Antioxidative Effects of Mushroom Extract and Fermented Milk Containing Its Extract on in vivo and in vitro Lipid Peroxidation (버섯 추출물과 이를 함유한 유산균 발효유가 in vivo 및 vitro 과산화지질에 미치는 영향)

  • The antioxidative effects of fermented milk, mushroom extract and fermented milk containing its extract (Lentinus edodes, Ganoderna lucidum, and Pleurotus ostreatus) on the lipid peroxidation in the tissues of female Sprague-Dawley rats and on the DPPH ($\alpha,\alpha$' -diphenyl-$\beta$-picrylhydrazyl) radical donating ability were studied. The total concentrations of polyphenolic compound in Lentinus edodes, Ganoderma lucidum and Pleurotus ostreatus were 0.34, 0.20 and 0.34%, respectively. The DPPH donating abilities of mushroom extract, fermented milk, fermented milk containing its extract and BHT (butylated hydorxytoluene) as standard were 33.9, 34.9, 51.9 and 95.6%, respectively. Experimental diet groups were divided into five groups: the normal diet (ND), the cholesterol diet (CD), and cholesterol + fermented milk diet (CDFM), cholesterol + mushroom extract diet (CDME) and cholesterol + fermented milk containing mushroom extract diet (CDFMME). The concentrations of lipid peroxide in liver and its microsome were significantly lower in both CDFM and CDFMME groups than in the other groups. The kidney concentration of lipid peroxide was significantly higher in the CD group than in the ND group, but this rise were significantly decreased in the CDFM and CDFMME groups. Meanwhile, the concentrations of heart and spleen and their fractions were not significantly different among dietary groups. This study was suggested that the fermented milk diet containing mushroom extract effectively reduced the lipid peroxidation in liver and kidney of cholesterol-fed female rats.

Studies on the development of mushroom medium of Pleurotus ostreatus using ginkgo nutshell (은행껍질을 이용한 느타리버섯 배지 개발)

  • Ten to thirty percent of ginko nut shell(GNS) were added to poplar sawdust to investigate mycelial growth. Mycelial growth on the medium with GNS was 93~97(mm/28days) and was slower than that of the control without GNS as that of 101(mm/28 days). Mycelial growth time on medium of sawdust with 10% GNS was same with control without GNS as 28 days but that on medium with 20 to 30% GNS was delayed for 2 days. The time of pinhead formation of sawdust with 10 to 30% GNS was 7 days, the growth time to harvest was 5 days. Yields per volume of GNS added treatments were 80.5g to 91.8g, which was increased 14 to 30% compared to 70.8g of the control without GNS.

Cultural characteristics of oyster mushroom (Pleurotus ostreatus) on addition rate of Cudrania tricuspidata (기능성자원 구지뽕나무(Cudrania tricuspidata) 톱밥의 첨가량에 따른 느타리버섯의 재배적 특성)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Cudrania tricuspidata for production of functional oyster mushroom. Total nitrogen and carbon source of Cudrania tricuspidata was 0.27% and 40.9%, respectively and C/N ratio was 152. Total nitrogen source and pH of substrates mixed with Cudrania tricuspidata were 2.7~2.8 and 4.9~5.1, respectively. The contents of $P_2O_5$, CaO, MgO and $Na_2O$ were no significant difference. Mycerial growth was faster at Cudrania tricuspidata 10% than that of control, but the other treatment was slower. Yields of fruiting body was the highest at Cudrania tricuspidata 20%, and diameter and thick of pileus were no significant difference to increase of Cudrania tricuspidata addition ratio. The L value of pileus was the highest at the Cudrania tricuspidata 20% during mushroom harvest, but there was no significant difference in the a-value and the b-value. Chemical contents of fruiting body were the highest at the Cudrania tricuspidata 30%.

  • In this study, portective effect on DNA damage several mushrooms (Pleurotus ostreatus, Flammulina velutipes, Lentinula edodes) according to cooking methods was investigated using Comet assay. Three edible mushrooms were cooked by grilling, blanching, pan-frying, or by preparing 'Jeon' (traditional Korean pancake). Cells were incubated in medium with 4 kinds of samples for 48 h ($37^{\circ}C$) were further treated with hydrogen peroxide ($H_2O_2$) for 5 min as an oxidative stimulus. Oxidative damage was evaluated by single-cell gel electrophoresis (Comet assay) and quantified by tail DNA% (TD), tail length (TL), tail moment (TM). Though oxidative DNA damages expressed as TD, TL, TM of 4 cooked samples were higher than raw sample, which means lower protective activities, all samples including raw sample had significantly higher protective effects than the positive control (p<0.05). The protective effect on DNA damage of cooked samples decreased much more when soybean oil added, likely due to the thermal oxidation of oil during cooking. Although heat treatment could degrade protective effect on DNA damage of mushrooms, the cooked mushroom had significant effect on oxidative stress. In conclusion, grilling and blanching were the most advantageous cooking methods to protect oxidative DNA damage induced by $H_2O_2$.

  • Comparison of fruit bodies of Pleurotus ostreatus cultivar chunchu No .2 grown on the sawdust, rice straw and cotton waste substrates revealed differences in the pattern of differentiation of hyphal compartments. Required period for primordium induction of fruit bodies grown on sawdust substrate was 13 days. Physical structure shown as hardness of stipes grown on the sawdust substrate, fruit bodies were harden than control. Pileocystidia were well developed on the surface of pileus in the fruit body cultivated on rice straw. Microstructures of fruit body grown on the sawdust and cotton wastes substrates shown fast-discharge of basidiospore and sytoms ageing. Hyphae of fruit bodies formed on sawdust substrate had less stainable cytoplasmic material and many more vacuoles than hyphae of fruit bodies formed on synthetic substrate with 50% of pine sawdust, 30% of cotton seed hull and 20 of beet pulp(control).

  • 'Miso', a new variety of oyster mushroom was developed for the bottle culture at the Chungnam Agricultural Research & Extension Services. Its mycelium grew rapidly with $8.4{\sim}8.6\;mm/day $ at $25{\sim}28^{\circ}C$ on PDA medium. The optimum pH of the mycelial growth was pH 5.0. It took 24 days for the primitive primordium formation after inoculation on pine sawdust media mixed with 20% wheat bran. Fruiting body color was white, and the shape of pileus was convex-umbonate. In the bottle culture, the yield was 115.7 g per 850 ml bottle. Stipe length was about 54 mm, the number of stipe per bottle was 18.1, the size of pileus was 28 mm, and gill was crowded. The moisture content of 'Miso' was lower than that of Jangan No.5 and Wonhyeong No.1, and contents of protein, ash and sugar of 'Miso' were higher than those of other varieties, and tannin acid content of 'Miso' was lower than that of others. On the basis of AFLP analysis, the 'Miso' was distinct not only from Wonhyeong No.1, but also from their closest relative, oyster mushrooms.

Comparison of amino acid and free amino acid contents between cap and stipe in Flammulina velutipes and Pleurotus ostreatus (팽이와 느타리버섯 자실체의 갓과 대에서 아미노산 및 유리아미노산 함량 비교)

  • The composition and content of amino acid and free amino acid were separately compared in cap and stipe of fruitbody obtained from different color strains in winter mushroom and oyster mushroom strains. The result showed different kinds and amount of amino acids according to the mushroom parts, strains and kinds. Tryptophane was not detected in any mushrooms. Alanine was also not detected in all oyster mushroom strains. The content of glutamic acid was the highest and that of methionine, phenylalanine and cysteine were very low in all mushroom strains. When compared by parts, the ratio of amono acids showed similar tendency in composition. Generally the content of amono acids showed higher in caps than in stipes. The profile and content of free amino acid showed a lot of variation between cap and strip in the intra and inter mushrooms.

Characteristics and breeding of a new oyster mushroom (Pleurotus ostreatus) variety 'Chunhwashim' by using cytoplasmic hybrid (세포질전환 기법에 의한 신품종 느타리 '천화심'의 육성 및 자실체 특성)

  • Hybrid strains was selected by crossing between dikaryotic strain ASI 0628 (Dagul) bred from ASI 2596 (Suhan No.3) and ASI 2782(Black pileus mutant) and monokaryotic strain ASI 2344-84. The strain 84 that shown the best cultural characteristics was selected to be a new cytoplasmic hybrid variety and named as 'Chunhwashim'. The 'Chunhwashim' shown incompatibility among them forming the confrontation growth line against parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho). Fruiting body produced about $161.4{\pm}4.8g$ per bottle. And also the individual generation of fruit body is multiple than chunchu2ho as 40.9. The 'Chunhwashim' was cytoplasmic hybrid included hybrid DNA bands of parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho) by Random Amplified Polymorphic DNA (RAPD) primer, and mitochondria DNA band of monokaryotic ASI 2344 (chunchu 2ho) using mitochondria microsatellite DNA marker. This new cytoplasmic hybrid variety 'Chunhwashim' of oyster mushroom is characterized by multiple of individual generation and deeply grey color of pileus.

Characteristics of virus-like particles in color mutants of oyster mushrooms (원형느타리백색변이체에 존재하는 바이러스의 특성)

  • This study was carried out to characterize virus-like particles (VLPs) in Pleurotus ostreatus. The white and the dark gray mutants frequently observed in mushroom farms of Pleurotus ostreatus (Wonhyeong-neutari). A 5.8kb segments of dsRNA was detected only in the white mutants but not in the dark gray mutants. The VLPs were purified from the fruit bodies by Polyethylene Glycol (PEG) and ultracentrifugation. Electron microscopy analysis showed that VLPs were isometric about 14, 20~45nm in diameter. Further study is needed to reveal the morphological and yield variations of mushroom strains including VLPs observed in the mushroom farms. Also it is needed to maintain fundamental research for taxonomy, diagnosis, and physiology of VLPs in the mushroom strains.

Biological Control of Mycophila speyeri Barnes (Diptera: Cecidomyiidae) using Korean Steinernema and Heterorhabditis Isolates in Pleurotus ostreatus Cultivation House (느타리버섯 재배사에서 한국산 Steinernema와 Heterorhabditis를 이용한 버섯혹파리(Mycophila speyeri)의 생물적 방제)

  • The potential of five entomopathogenic nematodes, Steinernema carpocapsae Pocheon strain, S. logicaudum Nonsan strain, S. glaseri Dongrae strain, Heterorhabditis bacteriophora Hamnyang strain, and Heterorabditis sp. Gyeongsan strain were evaluated as biological control agents against a mushroom fly, Mycophila speyeri in the mushroom, Pleurotus ostreatus cultivation house. Control effect of M. speyeri was significantly different according to nematode species. Heterorhabditis was more effective than Steinernema. H. bacteriophora Hamnyang strain showed the highest control effect representing $49.0\%$ (7 days), $89.5\%$ (14 days) and $89.1\%$ (21 days post-treatment) at the rate of $1\times10^6$ and $46.5\%$ (7 days), $76.6\%$ (14 days) and $85.4\%$ (21 days post-treatment) at the rate of $1.0\times10^5$ Ijs/$1.5 m^2$ in Changnyoung, Gyeongnam, respectively. In Jinju, Gyeongnam, control effects of the sa me species were $54.0\%$ (7 days), $74.5\%$ (14 days), and $79.8\%$ (21 days post-treatment) at the rate of $1\times10^6$ and $49.0\%$ (7 days), $76.6\%$ (14 days), and $61.1\%$ (21 days post-treatment) at the rate of $1.0\times10^5$ Ijs/$1.5 m^2$, respectively.
