolymer seemed to be linked to PGA(polyglutamic acid) which existed in the from of ${\gamma}$-peptide bond. By the results of Smith degradation of sugar components, glucose and glucosamine was bound by 1,3 glocosidic linkage. Therefore, this biopolymer was a glycopeptide, oligosaccaride ${\gamma}$-PGA. We concluded that the equivalent weight and the molecular weight of this biopolymer were estimated as about 171 and 5x105 dalton, respectively.

  • Methods for production of ${\gamma}$-Polyglutamic acid( ${\gamma}$-PGA ) by an alkalophilic Alcaligenes sP. were investigated for batch and continuous culture processes. Both quantity and productivity of ${\gamma}$-PGA by Alcaligenes sp. in batch culture were gradually increased with the increase of glucose concentration up to 50g/l , but the maximal production yield of 63% was obtained at 10g/l of glucose concentration. The highest specific growth rate was about $0.25hr^{-1}$ at 50un of glucose concentration, and substrate inhibition was observed at above 50g1f of glucose concentration. The highest ${\gamma}$-PGA formation about 11g/l in a batch system was obtained at 31'C, pH 10.0 and 87rpn Productivity of 2.80g/l/hr for continuous cultivation was 9 times higher than the productivity for batch cultivation.

  • A fusant FG-21 was selected on the basis of higher ${\gamma}-GTP$ activity following fusion process between SM-2 and SM-10 of Bacillus subtilis mutants. ${\gamma}-GTP$ activity of the mutant FG-21 was increased up to 612 U/mL when grown for 36 hr at $37^{\circ}C$ in culture media containing 1% glycerol 1% glycerol, 1% peptone, 0.1% citric acid, 5 mM $K_2HPO_4$, 1 mM $FeCl_3$, 1 mM $MgCl_2$, 1 mM $NH_4Cl$, pH 7.0. In fusnat FG-21, the ratio of protein to total sugar contents for biopolymer A was 38 to 59. for biopolymer B from parental strains it was 19 to 78. Fructose contents determined by HPLC were $573.7\;\mu\textrm{g}/mg\;and\;764.4\;\mu\textrm{g}/mg$ for biopolymer A and B, respectively. And glutamic acid content were $163.7\;\mu\textrm{g}/mg\;and\;94.6\;\mu\textrm{g}/mg$ for biopolymer A and B, respectively. In fusant FG-21, the ratio of fructose to glutamic acid contents for biopolymer A was 78 to 22. For biopolymer B from parental strains it was 89 to 11.

  • In order to study on the viscous substance produced during Chungkookjang fermentation, one strain which produce a lot of viscous substance was isolated from the rice straw and identified as Bacillus sp; This strain produced the most viscous substance in 48 hours at $42^{\circ}C$, in splite of little change of amino type nitrogen and ammonia type nitrogen when added M.S.G., while the yield of viscous substance and amino type nitrogen was increased but that of ammonia type nitrogen was decreased when added susrose. Molecular weight of viscous substance estimated between 15,000 and 65,000. Viscous substance was confirmed mostly composed of glutamate and fructose by G.C. and HPLC analysis. Composition of viscous substance would be controlled according to the changes of media.

Evaluation of the Probiotic Potential of Bacillus polyfermenticus CJ6 Isolated from Meju, a Korean Soybean Fermentation Starter

  • To evaluate the probiotic potential of Bacillus polyfermenticus CJ6 isolated from meju, a Korean traditional soybean fermentation starter, its functionality and safety were investigated. B. polyfermenticus CJ6 was sensitive to all antibiotics listed by the European Food Safety Authority. The strain was also non-hemolytic, carried no emetic toxin or enterotoxin genes, and produced no enterotoxins. The resistance of B. polyfermenticus CJ6 vegetative cells and spores to simulated gastrointestinal conditions was high (60-100% survival rate). B. polyfermenticus CJ6 produced high amounts (0.36 g as a purified lyophilized form) of ${\gamma}$-polyglutamic acid (PGA). We speculate that the improved cell viability and the production of ${\gamma}$-PGA have a significant correlation. Adhesion of the strain to Caco-2 and HT-29 cells was weaker than that of the reference strain (Lb. rhamnosus GG), but it was comparable to or stronger than those of reported Bacillus spp. When B. polyfermenticus CJ6 spores were given orally to mice, the number of cells excreted in the feces was 4-fold higher than the original inocula. This suggests the inoculated spores propagated within the intestinal tract of the mice. This idea was confirmed by field emission scanning electron microscopy, which revealed directly that B. polyfermenticus CJ6 cells germinated and adhered within the gastrointestinal tract of mice. Taken together, these findings suggest that B. polyfermenticus CJ6 has probiotic potential for both human consumption and use in animal feeds.

Evaluation of Radical Scavenging Activity and Physical Properties of Textured Vegetable Protein Fermented by Solid Culture with Bacillus subtilis HA According to Fermentation Time (고초균을 이용한 조직대두단백의 고체 발효 기간에 따른 라디칼 소거 활성 및 물성 평가)

  • Textured vegetable protein (TVP) was fermented by the solid-state fermentation using Bacillus subtilis HA and biologically active compounds were produced by fermentation for 7 days. The longer fermentation time resulted in the color change of fermented TVP with strong dark red and yellow color. Melanoidin production rapidly increased until fermentation for 48 hr, but did change afterwards. The 70% ethanol extract of TVP fermented for 24 hr showed higher DPPH radical scavenging effect with $IC_{50}$ of 0.99 mg/mL but longer fermentation did not increase its activity. Also, 70% ethanol extract of TVP fermented for 72 hr indicated higher ABTS radical scavenging effect with $IC_{50}$ of 1.68 mg/mL. Consistency index in TVP fermented for 48 hr was the highest values with 7.89 $Pa{\cdot}s^n$. Viscoelastic properties of TVP fermented for 48 hr were maximally enhanced, and viscous value (G") is higher than the elastic value (G'). The $\gamma$-polyglutamic acid (PGA) content was increased by increasing fermentation time with 37.72% of $\gamma$-PGA at 168 hr. However, levan content and molecular weight of PGA were decreased with increasing fermentation time from 7.83% to 3.91% and 1649.3 kDa to 1286.8 kDa, respectively.

Physicochemical Properties of a Biopolymer Flocculant Produced from Bacillus subtilis PUL-A (Bacillus subtilis PUL-A로부터 생산된 Biopolymer 응집제의 물리화학적 특성)

  • Soybean milk cake (SMC) was used for the solid-state fermentation by Bacillus subtilis PUL-A isolated from soybean milk cake. In the presence of 5% glutamate the maximum production of biopolymer (59.9 g/kg) was performed by fermentation at $42^{\circ}C$ for 24 hr. The recovered biopolymer was consisted of 87% $\gamma$-polyglutamic acid with molecular weight of $1.3{\times}10^6$ dalton and other biopolymer. The biopolymer solution showed the great decrease in consistency below pH 6.0, regardless of the molecular weight of PGA. Biopolymer solution has a typical pseudoplastic flow behavior and yield stress. The consistency of biopolymer solution was greatly decreased by increasing heating time and temperature in acidic condition compared to the alkaline condition. In kaolin clay suspension, the flocculating activity of biopolymer was the highest value with 15 mg/L biopolymer and 4.5 mM $CaCl_2$, but decreased greatly with $FeCl_3$. The flocculating activity of biopolymer was maximum at pH5, but decreased drastically by heating at $60{\sim}100^{\circ}C$. In particular, biopolymer with native PGA showed the efficient flocculating activity compared to that of modified biopolymer containing low molecular weight of PGA.

'Neulchan', a Middle-seed, Disease-resistant, and High-yield Soybean Cultivar for Soy-paste and Tofu (중립 내병 다수성 장류·두부용 콩 '늘찬')

  • The soybean cultivar 'Neulchan' was developed for production of soy-paste and tofu. SS91501-9-1-1 and SS96205 (F2) were crossed in 1998, and F3 to F7 were selected by the pedigree method. A preliminary yield trial (PYT) and an advanced yield trial (AYT) were conducted from 2006 to 2008, and a regional yield trial (RYT) in nine regions was conducted from 2009 to 2011. In the RYT, 'Neulchan' was stable in variable environments and generated high yield. 'Neulchan' was determinate with white flower, light brown pod color, yellow spherical seed, and yellow hilum. Its flowering date and maturity date were Jul. 30 and Oct. 9, respectively. The plant height was shorter than that of 'Daewonkong' (a standard cultivar). 'Neulchan' had the same node number (14), higher first-pod height (12 cm), and lighter seed weight (21.7 g/100-seed weight) than those of 'Daewonkong' (14, 11, and 24.2 g/100-seed weight, respectively). 'Neulchan' had high resistance to bacterial pustule, and its resistance to soybean mosaic virus was similar to that of 'Daewonkong'. The yield and color of 'Neulchan' tofu were similar to those of 'Daewonkong' tofu, but the hardness was lower than that of 'Daewonkong' tofu. The soybean malt scent, fermented soybean yield, and γ-polyglutamic acid (γ-PGA) of 'Neulchan' were 3, 215%, and 24.6 mg/g, respectively. Its yield in adaptable regions was 307 kg/10a, higher than that of 'Daewonkong'. 'Neulchan' was expected to be cultivated and used widely for soy-paste and tofu production. (Registration No. 4904).
