Effects of Castration and Ovariectomy on Plasma Hormone Concentration in rigs (돼지의 거세와 난소 적출이 혈중 호르몬 농도의 변화에 미치는 영향)

  • This study was conducted to investigate the effects of castration and ovariectomy on growth performance and plasma hormone concentration in pigs. A total of 48 pigs of 35 days of age were used. The results obtained in the present study are summarized as follows: 1. No significant difference was found in average daily gain between ovariectomy group (898.6g) and control gilt group (862.7g), and between castration group (926.0g) and control boar group (945.5g), respectively. Average daily gain of control boar group, however, was significantly higher than that of control gilt group (p<0.05). There was no significant difference in feed/gain between ovariectomy and control gilt group and between castration and control boar group, respectively. Backfat thickness was significantly (p<0.05) higher in ovariectomy or castration group than in control gilt or boar group, respectively. 2. Plasma concentration of IGF-I was significantly (p<0.05) increased during the period of 5 weeks of age (45.1 $\pm$0.72 ng/ml) to 15 weeks of age (356.3$\pm$3.05 ng/ml), and maintained constantly afterwards in control gilt group, as was in control boar group. That of ICF-I tended to be lower in ovariectomy or castration group than in control gilt or boar group, respectively. Regarding steroid hormones of estradiol-17$\beta$, progesterone, and testosterone, the concentration was extremely low at 5 weeks of age, however, increased from 11 weeks to 23 weeks of age in control gilt or boar group, while it was nearly under detection limit in ovariectomy or castration group. 3. Chemical compositions of pork loins were not affected by ovariectomy or castration, except that crude ash content was significantly (p<0.05) higher in castration group than in control boar group. These results indicated that ovariectomy or castration had no effects on growth performance and feed utilization. However, the concentration of sex steroid hormones was under detection limit in ovariectomy and castration group. Further studies, however, are needed to develope the techniques which minimize the stress related with castration or ovariectomy for the production of high quality pork.

Effect of LED Light Colors on Egg Production, Egg Quality and Reproductive Hormone Concentrations of Plasma and Oviduct in Brown Laying Hens Housed on Floor (LED 조명의 색이 평사 사육 갈색 산란계의 산란성적, 계란 품질 및 혈액과 난관 내 번식 호르몬 농도에 미치는 영향)

  • This study investigated the effect of LED light wavelength (color) on reproductive hormones and egg production of brown laying hens raised on floor. Red, blue, green and white colors of LED light were four treatments with four pens per treatment. One hundred forty four Hy-line brown laying hens (47 wks old) were allocated in a floor pen for six weeks trial. Egg production, egg quality, yolk cholesterol and hormones ($17{\beta}$-estradiol, progesterone) concentrations in plasma and oviduct were analyzed. Egg production of red group was higher (P<0.01) than that of green group. Haugh unit of eggs from red group was higher (P<0.01) than that of blue and green groups. Egg weight of green group was heavier (P<0.05) than that of red group. Shell of blue group was stronger (P<0.05) than that of red and white groups. Shell color of white group was browner (P<0.01) than that of blue and green groups. Yolk cholesterol of red group was higher (P<0.01) than that of others. Plasma $17{\beta}$-estradiol of red group was higher (P<0.05) than that of others at $3^{rd}$ week, but that of white group was highest (P<0.05) at $6^{th}$ week. Oviduct progesterone of green group was higher (P<0.01) than that of others. The result showed that the LED colors affect the reproductive hormone concentrations, egg production, egg weight and egg quality. This study suggested that red LED would be the most appropriate color for floor raising brown laying hens to sustain the egg production when it begins to decline with aging.

Effects of Chromium Picolinate on Growth Performance, Carcass Characteristics and Plasma Components in Holstein Bulls (홀스타인 수소에 있어서 Chromium Picolinate의 첨가가 성장성적, 혈액성상 및 도체품질에 미치는 영향)

  • We conducted two experiments to evaluate the effects of chromium picolinate(CrP) on growth performance, carcass characteristics and plasma components in Holstein bulls. In trial Ⅰ, eight finishing Holstein bulls(300${\pm}$6.99Kg) were allocated to 2 treatments(control and 0.05% CrP) with 4 replication for 10-months. In results, growth performance was not affected by CrP addition. The plasma insulin concentration in 0.05% CrP group was about 2 times higher than the control group of Holstein bulls. The levels of plasma NEFA were significantly decreased to 59.00 mEq/dl with 0.05% CrP treatment(P<0.05), but the levels of plasma glucose and PUN were not altered by 0.05% CrP treatment. The grade of carcass was not different between control and 0.05% CrP group, but back fat thickness in 0.05% CrP group was increased in 22.33% compared with control group. In trial 2, fifteen growing- finishing Holstein bulls(160${\pm}$4.63Kg) were allocated to 3 treatments(control, 0.025% CrP and 0.05% CrP) with 5 replication for 14-months. During the overall experimental period, growth performance was not affected by CrP levels. The levels of hormone and metabolites were not affected by CrP supplementa- tion. The carcass characteristics were not different between control and treatment. These results show that the CrP may have no effects for beef cattle production because of degradation of CrP conjugation in the rumen. However treatment of short term provide a possibility the effects of development for lipogenesis.

Comparisons of Estrus Induction with Anti-Prolactin and Hormones in Bitches (Anti-Prolactin과 다양한 호르몬 투여에 따른 개의 발정 유도 효율 비교)

  • Domestic bitches are non-seasonally monoestrus; spontaneously ovulate only once or twice occurs at anytime of the year. Estrus induction has been applied infrequent estrus, misleading ovulation, mating difficulties, failure to conceive after normal mating, pregnancy failure and biological research. Protocol of estrus induction which included variable hormones such as FSH, GnRH, and PMSG have been applied for the last decades. Recently, Bromocriptine, one of anti-prolatin/dopamine agonist has been occasionally applied for estrus induction. The study was carried out to investigate the effective method for the induction of estrus in bitches using different hormone treatments, and the initiation time of estrus from hormone treatment by assessments of cytological observation and blood plasma progesterone concentration. A total of 54 bitches on anestrus were selected for the study and divided randomly into 8 treatment groups as follow. Control, natural estrus; FSH (L), FSH (1.5 mg/kg, twice a day, $Falltrophin^{(R)}$, Vetrepharm); FSH (H), FSH (3.0 mg/kg, twice a day); GnRH+FSH, GnRH (5 ug/kg, once first day, $GNADON^{(R)}$, Dongbang)+FSH (3 mg/kg, SID); PMSG, PMSG (50 IU/kg, every third day $FOLLIGON^{(R)}$, Intevet); GnRH+PMSG, GnRH (5 ug/kg, only first day)+PMSG (50 IU/kg, every third day); GnRH, GnRH (5 ug/kg, only first day); Bromocriptine, bromocriptine (0.3 mg/kg, SID, $Parlodel^{(R)}$, Novartis). The bitches were evaluated clinical sign, cytological exam and $OVUCHECK^{(R)}$ Premate for assessment of estrus induction. Estrus induction rates were significantly (P<0.05) higher in GnRH+PMSG (100%) compared to others. PMSG and GnRH+PMGS (87.5 and 100%) and Bromocriptine (77.8%) were higher than others except GnRH+PMSG. Analysis of vaginal smear has proved to be effective a correct assessment of estrus induction with assay of progesterone concentration by $OVUCHECK^{(R)}$ Premate. Proestrus initiated by the $6^{th}$ after induction in most case. In conclusion, bromocriptine is an effective drug for estrus induction in bitches and assay of progesterone concentration by $OVUCHECK^{(R)}$ Premate with examination of vaginal smear that should be useful to detection of estrus induction of estrus induced bitches.

  • The present study was carried out to investigate morphologic changes in the corpus luteum of the pregnant rat by electron microscope after administration of prostaglandin F2$\alpha$(PGF2$\alpha$). Pregnant rates were treated with PGF2$\alpha$(1,500$\mu\textrm{g}$/rat) and their corpura lutea were observed morphologically. The results obtained in this study were summarized as follows ; 1. The weight of the ovaries and corpura lutea were decreased slightly at 8~24 hours after PGF2$\alpha$ administratin but no significant differences were observed. 2. The number of corpora lutea and luteal cells decreased slightly at 12~48 hours and 18~24 hours after PGF2$\alpha$ tretment but there were no signifciant differences between control and treatment. 3. The weight of uterus and the unmber of embryo decreased slightly at 96 hours and at 18~96 hours after PGF2$\alpha$ administration but no significant differences were obtained. 4. In the electron microscopic observatons, lipid droplets which are electron dense and appear in the cytoplasm moderately increased in number after PGF2$\alpha$ treatment. The lipid droplets were surrounded by mitochodria and appeared in the autophagic vacuoles. 5. Moderated and high electron dense mitochondria which are round or elongated in shape showed pleomorphism from 3 hours after PGF2$\alpha$ treatment. Destruction of tubular of vesicular cristae was observed at 6 hours after the treatment. Dense body and myelin figures in matrix of mitochondria were also appeared. 6. Well-developed smooth endoplasmic reticulum(sER) showed tubular or vesicular cisternae. A number of whorl membranes containing ribosomes, mitochondria and lipid droplets were observed at 1.5 hour after treatment. sER was abundant in luteal cells at 12 hours were treatment. 7. Well-developed Golgi pparatus appeared obviously 6 hours and more prominently at 12 hours. Those Golgi vesicles were remarkably dilated. 8. Generally, a few rough endoplasmic reticulum (rER) were appeared after treatment and cisternae showed slight dilatation. No differences among the treatments were observed. However, slight dilation of cisternae was observed at 1.5 hours after treatment. 9. Ribosomes composed of free and polyribosomes were abundant before treatment but polyribosomes were appeared at 12 to 24 hours after treatment. 10. Intercellular space were slightly extended at 3 hours and markedly extended at 12 hours. Numerous microvillous protrusions were observed at these times. Membranous multivesicular structures and autophagic vacuoles were also appeared in the intercellular space. 11. At 3 hours after the treatment, autophagic vacuoles appeared in the cytoplasm of the cell. They increased in number with time and were observed to transfer to the intercellular space. Lysosomal dense body appeared in the cytoplasm and the inclusion body was also observed in nucleus at 12 to 24 hours after treatment.

Non-Genomic Actions of Progesterone : Focussed on the Signaling Pathways in the Mammalian Ovary (프로게스테론의 비유전자 수준 작용 : 포유류 난소에서의 신호 전달 경로를 중심으로)

  • Progesterone(P4) is an important intermediate in the synthesis of androgens and estrogens. Furthermore, P4 itself plays a crucial role in ovulation, atresia and luteinization, and is essential for the continuation of early pregnancy in all mammalian species. In spite of the hormone's physiological importance, the exact action mechanism(s) of P4 in mammalian ovary has not been fully understood yet. In this context, a decades-long controversy regarding the identity of receptors that mediate non-genomic, transcription-independent cellular responses to P4 is presently attracting huge scientific interests. P4 may exert its action in mammalian ovary by several ways: 1) the well-documented genomic pathway, involving hormone binding to so-called classic cytosolic receptor(PGR) and subsequent modulation of gene expression by the ligand-receptor complex as transcription factor. 2) pathways are operating that do not act on the genome, therefore refered to as non-genomic actions. The prominent characteristics of the non-genomic P4 actions are: (i) rapid, (ii) insensitive to transcription inhibitors, (iii) transduced by membrane associated molecules. In particular, the non-genomic P4 actions could be mediated by: (a) classic genomic P4 receptor(PGR) that localizes at or near the plasma membrane, (b) a family of membrane progestin receptors(MPR $\alpha$, MPR $\beta$ and MPR $\gamma$), (c) progesterone receptor membrane component I(PGRMC1), and (d) a membrane complex composed of serpine I mRNA binding protein(SERBP1). The present review summarized these rapid signaling pathways of P4 in the mammalian ovary.

  • The syndrome of inappropriate secretion of the antidiuretic hormone (SIADH) is a well recognized paraneoplastic phenomenon related to impaired water excretion, and can result in dilutional hyponatremia as well as central nervous system symptoms. It is characterized by a decrease in plasma osmolarity with inappropriately concentrated urine. The causes of SIADH are associated with pulmonary and endocrine disorders, central nervous system diseases, and malignancies, including lung cancer. The other causes of SIADH include some drugs, particularly chemotherapy agents. Anticancer drugs, such as cisplatin, vincristine, and cyclophosphamide are well known causes of SIADH but the mechanisms are unclear. Recently, we encountered a patient with advanced non-small cell lung cancer who suffered from general weakness and altered mentality after an intravenous carboplatin and gemcitabine combination.

Ultrastructures of Oocyte Development and Electrophoretic Patterns of the Yolk Protein Following HCG Treatment in Korean Native Catfish (Silurus asotus)

  • During the rapid phase of gonadal development of the freshwater teleost, the catfish (Silurus asotus), the influence of hCG upon the inducement of final oocyte maturation and spawning was investigated electrophoretically and ultrastructurally. The electrophoretic patterns obtained were different in the presence and absence of some of the major or minor zones, because of the hormone level in catfish. The vitellogenin of hormone-treated fish was stained more intensively than that of sham-treated fish. These proteins showed some minor or main bands of egg extracts which migrated at positions corresponding to molecular weights of approximately 90,000. However, the thickness of electrophoretic band in molecular weight for hCG-treated fish was slightly lower than that for saline control. It seemed the plasma protein with molecular weight of approximately 45,000 in hCG-treated fish disappeared. In contrast to the control fish, the ovaries in the catfish treated with hCG shows a marked ultrastructural change under the electron microscope. No dilated profiles were seen in the granulosa cells of the mature oocyte before ovulation. After germinal vesicle breakdown (GVBD), the zona radiata interna (ZRI) becomes more compact, and there is a loss of all the processes from the pore canals. There is a wide space between the vitelline membrane and zona radiata. Also, during final maturation, the microvillar processes from the oocyte are seen no longer to penetrate deeply into the extracellular spaces of the overlying granulosa cells, and the reticulate patterns of the zona radiata interna becomes occluded, giving the zona radiata a more solid appearance. It has been possible to initiate 100% oocyte maturation in yolk granules and follicles in vivo by treatment with hCG and a high water temperature ($27^{\circ}C$). In hCG-treated fish, the percentages of successful artificial fertilization and hatching were maximal at 15 h after a single injection. It seems clear that a long acting preparation containing hCG can be successfully used in prespawning fish to advance the final events of gonadal maturation and initiate spawning. Further studies are necessary to evaluate the potential of hCG to either stimulate or inhibit the reproductive development of fish at other stages of the seasonal reproductive cycle.

Gonadal Development and Sex Steroid Hormone Levels of the Yellowfin Goby Acahthogobius flavimanus (문절망둑(Acanthogobius flavimanus)의 생식소 발달과 혈중 성 스테로이드호르몬 농도 변화)

  • Changes of sex steroid hormones in the plasma of yellowfin goby, Acanthogobius flavimanus were investigated in relation to the gonadosomatic index (GSI), the hepatosomatic index (HSI) and gonadal development. The GSI in females rose rapidly in November and remained high from December to May $(7.26\pm0.89­6.62\pm0.02)$. The Male's GSI also increased gradually from November and was highest in May $(0.16\pm0.08)$. The HSI in both sexes was in reverse correlation with the GSI, and the HSI was low during the spawning season (February-May). In females, the $estradiol-17{\beta}\;(E_2)$ level increased during vitellogenesis (November and December) and reached its maximum $(8.13\pm2.87 ng/mL)$ at the maturing period, in January. $17{\alpha},\;20{\beta}$-dihydroxy-4-pregnen-3-one$(17{\alpha}20{\beta}OHP)$ gradually increased from October $(0.063{\pm}0.02ng/mL)$ to March $(0.16{\pm}0.02ng/mL)$ and increased rapidly in May. The level of testosterone (T) showed a similar tendency of $E_2$. In males, T increased gradually during spermatogenesis from September to December $(0.14{\pm}0.06­0.26{\pm}0.10ng/mL)$ and peaked in January $(0.36{\pm}0.29 ng/mL)$ when the spermatozoa filled the testis. 11-KT also rose markedly in January and then decreased. On the other hand, $17{\alpha}29{\beta}OHP$ in males did not show any clear tendencies.

Responses in Hepatic Xenobiotic Metabolizing Enzymes and Sex Hormones of Yellowfin Goby Acanthogobius flavimanus in Nakdong Estuary (낙동강 하구에서 채집한 문절망둑 Acanthogobius flavimanus의 간장 약물대사효소계와 성호르몬 농도)

  • To assess effects of contaminants on fish in Nakdong river, feral yellowfin goby Acanthogobius flavimanus were caugt in two different sites and its hepatic monooxygenase enzyme, including cytochrome P450 (CYP), NADPH-cytochrome P450 reductase (P450R), NADH-cytochrome b5 reductase (b5R), ethoxyresorufin deethylase (EROD), glutathione S-transferase (GST) were quantitatively determined. Gonadosomatic index (GSI), hepatosomatic index (HSI) and three sex steroid hormone (17$\beta$-estradiol, E2; testosterone, T; 11-ketotestosterone, 11-KT) levels of the fish were also investigated. HSI of fish from polluted site (site 1) were significantly higher than that of unpolluted site (site 2), but GSI levels were significantly lower in polluted site. No significant differences in plasma 11-KT and T levels were observed in two sites surveyed. E2 level was, however, significantly (p<0.05) higher in female fish from site 1 than site 2. In addition, hepatic EROD activity and CYP level of site 1 fish were lower than those of site 2 fish, whereas relatively high levels of P450R, b5R and GST activities were found in site 1. The results imply that yellowfin goby, especially female fish in Nakdong river estuary are affected from contaminants disrupting sex steroid hormone system.
