Plasma-derived products are produced from plasma via fractionation and chromatography techniques, but can also be produced by other methods. In the performance of nucleic acid amplification tests (NAT) with plasma-derived products, it is necessary to include an internal control for the monitoring of all procedures. In order to avoid false negative results, we confirmed the usefulness of the bovine viral diarrhoea virus (BVDV) for use as an internal control in the detection of hepatitis C virus (HCV) RNA in plasma-derived products. These products, which were spiked with BVDV, were extracted and then NAT was performed. Specificity and sensitivity were determined via the adjustment of primer concentrations and annealing temperatures. BVDV detection allows for validation in the extraction, reverse transcription, and amplification techniques used for HCV detection in plasma-derived products.

HCV is transmitted via various plasma-derived medicinal products. The transmission of HCV could. however, be prevented by screening plasma pools with NAT and validating HCV viral clearance during the manufacturing of plasma derivatives, Although various screening methods including commercial kits are available. it is yet to develop an analytical method to detect HCV in both plasma and plasma derivatives. The objective of this study was to develop a reliable in house method for reliable for the HCV RNA detection from plasma and plasma derivatives. (omitted)
A kinetic assay was carried out in order to compare the ability of detection for prekallikrein activator(PKA) in plasma-derived products with that of an endpoint assay and a commercial method. Using these methods, 9 human albumin preparations were assayed and compared to each other. The coefficient of variation between the Kinetic assay and the end point assay was found within 6.6% and this result showed that two methods were highly correlative and the end point assay could act as a replacement of the kinetic assay. Another important goal of this study was to investigate the reproducibility among laboratories on the kinetic assay. A collaborative study was performed to validate the kinetic method with intra and inter assays. The coefficient of variation for the intra assay of each laboratory was less than 4% and that for between individuals in the inter assay was 4.1%. These results revealed that the kinetic assay showed good reproducibility. The contents of PKA in plasma-derived products were also determined by the kinetic assay. As a result, it was found that trace amounts of PKA were present in 32 human immunoglobulin preparations, however the average concentration of PKA in 171 albumin preparations was 5.8 IU/mL.
HCV는 HIV등과 함께 수혈이나 혈장된 획물질을 통하여 감염되는 주요 바이러스이다. 주로 혈액이나 혈장에서 HCV에 대한 항체를 검출함으로서 HCV의 감염을 방지하고 있다. 그러나 바이러스에 감염되었으나 항체가 생성되기 이전이나 항체의 양이 적은 경우에는 HCV의 검출이 어렵다. 따라서 핵산중폭시험(nucleic acid amplification tests, NAT)을 이용한 HCV 유전자를 검출하려는 시도들이 진행되고 있다. 이 연구의 목적은 혈장분획물질에서 HCV RNA를 검출할 수 있는 핵산증폭시험 방법을 개발하는 것이다. 5종류의 PCR primer를선별하여 실험에 이용하였다. 혈장분획물질의 HCV RNA 추출에는 컬럼 방법을 이용하는 것이 유용한 것으로나타났다. 핵산중폭시험의 결합 온도는 $48^{\circ}C$가 가장 적절한 것으로 나타났다. 또한 2차 PCR의 경우, 1차 PCR 산물 $1{\mu}l$와 30 pmol의 primer즐 사용하였을 때 높은 민감도와 특이성을 보이는 것을 알 수 있었다. 혈장 분획물질에 HCV를 주입하여 혈장중폭시험을 수행한 결과, 100 IU/ml까지 검출 할 수 있었다. 한편 근육주사용항체(IMIG)의 경우 핵산중폭시험을 통한 검출한계는 100IU/ml로 COBAS amplicor HCV2.0의 500 IU/ml 이상의 검출한계보다 민감도가 더 높은 것으로 나타났다. 이러한 결과들로 보아 본 실험에 이용된 핵산증폭시험이 혈장분획물질에서 HCV RNA를 검출하는데 유용한 방법으로 사용될 수 있을 것으로 생각된다.
The osteoporosis is a disease characterized by lower bone mineral content, deterioration of bone tissue and a reduction in the protein and mineral matrix of the bone. The bone becomes more porous leading to increased bone fragility and risk of fracture, particularly of the hip, spine and wrist. Osteoporosis can result in disfigurement, lowered self·esteem, reduction or loss of mobility, and decreased independence. Adequate calcium intake through milk and milk products in childhood and adolescence is a decisive marker for obtaining a maximum bone mass (peak adult bone mass) and f3r the prevention of osteoporosis. Calcium is one of the most critical nutrients associated with the osteoporosis. Dietary calcium is of great significance for healthy skeletal growth and development. The bone mineral content and bone mineral density of young adults is directly related to the calcium intake through milk and dairy products. Milk and milk products are the important sources of calcium as the richness and bioavailability of this nutrient is very high as compared to other food products. If enough calcium is not supplemented through diet, calcium from the bone will be depleted to maintain the blood plasma calcium level. The article focuses on the various issues related to osteoporosis manifestation and the role of dietary calcium especially calcium derived from dairy products.
Recently, marine organisms are emerging as a leading group for identifying and extracting novel bioactive substances. These substances are known to possess a potential regarding not only as a source of pharmaceutical products but also their beneficial effects on humans. Among the substances, antimicrobial peptides (AMPs) specifically have attracted considerable interest for possible use in the development of new antibiotics. AMPs are characterized by relatively short cationic peptides containing the ability to adopt a structure in which cationic or hydrophobic amino acids are spatially scattered. Although a few reports address novel marine organisms-derived AMPs, their antimicrobial mechanism(s) are still remain unknown. In this review, we summarized the peptides previously investigated, such as Pleurocidin, Urechistachykinins, Piscidins and Arenicin-1. These peptides exhibited significant antimicrobial activities against human microbial pathogens without remarkable hemolytic effects against human erythrocytes, and their mode of actions are based on permeabilization of the plasma membrane of the pathogen. Therefore, the study of antimicrobial peptides derived from marine organisms may prove to be useful in the design of future therapeutic antimicrobial drugs.
본 연구는 우리나라 성인 남녀에서 관상동맥질환의 주요 예측 지표인 AIP와 식품군 섭취의 관련성을 분석하였다. 본 연구에서는 40-79세의 성인 남녀 133,381명을 대상으로 AIP 5분위 그룹에 따른 식품군 섭취량을 제시하였으며, 그 결과 AIP가 높은 그룹에서 낮은 그룹에 비해 곡류, 김치/피클류의 섭취가 높았고, 콩류, 종실류, 육류, 난류, 생선류, 우유 및 유제품 등의 섭취가 낮은 경향을 보였다. 본 연구는 한국인의 식사와 관상동맥질환 위험도 간의 관계에 대한 중요한 정보를 제공하며, 관상동맥질환 예방을 위해 다양한 식품군이 포함된 균형 잡힌 식사가 중요함을 시사한다. 본 연구 결과는 관상동맥질환을 예방할 수 있는 식사지침 제시를 위한 기초자료로 활용될 수 있을 것이다. 추후 연구를 통해 한국인의 식사패턴을 분석하여 심혈관질환 발생 및 사망과의 관련성을 조사하고, 이를 통해 한국인의 심혈관질환 예방에 기여할 수 있을 것으로 기대한다.
We previously reported that leukotriene B4 (LTB4) contained in Trichomonas vaginalis-derived secretory products (TvSP) play an essential role in interleukin-8 (IL-8) production in human mast cell line (HMC-1 cells) via LTB4 receptor (BLT)-mediated Nuclear Factor-kappa B (NF-κB) activation. Dynamin, a GTPase, has been known to be involved in endocytosis of receptors for signaling of production of cytokine or chemokines. In the present study, we investigated the role of dynamin-mediated BLT1 endocytosis in TvSP-induced IL-8 production. When HMC-1 cells were transfected with BLT1 or BLT2 siRNA, TvSP-induced IL-8 production was significantly inhibited compared with that in cells transfected with control siRNA. In addition, pretreatment of HMC-1 cells with a dynamin inhibitor (Dynasore) reduced IL-8 production induced by TvSP or LTB4. TvSP- or LTB4-induced phosphorylation of NF-κB was also attenuated by pretreatment with Dynasore. After exposing HMC-1 cells to TvSP or LTB4, BLT1 was translocated from the intracellular compartments to the plasma membrane within 30 min. At 60 min after stimulation with TvSP or LTB4, BLT1 remigrated from the cell surface to intracellular areas. Pretreatment of HMC-1 cells with dynamin-2 siRNA blocked internalization of BLT1 induced by TvSP or LTB4. Co-immunoprecipitation experiments revealed that dynamin-2 strongly interacted with BLT1 60 min after stimulation with TvSP or LTB4. These results suggest that T. vaginalis-secreted LTB4 induces IL-8 production in HMC-1 cells via dynamin 2-mediated endocytosis of BLT1 and phosphorylation of NF-κB.

Spent $UO_2$ nuclear fuel discharged from a nuclear power plant (NPP) contains fission products, U, Pu, and other actinides. Due to neutron capture by $^{238}U$ in the rim region and a temperature gradient between the center and the rim of a fuel pellet, a considerable increase in the concentration of fission products, Pu, and other actinides are expected in the pellet periphery of high burnup fuel. The characterization of the radial profiles of the various isotopic concentrations is our main concern. For an analysis, spent nuclear fuels originating from the Yeonggwang-2 pressurized water reactor (PWR) were chosen as the test specimens. In this work, the distributions of some actinide isotopes were measured from center to rim of the spent fuel specimens by a radiation shielded laser ablation inductively coupled plasma mass spectrometer (LA-ICP-MS) system. Sampling was performed along the diameter of the specimen by reducing the sampling intervals from 500 ${\mu}m$ in the center to 100 ${\mu}m$ in the pellet periphery region. It was observed that the isotopic concentration ratios for minor actinides in the center of the specimen remain almost constant and increase near the pellet periphery due to the rim effect apart from the $^{236}U$ to $^{235}U$ ratio, which remains approximately constant. In addition, the distributions of local burnup were derived from the measured isotope ratios by applying the relationship between burnup and isotopic ratio for plutonium and minor actinides calculated by the ORIGEN2 code.
