약초(藥草) 중에 존재(存在)하는 배추좀나방에 대한 천연살충성(天然殺蟲性) 물질(物質)의 동정(同定) (Identification of natural insecticidal compound in medicinal plants against diamondback moth)

  • 약초 중에 존재하는 농약 활성 물질을 탐색하기 위하여 84종의 약초로부터 극성 및 비극성 분획을 얻고 주요 농업 해충에 대하여 살충성을 검토하였다. 일차 및 이차 검정 결과 참삽주 괴경 (창출)의 비극성 분획은 배추좀나방에 대하여 매우 높은 살충 효과를 보였다. 창출 비극성 분획내의 살충성 활성물질은 향류분배 분리, TLC, GC-MS, $^{1}H$- 및 $^{13}C-NMR$ 분석으로 $C_{15}H_{22}O$의 4,11-selinadien-3-one (${\alpha}$-cyperone)으로 확인되었다. 그러나 배추좀나방에 대하여 높은 살충활성을 보였던 다른 비극성 분획으로부터는 ${\alpha}$-cyperone을 검출할 수 없었다. 더욱이 ${\alpha}$-cyperone이 향부자로부터 최초로 분리 동정되었음에도 불구하고 우리나라에서 재배된 향부자에서는 ${\alpha}$-cyperone을 확인할 수 없었다.

  • Heterocyclic compounds with oxygen atoms are known to have potent biological effect. The flavonoids. isoflavonoids. and coumarins which form the bulk of these compounds are very polar and have limited use as drugs which have to pass through membranes. The non-polar property is increased by exchange oxygen to selenium as a part of heterocyclic compound. Our group is focused on synthesizing selenoheterocyclic compound with the above property. 활성탄관에 포집된 극성 및 비극성 유기용제 분석시 탈착용매 종류에 따른 탈착효율 비교 (A comparison of desorption efficiency by types of solvent for polar and non-polar organic compounds collected on activated charcoal tube)

  • This study was conducted to evaluate desorption efficiencies by types of desorption solvent for polar and non-polar organic compounds collected on activated charcoal tubes. Analytes tested were toluene, m-xylene, isobutyl alcohol, n-butyl alcohol, cellosolve acetate, and butyl cellosolve. Three different concentration levels of spiked sample were made. Types of cosolvent mixed with the main solvent, $CS_2$, were methanol, pentanol, and dimethylformamide (DMF) and the cosolvent for methylenechloride was methanol. The amounts of cosolvent added to the main solvent were 1, 5, and 10% by volume (v/v%), respectively. The results were as follows: 1. For all mixed solvents except 1% methanol and 1% pentanol with $CS_2$, desorption efficiency significantly increased, compared with that of $CS_2$ alone. 2. Desorption efficiency increased by increasing analyte loading on charcoal tube regardless of mixed solvents used and the material polarity. 3. For all cosolvents mixed with $CS_2$ by 1% and 5% volume, desorption efficiency for non-polar compound was significantly higher than that of polar compound. For the 10% mixed solvents and the methylenechloride mixed with methanol, the results were opposite. 4. The lowest mean percent bias of 4.79% was obtained from the 5% DMF-$CS_2$ mixed solvent, followed by 4.82% from the 10% DMF-$CS_2$ solvent while the highest bias of 23.26% was from the solvent of $CS_2$ alone. Based on the results of this study, in order to increase desorption efficiency, it is recommended to add such cosolvents as methanol, pentanol, and DMF to $CS_2$, preferably 5% by volume for analyzing polar compounds collected in charcoal tubes.

상동성 모델링을 이용한 Tricyclic Isoxazole 유도체와 ${\alpha}_{2c}$-Adrenoceptor의 상호작용 (Interactions of Tricyclic Isoxazole Analogues with ${\alpha}_{2c}$-Adrenoceptor by Homology Modeling)

  • Adrenoceptor has been considered to be an important target in psychiatric disorders. Based on x-ray structures of bovine rhodopsin, we established homology model of ${\alpha}_{2c}$-adrenoceptor (ADA2C_rat) and then analyzed docking from binding model of receptor-ligand complex with high-active compound No.29 in tricyclic isoxazole analogues (1-30). We observed that the N (1.907 $\AA$) and O (1.712 $\AA$) atoms of isoxazole ring on the docked ligand (No.29) formed H-bonding interaction with O-H of Ser5.32 and carmeron phenyl ring centroid of tricyclic isoxazole formed $\pi-\pi$ interaction at 3.342 $\AA$ distance with phenyl ring centroid of Phe6.52. According to predictions of blood-brain distribution (logBB) through penetration of blood-brain barrie (BBB) and polar surface area (PSA) of the ligands, the high-active compound No.29 has values of logBB=-0.203, PSA=67.50, respectively. These results suggest that the high-active compound No.29 is a novel anti-depressant with the characteristics such as dopamine and serotonin.

  • Flavonoids with oxygen atoms are known to have potent biological effect.They have been studied long as major antioxidants which protect cell membranes. Recent medical surveys show that increased intake of selenium decreases the risk of breast, colon, lung and prostrate cancer by preventing free radical generation. The flavonoids, isoflavonoids, and coumarins which form the bulk of these compounds are very polar and have limited use as drugs which have to pass through BBB(Brain Blood Barrier)The non-polar property is increased by exchange oxygen to selenium as a part of heterocyclic compound. 마이크로파를 이용한 비극성 고무컴파운드의 가황 효율 (Vulcanization Efficiency of Non-polar Rubber Compounds by Microwave)

  • 열원으로 열풍과 마이크로파를 이용하여 비극성 ethylene-propylene-diene terpolymer(EPDM)/카본블랙 컴파운드 제조에 있어서 시편의 두께 및 첨가제 함량에 따른 가황속도의 영향을 비교 조사하였다. 열풍가황의 경우, 컴파운드의 두께가 가장 중요한 인자로서 EPDM의 나쁜 열전도도 특성으로 인하여 컴파운드의 두께가 증가할수록 가황속도는 느려졌으며 두께 3 mm 컴파운드의 완전 가황은 $250^{\circ}C$에서 7분 이상이 소요되었다. 반면에 마이크로파 가황에서는 컴파운드의 두께에 의한 영향은 없었으나 극성첨가제인 카본블랙 함량에 큰 영향을 받았다. 카본블랙이 80 phr 이상인 컴파운드는 30초간 마이크로파 노출에 의해 거의 100% 가황이 일어남을 확인하였다. 이러한 결과는 극성물질을 포함하는 컴파운드의 가황열원으로서 마이크로파가 효과적임을 알 수 있었다.

  • Cetyl alcohol과 polyoxyethylene(20) sorbitan monooleate를 혼합한 유화제와 양친매성 resin을 용매에 용해시킨 분산상에 분산매인 물을 첨가하여 O/W 에멀젼을 제조하여 HLB(Hydrophilic Lipophilic Balance) 값과 온도변화에 따른 상변화를 관찰하였다. 양친매성 resin을 함유한 에멀젼은 이를 함유하지 않는 에멀젼보다 온도변화에 따른 상안정성이 좋다는 것을 알 수 있었다. 또한 양친매성 resin이 에멀젼 안정성에 미치는 영향을 파악하기 위해 양친매성 resin에 함유되어 있는 작용기중 하나의 작용기를 갖는 resin을 model compound로 하여 HLB값과 온도변화에 따른 에멀전 상거동을 관찰한 결과, Poly(acrylic acid)

Growth and characterization of periodically polarity-inverted ZnO structures grown on Cr-compound buffer layers

  • Periodically polarity inverted (PPI) ZnO structures on (0001) Al2O3 substrates are demonstrated by plasmas assisted molecular beam epitaxy. The patterning and re-growth methods are used to realize the PPI ZnO by employing the polarity controlling method. For the in-situ polarity controlling of ZnO films, Cr-compound buffer layers are used.[1, 2] The region with the CrN intermediate layer and the region with the Cr2O3 and Al2O3 substrate were used to grow the Zn- and O-polar ZnO films, respectively. The growth behaviors with anisotropic properties of PPI ZnO heterostructures are investigated. The periodical polarity inversion is evaluated by contrast images of piezo-response microscopy. Structural and optical interface properties of PPI ZnO are investigated by the transmission electron microcopy (TEM) and micro photoluminescence ($\mu$-PL). The inversion domain boundaries (IDBs) between the Zn and the O-polar ZnO regions were clearly observed by TEM. Moreover, the investigation of spatially resolved local photoluminescence characteristics of PPI ZnO revealed stronger excitonic emission at the interfacial region with the IDBs compared to the Zn-polar or the O-polar ZnO region. The possible mechanisms will be discussed with the consideration of the atomic configuration, carrier life time, and geometrical effects. The successful realization of PPI structures with nanometer scale period indicates the possibility for the application to the photonic band-gap structures or waveguide fabrication. The details of application and results will be discussed.

감자 유래 극성화합물의 생리활성 분석 (Biological Activity Analysis of Potato-derived Polar Compounds)

  • Natural substances have various physiological activities. Substances isolated from natural substances are known to be safer and more potent than pharmaceuticals. Potatoes not only act as energy sources but also contain active ingredients such as vitamins and minerals. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by chemical base anti-oxidation assay and cell based anti-oxidation assay. In the chemical base anti-oxidation assay, DPPH experiment showed activity in the order of Hongyoung, Haryung, Seohong, Sumi, and Jayoung. In the LPA experiment, IC50 was lower in the order of Jayoung, Seohong, Sumi, Hongyoung, and Haryung. In the cell based anti-oxidation assay, the smallest amount of ROS was generated when the compound was derived from Haryung and hongyoung, and strong SOD activity was observed in Sumi and Jayoung. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers.

마늘 Oleoresin 제조시 휘발성성분에 관한 추출용매효과 (Effect of Extraction Solvent on Volatile Compounds of Garlic Oleoresin)

  • 마늘의 이용성 증진에 관한 기초자료와 제품의 품질 향상에 기여하고자 마늘 oleoresin 제조과정 중의 휘발성 향기성분의 변화를 알기 위해 휘발성 향기성분을 4가지 용매로 추출하여 농축한 후, 가스크로마토그래피와 질량 분석 검출기 (mass selective detector)를 이용하여 polar(Supelcowax-$10^{TM}$)와 nonpolar capillary column(HP-5)에서 각각 분리, 동정하였다. 용매에 따른 oreoresin 추출을 보면 methanol을 사용하여 추출한 경우가 가장 peak area가 높았고 그외 acetone, methy acetate, hexane을 사용한 경우는 비슷하였다 Polar capillary column에서 총 41종이 분류되었고, pyran 및 그 유도체 물질 13종, 황화합물이 11여종, furan 및 그 유도체들이 6종, alcohol류 2종 분류되었고 그밖에도 2종의 heterocyclic compound가 각각 확인되었다. Nonpolar capillary column에서는 32종의 향기성분이 동정되었고, pyran및 그 유도체물질이 11종, 황화합물 11여 종, acid류 5종, pyran 및 그 유도체들이 3종 그밖에 eugenol 등이 나타났다. Nonpolar 보다 polar에서 휘발성성분의 다량 검출되었으며, 대부분의 휘발성 향기성분이 극성이 강한 methanol에서 잘 추출되었지만 마늘의 주요휘발성 성분인 황화합물 즉, 3,3'-thiobis 1-propene, methyl 2-propenyl disulfide, dimethyl trisulfide, di-2-propenyl -trisulfide, 2-thiophenecarboxylic acid는 비극성용매인 methyl acetate와 hexane에서 잘 추출되었다.
