Confusing a Pollen Grain with a Parasite Egg: an Appraisal of "Paleoparasitological Evidence of Pinworm (Enterobius Vermicularis) Infection in a Female Adolescent Residing in Ancient Tehran"

  • There is often the risk of confusing pollen grains with helminth eggs from archaeological sites. Thousands to millions of pollen grains can be recovered from archaeological burial sediments that represent past ritual, medication and environment. Some pollen grain types can be similar to parasite eggs. Such a confusion is represented by the diagnosis of enterobiasis in ancient Iran. The authors of this study confused a joint-pine (Ephedra spp.) pollen grain with a pinworm egg. This paper describes the specific Ephedra pollen morphology that can be confused with pinworm eggs.

Effect of Low Temperature Pretreatment on Pollen Dimorphism and Embryo Formation in Anther Culture of Platycodon grandiflorum (도라지 (Platycodon grandiflorum) 약배양에서 저온처리가 화분 2형현상 및 배형성에 미치는 영향)

  • In order to investigate the effect of low temperature pretreatment on pollen dimorphism and embryo formation in anther culture of Platycodon grandiflorum, the anthers with microspore at the uninucleate stage were cultured on Murashige and Skoog medium supplemented with 0.5mg/L NAA and 1.0 mg/L BA. The low temperature pretreatment have clear effect on the frequencies of S pollen grains, symmetrical binucleate microspores (B type of S pollen), multinucleate and multicelled pollen grains. Especially, after low temperature pretreatment at 8$^{\circ}C$ for 5 days increased the frequency of S pollen grain (20.6%) in vivo. In addition, the highest frequency of callus induction (54.9%) and embryo formation (9.9%) were obtained from the anther pretreatment at 8$^{\circ}C$ or 5 days. Three distinct pathways could be recognized in the androgenesis, one involving mainly the vegetative cell, the second starting with the vegetative and the generative cell, respectively, and the third accompaning with two equal vegetative type cells in the pollen grains.

Selection of Cultivars and Organic Solvents to Improve Fruit Set of Greenhouse Watermelon during Cold Period (저온기 시설수박 착과율 증진을 위한 품종 및 화분분리 유기용매 선발)

  • Poor fruit set during winter period is one of the biggest problem in plastic tunnel watermelon cultivation. Hand pollination is inevitable to maximize fruit set of the winter watermelon. Productivity and viability of pollen grain and organic solvents for pollen storage were investigated. All cultivars produced more than 10 mg/flower except for 'Kumchun' cultivar. Pollen amount per flower were 13.8 mg in 'Bok' and 12.1 mg in 'Speedkul'. Germination rate of pollen grains incubated at $30^{\circ}C$ right after soaking in pentane solvent were 76% in 'Kumchun' as the lowest and 92% in 'Apollokul' as the highest. The pollen of 'Bok' showed the highest germination rate by 75% after a 15-day storage in pentane. All cultivars showed their pollen germination rate below 25% after a 24-day storage. Among the cuitivars, speed of pollen tube growth in vitro were relatively lower in 'Kumchun' and 'Sambokkul' by below $50\;{\mu}m/hr$. Pollen tube of these cultivars tended to burst during its elongation on the medium. Pollen stored 24 hrs in organic solvents showed 45, 39, 34, 23, and 19% of germination in pentane, ethyl ether, n-hexane, ethyl acetate, and acetone, respectively. Compared with light condition, pollen viability was higher in darkness during pollen storage in organic solvents. Pollen grain was susceptible to the organic solvent. The viability of pollen grains seems to be influenced greatly by duration of soaking pollen in organic solvent and the polarity of solvents. Organic solvent damages surface of pollen grain and extent of damage was varied by the solvents.

  • Morphological changes of flowers and insect visitors were observed to investigate pollinator of Caesalpinia decapetala. The flowers of C. decapetala are protandrous. Functionally, the flower is changed from male to female. As a male, pollen grain is released after anther dehisced while style is immature. After completed pollen grain release, the style starts to lengthen. It helps the stigma to easily touch the carpenter bee's thorax covered with pollen grain. At this time, flower functions as a female. The majority of taxa and individuals observed were Hymenoptera. The most frequent visitor was the Xylocopa appendiculata circumvolans, carpenter bee. Carpenter bees exhibited only typical pollinator behavior among flower visitors, with touching reproductive organs and seeking nectar at the same time. The pollination behavior is as follows. Soon after carpenter bees perceived guide mark, they foraged rightward and grasped style and stamens with legs and they inserted proboscis into standard petal to seek nectar. With this behavior, the pollen grains of the male flower transfer to the ventral thorax of the carpenter bee. As the carpenter bee moves to another female flower, the deposited pollen grains are delivered to the stigma.

Interspecific and Intraspecific Variation in Pollen Grains of Quercus Subgenus Lepidobalanus Endl. in Korea

  • Palynological characters such as pollen grain polar axis length (PL), pollen equatorial diameter (ED), colpus length, colpus width, and P/E ratio of six species of Quercus subgenus Lepidobalanus Endl. from Korea were studied. A significant interspecific variation, unequal distances between the species, and various degree of intraspecific variations were found. The taxonomic value of the pollen morphology parameters measured was found to vary according to species. These results suggest a possible relationship between parameters measured and ploidy level of the Quercus species studied.

  • Attempts were made in order to make clear the meiotic irregularities, male gametophyte formation and spermatogenesis for the purpose of applying these embryological facts to taxonomic works. Followings are the results obtained : 1. Meiosis is extremely irregular, giving rise to giant, micro, and empty, pollens. Meiotic division in each PMC is observed synchronizing. 2. In the microspore immediately before the vegetative-generative nuclear division, the nucleus locates appressed to the inner wall, most of the other part of the cell being occupied by large vacuoles. 3. Spindle symmetry in the microspore nuclear division is of intermediate type. 4. Nuclear divisions in each micropore are not synchronized as in meiosis. 5. Generative nucleus is located in close contact with the inner wall, while the vegetative nucleus occupies the central part of pollen grain. The pollen in this stage looks somewhat like broad bean. Afterward the generative nucleus loses contact with the wall, the nuclear shape changing from lenticular to roundsh. 6. The generative nucleus in fully matured pollen grain usually takes the shape of crescence, those in abortive pollen, lenticular or ellipsoidal, etc.

Pollen Morphology of the Woody Fabaceae in Korea (한국에서 생육하는 콩과 목본식물의 화분형태)

  • The pollen morphology of 31 taxa, representing all woody species from 12 genera of Fabaceae in Korea, was examined by light microscope and scanning electron microscope. The pollen grains were monad and prolate spheroidal or rarely subprolate in equatorial view. The pollen grain aperture was trizonocolporate or rarely trizonooperculate, which does not occur in the other families of Fabales in Korea. The exine showed a great variety of sculpture pattern, i.e. psilate, microperforate, densely microperforate, perforate, polygonal perforate, macroperforate, macroperforate with granules in holes, polygonal macroperforate, verrucate with pitted perforate striae, verrucate-perforate, granulate, rugulate, reticulate or refined reticulate. The pollen morphological data supported the idea that the genera Echinosophora and Sophora were congeneric. Caragana (Galegeae) was quite close to the Sophora group (Maackia, Sophora and Echinosophora) of the Sophoreae in each measured pollen parameter and exine sculpture pattern. Eleven genera of Fabaceae were recognized based on differences in aperture type, pollen grain shape and exine pattern, and a generic pollen key of the woody Fabaceae in Korea was developed. A key to pollen types of Lespedeza was also suggested. The examined taxa of Coronilla and Wisteria were recognized by their exine patterns.

  • Pollen and spores as well as their morphological characteristics are biological stability materials in which contains historical information about vegetation and climate changes. As noted researcher Neishtadt in 1971, the lake and swamp deposits were developed only in Holocene period. The geological history of Darkhad depression which is situated in the Northern end of Mongolia from ancient time become under scientific interest of many researchers. Our investigation was focused on pollen analysis of lake sediment of Darkhad depression which caused bottom of paleolake. In Mongolia the palynology science is beginning to develop since 1990 and in 2000 the laboratory of palynology was established at the department of Biology, State Pedagogical University of Mongolia. Researchers from this laboratory working on pollen morphology of present flora in order to classify plants taxonomy. Another part of our investigation is mellitopalynology, which is studying of pollen grains in a honey. Although, this research has been extending by pollen analysis of sediment and lake deposits in past few years on the basis of high sensitive microscope and modem technology.

Comparative study on the external micro-morphology of 3 kinds of minute pollen and spore Herbs (Pini Pollen, Typhae Pollen, Lygodii Spora) utilizing scanning electron microscope (주사전자현미경을 이용한 3종 화분포자류(花粉胞子類) 한약재(송화분(松花粉), 포황(蒲黃), 해금사(海金沙))의 미세형태 비교연구)

  • Objectives : We tried to establish standards for genuine by discriminating 3 kinds of pollen and spore herbs that are highly to be mixed with each other. Using an scanning electron microscope, we prepare a standard for discriminating external morphological characters of minute herbs, thereby contributing to the stable supply of genuine herbal medicines. Methods : Standard samples were confirmed by literature review on external morphological characteristics of original plants and herbal medicines, and collection and identification of original plants. The herbal medicines on the market were purchased and classified with using naked eye observation and magnifier. Finally, micromorphological identifications were conducted using an scanning electron microscope. Results : 1. Pini Pollen was clearly distinguished by its relatively medium size and a pollen grain with two swollen reticulate sacci at both ends. The verrucate ornamentation on the exine surface of the corpus and a sunken leptoma germ pore may be used as a discrimination criteria. 2. Typhae Pollen was distinguished by its relatively small size and a saccus on the end of a pollen grain. Reticulate ornamentation of exine surface of the corpus, and a slightly clear ulcerate germ pore can be used as a discrimination criteria. 3. Lygodii Spora was distinguished by its relatively large triangular-ovate shape and trichotomous fissure. Verrucate-tuberculate ornamentation of exine surface and trilete aperture could be used as a discrimination criteria. Conclusion : These results indicate that the use of electron microscopy is very effective for discriminating the external morphology of minute herbal medicines.

Characteristics and Temporal Distribution of Airborne Pollen in an Urban Area of Japan

  • Using a sampling device of our own making, airborne pollen has been monitored in Kyoto, Japan from the middle of February to the end of May 2004. From the morphological analysis of pollen grains by Scanning Electron Microscope (SEM), it was possible to identify some pollen types like Cryptomeria, Pine, Alder, Cyclobalanopsis, Chamaecyparis, and Equisetum. Daily average airborne pollen counts show strong variations from the day to day which makes the appropriate daily forecasts that could be of practical use for patients difficult. Diurnal variation of airborne pollen grains at our local sampling site is very irregular and shows no similarity between pollen types. The highest concentrations of Cryptomeria and Alder pollens in the south -west wind directions might be attributed to the airborne pollen transport, while the increase in Pine pollen grain in the southern wind direction was probably due to the local spread. Prevailing wind direction (SW) during the pollinating periods of Cryptomeria and Alder pollens could suggest a long-distance transport from a distant mountain.
