Distribution of Cd and Pb Accumulated in Medicinal Plant Roots and Their Cultivation Soils

  • In general, plant roots accumulate more heavy metals than the above ground organs such as leaf, stem, and fruit. This implies that root medicinal plants would be an issue with excessive heavy metal accumulation. Therefore, the current study was carried out to investigate the distribution of heavy metal (focused on Cd and Pb) concentrations in soils and medicinal plant roots grown in different region of Korea. Total 293 samples for each soil and plant were collected along the national wide. Soil pH, total and phytoavailable metal concentrations (1 M $NH_4NO_3$ extracted) in soils were determined and heavy metal concentrations in root of the medicinal plants were analyzed. Heavy metal concentrations of the soil samples studied were not exceeded standard limits legislated in 'Soil Environmental Conservation Act', except 2 samples for Cu. However, substantial amount of Cd was accumulated in medicinal plant roots with 29% samples exceeding the standard limit legislated in 'Pharmaceutical Affairs Act' while all plant samples were lower than the standard limit value for Pb. Also the current study demonstrated that cadmium concentrations in the roots were governed by the phytoavailable Cd in soils, which decreased as soil pH increased. From this result, application of heavy metal immobilization technique using a pH change-induced immobilizing agents can be suggested for safer root medicinal plant production.

Antagonistic Potential of Fluorescent Pseudomonads and Control of Crown and Root Rot of Cucumber Caused by Phythophtora drechsleri

  • In this study, 200 isolates of fluorescent pseudomonads were isolated from different fields of East and West Azarbaijan and Ardebil provinces of Iran. These bacterial isolates were screened on the basis of a dual culture assay, the presence of known antibiotic genes, and their ability to successfully colonize roots and to promote plant growth. Twelve isolates exhibited 30% or more inhibition of mycelia growth of $P.$ $drechsleri$. Genes encoding production of the antibiotics 2,4-diacetylphloroglucinol, phenazine-1-carboxylic acid, and pyoluteorin were detected in some strains but none of the strains possessed the coding gene for production of antibiotic pyrrolnitrin. In an $in$ $vitro$ test for root colonization, the population density on roots of plants treated with most of the above strains was more than 6 $\log_{10}$ CFU $g^{-1}$ roots, with a maximum of 7.99 $\log_{10}$ CFU $g^{-1}$ roots for strain 58A. Most of the strains promoted significant plant growth in comparison to non-treated controls. In green house studies, the percentage of healthy plants in pots treated with strains 58A and 8B was 90.8% and 88.7%, respectively. The difference between these treatments and treatment with the fungicide metalaxyl was not significant.

Effects of Pseudomonas aureofaciens 63-28 on Defense Responses in Soybean Plants Infected by Rhizoctonia solani

  • The objective of this work was to investigate the ability of the plant growth-promoting rhizobacterium Pseudomonas aureofaciens 63-28 to induce plant defense systems, including defense-related enzyme levels and expression of defense-related isoenzymes, and isoflavone production, leading to improved resistance to the phytopathogen Rhizoctonia solani AG-4 in soybean seedlings. Seven-day-old soybean seedlings were inoculated with P. aureofaciens 63-28, R. solani AG-4, or P. aureofaciens 63-28 plus R. solani AG-4 (P+R), or not inoculated (control). After 7 days of incubation, roots treated with R. solani AG-4 had obvious damping-off symptoms, but P+R-treated soybean plants had less disease development, indicating suppression of R. solani AG-4 in soybean seedlings. Superoxide dismutase (SOD) and catalase (CAT) activities of R. solani AG-4-treated roots increased by 24.6% and 54.0%, respectively, compared with control roots. Ascorbate peroxidase (APX) and phenylalanine ammonia lyase (PAL) activities of R. solani AG-4-treated roots were increased by 75.1% and 23.6%, respectively. Polyphenol oxidase (PPO) activity in soybean roots challenged with P. aureofaciens 63-28 and P+R increased by 25.0% and 11.6%, respectively. Mn-SOD (S1 band on gel) and Fe-SOD (S2) were strongly induced in P+R-treated roots, whereas one CAT (C1) and one APX (A3) were strongly induced in R. solani AG-4- treated roots. The total isoflavone concentration in P+Rtreated shoots was 27.2% greater than the control treatment. The isoflavone yield of R. solani AG-4-treated shoots was 60.9% less than the control.

  • 최근 인공지반녹화가 주변 생활공간의 쾌적화, 에너지 절약효과, 도시 미관의 환경개선 등의 장점을 갖고 있어 콘크리트 위에 녹화 공간 조성이 증가하고 있다. 이에 따라 건축공사 표준 시방서, 건축물 녹화 설계기준 등에서는 방수층의 수밀안정성 확보를 위해 일정 품질 이상의 방근층 설치를 규정하고 있으나, 실제 우리나라의 많은 현장은 방수층 위에 누름콘크리트 타설 후 인공녹화를 시행하고 있는 실정이다. 이에 본 연구에서는 방근층을 시공하지 않은 현장을 대상으로 지표를 설정하였으며, 누름콘크리트의 균열에 따른 식물의 뿌리가 균열부로 침입 및 관통하여 누름콘크리트 하단부의 아스팔트 방수시트에 미치는 영향을 평가하였다.

Triterpenoid production and phenotypic changes in hairy roots of Codonopsis lanceolata and the plants regenerated from them

  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of triterpenoid saponin with high medicinal values. In this work, we induced transgenic hairy roots of C. lanceolata and analyzed triterpenoid saponins from the hairy roots and hairy root-derived transgenic plants. Hairy roots were obtained from leaf explants by the transformation of Agrobacterium rhizogenes R1000. Transgenic hairy root lines were confirmed by the transcriptional activities of rolA, B, C, and D genes by RT-PCR. Transgenic root lines actively proliferated on hormone-free medium but not in nontransformed roots. Hairy roots contained richer triterpenoids (lancemaside A, foetidissimoside A, and aster saponin Hb) than nontransformed roots. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. They showed phenotypic alterations such as shortened shoots and an increased number of axillary buds and adventitious roots. The transgenic plants also contained higher triterpenoid levels than wild-type plants. These results suggest that hairy roots and transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

A Reliable "Direct from Field" PCR Method for Identification of Mycorrhizal Fungi from Associated Roots

  • A very reliable and specific method for the identification of fungi in ectotrophic mycorrhizal symbiosis was developed using a specific PCR assay based on the amplification of the ITS1 region. To obtain specific data, an ITS-diagnostic assay was carried out that reveals genera and species specific sequences. Here, an application of one method is presented, which covers the identification of pure mycelia, basidiocarps as well as mixed samples such as ectomycorrhizal roots that were mingled with remains of the host plant. For this purpose a protocol was established that allowed the extraction of DNA from single mycorrhizal roots. In order to perform a specific ITS analysis we generated a new ITS-primer(ITS8) by a multiple alignment of five different genera and species of mycorrhizal fungi. The utilization of ITS1 and ITS8 resulted in specific PCR amplicons, which were characterized by sequencing without purification steps, even when the template DNA was associated with roots.

해녀콩(Canavalia lineata)의 잎, 뿌리 및 도관액에서 Canavanine의 함량분석 (The Analysis of Canavanine Content in Leaves, Roots, and Xylem Exudate of Canavalia lineata)

  • The content of canavanine was measured and analyzed in leaves, roots and xylem exudate of Canavalia lneata. In non-nodulated plants, the cotyledons were removed after a week of sowing and the plants were grown for 3 weeks. The quantity of canavanine measured by canavanine specific-PCAF colorimetric assay was 9-10 $\mu$mol/g fresh wt. in leaves, 5-6 $\mu$mol/g fresh wt. in roots, and 0.3-0.5 $\mu$mol/ml in xylem exudate. When free amino acids of leaves, roots, and xylem exudate were analysed by HPLC, the relative proportion of asparagine plus glycine was the highest and canavanine was high secondarily. And the relative proportion of canavanine among total free amino acids was 30-35% in leaves and roots, and 12-13% in xylem exudate. In non-nodulated plants grown for 8 weeks, the canavanine content of each part was similar to that of 3-week-old plants. By the formation of nodules, the canavanine content of leaves, roots, xylem exudate, and nodules decreased apparently. In xylem exduate, the nitrogenous compounds were also analyzed. The relative contents of NO3-, free amino acids, and ureides(allantoin and allantoic acid) were 60-80%, 20-30%, and 5%, respectively. From these results, it can be assumed that canavanine is synthesized in the root of plant and nodulation affects the canavanine content. It is obvious that canavanine is considered one of the reduced-N forms transported via xylem.

  • The rice variety, Kwanok, was reared in the water, land and salt seed beds and transplanted to the reclaimed soil area having an average of 0.48% salt content (0.67% at the end of April). The plant height of land bel seedlings at transplanting stage was short but the dry-weight/plant-height ratio was large and the rooting ability was vigorous remarkably after transplantation in the salty area. The central cylinder, vessels, sclerenchyma, endodermis and other mechanical tissues of the root of land bed seedlings were well developed while the size of cortical cell layers were small. The cytoplasm of the cortical parenchyma at the root tips seemed to be most abundant in the land bed seedlings. The formation of the aerial cavity in the cortex of primary root was rapid and it seemed that the developmental mechanism of the aerial cavity in the rice plant roots was related to the development of the lateral roots.

식물의 뿌리와 근권으로부터 Salicylic acid를 생성하는 근권세균의 검색 (Screening of Salicylic acid Producing Rhizobacteria Isolated from Plant Roots and Rhizosphere)

  • Twenty two rhizobacteria were isolated from the roots and rhizosphere of radish, carnation, potato and tomato. There isolates produced a fluorescent pigment in King's B medium and identified as Pseudomonas spp. These isolates colonized roots and rhizosphere of the host plants. In the study of cultural characteristics of the bacteria, the pH of the culture broth was changed from neutral (7.0) to alkali (8.8∼9.41) and the numbers of cells were increased from 106 to 108 after 40 hr of incubation in basal standard succinate medium. The salicylic acid production identified by pink color reaction were observed in 7 bacteria. Out of these 7 salicylic acid producing bacteria, only 2 strains of bacteria such as Pseudomonas fluorescens RS006, and Pseudomonas sp. EN401 were confirmed as salicylic acid producers by optical density measurement. Therefore, for screening of salicylic acid producing bacteria from the roots and rhizosphere, color reaction of the culture medium should be done in the first step, and then optical density measurement of culture extract should be made for the confirmation of salicylic acid production.

Community Structure of Arbuscular Mycorrhizal Fungi in Upo Wetland, Korea

  • Arbuscular mycorrhizal fungi (AMF) are one of the most widespread symbionts globally. Owing to their enhanced nutrient absorption capacity, AMF significantly contribute to the survival of individual plants and the ecosystem functioning. Community structures of AMF are affected by many environmental factors Inland wetlands have a different environment from common forest soils, therefore, plants inhabiting wetlands may have characteristic AMF communities. The purpose of this study was to compare the AMF communities in wetlands, among the species of host plants. We sampled the roots of 3 host plant species, Phragmites communis, Miscanthus sacchariflorus, and Trisetum bifidum with rhizospheres from 3 isolated areas in Upo wetland, Korea. We extracted DNA from AMF spores in rhizospheres and the roots of 3 plant species. We amplified 18S rDNA of AMF using AMF specific primer. As a result, we confirmed 9 species from 5 genera in AMF spores, and 5 species from 3 genera in plant roots. Funneliformis caledonium was the most dominant species in field soils, on the other hand, Diversispora aurantia was the most dominant species in plant roots. We confirmed that species diversity and abundance of AMF communities were different among host plant species. These results showed that the AMF community had specific to host plants in the inland wetland.
