  • The green colonial algae Botryococcus braunii is characterized by unusual high hydrocarbon contents, ranging from 15 to 75% of dry weight, as long-chain unsaturated hydrocarbons. In two-phase bubble column using various organic solvents, poor recovery 08 - 32%) of hydrocarbon seems to be caused by insufficient mixing between two phases, which was operated using only aeration on the narrow interface between hydrophobic solvent and cell suspension. In addition, hydrocarbon was entrapped tightly in cell-matrix (formed by exopolysaccharide) of algal colony, which make difficult to extract using two-phase system. To improve recovery efficiency, mixed-solvent of extractive solvent (octane) and biocompatible solvent (octane) was tested in two-phase column for in situ extraction. In two-phase extraction culture using mixed-solvent, the algal growth was intensely inhibited even at low concentration of polar octanol solvent. the hydrocarbon recovery in two-stage cell-recycle extraction showed a 2.9 fold increase (57%) over that in two-phase extraction. Up to 60 % of hydrocarbon could be recovered without serious cell-damage in the case of downstream separation for 6 h at the high recycle flow rate using this process after batch culture.

Zooplankton Community and Distribution in Relation to Water Quality in the Saemangeum Area, Korea: Change in Zooplankton Community by the Construction of Sea Dyke (새만금 수질 환경과 동물플랑크톤 군집 분포: 방조제 건설에 따른 군집 변화)

  • In order to examine the transition of zooplankton community by the Saemangeum sea dyke, the temporal and spatial distribution of zooplankton community with relation to environmental variables was investigated using data collected in 2004 and 2005. Sixty-one zooplankton taxa were identified. Average abundance (except Noctiluca scintiliam) ranged from 236 to 1810 indiv. $m^{-3}$, and was the highest in May 2005 and the lowest in february 2005. Dominant species were Acartia hongi and Paracalanus indicus, and cirripedia nauplii and zoea were dominant groups. After the closure of the 4th sea dyke, brackish species such as Tortanus derjugini and Pseudodiaptomus inopinus are widely distributed while the abundance of N. scintillans decreased in the northern area inside the dyke. In canonical correspondence analysis (CCA) for examining the relationships among zooplankton, stations and environmental variables, the northern area inside the dyke was distinguished from the other areas and was represented by Acartia spp, and brackish copepods.; Also, this area was characterized by high chlorophyll a concentration and COD, and low diversity.

  • Seasonal and tidal variations of density stratification in the Saemangeum waters are investigated based on synoptic CTD observations between July 2003 and September 2005. CTD data used in this study are those obtained after closing the dike No. 4 and before closing the two final gaps, the Sinsi and the Garyeok, on the Saemangeum tidal harrier. A total of 19 field campaigns comprehend a wide temporal spectrum, that is, few seasons, spring and neap tides, and high and low waters. In addition, ADCPs were anchored and CTDs were cast at three stations for 25 h in July 2005. Water columns are vertically homogeneous in autumn and winter. The vertical homogeneity persists in spring but with an occasional weak stratification in i:he northern part of the Gogunsan Islands. Increased reshwater runoff tends to stabilize the water columns and strong density stratification is established in summer. The mean potential energy anomaly (PEA) in summer used as a stratification parameter is the largest $(27.7\;J\;m^{-3})$ in the northern part of the Gogunsan Islands where the Geum River discharge dominates, the smallest $(16.9\;J\;m^{-3})$ is in the inner area of the barrier, in between the two $(21.6\;J\;m^{-3})$ in the southern part of the Gogunsan Islands. Whereas the stratification is generally strengthened in summer, strong winds or large tidal currents over the shallow depths frequently destratify the water column near the mouth of river runoff inside the tidal barrier. Periodic stratification, the development of stratification on the ebb and its breakdown on the flood, occurs in the mid-area inside the barrier induced by the tidal straining, which can also be found in the results of 25 h observation.

Ecological Status Evaluation using Seaweed Community Structures of Taean Coastal Areas in Korea

  • To evaluate the relative ecological quality of Taean coastal areas in terms of various seaweed community indices, seasonal samplings were taken at the Hakampo, Padori, Chaeseokpo, Mongsanpo and Bangpo shores from March 2006 to January 2007. A total of 105 species were identified; species richness ranged from 37~72 species spatially and from 65~75 species seasonally over the study period. Coarsely-branched seaweeds were dominant in functional group and ESG I (ecological state group I) made up 61 species (58.10%) of the identified macroalgae. The average seaweed biomass at the five study sites was $56.63g\;dry\;wt./m^2$ (range, 36.66 at Hakampo $-73.89g/m^2$ at Mongsanpo). Seaweeds were generally abundant in mid and low intertidal zone. Corallina pilulifera, Ulva australis, Sargassum thunbergii, Neorhodomela aculeata, and Symphyocladia latiuscula were the dominant species across all five study sites. Species diversity was between 1.24~2.30, while species evenness was between 0.40 and 0.61. The dominance index ranged from 0.43 at Padori to 0.64 at Mongsanpo. Given the community indices and shore descriptions, the five study sites were divided into two groups based on ecological quality: moderate (Chaeseokpo and Mongsanpo) and good (Hakampo, Padori and Bangpo).

Seasonal Variation of Zooplankton Communities in the Southern Coastal Waters of Korea (남해 연안 동물플랑크톤 군집의 계절변동)

  • The seasonal variations in the zooplankton community of the southern coastal waters of Korea were investigated seasonally in May, August, November 2005, and February 2006. A total of 74 taxa were sampled, with an average abundance ranging from 2,426~23,793 indiv./$m^3$, among which Noctiluca scintillans predominated. Noctiluca scintillans, Acartia omorii, Acartia erythraea, Paracalanus parvus s. l., Centropages abdominalis, Tortanus forcipatus, and Pseudevadne tergestina were the most abundant species detected. Zooplankton diversity was high around the inner regions during the summer, but it was relatively low in the stations located in the outer regions in the autumn. Non-metric multidimensional scaling (nMDS) revealed significant differences in the structures of the zooplankton community among the three regions. Our results showed that the seasonal variations in zooplankton communities in the southern coastal waters of Korea were attributable to seasonal changes in temperature, salinity, chlorophyll a concentrations, and N. scintillans blooms; additionally, this particularly study area might have been specifically influenced by the appearance of the Tsushima Warm Current.

A Preliminary Study on UAV Photogrammetry for the Hyanho Coast Near the Military Reservation Zone, Eastern Coast of Korea (동해안 군사시설보호구역 주변 향호 연안역을 대상으로 무인항공사진측량에 관한 예비 연구)

  • To evaluate the accuracy of UAV photogrammetry for Hyangho coast, eastern coast of Korea, we conducted a field experiment wherein UAV photogrammetry test was repeated three times. Since the Haygho coast is located within a military reservation zone, it was necessary to obtain permission to gain access to the beach and to have sensitive aerial photographs showing military facilities inspected and cropped. The standard deviation of the UAV shooting position between the three tests was less than 1 m, but repeatability of footprint on the ground was low due to wind-driven variability of the UAV pose. Self-calibrating bundle adjustment(SCBA) of implementing non-metric camera calibration was failed in one test. In two tests, the vertical error was twice as large as the pixel size except for those areas that were subject to security inspection and cropping. Given the problems that can arise with regard to the repeatability of the shooting area as well as the possibility of failure with regard to SCBA, we strongly recommend that UAV photogrammetry in coastal areas needs to be repeated at least twice.

Changes in the Spawning Ground Environment of the Common Squid, Todarodes pacificus due to Climate Change (기후변화에 따른 살오징어(Todarodes pacificus) 산란장 환경 변화)

  • This study analyzed the influence of climate change on the spawning ground area of the common squid, Todarodes pacificus. To estimate long term changes in the area of the spawning ground of the common squid, water temperature at 50 m deep that can be inferred from sea surface temperature (SST) based on both NOAA/AVHRR (1981.07-2002.12) and MODIS/AQUA (2003.01-2009.12) ocean color data was analyzed. In addition, five climate indices, Arctic Oscillation Index (AO), Siberian High Index (SH), Aleutian Low Pressure Index (ALP), East Asia Winter Monsoon Index (EAWM) and Pacific Decadal Oscillation (PDO) which are the main indicators of climate changes in the northwestern Pacific were used to study the relationship between the magnitude of the estimated spawning ground and climate indices. The area of the estimated spawning ground was highly correlated with the total catch of common squid throughout four decades. The area of the estimated spawning ground was negatively correlated with SH and EAWM. Especially, PDO was negatively correlated with the area of the spawning ground in the northwestern Pacific (r = -0.39) and in the southern part of the East Sea (r = -0.38). There was a positive relationship between the AO and the area of the spawning ground in the northwestern Pacific (r = 0.46) as well as in the southern part of the East Sea (r = 0.32). Temporally, the area of the winter spawning ground in the southern part of the East Sea in the 1980s was smaller than those areas in the 1990s and 2000s, because the area was disconnected with the western coastal spawning ground of Japan in the 1980s, while the area had been made wider and more continuous from the Korea strait to the western coastal water of Honshu in the 1990s and 2000s.

Inhella inkyongensis gen. nov., sp. nov., a New Freshwater Bacterium in the Order Burkholderiales

  • A freshwater bacterium, designated $IMCC1713^T$, was isolated from a highly eutrophic artificial pond. Cells of the strain were Gram-negative, chemoheterotrophic, poly-$\beta$-hydroxybutyrate granule containing and obligately aerobic short rods that were motile with a single polar flagellum. The 16S rRNA gene sequence similarity analysis showed that the novel strain was most closely related to the species Roseateles depolymerans (96.3%), Mitsuaria chitosanitabida (96.2%), Ideonella dechloratans (96.2%), and Pelomonas saccharophila (96.1%) in the Sphaerotilus-Leptothrix group within the order Burkholderiales. Phylogenetic trees based on 16S rRNA gene sequences indicated that the isolate formed an independent monophyletic clade within the order Burkholderiales. The relatively low DNA G+C content (57.4mol%), together with several phenotypic characteristics, differentiated the novel strain from other members of the Sphaerotilus-Leptothrix group. From the taxonomic data, therefore, the strain should be classified as a novel genus and species, for which the name Inhella inkyongensis gen. nov., sp. nov. is proposed. The type strain of the proposed species is strain $IMCC1713^T$ (=KCTC $12791^T$=NBRC $103252^T$=CCUG $54308^T$).

Conceptual Design of a Solid State Telescope for Small scale magNetospheric Ionospheric Plasma Experiments

  • The present paper describes the design of a Solid State Telescope (SST) on board the Korea Astronomy and Space Science Institute satellite-1 (KASISat-1) consisting of four [TBD] nanosatellites. The SST will measure these radiation belt electrons from a low-Earth polar orbit satellite to study mechanisms related to the spatial resolution of electron precipitation, such as electron microbursts, and those related to the measurement of energy dispersion with a high temporal resolution in the sub-auroral regions. We performed a simulation to determine the sensor design of the SST using GEometry ANd Tracking 4 (GEANT4) simulations and the Bethe formula. The simulation was performed in the range of 100 ~ 400 keV considering that the electron, which is to be detected in the space environment. The SST is based on a silicon barrier detector and consists of two telescopes mounted on a satellite to observe the electrons moving along the geomagnetic field (pitch angle $0^{\circ}$) and the quasi-trapped electrons (pitch angle $90^{\circ}$) during observations. We determined the telescope design of the SST in view of previous measurements and the geometrical factor in the cylindrical geometry of Sullivan (1971). With a high spectral resolution of 16 channels over the 100 keV ~ 400 keV energy range, together with the pitch angle information, the designed SST will answer questions regarding the occurrence of microbursts and the interaction with energetic particles. The KASISat-1 is expected to be launched in the latter half of 2020.

Desorption characteristics of Activated Carbon and Activated Carbon Fiber by Development of Sorbent Tube for Measurement of Organic Solvent (유기용제 측정용 흡착관 개발을 위한 AC 및 ACF의 흡착특성)

  • Charcoal $tube/CS_2$ method are more popularly used than any other in the measurement of the working environment for the exposure evaluation of organic solvent, but it is some weak points that the lower accuracy can be obtained on the polar materials and within the range of the low concentration. Thus solvent desorption method has been developed to make accuracy higher and to overcome some weak points. However, because of high price of adsorption tube for thermal desorption and the short of study on its application to the working environment, it is not popularly used in the domestic industrial hygiene fields. This dissertation aims to develop thermal desorption and adsorption tubes for measuring organic solvents in the working environment, by comparing and analyzing breakthrough condition and adsorption capacity with ACF. Specific surface area of ACF used in this study is wider than the one of AC and micropore of ACF related with adsorption has been developed, and adsorption velocity and adsorption amount are very excellent by linking a pore of surface and an inside well into micropore. 1. Result of analysis on physical characteristics of adsorbent, the specific surface area of ACF was 1.3 times higher than that of AC. Distribution ratio of micropore related to adsorption was 94% on ACF and AC. Result of SEM, micropore of the AC is opened to the surface. In contrast, ACF shows that extremely fast adsorption speed. Because of micropore are exposed on the surface and penetrate through each other. 2. Breakthrough characteristics of adsorbents was not different from slop of breakthrough curve. The effluent concentration reaches 10% of initial concentration($C_{out}/C_{in}=0.1$, break point) of ACF was 30~316min longer than that of AC. Therefore, the adsorption capacities of ACF was 1.1~4.6 times higher than that of AC. ACF can be used as a proper adsorbent for measurement of organic solvent.
