고체분산체로부터 비페닐디메칠디카르복실레이트의 용출 특성 및 토끼의 십이지장 점막 투과 (Dissolution Characteristics of Biphenyl Dimethyl Dicarboxylate from Solid Dispersions and Permeation through Rabbit Deuodenal Mucosa)

  • To increase the dissolution rate of practically insoluble biphenyl dimethyl dicarboxylate (DDB), various solid dispersions were prepared with water soluble carriers, such as povidone (PVP K-30), poloxamer 407, sodium deoxycholate (SDC) and polyethylene glycol (PEG) 6000, at drug to carrier ratios of 1:3, 1:5 and 1:10 (w/w) by solvent or fusion method. Dissolution test was performed by the paddle method. The dissolution rate of DDB tablets (25 mg) on market was found to be very low (11.44, 9.02 and 6.42% at pH 1.2, 4.0 and 6.5 after 120 min, respectively). However, dissolution rates of DDB from various solid dispersions were very fast and reached supersaturation within 10 min. DDB-PEG 6000 solid dispersion appeared to be better in enhancing the in vitro dissolution rate than others. Furthermore, the incorporation of DDB and phosphatidylcholine (PC) into ${\beta}-cyclodextrin$ at ratios of 1:2:20, 1:5:20 and 1:10:20 resulted in a 4.9-, 11.2- and 19.6-fold increase in DDB dissolution after 120 min as compared with the pure drug, respectively. This might be attributed to the formation of lipid vesicles which entrapped a certain concentration of DDB during dissolution. On the other hand, the permeation of DDB through rabbit duodenal mucosa was examined using some enhancers such as SDC, sod. glycocholate (SGC) and glycyrrhizic acid ammonium salt (GAA). Only trace amounts of DDB were found to permeate through deuodenal mucosa in the absence of enhancer. SDC was found to markedly decrease the permeation flux of DDB, however, SGC and GAA (5 mM) enhanced the flux of DDB 1.6 and 2.4 times higher as compared with no additive, respectively.

Cellulomonas속 원형질체 재생과 종간 융합조건 (Protoplast Regeneration and Interspecific Fusion of the Genus Cellulomonas)

  • Cellulose를 이용할 수 있는 Cellulomonas속 균주의 종간 세포의 융합방법을 확립하기 위해 원형질체 재생 및 융합조건에 대해 검토하였다. Cellulomonas속 균주의 원형질체는 osmotic stabilizer를 포함하는 재생배지에서 유동성 한천배지로 중층하여 재생시켰고 재생 확인은 주사전자현미경으로 하였다. 원형질체 융합은 항생물질 내성과 영양요구성 돌연변이주의 유전자 표지에 확인하였다. Lysozyme을 처리하여 형성된 C. flavigena 원형 질체의 세포벽 재생은 osmotic stabilizer로 0.4M sorbitol을 포함하는 재생배지에서 약 15% 수준이었으며 여기에 Polyvinyl Pyrrolidone(PVP) 3% 첨가로 재생율을 약 3배정도 증가시킬 수 있었다. Cellulomonas속 균주의 변이주간의 원형질체 융합은 융합유도제로 PEG 6000을 취하여 최적농도 40% (w/v), 치적 처리시간 15분, Ca농도 25mM에서 약 2.0$\times$$10^{-4}$ - 4.0$\times$$10^{-4}$의 융합빈도를 얻을 수 있었다.

  • 섬유소 분해효소를 생산하는 P. verruculosum으로부터 유도된 영 양요구성 돌연변이주의 원형질체 융합을 위한 조건을 검토하였다. 18-20시간 배양한 각 영양요구성 돌연변이주 균사체에 Novozym 234를 처리하여 원형질체를 추출할 수 있었으며, 원형질체 생성량은 각 영양요구성 돌연변이주의 균사체 40mg (dry weight) 당 2.4-3.0$\times$10/suo 7/수준이었고, 원형질체 재생 완전 배지상에서의 환원율은 26.6-42.4% 수준이었다. 원형질체 융합을 위한 Polyethylene glycol (PEG) 6000의 최적농도는 20%였으며, PEG 최적 처리시간은 10분, CaCl$_2$의 최적 첨가농도는 10mM, 최적 pH는 5.5였고, 원형질체 융합 최적조건 하에서의 융합율은 1.8$\times$$10^{-3}$ 수준으로 나타났다. 모균주와 융합체로부터 측정된 DNA 양의 차이로 보아 융합체의 염색체는 aneuploid 상태임을 알 수 있었으며, filter paper를 기질로 한 Cellulase 활성측정에서 융합체가 야생형 균주보다 2배이상, 모균주 보다는 1-3배 이상 향상되었다.

새로운 기법에 의한 치코리 (Cichorium intybus L.) 의 사료화에 관한 연구 (Devloping Chcory for Forage Crop by New Technology)

  • Seeds of chicory were collected and selected with good germination, emergence and productivity. To develop chicory as a new forage crop seeds were primed with polyethylene glycol (PEG 6000) and coated with some chemicals, and tested in laboratory and field. The results obtained are summarized as follows: 1. Among the seed lines collected, PUlgAH, PU21TH, PU37CH and PU30TK were germinated more quickly and took less time for germination than the others. 2. PUI8AH was the best in emergence and growth although PU21TH showed superior germinative ability among the lines collected. 3. Chicory seeds primed with 20~25% PEG solution for three days at $10^{\circ}C$, or the seeds treated with 25% PEG solution at $15^{\circ}C$ regardless the treatment period showed good germination performances. 4. Osmotic priming with PEG accelerated the germination of chicory, but not in the field. 5. Coated seeds geminated more slowly in Petridishes but showed superior emergence and growth to those of primed or untreated seeds. Treatment 3 showed the best performances in pot experiment. 6. Coated seeds showed remarkable increase in establishment and dry matter yield especially in sod sowing compared to the other sowing method. 7. Most nutrient contents except iron and manganese of chicory were much higher than those of mixed pasture .

  • The dissolution characteristics of DDB were markedly enhanced by preparing solid dispersions of drug with polyethylene glycol 6000. Solid dispersions of various weight fraction were formed by a melting method. And various tablets$(A{\sim}E)$ were prepared from these solid dispersions with excipients (lactose, com starch, Avicel and PVP) by wet granulation method. There were no significant differences in dissolution rates between physical mixture and DDB alone. But dissolution rates of solid dispersions were $1.4{\sim}2.0$ times greater than that of DDB alone and $1.2{\sim}1.8$ times greater than those of a commercial tablet.

리기테다 소나무 종자(種子)의 피복(被覆)과 전처리(前處理)에 의한 발아(發芽) 및 유묘(幼苗) 생장(生長) 촉진(促進) (Improving Germination and Seedling Growth of Rigitaeda Pine Tree by Coating and Prime Treatment)

  • 리기테다 소나무 종자(種子)에 삼투압처리 및 피복(被覆)처리를 하여 균일(均一)한 발아(發芽)와 유묘생육(幼苗生育)을 촉진(促進)하기 위하여 이들 처리(處理) 종자(種子)의 발아(發芽)와 유식물(幼植物) 생육(生育)을 비교(比較) 검토(檢討)하였다. 1. Polyethylene glycol(PEG) 처리농도가 높아질수록 총발아율(TGP)도 높아졌으며 $10^{\circ}C$보다는 $15^{\circ}C$$20^{\circ}C$에서 총발아율이 향상되었다. 2. PEG처리기간이 길수록 또한 처리농도가 높을수록 발아속도(GR)가 양호하였으며, 처리농도와 온도간에는 상호 상관관계가 인정되지 않았다. 3. 최대 발아속도(MGR)는 PEG처리기간이 길수록 높았으며, $20^{\circ}C$에서 더욱 뚜렷하게 나타났다. 4. $20^{\circ}C$에서 12일간 PEG 처리시 약 2일 정도의 발아기간(發芽期間) 단축효과가 있었다. 5. PEG 처리는 저수분상태(低水分狀態) 하에서도 발아가 양호하였으며, 피복처리(被覆處理)된 종자는 약해(藥害)로 인한 발아(發芽) 장애현상(障碍現象)이 나타났다. 6. 조파(條播)와 직파(直播)간에는 건물생산량(乾物生產量)에 큰 차이가 있었으며, PEG 처리가 가장 양호한 성적을 보여 주었다. 적당한 토양수분 상태(포장용수량의 약 75%)보다 건조한 토양조건(포장용수량의 약 50%)에서 피복처리효과가 더욱 뚜렷하였다.

수분 장애시 종자 전처리가 오차드그라스의 발아에 미치는 영향 (Accelerated Germination of Orchardgrass(Dactylis glomerata) Seed Under Water Stress by Seed Pre-treatments)

  • 수분장애조건에서 오차드그라스(cv. Potomac) 종자의 발아력을 향상시키기 위하여 Polyethylene glycol 6,000(PEG) 및 생장조절제(GA$_3$, Kinetin, NAA)를 종자에 전처리하여 0 bar와 -5 bar의 두 수분조건하에서 17일간 발아양상을 비교, 검토하였는데 그 결과는 다음과 같다. 1. 전처리를 하지 않은 종자는 수분 삼투압 -5bar 조건에서 대조구(0 bar)에 비해 총 발아율은 감소되었으며 평균발아 소요일수와 발아종료 일은 지연되었다. 2. 대조구에서 PEG 종자 전처리는 무처리에 비해 발아율에 영향을 미치지 못하였으나 평균발아소요일 수는 -10 bar와 -15 bar에서 각각 5일 및 7일 처리한 것이 효과가 있었다. 3. 수분삼투압 -5 bar 조건에서는 PEG 종자 전처리시 무처리에 비해 발아율이 향상되었고 평균발아 소요일수가 단축되었는데 특히, -10 bar에서 7일 전처리 했을 경우 무처리에 비해 발아율이 1.9배로 향상되었고, 평균발아 소요일수는 모든 PEG 처리구에서 5일과 7일 전처리한 것이 단축되었다. 4. 대조구에서 생장조절제의 전처리는 GA$_3$와 NAA 공히 발아율에 영향을 미치지 못하였으나 Kinetin은 농도가 증가할수록 발아율을 감소시켰다. 평균발아 소요일수는 NAA와 Kinetin 전처리시 효과가 없었으나, GA$_3$ 전처리시에는 모든 처리구에서 단축되었다. 5. 수분삼투압 -5 bar 조건에서는 각각의 생장조절제가 공히 발아율과 평균발아 소요일수에 있어서 무처리에 비해 유의차가 인정되었는데, Kinetin과 NAA는 농도가 증가할수록 발아율을 감소시켰고 반면, GA$_3$는 농도에 관계없이 무처리에 비해 발아율이 향상되었다. 평균발아 소요일수에 있어서도 Kinetin 과 NAA는 무처리보다 지연되는 경향이었으나 GA$_3$는 100 ppm 전처리시 무처리 대비 2.0일이 단축되었다. 6. PEG나 GA3의 종자 전처리는 수분장애 조건에서 공히 무처리에 비해 발아율이 향상되었고 평균발아 소요일수도 단축되어, 수분장애시 오차드그라스의 균일한 발아 유도와 발아속도를 개선하는데 효과가 있을 것으로 생각되나, 어떠한 처리에 있어서도 대조구의 무처리구 발아율(79.0%)을 능가하지 못하여 오차드그라스의 발아에 관한 수분장애 요소를 완전히 극복할 수는 없었다.

Physiological Evaluation of Transgenic Rice Developed for Drought Tolerance

  • Evaluation of physiological performance of trehalose-producing transgenic rice line was conducted to investigate drought tolerance at early growth stage. Under artificially induced drought condition of 8% polyethylene glycol 6000, this transgenic rice line had leaf photosynthetic rate of 11.08 uml CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 8.38 mmol $H_2O$ $m^{-2}s^{-1}$ and leaf water potential of -1.12 MPa after 96 hours of treatment. Nakdongbyeo, the parent of this tyansgenic rice line, had photosynthetic rate of 15.42 $\mu$mol CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 8,04 mmol $H_2O$ $m^{-2}s^{-1}$ and leaf water potential of -0.88 MPa. The other variety used in this experiment for comparison, IR 72, showed higher values than both tyansgenic rice line and variety Nakdonbyeo on all three parameters; leaf photosynthetic rate of 20.61 $\mu$mol CO$_2$ $m^{-2}s^{-1}$, leaf transpiration rate of 12.88 mmol $H_2O$ $m^{-2}s^{-1}$, and leaf water potential of -0.82 MPa. So this transgenic rice line did not show superior performance in leaf transpiration rate, leaf photosynthetic rate and leaf water potential compared to variety Nakdongbyeo. This result along with visual observation on leaf rolling and drying during the experimental period indicated poor physiological performance of this transgenic rice line. Further studies on metabolic status of stress-induced trehalose, along with study on physiological response of this transgenic rice line during drought stress would shed more light on overall physiological performance of this transgenic rice line.

Chemical Composition, Degradation Characteristics and Effect of Tannin on Digestibility of Some Browse Species from Kenya Harvested during the Wet Season

  • A study was conducted with the objective of evaluating the nutritive value of some browse species from Kenya. The species evaluated included: Bauhinia alba, Bauhinia variegata, Bridelia micrantha, Calliandra calothyrsus, Carisa edulis, Cratylia argentea, Gliricidia sepium, Lantana camara, Maerua angolensis, Sesbania micrantha and S. sesban. The browses were evaluated by their chemical composition including phenolics, in vitro gas production and tannin activity (tannin bioassay). All the species had high crude protein content (149-268 g/kg DM) and low NDF content (239-549 g/kg DM). The feeds had varying contents of total extractable tannins (TET) ranging from low (3-22 mg/g DM), moderate (42-58 mg/g DM) and high (77-152 mg/g DM). Calliandra calothyrsus had the highest tannin content. Significant (p<0.05) variation in gas production was recorded among the species. Sesbania micrantha had the highest (p<0.05) potential gas production while Gliricidia sepium had the highest (p<0.05) rate of gas production. Use of polyethylene glycol (PEG 6000), to assess the adverse affect of tannins, indicated that tannins in browse species with high tannin content had inhibitory effects on rumen microbial fermentation as indicated by the gas production. Estimated organic matter digestibility and metabolizable energy also increased with PEG addition. The results of this study indicate that such Kenyan browse species have the potential to be used as feed supplements for ruminant animals.

Phenolic Composition, Fermentation Profile, Protozoa Population and Methane Production from Sheanut (Butryospermum Parkii) Byproducts In vitro

  • Sheanut cake (SNC), expeller (SNE) and solvent extractions (SNSE) samples were evaluated to determine their suitability in animal feeding. The CP content was highest in SNSE (16.2%) followed by SNE (14.7%) and SNC (11.6%). However, metabolizable energy (ME, MJ/kg) was maximum in SNC (8.2) followed by SNE (7.9) and SNSE (7.0). The tannin phenol content was about 7.0 per cent and mostly in the form of hydrolyzable tannin (HT), whereas condensed tannin (CT) was less than one per cent. The in vitro gas production profiles indicated similar y max (maximum potential of gas production) among the 3 by-products. However, the rate of degradation (k) was maximum in SNC followed by SNE and SNSE. The $t^{1/2}$ (time taken for reaching half asymptote) was lowest in SNC (14.4 h) followed by SNE (18.7 h) and SNSE (21.9 h). The increment in the in vitro gas volume (ml/200 mg DM) with PEG (polyethylene glycol)-6000 (as a tannin binder) addition was 12.0 in SNC, 9.6 in SNE and 11.0 in SNSE, respectively. The highest ratio of $CH_4$ (ml) reduction per ml of the total gas, an indicator of the potential of tannin, was recorded in SNE (0.482) followed by SNC (0.301) and SNSE (0.261). There was significant (p<0.05) reduction in entodinia population and total protozoa population. Differential protozoa counts revealed that Entodinia populations increased to a greater extent than Holotricha when PEG was added. This is the first report on the antimethanogenic property of sheanut byproducts. It could be concluded that all the three forms of SN byproducts are medium source of protein and energy for ruminants. There is a great potential for SN by-products to be incorporated in ruminant feeding not only as a source of energy and protein, but also to protect the protein from rumen degradation and suppress enteric methanogenesis.
