The Effects of Dietary Shellfish on Lipid Metabolism in Rats Fed High-Fat Supplement (고지방식이를 급여한 흰쥐의 패류식이 급여 효과)

  • This study was conducted to investigate the effects of shellfish on lipid metabolism in rats fed high fat supplements. Male sprague-dawley rats weighting approximately 165g were fed a basal diet, a high fat diet, or a high fat diet plus shellfish for 4 weeks. The shellfishes on the were oyster, hard-shelled mussel, little neck clam, and march clam. Alfter 4 weeks high fat diet, supplementation of 20% lard significantly increased plasma. GOT, GPT, ${\gamma}$-GTP , and liver triglyceride (TG). Plasma GOT, GPT, ${\gamma}$-GTP , triglyceride, and total cholestrerol levels were significantly lower in shellfish groups than in basal and high-fat groups regardless of high-fat supplementation (p<0.05). The total lipid and cholesterol content in liver showed similar results(p<0.05). There were no differences in glucose, HDL-cholesterol in plasma and total cholesterol and total lipid in liver between basal and high-fat supplemented diets. Liong chain fatty acids, specific components of shellfishes group, were exclusively higher than in basal and high-fat diets, and were most well-reflected in liver and plasma. From the above results, the hypolipidemic effects of shellfish were detected in the process of inducing hyperlipidemia by high-fat supplement.

Hypoglycemic Effect of Exo- and Endo-biopolymers Produced by Submerged Mycelial Culture of Ganoderma lucidum in Streptozotocin-Induced Diabetic Rats

  • The hypoglycemic effect of an exo-biopolymer (EXO) and endo-biopolymer (ENDO) produced from submerged mycelial culture of Ganoderma lucidum was investigated in streptozotocin (STZ)-induced diabetic rats. Both the EXO and ENDO showed hypoglycemic potential, however, the former proved to be more potent than the latter. The administration of the EXO at the dose of 100 mg/kg body weight (BW) significantly reduced the plasma glucose level (23.5%) and increased the plasma insulin level (2.2 fold) in the diabetic animals. The EXO also lowered the plasma total cholesterol, triglyceride, low-density lipoprotein (LDL) cholesterol, and athrogenic index by 14.7, 31.4, 24.1, and 45.4%, respectively, and reduced the liver total cholesterol and triglyceride levels by 6.7 and 25.8%, respectively. It increased the plasma high-density lipoprotein (HDL) cholesterol (37.7%), compared to the control group. Furthermore, the alanine transaminase (ALT) and aspartate transaminase (AST) showed lower activities in the EXO administered groups than the other experimental groups. Taken together, these results suggest that the exo-biopolymer may alleviate the blood glucose level by increased insulin secretion.

Effect or Cornus officinalis Sieb. et Zuccha Extracts on Physiological and Antioxidative Activities in Streptozotocin Induced Diabetic Rats

  • This investigation was performed to study the antioxidant activities of Cornus officinalis Sieb extracts and the effect of Cornus officinalis Sieb extracts on glucose, lipid metabolism in diabetic rats. DPPH free radical scavanging activity and superoxide anion radical scavenging of Cornus officinalis Sieb extracts were 94.7% and 92.1%, respectively. Streptozotocin (45 mg/kg body weight, i.p.) induced diabetic rats showed a significant increases of plasma glucose, triglyceride and total cholesterol, concomitantly significant decrease of plasma high density lipoprotein. Glutathione level were decrease in cytosol of liver, lung and brain tissue of rats. Lipid peroxide were increase in microsome of liver cells. Group 1 and 2 were treated with Cornus officinalis Sieb extracts 200 mg/kg body weight and 100 mg/kg body weight for 24 days, individually. Group 1 and 2 rats showed decreased plasma glucose, triglyceride, total cholesterol and lipid peroxide in microsome of liver, and increased plasma high density lipoprotein and glutathione in cytosol of liver, lung and brain. The result suggest that Cornus officinalis Sieb extracts may normalize the Impaired antioxiants status in streptozotocin induced diabetic rats. Cornus officinalis Sieb extracts were used to improve the imbalance between free radicals and antioxidant system due to the diabetes.

Effect of Mixed Onion Pills on Plasma Triglyceride and Total Cholesterol Levels in Overweight or Obese Men (혼합 양파환이 과체중 및 비만 남성의 혈중 지질 농도에 미치는 영향)

  • The effect of mixed onion pills on body composition, blood pressure, and concentrations of blood glucose and lipids were studied in 10 men ($38.11{\pm}2.82$ years old). The mixed onion pills was produced by mixing quercetin, onion, mulberry leaves and buckwheat. Ten volunteers (male, BMI 23.0~35.4 kg/$m^2$) were chosen for this study. The volunteers consumed 1.2 g of mixed onion pills (containing about 30 mg of quercetin) per day for 2 months. The administration of the mixed onion pills did not produce any weight and BMI changes in the subjects. The systolic blood pressure and the level of blood glucose were slightly lowered by the supplementation of the mixed onion pills. After 2 months both the concentration of plasma triglycerides and total cholesterol of the subjects were lowered by 12.4% and 5.0%, respectively (triglyceride; pre $251.67{\pm}45.63$ mg/dL vs. post $220.44{\pm}44.67$ mg/dL and total cholesterol; pre $207.25{\pm}10.39$ mg/dL vs. post $196.63{\pm}10.47$ mg/dL, p<0.05). These results suggest that the mixed onion pills may possibly lower the level of plasma lipids in overweight or obese men (BMI greater than 23 kg/$m^2$).

The Effect of Vitamin B6 Deficiency on Energy Metabolite in Streptozotocin-induced Diabetic Rats (Vitamin B6 결핍이 Streptozotocin 유발 당뇨 흰쥐의 에너지 대사물 농도에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin B6 deficiency on the concentration of energy metabolite in streptozotocin-induced diabetic rats. Thirty rats were fed a vitamin B6 deficient diet(-B6) or a control diet(+B6) for 5 weeks and then subdivided into 3 groups respectively ; base group, one day diabetic group and three day diabetic group. Diabetes of rats were induced by streptozotocin injection into the tail vein. Glucose, glycogen, protein, alanine, triglyceride and free fatty acids were compared in plasma, liver skeletal muscle of rats. Also, the total urinary nitrogen and glucose excretion were compared. Compared with +B6 rats, the increase of plasma glucose in -B6 rats due to the diabetes was smaller. After diabetes was induced, the level of plasma alamine was not changed in -B6 rats while increased significantly(p<0.05) in +B6 rats. The increase of urinary nitrogen excretion was smaller and the increase of muscle protein was larger in -B6 rats at the first day diabetes was induced. The levels of plasma free fatty acid and liver triglyceride were significantly (p<0.05) higher in -B6 rats after diabetes was induced. These results suggest that vitamin B6 deficiency may impair the adaptation of animals to the energy metabolism related due to a decrease of the body protein catabolism of fatty acid oxidation in diabetes and aggravate fatty liver which is one of the chronic complications of diabetes.

Effects of Powdered Siho(Bupleuri Radix) on Serum and Liver Lipid Composition and Antioxidative Capacity in Rat Fed High Oxidized Fat (시호(Bupleuri Radix)분말이 과산화지질을 급여한 흰쥐의 혈장 및 간장지질구성과 항산화능에 미치는 영향)

  • Effects of powdered siho(bupleuri radix) on serum and liver lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Sixty male Sprague-Dawley rats weighing 161.25$\pm$2.51g were blocked into four groups according to body weight and raised seven weeks with basal diet(normal group), basal diet and 10% oxidized fat(control group), basal diet, 10% oxidized fat and 2% powdered siho(2% powdered siho group) and basal diet, 10% oxidized fat and 3% powdered siho(3% powdered siho group). Feed intake, body weight gain and feed efficiency were not significantly different among oxidized fat diet groups. The values of total cholesterol and triglyceride in plasma and liver showed no significantly different(p>0.05) in the oxidized fat diet groups. Plasma HDL-cholesterol showed atendency to increase in powdered siho diet groups. The values of thiobarbituric acid in plasma and liver were lower in the powdered siho diet groups than control group. Plasma GOT and GPT activity showed a tendence to decrease in powdered siho groups. Liver glutathione peroxidase activity showed a tendence to increase in powdered siho diet groups. Thus, it seems that powdered siho had some antioxidative elements.

Effects of Inonotus obliquus Mycelia on the Level of Plasma Glucose and Lipids in Streptozotocin-induced Diabetic Rats (차가버섯 균사체가 Streptozotocin으로 유발된 당뇨 흰쥐의 혈장 포도당과 지질 농도에 미치는 영향)

  • This investigation was undertaken to study the effects of oral administration of Inonotus obliquus mycelia produced by a submerged culture on plasma glucose level and other biochemical parameters in streptozotocin (STZ)-induced diabetic rats. The mycelia, at the dose of 200 mg/kg body weight (BW), substantially reduced the plasma glucose level by as much as 23.1% as compared to the control group. The levels of total cholesterol and triglyceride in plasma were reduced to the extent of 12.6% and 22.6%, respectively. The activities of alanine transaminase (ALT) and aspartate transaminase (AST) were decreased by 27.6% and 21.9%, respectively, under the influence of I. obliquus mycelia. The general components of I. obliquus mycelia were found to contain 5.55% crude ash, 2.35% crude fat, 28.29% crude protein, 9.53% carbohydrate, and 54.28% dietary fiber.

Effect of Dried Powders or Ethanol Extracts of Onion Flesh and Peel on Lipid Metabolism, Antioxidative and Antithrombogenic Capacities in 16-Month-Old Rats (양파의 육질과 껍질의 건분 및 에탄을 추출물에 노령흰쥐의 지방대사와 항산화능 및 항혈전능에 미치는 영향)

  • This study was performed to investigate effect of dried powder or ethanol extracts of onion flesh and peel intakes on lipid metabolism, antioxidative and antithrombogenic capacities in l6-month -old rats. Total of 40 Sprague-Dawley male rats of l6-month-old and weighing 816$\pm$6g were blocked into 5 groups according to body weight and raised for three months with control and experimental diets containing 5% (w/w) of dried powders of onion flesh or peel or ethanol extracts from equal amount of each dried powder. Contents of total flavonoids and total dietary fibers in peel powder were highest among onion preparations. Body weight gain and epididymal pad fat weight were lower in peel powder group than other groups. Plasma total lipid, triglyceride and total cholesterol concentrations of onion-containing groups were lower than control group. Above all, peel ethanol extract intake decreased them most remarkably. Plasma HDL-cholesterol concentrations in onion-containing groups were higher than control group, especially that of flesh powder group was the highest among groups. Liver total lipid, triglyceride and total cholesterol concentrations were not significantly different among all experimental groups. However, liver total lipid and triglyceride concentrations were tended to be lower in onion-containing groups than control group. Thiobarbituric acid reactive substances (TBARS) concentrations in LDL + VLDL fraction was not significantly affected by onion intakes, However peel powder group showed the lowest concentration, Plasma TX $B_2$ concentrations in onion flesh powder, peel powder and peel ethanol extract groups were lower than control group, while plasma 6-keto-PG $F_{1a}$ concentrations in these same groups were higher than control group. Clotting time was tended to be increased in peel ethanol extract group. In conclusion onion diets seemed to improve lipid metabolism and antithrombogenic capacity while effect on antioxidative was not significant.t.

The Relationship between Serum Cholesterol Levels and Dietary Fatty Acid Patterns, Plasma Fatty Acids, and Other Lipid Profile among Korean Adults (성인 남녀의 혈청 콜레스테롤의 수준과 지방산 섭취 양상, 혈중 지질 및 지방산 조성의 상관 관계 연구)

  • The purposes of this research were to assess dietary fatty acid patterns and to elucidate the relationship between the serum cholesterol levels and dietary fatty acid patterns, plasma fatty acid compositions, BMI (body mass index), and other lipid profile. The subjects were 151 adults aged 23 to 80 years, selected from the Outpatient Clinic and Cardiova-scular Department of the Seoul Municipal Hospital. Dietary data were obtained using three day food records. Sixteen dietary fatty acids were analyzed using Korean and US nutrient databases. The subjects were divided into three serum cholesterol levels: desirable (< 200 mg/dl, N = 44), borderline-risk ($\geq$ 200 - < 240 mg/dl, N = 35), and high-risk ($\geq$ 240 mg/dl, N = 72) groups. The high-risk group had higher BMI, waist, and waist to hip ratio (WHR) than the desirable and borderline-risk groups. Serum concentrations of triglyceride, LDL cholesterol and LDL/HDL cholesterol ratio were significantly higher in the high-risk group as compared to those in the other two groups. The serum cholesterol levels were highly correlated with BMI (r = 0.435), triglyceride (r = 0.425) and LDL/HDL cholesterol (r = 0.870) ratio. The highest fatty acid intake was from oleic acid (33 - 34% of total fatty acid intakes), which was followed by linoleic acid (27%), palmitic acid (19%), and stearic acid (7%). There was no correlation between the serum cholesterol levels and the dietary fatty acid intakes, polyunsaturateumonounsaturateusaturated fatty acids (P/M/S) and $\omega$6/$\omega$3 ratios. The correlation between plasma fatty acids such as myristic acid, oleic acid, linoleic acid, and docosahexaenoic acid and serum cholesterol levels was also weak. Effect of FC-GT Supplement on Body Fat and Lipid Metabolism in Rats

  • The objective of this study was to investigate the effects of supplementation of an antiobese functional formula (FC-GT) on body weight and lipid metabolism in rats fed a high-fat diet. Three groups of male Sprague-Dawley rats were fed different diets for 6 weeks: normal control (NC), high-fat (HF), and high-fat supplemented with powdered antiobese functional formula (FC-GT) (5% wt/wt) groups. Although body weight was not significantly different among the groups, relative weights of epididymal and perirenal white adipose tissues were significantly lower in the FC-GT group than in the HF group. FC-GT supplementation significantly lowered the plasma total cholesterol and triglyceride concentrations, whereas it elevated the ratio of HDL-C/total-C and improved the atherogenic index. Hepatic cholesterol and triglyceride concentrations were significantly lowered in the FC-GT group compared to the HF group. The accumulation of hepatic lipid droplets and the epididymal white adipocyte size of the FC-GT group were diminished compared to the HF group. Hepatic HMG-CoA reductase activity was significantly lower in the FC-GT group than in the HF group. Plasma GPT activity was significantly lowered in the FC-GT group compared to the HF group. Additionally, fecal weight was significantly increased in the FC-GT group than in the HF group. In addition, contents of fecal triglyceride and cholesterol were significantly higher in the FC-GT group compared to the other groups. The antioxidant activities of hepatic SOD, CAT, and GR were significantly increased in the FC-GT group compared to the HF group. Hepatic mARS and plasma mARS levels were significantly lowered in the FC-GT group compared to the NC group. Accordingly, we conclude that supplementation of FC-GT improves plasma and hepatic lipid levels in high-fat fed rats.
