남극 남쉐틀랜드군도 주변 해역의 영양염과 식물플랑크톤 생물량 분포 (Distribution of Nutrients and Phytoplankton Biomass in the Area Around the South Shetland Islands, Antarctica)

  • 2000년 1월에 남극 남쉐틀랜드군도(South Shetland Islands) 주변해역에서 수심 200 m 까지 수온, 염분, 영양염, 엽록소, 일차생산력 등을 측정하였다. 표층수온은 남쉐틀랜드군도 북쪽 드레이크해협(Drake Passage)에서 높았고 남극반도 북동해역에서 낮았다. 반대로 염분은 드레이크해협에서 낮았고 남극반도쪽으로 갈수록 점차적으로 증가하여 남극반도 북동해역에서 최고 값을 보였다. 표층해수의 영양염 농도는 대체로 드레이크해협에서 낮은 값을 보였고 남쉐틀랜드군도 인접해역에서 높은 값을 보였다. 엽록소 농도는 드레이크해협과 남극반도 인접해역에서 낮았고 킹조오지섬(King George Island) 북쪽해역에서 높았다. 연구해역은 T-S diagram의 특징적인 형태에 따라 4개 해역으로 구분된다; 드레이크해협, 브랜스필드해협 (Bransfield Strait), 혼합 해역(Mixed zone), 웨델해(Weddell Sea) 해역. 이들 4개 해역들은 각각 특징적인 물리, 화학, 생물학적 특성을 보였다. 식물플랑크톤 생물량은 드레이크해협과 웨델해 해역에서 상대적으로 낮았고 브랜스필드해협과 혼합 해역에서 높았다. 웨델해 해역에서 식물플랑크톤 생물량이 낮은 것은 낮은 수온과 수심 200 m 이상의 깊은 표층혼합 때문이라고 생각되고 드레이크해협 해역에서는 높은 동물플랑크톤 포식압과 낮은 철농도 때문인 것으로 생각된다.

Distribution and Vertical Structures of Water Masses around the Antarctic Continental Margin

  • Spatial distribution and vertical structures of water masses around the Antarctic continental margin are described using synthesized hydrographic data. Antarctic Surface Water (AASW) over the shelf regime is distinguished from underlying other water masses by the cut-off salinity, varying from approximately 34.35 to 34.45 around Antarctica. Shelf water, characterized by salinity greater than the cut-off salinity and potential temperature less than $-17^{\circ}C$, is observed on the Ross Sea, off George V Land, off Wilkes Land, the Amery Basin, and the Weddell Sea, but in some shelves AASW occupies the entire shelf. Lower Circumpolar Deep Water is present everywhere around the Antarctic oceanic regime and in some places it mixes with Shelf Water, producing Antarctic Slope Front Water (ASFW). ASFW, characterized by potential temperature less than about $0^{\circ}C$ and greater than $-17^{\circ}C$, and salinity greater than the cut-off salinity, is found everywhere around Antarctica except in the Bellingshausen-Amundsen sector. The presence of different water masses over the Antarctic shelves and shelf edges produces mainly three types of water mass stratifications: no significant meridional property gradient in the Bellingshausen and Amundsen Seas, single property gradient where ASFW presents, and a V-shaped front where Shelf Water exists.

대류권계면 접힘에 의한 중규모 강설 발달에 대한 사례 연구 (A Case Study of Mesoscale Snowfall Development Associated with Tropopause Folding)

  • A case study of mesoscale snowfall with polar low signature during 25~26 December 2010 in South Korea is presented. The data used for analysis include surface and upper level weather charts, rain gauge, sea surface temperature, satellite imagery, sounding, and global $1^{\circ}{\times}1^{\circ}$ reanalysis data. The system initiated with a surface trough near the bay of Bohai but quickly intensified to become a polar low within 12 hours. The polar low moved southeastward bringing snowfall to southwestern Korea. There was strong instability layer beneath 800 hPa but baroclinicty was weak and disappeared as the low progressed onto land. Shortwave at 500 hPa and the surface trough became in-phase which hindered the development of the polar low while it approached Korea. However, there were strong tropopause folding (~500 hPa) and high potential vorticity (PV), which allowed the system to maintain its structure and dump 20.3 cm of snow in Jeonju. Synoptic, thermodynamic, dynamic, and moisture analyses reveal that polar low developed in an area of baroclinicity with strong conditional instability and warm air advection at the lower levels. Further, the development of a surface trough to polar low was aided by tropopause folding with PV advection in the upper level, shortwave trough at 500 hPa, and moisture advection with low-level jet (LLJ) of 15 m $s^{-1}$ or more at 850 hPa. Maximum snowfall was concentrated in this region with convection being sustained by latent heat release.

Stem-leaves of Panax as a rich and sustainable source of less-polar ginsenosides: comparison of ginsenosides from Panax ginseng, American ginseng and Panax notoginseng prepared by heating and acid treatment

  • Background: Ginsenosides, which have strong biological activities, can be divided into polar or less-polar ginsenosides. Methods: This study evaluated the phytochemical diversity of the saponins in Panax ginseng (PG) root, American ginseng (AG) root, and Panax notoginseng (NG) root; the stem-leaves from Panax ginseng (SPG) root, American ginseng (SAG) root, and Panax notoginseng (SNG) root as well as the saponins obtained following heating and acidification [transformed Panax ginseng (TPG), transformed American ginseng (TAG), transformed Panax notoginseng (TNG), transformed stem-leaves from Panax ginseng (TSPG), transformed stem-leaves from American ginseng (TSAG), and transformed stem-leaves from Panax notoginseng (TSNG)]. The diversity was determined through the simultaneous quantification of the 16 major ginsenosides. Results: The content of ginsenosides in NG was found to be higher than those in AG and PG, and the content in SPG was greater than those in SNG and SAG. After transformation, the contents of polar ginsenosides in the raw saponins decreased, and contents of less-polar compounds increased. TNG had the highest levels of ginsenosides, which is consistent with the transformation of ginseng root. The contents of saponins in the stem-leaves were higher than those in the roots. The transformation rate of SNG was higher than those of the other samples, and the loss ratios of total ginsenosides from NG (6%) and SNG (4%) were the lowest among the tested materials. In addition to the conversion temperature, time, and pH, the crude protein content also affects the conversion to rare saponins. The proteins in Panax notoginseng allowed the highest conversion rate. Conclusion: Thus, the industrial preparation of less-polar ginsenosides from SNG is more efficient and cheaper.

Heterologous Gene Expression System Using the Cold-Inducible CnAFP Promoter in Chlamydomonas reinhardtii

  • To increase the availability of microalgae as producers of valuable compounds, it is necessary to develop novel systems for gene expression regulation. Among the diverse expression systems available in microalgae, none are designed to induce expression by low temperature. In this study, we explored a cold-inducible system using the antifreeze protein (AFP) promoter from a polar diatom, Chaetoceros neogracile. A vector containing the CnAFP promoter (pCnAFP) was generated to regulate nuclear gene expression, and reporter genes (Gaussia luciferase (GLuc) and mVenus fluorescent protein (mVenus)) were successfully expressed in the model microalga, Chlamydomonas reinhardtii. In particular, under the control of pCnAFP, the expression of these genes was increased at low temperature, unlike pAR1, a promoter that is widely used for gene expression in C. reinhardtii. Promoter truncation assays showed that cold inducibility was still present even when pCnAFP was shortened to 600 bp, indicating the presence of a low-temperature response element between -600 and -477 bp. Our results show the availability of new heterologous gene expression systems with cold-inducible promoters and the possibility to find novel low-temperature response factors in microalgae. Through further improvement, this cold-inducible promoter could be used to develop more efficient expression tools.

활성탄관에 포집된 극성유기용제의 탈착효율에 관한 연구 (A Study on Desorption Efficiency for Polar Solvents Collected on Charcoal Tube)

  • This study was performed to evaluate factors affecting desorption of organic solvents collected on charcoal tube and to find out the optimum condition. Desorption efficiency for polar analytes was improved when several polar desorption solvents such as methanol, dimethylformamide(DMF), 2-(2-butoxyethoxy)ethanol were added to carbon disulfide($CS_2$). The best improvement was achieved when 10% dimethylformamide(DMF) in $CS_2$ was used as desorption solvent. During storage of polar analytes, recovery was greatly reduced. Especially, the recovery of cyclohexanone was decreased to 18.1 % after a month storage at $34^{\circ}C$. After two weeks storage, recovery of polar analytes was sharply decreased. Water adsorbed on charcoal interfered the recovery of polar analytes but didn't interfere that one of nonpolar solvent, toluene. When 10% DMF in $CS_2$ was used as desorption solvent, the effect of water on recovery was decreased, comparing with Desorption efficiency increased when analyte loading increased, and usage of 10% DMF in $CS_2$ decreased the loading effect. Increasing volume of desorption solvent was not effective to improve desorption efficiency of analytes when 10% DMF was used. Continuous shaking and sonication is not helpful to increase the desorption efficiency of analytes except cyclohexanone using 10% DMF. When silica gel used as adsorbent, methanol was better desorbent than dimethylsulfoxide. Analytes adsorbed on silica gel showed high recovery in low concentration and less affected by humidity. On the basis of this study, the following conclusions have been drawn. To improve the recovery of polar organic materials in air samples, it is necessary to analyze samples as soon as possible after they were collected. Otherwise, samples must be stored at low temperature. Using two components of desorption solvents, such as 10% DMF in $CS_2$, the effects of loading and humidity decreased for polar analytes such as methyl ethyl ketone and methyl isobutyl ketone. When work place has high humidity with low concentration of polar organic solvents, silica gel can be used as adsorbent, because it produces quantitative recovery for polar analytes at this condition. But it should be noted that high humidity makes breakthrough easy in silica gel samples.

Excitation Processes of the CH4 Aurorae of Jupiter and Saturn

  • Recently, an analysis of 3-micron spectra of CH4 line emission from our Gemini/GNIRS observations of Jupiter's polar regions yielded an unexpected result: The homopause (~1 microbar pressure level) located directly above the long-lasting 8-micron CH4 north-polar hot spot (Great 8-micron Hot Spot: GHS) is cool compared with the temperatures of nearby auroral regions (Kim et al. 2017). Most of the 8-micron emission of the GHS originates from CH4 at the ~1 mbar level (i.e., deeper in the stratosphere, where cooling time is several years), much longer than at the altitude of the homopause. We propose a mechanism to explain the temperature difference: locally-fixed and transient, but energetic auroral particles, which can penetrate to the 1 mbar level and deposit energy there creating and maintaining the GHS. For Saturn, thus far we have not detected distinctive 8-micron nor 3-micron CH4 hot spots in the polar regions. We will present a possible implication for this difference between Jupiter and Saturn.

Westerly Winds in the Southern Ocean During the Last Glacial Maximum Simulated in CCM3

  • We investigated the response of the westerly winds over the Southern Ocean (SO) to glacial boundary conditions for the Last Glacial Maximum using the CCM3 atmospheric general circulation model. In response to glacial boundary conditions, the zonally averaged maximum SO westerly winds weakened 20-35% and were displaced toward the equator by 3-4 degrees. This weakening of the SO westerly winds arose from a substantial increase in mean sea level pressure (MSLP) in the southern part of the SO around Antarctica relative to the northern part. The increase in MSLP around Antarctica is associated with a marked temperature reduction caused by an increase in sea ice cover and ice albedo feedback during the glacial time. The weakened westerly winds in the SO and their equator-ward displacement might play a role in reducing the atmospheric $CO_2$ concentration by reducing upwelling of the carbon rich deep water during the glacial time.

Dynamic Behavior of Photoinduced Birefringence of Copolymers Containing Aminonitro Azobenzene Chromophore in the Side Chain

  • Photoresponsive side chain polymers containing aminonitro azobenzene were synthesized for studying optically induced birefringence. Four different copolymers were prepared using methacrylate, a-methylstyrene, and itaconate monomer. Two copolymers are totally amorphous and the other two are liquid crystalline in nature. Trans-to-cis photoisomerization was observed under the exposure of UV light with UV-VIS absorption spectroscopy. Reorientation of polar azobenzene molecules induced optical anisotropy under a linearly polarized light at 532 nm. The dynamic parameters of optically induced birefringence let us compare the effect of polymeric structure on the rate of growth and decay of the birefringence. Besides the effect of glass transition temperature on the dynamics of photoinduced birefringence, we focused our interests on the geometrical hindrance of polar azobenzene molecules and cooperative motion of environmental mesogenic molecules in the vicinity of polar azobenzene moiety.
