The Hypocholesterolemic Effect of Platycodi radix Saponin in Rats Fed a Hypercholesterol Diet (길경 saponin이 고콜레스테롤식이를 공급한 흰쥐의 지질대사에 미치는 영향)

  • The objective of this study is to find the effect of improving hyperlipidemia through Platycodi radix saponin. A hypercholoesterol diet and Platycodi radix saponin methanol extract were supplemented to the 6 week old male Spargue Dawley rats for four weeks. Results showed that the serum total cholesterol, triglyceride and LDL-cholesterol level decreased in the group supplemented with Platycodi radix saponin methanol extract(p<0.05), however, the serum total lipid and HDL-cholesterol did not show any difference from the control group according to the supplement of Platycodi radix saponin methanol extract. In the group supplemented with Platycodi radix saponin methanol extract, the total cholesterol decreased by 20%, triglyceride by 36% and LDL-cholesterol by 25% respectively, and there was no difference according to the level of Platycodi radix saponin methanol extract. AI(atherogenic index) and CRF(cardiac risk factor) were low in the group supplemented with Platycodi radix saponin methanol extract. Total lipid, total cholesterol and triglyceride in liver and feces were significantly low in the group supplemented with the Platycodi radix saponin methanol extract compared with the control group(p<0.05). According to this result, it is expected that the Platycodi radix saponin methanol extract supplemented in the hypercholesterol diet can lower atherosclerosis, the cause of cardiovascular disease, by reducing not only serum cholesterol and triglyceride but also liver cholesterol and triglyceride.

  • This study was conducted to investigate the effect of the Platycodi radix powder (PRP) and Platycodi radix saponin(PRS) on the reduction of lipid status In rats fed on high fat diet for 6 weeks after which lipid contents were measured in liver. And also by carrying out the histological examination throughout light microscope to observe the effects of fat accumulation reduction. The results obtained from this study are as fellows. In the levels of total lipid in liver, PRS Group significantly decreased compared with Contred Group, but PRP Group was not significantly changed. The content of triglyceride was tended to be slightly decreased in the PRP and PRS groups compared to the control group, which was not significant. It was observed from photomicrographs of hepatic tissue in rats that the PRP and PRS groups inhibits the lipid accumulation induced by high fat diets.

  • This study was conducted to investigate the effect of Platycodi radix Saponin(PRS) on the reduction of lipid status in rats fed on high fat diet for 6 weeks after which lipid contents were measured in serum , liver and feces. The results obtained from this study are as follows : That the levels of total lipid, total cholesterol and triglyceride in serum and liver were significantly lower in the PRS group as compared with the control group. The contents of total lipid and total cholesterol excreted in the feces were tended to be slightly increase in PRS group compared to the control group, which were not significant.

  • The extract of Platycodi radix has long been used as an anti-inflammatory traditional medicine in Korea for the treatment of bronchitis, asthma and acute respiratory disease. In this study, the anti-inflammatory actions of the methanol extract of Platycodi radix administered via oral administration were evaluated. The results revealed that the extracts exhibited potent anti-inflammatory action. Of the hexane, chloroform, ethyl acetate, butanol and water fractions, the butanol fraction had the most potent anti-inflammatory action. Additioinally the sapogenin obtained through acid hydrolysis of the butanol fraction did not show anti-inflammatory action. These findings confirm the value of Platycodi radix as a traditional Korean medicine.

In Vitro Inhibitory Effect of Triterpenoidal Saponins from Platycodi Radix on Pancreatic Lipase

  • In the process of investigating anti-obesity effect of Platycodi Radix, we found that aqueous extract of Platycodi Radix might inhibit intestinal absorption of dietary fat by inhibiting pancreatic lipase (PL) activity. In order to clarify the anti-obesity mechanism of Platycodi Radix, activity-guided isolation was performed to find active components. The total saponin fraction of Platycodi Radix appeared to have a potent inhibitory activity against the hydrolysis of triolein emulsified with phosphatidycholine by pancreatic lipase in vitro. Based on these results, further purification of active components yielded 10 known triterpenoidal saponins, among these compounds, platycodin A, C, D, and deapioplatycodin D exhibited significant inhibitory effects on PL at the concentration of $500\;{\mu}g/mL$ with 3.3, 5.2, 34.8, and $11.67\%$ pancreatic lipase activity vs control, respectively. Platycodin D was found to inhibit the PL activity in a dose-dependent manner. Therefore, the anti-obesity effect of Platycodi Radix might be due to the inhibition of pancreatic lipase by its saponins.

Variation of Saponin Content in the Decoctions of Platycodi Radix (길경(桔梗)의 추출조건에 따른 사포닌 함량의 변화)

  • We investigated the contents of crude saponin in the decoctions of Platycodi Radix prepared by the different extraction processes. Each lyophilized decoctions prepared by the different protocol were suspended in water and partitioned with ethylacetate (EtOAc) and n-butanol (BuOH), which gave the EtOAc fraction, BuOH fraction and the remaining water fraction, respectively. The content of crude saponin, which was estimated as the amount of BuOH fraction, and the HPLC profiles of the BuOH fraction were analyzed, and compared each others. The crude saponin content in the decoctions were increased proportionally to the increment of the extraction time, amount of water, extraction temperature and the number of repetition for extraction. Consequently, the optimized conditions were deduced to be suitable and recommendable for the preparation of Platycodi Radix ; the water amount needed for the extraction be 15-20 volumes to crude material, temperature for extraction be optimal at $85{\sim}100^{\circ}C$, extraction time be more than 5 hours and the repetition of extraction was 2 times.

A Saponin Complex, KPRG-C, and Its Sapogenin Complex, KPRG-D, Reduce Nociception and Inflammation in Animals

  • To develop a clinically available saponin- or sapogenin complex from Oriental medicines, the EtOH extract (KPRG-A) was obtained by extracting from the four crude drugs, Kalopanacis Cortex, Platycodi Radix, Rubi Fructus and Glycyrrhizae Radis. The BuOH fraction (KPRG-B), a crude saponin complex, was prepared by fractionating KPRG-A, which were further completely hydrolyzed to afford the sapogenin complex (KPRG-D). In an attempt to find the antinoicpetive effects of the saponin complex and sapogenin complex, KPRG-C, and -D, were assayed by writhing-, hot plate-, and tail-flick tests using mice or rats. The three samples were also subjected to antiiflammatory tests using serotonin-induced and carrageenan-induced hind paw edema mice and rats, respectively. The three samples significantly reduced inflammations and pains of the experimental animal. The potency were found in the order of KPRG-D> KPRG-C> KPRG-B. The most active sample, KPRG-D, caused no death, no body increase or no anatomical pathlogic change even at 2,000 mg/kg dose. These results suggest that a sapogenin complex, KPRG-D, which was found to contain mainly hederagenin, platycodigenin, polygalacic acid, 23-hydroxytormentic acid, glycyrrhetic acid together with minor triterpene acids, could be a potential candidate for antiinflammatory therapeutics.

Compositional Analysis of Major Saponins and Anti-inflammatory Activitiy of Steam-Processed Platycodi Radix under Pressure

  • Platycosides are the saponins in Platycodi Radix and they have several beneficial effects such as antiinflammatory and anti-obesity activities. This study was designed to determine the changes in the saponin composition in Platycodi Radix (platycosides) after being processed under steam and pressure and to investigate the anti-inflammatory effects of their extracts. The change of the platycoside compositions was investigated after 1, 2, 3, 6 and 9h heat processing of Platycodi Radices by using HPLC coupled with an evaporative light scattering detection (ELSD) system. After heat treatment ($125^{\circ}C$, 1, 2, 3, 6 and 9 h), the contents of several platycosides such as platycoside E, platycodin $D_3$, platycodin D, polygalacin D, and platycodin A decreased as the processing time was longer. While the total contents of the saponins decreased, the contents of deapi-forms of deapiplatycoside E, deapi-platycodin $D_3$, and deapi-platycodin D increased relatively. These results indicate that the linkage between apiose and xylose located at C-28 is labile to heat and pressure. The LPS-induced iNOS inhibitory activities of the samples treated for 1 and 2 hours were enhanced and after then, the activities were reduced. These results suggested that heat treatment of the samples affect the content of the total saponins and the saponin content may be the important criteria representing the anti-inflammatory activity.

Pharmacognostical Studies on Codonopsis lanceolata (더덕(사삼(沙蔘))의 생약학적 연구)

  • Attempts were made to determine the characteristics of the morphology of Codonopsis lanceolata $T_{RAUT's}$ root. Its saponin and crude powder were examined by foaming and hemolytic tests and by observing transverse sections: 1) The cork-layer was consisted of multiple cells, and some of the cork cells contained monoclinic prism crystals of calcium oxalates. 2) The vascular bundle was formed of radial arrangement by the preminentic bursting pith and the irregular cambium ring with the abnormal growth. 3) The latex vessel and latex of the cortex were brown-gray or yellowish-brown. The latex vessel and sieve tube were consisted of the independent aggregates which had a number of layers. It had much more independent aggregates than Platycodi Radix. 4) The xylem was less than the phloem portion. The thick-walled vessels which had one to five lines ware more distributed than in Platycodi Radix. 5) The inulin distribution on a piece of section which is dipped in alcohol was observed through out the tissues, but starch grain was not found in it. 6) The duration time of foaming reaction of the crude powder in test tubes was shorter than that of Platycodi Radix. Foaming index of the foaming test of five percent decoctions was 3.33, 7) In the hemolytic test which used 10 percent decoction and one percent saponin solution, hemolytic action was very weak.

HPLC Analysis of Saponins in Platycodi Radix (HPLC를 이용한 길경(桔梗) 사포닌 분석법(分析法))

  • A rapid and practical HPLC assay was developed for quantitative analysis of saponins in Platycodi Radix. Seven saponin components in Platycodi Radix, i.e., deapioplatycoside E, platycoside E, deapioplatycodin D$_3$, platycodin D$_3$, polygalacin D$_2$, platycodin D$_2$ and platycodin D were successfully resolved on C18 column and detected by ELSD. Standard curves were linear over the concentration range 1 ${\sim}$2,000 ${\mu}$g/ml (r$^2$>0.992). Intra- and inter-day coefficients for variation of seven saponin components were < 10% and limit of quantification of them were around 0.7${\sim}$1.5 ${\mu}$g/ml, respectively. Using this method, contents of seven saponins in various plant materials under different cultivating conditions were estimated.
