Inhibition of SV40 DNA replication in simian cell by bacteriophage M13 DNA sequences (원숭이 신장 세포에서 M13 DNA에 의한 SV40 DNA 복제 억제 현상에 대하여)

  • Bacteriophage M13 DNAs carrying the wild type or base substituted SV40 DNA replication origins were used for replication assay. In vivo and in vitro assay with African green monkey cell line COS-1 showed that the replication of M13-SV40 recombinant DNAs was restricted like a pBR322 SV40 recombinant DNA(Lusky and Botchan, 1981). Furthermore, recombinant phage DNAs isolated from the transfected siminan cells subsequently show a reduced ability to retransform E. coli. But pATSV-W(Kim et al., 1988) was replicated in COS-1 cells normally. We think that a poison sequence may exist on bacteriophage M13 DNA like pBR322.

A fast adaptive numerical solver for nonseparable elliptic partial differential equations

  • We describe a fast numerical method for non-separable elliptic equations in self-adjoin form on irregular adaptive domains. One of the most successful results in numerical PDE is developing rapid elliptic solvers for separable EPDEs, for example, Fourier transformation methods for Poisson problem on a square, however, it is known that there is no rapid elliptic solvers capable of solving a general nonseparable problems. It is the purpose of this paper to present an iterative solver for linear EPDEs in self-adjoint form. The scheme discussed in this paper solves a given non-separable equation using a sequence of solutions of Poisson equations, therefore, the most important key for such a method is having a good Poison solver. High performance is achieved by using a fast high-order adaptive Poisson solver which requires only about 500 floating point operations per gridpoint in order to obtain machine precision for both the computed solution and its partial derivatives. A few numerical examples have been presented.

Isolation, Identification and Optimal Culture Conditions of Streptomyces albidoflavus C247 Producing Antifungal Agents against Rhizoctonia solani AG2-2

  • Streptomyces albidoflavus C247 was isolated from the soil of the Gyeongsan golf course in Korea. Physiological, biochemical and 16S rDNA gene sequence analysis strongly suggested that the isolate belonged to Streptomyces albidoflavus. Preliminary screening revealed that the isolate was active against fungi and bacteria. Self-directing optimization was employed to determine the best combination of parameters such as carbon and nitrogen source, pH and temperature. Nutritional and culture conditions for the production of antibiotics by this organism under shake-flask conditions were also optimized. Maltose (5%) and soytone (5%) were found to be the best carbon and nitrogen sources for the production of antibiotics by S. albidoflavus C247. Additionally, 62.89% mycelial growth inhibition was achieved when the organism was cultured at $30^{\circ}C$ and pH 6.5. Ethyl acetate (EtOAc) was the best extraction solvent for the isolation of the antibiotics, and 100 ${mu}$/ml of EtOAc extract was found to inhibit 60.27% of the mycelial growth of Rhizoctonia solani AG2-2(IV) when the poison plate diffusion method was conducted.

A Philological Study on The Portulacea (마치현에 대(對)한 문헌적(文獻的) 고찰(考察))

  • Objectives : The aim of this study is to find the therapeutic meaning of the Portulacea in herbal medication. Methods : About the origin, the component, the processing the drug, the properties and tastes of drugs, the meridian tropism, the effects, the treating disease, the contraindication and the method of administration, I have researched thirty literatures to mention the Portulacea in time sequence. Results : 1. The Portulacea belongs to the Portulacaceae herbs and it consists of noradrenaline, potassium sometimes containing small amounts of dopamine, dopa, malic acid, citric acid, glutamine acid, asparagin acid, alanine, cane sugar, grape sugar, fruit sugar. 2. The processing of drug is wash of water clealy, and remove the foreign substance. Then the drug is cutting to use. The method of burnt to ashes is used, too. 3. The properties and tastes of drugs is acid, cold, nontoxic. The meridian tropism is mainly liver and the large intestine meridian. 4. From old times, Portulacea has come into general use to treat eczema, the rose erysipelas, an acne, hemorrhoids, discharge from the womb etc. because it is effective on neutralizing poison, reduce a swelling, a tumor, an abscess and stopping of bleeding 5. Portulacea must be stoped When person have a weak digestive organ becase it is cold herba. And don't take use with Fish and shellfish. 6. Portulacea is useful method to external care. To use the herba, pulverize amount of property and then apply to the the affected part Conclusions : This study showed that the Portulacea is useful herb to treat of skin disease and useful method to external care.
