This study was designed to investigate the suitability of the pubertal assay and the enhanced TG 407 as methods for detection of endocrine-mediated effects, especially thyroid function. Male and female Sprague-Dawley rats were gavaged daily with 0, 12.5, 25, 50 mg/kg alachlor in corn oil during 30 days. The effects of alachlor on thyroid gland, the genital organs and thyroid hormone were measured in male and female rats. Dose of alachlor 25, 50 mg/kg/day increased relative weight of testis and thyroid gland in exposed male rats and decreased relative weight of vagina in exposed female rats. Relative weight of thyroid gland was decreased in alachlor 25 mg/kg/day exposed female rats. Dose of alachlor 25, 50 mg/kg/day decreased plasma T4 and testosterone in female rats. Another purpose of this study was to investigate the effects of endocrine disruptors as like thyroid hormone in vitro. Luciferase activity was measured to dectect reaction of test chemicals and thyroid hormone response elements in HeLaTRE cell. Dose of alachlor 1 nM-1000 nM increased 100-134% luciferase activity compared with control.
The objective of this study was to examine the effect of plasma leptin and insulin concentrations on fat traits in Hanwoo. If a biological indicator such as plasma leptin and insulin was identified, it would be a useful biological marker that can be predicted marbling score in young animal. The relationship between plasma hormone (leptin and insulin) and fat traits (marbling score, back fat thickness and P8 fat thickness) was investigated. The experiment studies 100 Hanwoo that were randomly sampled from Hanwoo Experimental Station Herd. The concentration of plasma insulin was significantly associated with marbling score (P=0.02) but was not significantly with back fat thickness (P=0.07) and P8 fat thickness (P=0.09). Statistical model determinant that plasma insulin concentration account for phenotypes was moderate on marbling score (5%), back fat thickness (3%) and P8 fat thickness (9%). On the other hand, plasma leptin concentration was significantly associated with marbling score (P=0.03) and back fat thickness (P=0.02), but was not significant on P8 fat thickness (0.07). Statistical model determinant that plasma leptin concentration accounting for phenotypes was moderate effect on marbling score (3%) and back fat thickness (2%), but it has a slightly bigger effect on P8 fat thickness (7%). In conclusion, the plasma leptin and insulin seems to have an effect on fat traits (marbling score, backfat thickness and P8 fat thickness) in Hanwoo.

Aging process can be explained by many factors. In this study, we counted complete Blood Cells (CBC) such as WBC, Lymphocytes, Monocytes, Granulocytes, RBC, HGB, and HCT of both SAM P6 and SAM P1 during the aging process. Plasma albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea concentrations were also measured at the same time. In addition to these, plasma concentrations of cortisol, total T3, and total T4 were analyzed by Chemiluminescent Immunoassay. There were no change in CBC counts of SAM R1 and SAM P6 during this study. Plasma concentratins of albumin and glucose decreased significantly in SAM R1. However, plasma alkaline phosphatases and creatinine concentration in SAM P6 decreased significantly at 16 week after birth comparing to the control. Total T4 levels were siginificantly increased although cortisol and total T3 concentrations were the same in SAM R1 groups. Especially, the after birth of creatinine, alkaline phosphatase, T4 of SAM P6 at 16 week were significantly different form those of SAM R1. At 12 week after birth, pilose antler extract was given 5g/kg/day p.o. for 0, 7, 14, 21, and 30 days each in both SAM R1 and SAM P6. The RBC, HGB, and HCT levels started to increase significantly from 7 days after the dose at SAM P6 only. Total T4 conectrations were elevated gradually during the study although the antler extract administration did not prevent or inhibit the increase in total T4 concentration during the study. Therefore, the elevation of erythrocytes after administration of the extract needs to be studied in future.
The study was conducted on six multiparous Murrah buffaloes which were earlier artificially induced into lactation. During the experimental period of 15 days, buffaloes were managed in a loose housing system. All the buffaloes were administered a single injection of bromocryptine (@ $100{\mu}g/kg$ body weight) subcutaneously in the neck region at 08:30 A.M., 50 days postpartum (early lactation). Blood samples were collected from four buffaloes for a period of 5 days before the administration of bromocryptine i.e. on days -5, -4, -3, -2, -1, on day of treatment (day 0) and thereafter daily for a period of 9 days i.e 1, 2, 3, 4, 5, 6, 7, 8 and 9 to determine the hormones and blood metabolites. Homogeneous milk samples from all the buffaloes were collected at morning and evening milkings on days coinciding with the days of blood sampling for analysis of milk constituents. Administration of bromocryptine resulted in a significant inhibition of plasma prolactin within 24 hrs of treatment, but the response in all the buffaloes was not uniform. The effect of bromocryptine on plasma prolactin hormone lasted for 1-4 days but Cortisol concentration were not altered. Administration of bromocryptine neither affected blood glucose nor plasma non-esterified fatty acids concentration. Irrespective of level of milk production from different buffaloes, there was no effect of bromocryptine on milk yield which indicated that prolactin is not required for milk secretion during early lactation in buffaloes. Milk constituents like fat, protein and lactose were not affected by bromocryptine may be due to no effect of bromocryptine of milk yield.
An experiment was conducted to evaluate the physiological changes of laying hens subjected to feed removal during induced molting while received probiotics in the drinking water. Post-molt performance and egg quality criteria were also studied. Ninety 78-week-old Hy-line W36 laying hens were divided into two treatment groups according to equal body weight and subjected to induced molting by continuous feed removal until around 30% BW reduction. The experiment lasted 12 wks consisting of 4-wk molting and 8-wk post-molt periods. Treatment 1 received no probiotics and was considered as the control. Treatment 2 was similar to the control except that hens received probiotics in the drinking water at 400 mg/L during feed deprivation. The results indicated that hens in both groups went out of production by Day 5. However, hens received probiotics reached 5 and 50% egg production sooner than the control (30 and 52 days vs. 31 and 54 days). Starvation during molting increased heterophil to lymphocyte (H/L) ratio, hematocrit and plasma T4 and $Na^+$ levels while plasma T3 and Cl- levels were decreased. Probiotics had no significant impact on BW reduction during molt. Post-molt egg production and egg mass were higher in hens which previously received probiotics, but these responses were not significant. However, feed conversion ratio was significantly better in hens which received probiotics. Hematocrit, plasma thyroid hormone concentrations (T3 and T4) and plasma $Na^+$, $K^+$ and Cl- levels during molting were not significantly influenced by supplementation of probiotics. However, H/L ratio showed a significant (p<0.05) reduction in birds which received probiotics suggesting beneficial effects of this product for feed-deprived laying hens. No significant difference was observed in post-molt egg quality criteria.
The efficiency of artificial insemination (AI) for horses remains unsatisfactory. It is mainly because each process of AI causes a detrimental effect on semen quality. To sustain quality of semen properly, several factors including libido of stallions and sperm damage during sperm processing and preservation should be considered. Stallions with decent libido produce a high ratio of sperm to seminal plasma in their ejaculates, which is the ideal semen composition for maintaining sperm quality. Thus, to maximize the fertility rate upon AI, stallions should be appropriately managed to enhance their libido. Seminal plasma should have a positive effect on horse fertility in the case of natural breeding, whereas the effects of seminal plasma on both sperm viability and quality in the context of AI remain controversial. Centrifugation of semen is performed during semen processing to remove seminal plasma and to isolate fine quality sperm from semen. However, the centrifugation process can also result in sperm loss and damage. To solve this problem, several different centrifugation techniques such as Cushion Fluid along with dual and single Androcoll-E$^{TM}$ were developed to minimize loss of sperm and to damage at the bottom of the pellet. Most recently, a new technique without centrifugation was developed with the purpose of separating sperm from semen. AI techniques have been advanced to deliver sperm to optimal region of female reproductive tract at perfect timing. Recombinant equine luteinizing hormone (reLH) and low dose insemination techniques have been developed to maximize both fertility rate and the efficiency of AI. Horse breeders should consider that the entire AI procedure should be optimized for each stallion due to variation in individual horses for a uniformed AI protocol.
Eighteen crossbred goats were selected from the Institute's goat herd to determine the changes in hormones, blood metabolites and yield and composition of milk during lactation. The blood and milk samples were collected from each goat in a heparinized vacutainer tubes at fortnightly interval for a period of 150 days. In milk samples, fat, protein and lactose contents were estimated while in blood plasma hormones viz., prolactin, GH, cortisol, insulin, $T_4$ and $T_3$ were measured using radioimmunoassay methods. The plasma concentration of prolactin, GH and cortisol were high during early lactation when the goats acquired peak milk yield. During remainder of lactation their concentration varied. The high NEFA concentration during early lactation indicated mobilization of body reserves as the body weights also decrease during early lactation. However, with the advancement of lactation, the body weights of the goats and the concentration of NEFA declined which indicated utilization of NEFA for energy yielding purposes in addition to fatty acid synthesis. The ambient temperatures did not influence plasma concentration of prolactin, GH, insulin, $T_3$ and $T_4$ during the lactation cycle. The fat content of milk varied significantly (p<0.01) but protein and lactose content of milk remains unchanged during different stages of lactation. Growth hormone was positively correlated with insulin (p<0.05) during lactation while prolactin had a positive correlation with lactose and plasma NEFA (p<0.01) and negative correlation with $T_3$ (p<0.05).
The effects of electroacupuncture(EA) on blood concentration of endocrine substances were investigated in 6 horses. Three acupuncture points ; Guan Yuan Shu(BL-26), Wei Shu(BL-21) and Da Chang Shu(BL-25) were stimulated for 20 minutes by EA at separate occasions under varying condition ; 2V-1Hz, 2V-5Hz, 2V-30Hz, 4V-1Hz, 4V-5Hz and 4V-30Hz. Plasma levels of adrenocorticotropic hormone(ACTH), ${\beta}$-endorphin, epinephrine, norepinephrine and serum levels of gastrin were analysed. Blood samplings were carried out before, 0, 20 and 40 minutes after the EA stimulation. The serum gastrin levels were increased by 2V-5Hz stimulation on the Wei Shu. Plasma ACTH levels were decreased by 2V-1Hz stimulation on the Wei Shu, but largely increased by 4V-30Hz stimulation on the Guan Yuan Shu. Plasma ${\beta}$-endorphin levels were slightly increased or decreased by 2V-1Hz stimulation, but largely increased by 4V-30Hz stimulation on the Guan Yuan Shu. Plasma levels of epinephrine and norepinephrine were not so much changed by 2V-1Hz or 5Hz stimulation, but tended to increase by 4V-30Hz stimulation on Guan Yuan Shu. These results suggest that the low voltage-low frequence EA stimulation increased blood concentration of gastrin, but decreased ACTH, ${\beta}$-endorphin, epinephrine and norepinephrine, whereas high voltage-high frequence EA stimulation induced opposite results. Accordingly, there appears to be a close relationship between the changes of gastrointestinal motility and the changes of blood concentration of endocrine substances by EA stimulation.
Ghrelin is a novel growth-hormone-releasing acylated peptide, which has been purified and identified in rat stomach. In the present study, the full-length sequence of bovine ghrelin cDNA was cloned by RT-PCR. The bovine ghrelin cDNA sequence derived in the present study included a 348 bp open reading frame and a 137 bp 3'UTR. The putative amino acid sequence of bovine prepro-ghrelin consisted of 116 amino acids, which contained the 27-amino acid ghrelin. The sequence analysis of the bovine ghrelin gene revealed that an intron existed between Gln$^{13}$ and Arg$^{14}$ of ghrelin. This exon-intron boundary matched the GT-AG rule of the splicing mechanism. Compared with rats, which have two tandem CAG sequences in the 3'end of intron, bovine ghrelin genome has only one CAG sequence. Therefore, although rats can produce 28 amino acid-ghrelin and 27 amino acid-des-Gln$^{14}$-ghrelin by alternative splicing, ruminant species, including bovines, might be able to produce only one type of ghrelin peptide, des-Gln$^{14}$-ghrelin. The influence of aging on plasma ghrelin concentration was also examined. Plasma ghrelin concentration increased after birth to approximately 600 days of age, and then remained constant.

Postmenopausal women or ovariectomized rats are associated with increased cholesterol levels, which are risk factors of metabolic syndrome and cardiovascular diseases. Increased prevalence of metabolic syndrome after menopause might be associated with estradiol deficiency. Harmful effect of estradiol hampers the casual usage of hormone to prevent the metabolic syndrome. Soy protein has been reported to show several beneficial effects on health, however it is unclear which components of soy protein is responsible for anti-obesity and hypocholesterolemic effects. Soy isoflavones, gem-stein and daizein, are suggested to have anti-obesity and hypocholesterolemic effects but with inconsistency. The present study investigated the effect of supplementation of genistein (experiment I) and soy protein containing isoflavones (experiment II) to high fat diet on body weight gain, food intake, liver and fat tissue weight and the lipid levels in ovariectomized rats. Plasma and hepatic lipid contents and the mRNA levels of genes encoding lipid metabolism related proteins, such as CPT1 and HMGR were measured. Ovariectomy increased body weight, fat tissue weight and plasma and hepatic lipid levels which increase the risk of metabolic syndrome. Soy protein could improve plasma and hepatic lipids levels. Soy protein also increased hepatic CPT1 and HMGR mRNA levels. Plasma and hepatic lipids levels could not be decreased by dietary genistein alone. In contrast, lipids levels could be decreased by isoflavone-fortified soy protein, suggesting that the ingestion of soy protein enriched with isoflavone gives more benefit for protecting postmenopausal women from metabolic syndrome.
