Characteristics of Escherichia coli isolated from piglets with diarrhea ; antimicrobial susceptibility, genotypes of enterotoxins and pili and plasmid profiles (설사 자돈으로부터 분리한 Escherichia coli의 특성에 관한 연구 ; 항균제 감수성, 장독소 및 섬모의 유전형의 분포 및 plasmid profiles)

  • The antimicrobial susceptibility, genotypes of enterotoxins(LT, STa) and pili(K88, 987P), and plasmid profiles were investigated with 102 Escherichia coli isolated from piglets showing diarrhea in Korea. Almost of them were susceptible to ceftiofur(99%), cefquinone(97.1%). However they showed resistance to bacitracin(100%), streptomycin(98%), vancomycin(97%), trimethoprim/sulfamethoxazole(87.2%), tetracycline(84.3%) in antimicrobial susceptibility test. Moreover, all of the isolates demonstrated resistance to more than 2, and 78% of them were resistant to more than 8 of total 17 drugs. Multiplex PCRs for genotyping of enterotoxins(LT, STa) and pili(K88, 987P) were established with primers designed based on sequences from Genebank. Seventeen strains(16.6%) of the isolates had STa gene, 11 strains(10.8%) of them had both STa and LT genes, and 18 strains(17.8%) had K88 gene. But none of the isolates harbored a gene exclusively encoding LT. The gene encoding 987P pili was not found in all isolates. Fifty-four strains of 102 isolates(52.9%) had plasmid with various sizes ranging from 125kb to 1.1kb. Numbers of plasmid per isolates were also various, from 1 to 9. Distinctive relationship between plasmid profiles and genotypes of enterotoxins and pili in the isolates was not found. These results might provide the basic knowledge to establish strategies for the treatment and prevention of colibacillosis in piglets.

Antibiotic resistance and plasmid profile of salmonella spp isolated from swine in Kyoungbuk province (경북지방 돼지유래 salmonella 속균의 약제내성과 plasmid profile)

  • This study was conducted to investigate the antibiotic resistance and plasmid profiles of 58 salmonella spp isolated from mesenteric lymphnodes of slaughter pigs in Kyoungbuk province during the period from September 1997 to June 1998. The results obtained are as follow that all isolates were susceptible to amikacin, gentamicin, ciprofloxacin and enrofloxacin, and the majority of isolates were highly susceptible to norfloxacin, colistin, nalidixic acid and apramycin while they were moderately susceptible to kanamycin, cephalothin, chloramphenicol, ampicillin, trimethoprim and penicillin. The majority of isolates were over 90% resistant rates to lincomycin, sulfadimethoxine, vancomycin, methicillin and erythromycin. The plasmid profiles of 58 salmonella spp are developed 1 to 4 fractions, 0.9 to 29.5 Kb molecular range sizes and U strains (45.5%) were showed plasmid profiles by agarose gel electrophoresis. 5 derby harbored 29.5 Kb and 7 Kb, and S schwarzengrund had 14 Kb and 0.9 Kb harboring sizes. Four of 10 S agona and 2 of 4 S typhimurium were harbored 3.1 Kb and n.5 Kb, respectively. Thirty-five untypable strains are developed variable size fractions its showed small size plasmid profile less than 6 Kb and 22 (62.8%) of them had no detectable plasmids.

  • Antimicrobial susceptibility, plasmid profiles and distribution of toxA gene were investigated in Pasteurella multocida isolated from pneumonic lung lesions of swine. The bacteria were highly susceptible to norfloxacin, cabenicillin, enrofloxacin and chloramphenicol, but resistant to colistin, sulfamethoxawle/trimethoprime, bacitracin, streptomycin. Sixty percentage of the isolates was resistant more than 2 drugs used in this experiment and 21 strains (23.6%) were resistant more than 5 drugs. This phenomenon meant that they had highly multi-drugs resistance. In the analysis of plasmid profiles, nineteen strains (47.5%) of 40 P multocida isolates harbored plasmids, ranging from 53.3kb to 2.49kb in size and the plasmid profiles could be classified into 5 groups. However, there was no relationship between the size and the profile of plasmid and the resistance pattern of antimicrobial agents. Thirty strains of 39 P multocida isolates (77%) investigated by PCR harbored toxA gene. This result suggested involvement of the ToxA protein expressed from the gene in pneumonic pasteurellosis of swine.

Characteristics of the Nisin-Resistant Transformants of Lactococcus lactis subsp. lactis LM0230

  • To investigate the nature and location of the nisin-resistance determinant of Lactococcus lactis subsp. lactis 7962 (L. lactis 7962), a total plasmid DNA prepared from L. lactis 7962, a nisin producer, was used to transform L. lactis subsp. lactis LM0230, a plasmid-free and nisin-sensitive strain, by protoplast mediated transformation procedures. All of the nisin-resistant transformants acquired the ability to utilize sucrose at the same time, confirming the close linkage between these two determinants in L. lactis 7962. The plasmid DNA profiles of a few selected nisin-resistant transformants were examined by agarose gel electrophoresis. No common plasmid was found among the transformants and some small plasmids previously not present in L. lactis 7962 were detected. These transformants were named as L. lactis KL1, KL2, KL3, KL4, or KL5, respectively based on their plasmid profiles. Growth curves of all transformants were similar to that of L. lactis LM0230, but different from that of L. lactis 7962. L. lactis KL5 showed the highest level of resistance to nisin, growing up to 1, 200 IU nisin/ml after 40 hr incubation. Some nisin-sensitive derivatives of KL1 or KL2 were obtained by plasmid curing experiments. The plasmid DNA profiles of the nisin-sensitive KL1 derivatives were apparently the same as that of the KL1. All of the nisin-sensitive KL2 derivatives were plasmid-free, but a nisin-resistant strain with no apparent plasmid was also obtained. These results indicate that the nisin-resistance of the $Nis^r$ transformants is presumably mediated by the chromosomally located gene(s) rather than plasmid-encoded gene(s).

  • A total of 166 strains of Salmonella (S) typhimurium var copenhagen isolated from pigeons (164 strains) and aquatic birds (2 strains) were examined for the biochemical characteristics and plasmid profiles. All the strains were sensitive to ampicillin, chloramphenicol, gentamicin, kanamycin and sulfadimethoxine. But 13 strains(7.8%) were resistant to streptomycin (Sm), 2 (1.2%) to tetracycline, 2 (1.2%) to rifampicin, and 1 (0.6%) to nalidixic acid. Among drug resistant strains, only one strain resistant to Sm contained conjugative R plasmid which was fertility inhibition and incompatibility group $I_{\alpha}$. All the strains were sensitive to cobalt chloride, cupric sulfate, lead nitrate, mercuric chloride and silver nitrate. Of 166 isolates, 6 (3.6%) were resistant to sodium arsenate and 1 (0.6%) to potassium tellurite. Among 166 isolates, 1 (0.6%) was colicinogenic, 12 (7.2%) sucrose fermenters, and 166 (100%) maltose fermenters. Plasmid profiles were confirmed as being 4 or 5 plasmids, and their molecular weight ranged 3.2 to 60 megadalton (MD). All the strains harbored 60 Md plasmid. There are three patterns by the plasmid profile, 150 isolates (90.4%) were pattern I (3.2, 3.5, 33, 60Md), 14 (8.4%) pattern II (3.2, 3.5, 29, 60Md), and 2 (1.2%) pattern III (4.2, 7.8, 8.5, 15, 60Md). S typhimurium var copenhagen strains containing 60Md plasmid were resistant to killing by 90% normal guinea pig serum.

Growth Characteristics and Plasmid Profiles of Yersinia enterocolitica lsolated from Springs Water (약수터수로부터 분리한 Yersinia enterocolitica의 성장특성 및 Plasmid 유형)

  • The studies were conducted to explore the dffects of growth or survival against various factors and plasmid profiles of 49 Y. enterocolitica isolated from springs water. In the presence of calcium hypochlorite, y. enterocolitica was entirely extinguished by exposure for 33 hours at 0.8 ppm concentration, and was grown up to 7% NaCl, but not at 95 NaCI. Y. enterocolitica was presented optimal growth at pH 7.0 anad 9.0, and not allowth the growth at pH3.0, 5.0 and 11.0. The optimal temperature for growth of Y. enterocolitica was 25$\circ$C and 35$\circ$C, and allowed the growth at refrigerant temperature, 5$\circ$C. Y. enterocolitica was remarkably decreased by exposure for 30 seconds under UV light, and entirely extinguished by exposure for 90 seconds. Therefore, UV light was effective for sterilization of Y. enterocolitica. Fourty-nine strains of Y. enterocolitica were harbor plasmid DNA of approximately 46 Kb molecular weight.

Proteome analysis between diverse phenotypes of Salmonella enterica subspecies enterica serovar Typhimurium (S. Typhimurium)

  • Protein expression patterns in Salmonella enterica subspecies enterica serovar Typhimurium (S. Typhimurium) strains with diverse phenotypes, such as phage type, antibiotic resistance pattern and plasmid profiles were examined. For detailed analysis of proteins expressed by different S. Typhimurium strains, protein fractions were divided into detergent-rich phase (DP) and aqueous phase (AP) using triton X-114 detergent. The two phases were subjected to two-dimensional gel electrophoresis (2-DE), followed by protein identification using peptide mass fingerprinting (PMF). In the results, PMF showed that DP fractions consisted mainly of outer membrane proteins, whereas the AP fractions included cytosolic proteins. Comparison of 2-DE profiles of DP did not show any distinct protein spots which could be correlated with phage type, antibiotic resistance pattern or plasmid profile. However, comparisons of 2-DE profiles of the AP revealed differences in the protein spots, which could be correlated with the plasmid profile and phage types. Among these protein spots, flagellin was specific for strains containing a 90 kb plasmid. Compared to DT193 phage type, three protein spots in the range of pI 5.0-5.5 and MW 8-15 kDa of AP 2-DE profiles were absent in the DT104 phage types. Additionally, a protein spot with PI in the range of 4.5-5.0 and molecular weight (MW) between 51-69 kDa was specific for phage type DT104, while a protein spot with pI in the range of 4.0-4.8 and MW between 18-20 kDa was specific for DT193 phage type. These protein spots may be useful for discriminating phage types of S. Typhimurium.

  • The purpose of this study was the examination for presence of pilus antigen, O serogroups, colicin production, antibiotic susceptibility and plasmid profiles among E coli isolated from diarrheal piglets and fattening pigs in Taegu province. Of 145 E coli isolated, 98 strains (67.4%) possesed pilus antigens which belonged to either K88 (47.6%), K99 (11.7%) or 987P (8.3%) types. Fifty-nine strains (40.7%) were classified into tenO serogroups and their types were O8 (22.0%), O20(16.9%), O141(15.3%), O9(10.2%), O45(10.2%), O139(8.5%), O064(6.8%), O149(5.0%), O157(3.4%), and O115(1.7%). Thirty-three strains (22.8%) were colicinogenic and 6 strains (4.1%) were hemolytic. One hundred and thirty-nine strains (95.9%) of 145 E coli isolates were resistant to ampicillin, chloramphenicol, gentamicin, kanamycin, streptomycin, tetracycline, rifampicin and nalidixic acid, alone or in combination thereof. Ninety strains (64.7%) of 139 drug resistant strains carried R factor (R) which were transferable to the recipient by conjugation. In gel electrophoresis for the isolation of plasmid DNA, the number of plasmid DNA band varied from 2 to 11 in 16 E coli with pilus antigen. It's molecular weight ranged from 1.0 to 60.0 megadalton.

Virulence and Plasmid Profiles of Salmonella gallinarum Isolated from Chickens (닭에서 분리한 Salmonella gallinarum의 병원성 및 Plasmid Profile)

  • The present study was conducted to investigate the antibiotic resistance of 24 Salmonella gallinarum isolated from 48 chicken samples of diagnosed fowl typhoid cases during the period from November 1998 to November 1999. And the isolates of S gallinarum were also tested for their invasion abilities to experimental infection as one of virulence tests, and the presence of virulence-related plasmid in S gallinarum isolates. The results obtained through this study were summarized as follows; 1. All of isolates from 48 cases of 24 farms were identified S gallinarum by biochemical and serological tests.2. Antimicrobial drug resistance test against 24 isolates showed that the isolates were resistant to Colistin(95.8%), and Penicillin(79.2%), Polymyxin B(75.0%), Streptomycin (65.2%), Gentamycin(54.2%), and Tetracycline(33.3%). 3. Mortality in chicken following peroral inoculation of four isolates of S gallinarum during 14-days inoculation pecked at 5 days(40%) after inoculation and all of experimental chickens died within 13 days after inoculation.4. Based on the pattern and number of isolated plasmid from each isolate, plasmid profiles were divided into five groups; group I with 3 plasmids, group II to group IV with 4 plasmids and group V with 5 plasmids.

Plasmid profiling of multi-drug resistant Vibrio sp. isolated from influent and effluent water samples of fish farms in Jeju, South Korea (제주 양식장 유입수과 방출수에서 분리한 다제내성 Vibrio균 플라스미드 프로파일링)

  • The objective of this study was to investigate the plasmid profiling of multi-drug resistant (MDR) Vibrio in influent (inflow) and effluent (discharged) water samples of fish farms in Jeju, South Korea. MDR isolates identified through disc diffusion susceptibility tests, were subjected to plasmid profiling. One hundred fifty Vibrio isolates were obtained from each influent and effluent water sample. All MDR isolates were subjected to plasmid profiling. Greater number of bacteria were enumerated from effluents (61%) comparing to influents (39%). High incidence of neomycin, sulfamethoxazole, amoxicillin and oxytetracycline resistance was observed among the isolates, which was higher in effluent samples. In contrast, Vibrio isolates were more susceptible to florfenicol, chloramphenicol, ciprofloxacin, and nalidixic acid. Among 99 (influent 39 and effluent 60) MDR isolates, a total of 58 (influent 38 and effluent 20) were found to bear plasmids ranging from 1.7 kb to >10 kb and showed 19 different antibiograms according to the size of plasmids. MDR isolates showed six and four distinct plasmid profiles in influent and effluent, respectively. Effluent samples contained more plasmid-carrying MDR Vibrio isolates with more diverse plasmid profiles and antibiograms, suggesting that fish farm tanks may serve as a reservoir of antibiotic resistance genes. The presence of plasmid-carrying MDR Vibrio isolates in fish farm effluent water may contribute to the dissemination of antibiotic resistance genes to the environments, which ultimately poses threat to human health.
