  • In an effort to optimize tissue culture responses of tall fescue, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. The MS basal medium was superior to $N_{5}$ 6/ and $B_{medium}$ in enhancing callus growth and plant regeneration. To determine the effect of carbon sources on plant regeneration, the seeds were cultured on medium with 30 g/L sucrose and maltose, respectively. Medium supplemented with 30 g/L sucrose resulted in regeneration of shoots from 50% of the calli. The genotypic difference in plant regenerability was obvious among five cultivars of tall fescue tested. \\`KY31\\` and \\`Hokuryo\\` showed to have higher regenerability with the frequency of 33% and 51%, respectively.

  • The effects of several additives on plant regeneration were investigated from leaf disc culture of S. tuberosum cv. Atlantic which is known as poor in regeneration ability. The presence of 2 g/L casein hydrolysate significantly enhanced shoot regeneration. Addition of 10~20 $\mu$M $AgNO_3$, not only increased the frequency of shoot regeneration but also maintained the leaf disc green presumably by the inhibitory action of ethylene accumulation in vitro. Decrease of sucrose levels to below 3% significantly increased the degree of regeneration. The addition of CuSO$_4$had no effect on shoot regeneration.

Efficient Callus Induction and Plant Regeneration from Immature and Mature Embryo Culture of Korean Wheat Genotypes

  • Immature and mature embryos of 18 Korean wheat genotypes were cultured in vitro to develop an efficient method of callus formation and plant regeneration, and to compare the responses of both embryo cultures. Immature and mature embryos were placed on a solid agar medium containing the MS salts and vitamins, 30g/l maltose, 2 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D), and amino acids. The developed calli were maintained on regeneration medium containing MS salts and B5 vitamins, 20 g/l sucrose, and the combination of two plant growth regulators, 6-benzylaminopurine (BAP) and indole-3-acetic acid (IAA). Immature embryos in most genotypes showed high efficiency of callus induction except three genotypes; Eunpamil, Chunggemil, and Namhaemil, and significant differences among the genotypes. Plant regeneration of calli induced from immature embryos showed high efficiency in Geurumil (56.5%), Tapdongmil (50.5%), Gobunmil (45.5%), and Urimil(42.2%). The analysis of variance showed significant differences for regeneration frequency among the genotypes. Mature embryos showed low callus induction frequency compared with that in immature embryos, and significant differences among the genotypes. Plant regeneration of calli induced from mature embryos showed high efficiency in Keumkangmil (33.33%), Tapdongmil(28.13%), and Geurumil (27.78%). The analysis of variance showed significant differences for plant regeneration frequency among the genotypes.

Influence of formulated organic Plant tissue culture medium in the shoot regeneration study of Brassica juncea (l.) - Indian mustard

  • Efficient protocol for plant shoot regeneration of Brassica juncea L. CZERN was established by using organic media components and growth stimulating factors of the vermicompost and coelomic fluids. Formulated organic plant tissue culture media (Vermicompost (30%) extracts supplemented with 20 mL/L coelomic fluid) have shown maximum shoot regeneration when compared with the Murashige and Skoog (MS) medium, which were supplemented with 1 mg/L 6-benzyladenine (BA) and 0.1 mg/L of Naphthaleneacetic acid (NAA). Cotyledon explants produced the highest shoot regeneration frequency from fourday-old germinated seedlings in comparison with non-germinated seedlings. The vermicompost extracts have proved to be the best organic plant growth media to induce shoots from cotyledons compared to the MS media. Statistically significant difference (P = 0.008) for the root length, shoot length (P=0.000350) and the leaves (P=0.375) of the mustard plantlets were analyzed successfully. The survival rate was 98% in the mustard cotyledons on the Vermicompost extract media and 63% on MS media respectively. The coelomic fluid also is much suitable to induce shoots from cotyledons at lower concentrations. It was also shown that the vermicompost extract, which comprised of humic acids along with coelomic fluid, affected shoot regeneration from the cotyledons. An efficient and organic shoot regeneration study was standardized and it can be applicable in the improvement of the economically important crops.

Partial Desiccation of Embryogenic Calli Improves Plant Regeneration in Sugarcane (Saccharum Spp.)

  • Partial desiccation of embryogenic calli cultures or somatic embryos leads to different physiological changes and maturation of somatic embryos, leading to improved plant regeneration. Embryogenic calli was induced from immature inflorescence segments and young leaf rolls of sugarcane (Saccharum officinarum hybrids CoC-671) on Murashige and Skoog's basal medium enriched with different concentrations of 2,4-D ($1-4\;\cal{mg/l}$), L-glutamine ($100\cal{mg/l}$), malt extract ($100\cal{mg/l}$), casein hydrolysate ($1000\;\cal{mg/l}$) and coconut milk ($5\%$) and solidified with $0.2\%$ gel rite. The embryogenic calli were subjected to desiccation for 1-8 h. Desiccation of the calli for 6-7 h resulted in enhancement of plant regeneration frequency ($83-96\%$) as compared to control ($12\%$). Plantlets exhibited vigorous growth to maturity in the greenhouse. Partial desiccation of embryogenic calli offers as a simple method for improving plant regeneration frequency in sugarcane.

  • In vitro shoot regeneration from cotyledon and hypocotyl explants derived from germinating mature Impatiens seeds. The induction of organogenetic tissue was also influenced by the cotyledon and hypocotyl. Multiple shoot induction was higher in hypocotyl than in the cotyledon explant with Thidiazuron and a NAA medium.

  • To identify the effects embryo developmental stage and gelrite concentration on plant regeneration in seed culture of rice, mature and immature seeds of rice were cultured on the $N_{6}$ medium supplemented with2 mg/$\ell$ 2.4-D and different levels of gelrite(0.2~1.0%). The calli formed immature embryos were produced more plants than those from mature embryos. The maximum frequency of plant regeneration was achieved in the culture of the calli of immature embryos which was harvested at the 21$^{th}$ day after pollination. The plant regeneration on the medium with gelrite was more accelerate than that on the medium with agar. The highest frequency(55%) of plant regeneration was obtained from the calli transferred to the medium with 6g/$\ell$ gelrite.

  • An efficient genetic transformation protocol is a fundamental requirement for high regeneration capacity from cultivated durum wheat (Triticum durum) varieties. In this study, wereportedtheeffectsoftwoauxins,2,4-dichlorophenoxyaceticacid(2,4-D)and4-amino-3,5,6-trichloropicoli nicacid(picloram), at a concentration of 2 mg/Laloneandincombination on the embryogenic callus and plantlet regeneration of four durum wheat varieties (Amria, Chaoui, Marouane, and Tomouh) using mature embryos (MEs) and immature embryos (ImEs). Significanteffectsofvariety,culturemedium(theauxinused),andvariety-mediuminteraction were observed on the callus weight and plantlet regeneration of both MR and ImE explants. The medium used for callus induction significantly affected plantlet regeneration (p < 0.001). Comparedto2,4-D, picloram led to a higher plantlet regeneration rate in both ME and ImE explants (19.8% and 40.86%, respectively). Plantlet regeneration also varied significantly depending on the variety and medium used. PicloramledtohighplantletregenerationofbothME and ImE explants in all varieties except Tomouh, which showed high plantlet regeneration of ME explants in 2,4-D. A comparison of ME and ImE responses indicated that ImEs are the best explants for high plantlet regeneration in durum wheat. Ourfindingssuggestthatpicloramisthebestauxin and should be used instead of 2,4-D due to its positive effect on increasing plant regeneration of durum wheat ME and ImE explants.

  • In order to optimize the conditions of callus induction and plant regeneration in seed culture of Italian ryegrass, the effects of basic medium and carbon sources on seed culturability and genotypic difference of plant regenerability were investigated. MS medium was better than N6 and B5 medium in enhancing callus growth and plant regeneration. Sucrose was superior to maltose in plant regeneration as carbon source in the medium. The genotypic difference in plant regenerability was obvious among four cultivars of Italian ryegrass tested. 'Rio' and 'Jeanne' showed to have higher regenerability with the frequency of 38% and 56%, respectively.

Plant Regeneration and Bulblet Formation of Allium wakegi Araki

  • Allium wakegi was cultured shoot tip in the condition of light culture. The Allium wakegi added plant growth regulator was observed of plant regeneration and bulblet formation. Callus Induction and growing rate was the best of 78% when added alone 2,4-D 0.5mg/L. In the formation of shoot, its regeneration rate was 96% when added BA 0.5mg/L in the light culture condition. When BA 0.5mg/L and NAA 0.5mg/L mixed and BA 0.5 mg/L and NAA 1.0mg/L mixed, the rates were 99% and 97% respectively, and these conditions were suitable for forming shoot. In the formation of roots, when added NAA 2.0mg/L in the light culture condition, the regeneration rate was 90.6 % and the roots were abnormal. When added NAA 1.0mg/L, the rate was 82 % and the highest. In the formation of bulbs, when BA 05mg/L and NAA 1.0mg/L mixed, the root generantion and its size in the bulbs was the best compare to other treatment experiments.
