Rapid Regeneration of Plants on N6 Medium from Orchardgrass (Dactylis Glomerate L.) Calli (N6 배지에서 오차드그래스 캘러스로부터 빠른 재분화)

  • We confirmed conditions for callus formation and plant regeneration of five varieties of orchardgrass (Dactylis glomerma L.). Among five varieties of orchardgrass, "Hapsung 19" expressed the highest rate for both of callus formation and plant regeneration. Otherwise, among SH (Schenk and Hildebrandt), MS (Murashige and Skoog) and N6 medium (Chu), MS and N6 medium were highest degree of efficiencies in callus formation and plant regeneration, respectively. In this study, we determined volume of hormones appended in media; $3\;mg/\;{\ell}$ ofdicamba for callus formation and $1\;mg/\;{\ell}$ of NAA (I-naphtalene acetic acid) and $5\;mg/\;{\ell}$ kinetin (6-furfurylaminopurine) for plant regeneration were appended in their media. We obtained orchardgrass plants from callus about 50~80 days after transferred to regeneration media.ation media.

  • Optimum culture conditions for high frequency plant regeneration from leaf explants of Kalanchoe daigremontiana Hamet &Perrier were established. Shoot regeneration was achieved from leaf explant cultures using MS medium supplemented with indole-3-acetic acid (IAA) and thidiazuron (TDZ) or benzyladenine (BA). Percent regeneration was influenced by plant growth regulators and source of explants. MS medium supplemented with TDZ (1.0 mg/l) and IAA (0.4 mg/l) was the most effective, providing shoot regeneration for 76.7 % of ex vitro leaf explants associated with a high number of shoots per explant (9.5 mean shoots per explant), whereas 100% shoot regeneration associated with 12.4 shoots per explant occurred from in vitro leaf explants on the same medium. Clusters of shoots were multiplied and elongated on MS medium containing several concentrations of BA. MS medium supplemented with 0.25 mg/l BA was proved as the most effective shoot elongation medium. Elongated shoots (2-3 cm) were rooted at 100% on half-strength MS medium. Rooted plantlets were then transferred to potting soil. Regenerated plants were established in the soil with 90% success.

  • This study was conducted to develop an effective production of callus induction and plant regeneration system for garlic transformation. The best callus production occurred on in vitro root segment initially cultured on MS medium with 1.0 mg/L 2,4-D and 0.2 mg/L IAA in both ‘Danyang' and ‘Euseong'. The frequency of callus formation were 81.2% ‘Danyang' and 76.1% ‘Euseong'. Eight weeks after callus induction, callus lines were transferred to regeneration medium during 7 weeks. The best shoot regeneration medium was MS supplemented with 5 mg/L Kinetin and 1 mg/L NAA for ‘Danyang' and MS supplemented with 10 mg/L BAP for ‘Euseong'. The frequency of shoot regeneration were 51.5% ‘Danyang' and 56.6% ‘Euseong' The plantlets were acclimatized and transferred to the greenhouse with almost survival. This in vitro regeneration system should be useful for garlic transformation.

  • New types of cytoplasmic male sterility in Brassica species would be very useful for the production of F$_1$, hybrid seeds. Leaves and stems of rapid cycling stock of B.juncea (CrGC4-3) containing Anand CMS were used as experimental materials for plant regeneration from protoplast culture. Very high plant regeneration rate (85%) was found in the Kao & Michayluk medium supplemented with 2 mg/L zeatin, 0.5 mg/L BAP, and 1 mg/L NAA when only leaf, not stem, segments were cultured. Protoplasts were isolated from leaves using mixtures of enzymes (1% Cellulycin, 0.5% Macerozyme) in 0.4 M mannitol and 50 mM $CaCl_2$.$2H_2$O. Mcrocalli induced from protoplasts were transferred to the shoot regeneration medium containing 2 mg/L BAP, 2 mg/L zeatin, and 0.5 mg/L NAA. After 60 days of initial protoplast culture, regenerated plantlets were obtained, acclimatized, transplanted into the pots, and grown up to the flowering stage.

  • Commercial cultivars and elite germplasms of barely (Hordeum vulgare L) are still recalcitrant to genetic transformation because of the lack of an efficient regeneration system. In this study, we established an efficient plant regeneration procedure from embryogenic calli derived from mature embryos. Callus induction from germinated mature embryos was best as over 95% in CIM medium (CI medium containing $2.5mg/{\ell}$ dicamba) under dark incubation. Development of embryogenic callus was highest as over 50% in CI3D medium (EC medium supplemented with $3mg/{\ell}$ 2,4-D). The highest regeneration of plants from embryogenic callus (40%) was obtained with CIS medium ($SI+1mg/{\ell}IAA\;and\;2mg/{\ell}\;BA$). These plant regeneration conditions could be useful in improving barley transformation efficiency.

  • This study was carried out to improve the ability of embryo formation and the efficiency of plant regeneration from suspension cultured cells of seed derived calli of orchardgrass (Dactylis glomerata L.). The frequency of formation of round cell and cell colonies was highest at 50 days after suspension culture in $N_6$ medium supplemented with $4\;g/{\ell}$ casein hydrolysate (CH), $20\;g/{\ell}$ sucrose and $30\;g/{\ell}$ sorbitol. The highest frequency of plant regeneration and somatic embryo formation was obtained from suspension cultured cells of 60 days. Addition of CH ($4\;g/{\ell}$) in suspension culture medium gave the highest frequency of embryo formation (39.6%) and plant regeneration (73.0%).

In this study, we determined volume of hormones and other compounds appended in media. For callus formation, only $5\;mg/\;{\ell}$ of 2,4-D (2,4-dichlorophenoxy acetic acid) was appended in their media. For plant regeneration, we used MS medium containing $1.0\;mg/\;{\ell}$ of BA and $0.1\;mg/\;{\ell}$ of NAA.

  • Apical meristems tissue were cultured on LS medium with different zeatin and NAA concentrations to compare their potential to regenerate shoots, roots and bulblet formation. Shoot regeneration from apical meristem was effective on LS medium added with zeatin 0.1, 0.5, 1.0 mg/L alone treatment or zeatin 0.5 mg/L and NAA 1.0 mg/L combination treatment. A high frequency of root regeneration was obtained on LS medium supplemented with zeatin 0.5 mg/ and NAA 1.0 combination treatment. Linsmaier and Skoog(LS) medium with NAA 3.0 mg/L and zeatin 1.0 mg/L combination treatment gave the best results for normal bulblet formation#KW=Allium wakegi ; plant regeneration ; bulblet formation

Optimal Medium Compositions for Plant Regeneration via Adventitious Shoot Formation Using 'Fuji' Apple Leaf Explants (사과 '후지'의 잎 절편체로부터 신초 기관형성을 통한 식물체 재생에 적합한 배지조성)

  • Plant regeneration protocols for adventitious shoot organogenesis from apple (Malus domestica 'Fuji') leaf explants were developed in the present study. The effects of different basal media, types and concentrations of carbon sources, and concentrations of plant growth regulators were evaluated to determine the optimal shoot regeneration conditions for 'Fuji' apple leaf explants. On different treatments involving combinations of basal media, LS and N6 media, and different types and concentrations of cytokinins, 6-benzyl-adenine (BA) and thidiazuron (TDZ), shoot regeneration rates were the highest in the N6 medium combined with BA. Among the plant growth regulator and carbon source combination treatments, 5.0 mg/L BA, and 0.1 mg/L α-naphthalene acetic acid (NAA) with 40 g/L sorbitol was the optimal combination for shoot regeneration. In addition, the optimal sorbitol concentrations for shoot regeneration were 40 g/L and 60 g/L. The highest regeneration (81.8%) was achieved using 40 g/L sorbitol. The regenerated shoots elongated and rooted on rooting medium, consisting of 1/4 MS medium with 0.2 mg/L indole-3-butyric acid (IBA). The plantlets were acclimatized and the regenerated plants exhibited normal phenotypes.

Shoot induction and regeneration using internodal transverse thin cell layer culture in Sesamum indicum L.

  • An efficient protocol for shoot regeneration was developed for sesame (Sesamum indicum L.) internodes using the transverse thin cell layer (tTCL) culture method. The frequency of shoot regeneration and the number of adventitious buds produced from regenerated shoots depend significantly on explant age, thickness of the tTCL sections, and the phytohormones supplemented to the culture medium. A combination of 6-benzyladenine (2.0 $mg\;l^{-1}$) and a-naphthaleneacetic acid (0.5 $mg\;l^{-1}$) was found to be the best phytohormone combination for shoot bud induction, with the maximum number of shoots obtained when the tTCL sections were 0.5-1.0 mm thick and derived from 4- to 6-week-old seedlings of sesame. Well-developed shoots were rooted on MS medium without phytohormones, and 80% of the regenerated plantlets were successfully established in soil.
