The allopolyploid origin of Euphorbia stevenii and E. boöphthona (Euphorbiaceae)

  • To elucidate the ancestry of the allopolyploids E. stevenii and E. boöphthona, I examined eleven isozyme loci and 24 morphological characters from 28 populations representing five related Euphorbia species from Australia. According to an analysis of genetic and morphological data, three diploid species differentiated recently, but two independent polyploid species are estimated to have differentiated a relatively long time ago. Fixed heterozygosity for most isozymes in E. stevenii and E. boöphthona strongly suggests that these two species are allopolyploids rather than autopolyploids. The isozyme profiles of E. stevenii indicate that it is an allopolyploid that evolved from interspecific hybridization between the diploid E. tannensis and unidentified or extinct tetraploid species. In addition, isozyme patterns strongly suggest that E. stevenii was one of the ancestors of E. boöphthona. However, E. boöphthona showed a large number of fixed alleles that were not detected in any other Australian Eremophyton species. The most likely hypothesis for the origin of E. boöphthona is that it was formed by hybridization and chromosomal doubling between an extinct diploid species and the hexaploid E. stevenii.

  • As a part of our ongoing research to discover novel monamine oxidase (MAO) inhibitors of plant origin, we found that a MeOH extract from the whole plant of Cayratia japonica (Vitaceae) strongly inhibited the MAO activity in mouse brain. The EtOAc-soluble fraction was. therefore, subjected to the bioactivity-guided fractionations to isolate the active compounds. (omitted)

Phenylpropanoids of Plant Origin as Inhibitors of Biofilm Formation by Candida albicans

  • Biofilm-related infections of Candida albicans are a frequent cause of morbidity and mortality in hospitalized patients, especially those with immunocompromised status. Options of the antifungal drugs available for successful treatment of drug-resistant biofilms are very few, and as such, new strategies need to be explored against them. The aim of this study was to evaluate the efficacy of phenylpropanoids of plant origin against planktonic cells, important virulence factors, and biofilm forms of C. albicans. Standard susceptibility testing protocol was used to evaluate the activities of 13 phenylpropanoids against planktonic growth. Their effects on adhesion and yeast-to-hyphae morphogenesis were studied in microplate-based methodologies. An in vitro biofilm model analyzed the phenylpropanoid-mediated prevention of biofilm development and mature biofilms using XTT-metabolic assay, crystal violet assay, and light microscopy. Six molecules exhibited fungistatic activity at ${\leq}0.5mg/ml$, of which four were fungicidal at low concentrations. Seven phenylpropanoids inhibited yeast-to-hyphae transition at low concentrations (0.031-0.5 mg/ml), whereas adhesion to the solid substrate was prevented in the range of 0.5-2 mg/ml. Treatment with ${\leq}0.5mg/ml$ concentrations of at least six small molecules resulted in significant (p < 0.05) inhibition of biofilm formation by C. albicans. Mature biofilms that are highly resistant to antifungal drugs were susceptible to low concentrations of 4 of the 13 molecules. This study revealed phenylpropanoids of plant origin as promising candidates to devise preventive strategies against drug-resistant biofilms of C. albicans.

A test of the hybrid origin of Korean endemic Sedum latiovalifolium (Crassulaceae) (한국특산 태백기린초(돌나물과)의 교잡 기원에 대한 검증)

  • Eighteen morphological characters from 40 populations and ten isozyme loci from 35 populations of Sedum latiovalifolium and related species were examined to investigate the degree of morphological and genetic variation. The high-frequency marker alleles $MDH-2^a$ and $PGI-1^a$ in S. aizoon, S. kamtschaticum, and S. zokuriense did not appear in the populations of S. latiovalifolium. In addition, the high-frequency allele ($MDH-2^c$) in S. latiovalifolium appeared at a very low frequency in other subg. aizoon species. Thus, the isozyme data did not support a hybrid origin of S. latiovalifolium from S. aizoon with S. kamtschaticum. The results of an unweighted pair-group method using the arithmetic average method and a principal components analysis using morphological data also did not support a hybrid origin of S. latiovalifolium. However, our data strongly suggest that some individuals from the populations found in the Gumdaebong area were most likely hybrids due to introgression between S. latiovalifolium and S. kamtschaticum or S. aizoon and S. kamtschaticum.

Synonyms, Origin, Formation mechanism, Description and Efficacy of Hwangnapchim in Elucidation of Materia Medica (《본초연의》에 수록된 황납침(黃蠟沈)의 이명(異名)과 기원, 생성기전, 성상, 효능)

  • Objectives : The purpose of this study was to fix the synonyms, origin, formation mechanism, description, and efficacy of Hwangnapchim in Elucidation of Materia Medica. Methods : Through searching histories and herbal classics, We categorized the contents, and compared with Aquilariae Lignum Resinatum. Results : The synonyms of Hwangnapchim were classified by 3 categories : Napchimhyang-type, Ganamhyang-type and Ginamhyang-type. In early period, it was called by Napchimhyang-type, named after its description that was similar to beeswax. Afterward it was called by Ganamhyang-type or Ginamhyang-type, because of the name "Kynam" that was called by in its producing region. And there were records of producing region of Vietnam, Cambodia, and being from abroad, while those of Vietnam were most frequent. According to custom duties of Ming Dynasty, Hwangnapchim was 17.5 times as expensive as Agarwood. Hwangnapchim shared main producing region, original plant, and process of formation with Agarwood. Therefore, its original plant was Aquilaria crassna, but added honey during formation process. Hwangnapchim was classified 5 types according to its description, the highest quality was Yu-gyeol, resembled beeswax, and easily recovered if get a scar. Hwangnapchim had a fragrance at room temperature, a spicy flavor, and the efficacy of blocking urine and feces. Conclusions : These results showed that Vietnam was major producing area of Hwangnapchim, and that it shared same origin with Agarwood, but it had different description, efficacy, and higher prices because of being added honey during formation.

Plant Regeneration from Immature Embryo and Bulb Scale Tissue of Hippeastrum hybridum (아마릴리스의 미숙배와 인편조직으로부터 식물체 재분화)

  • Immature hybrid embryos of H. hybridum, 'Picottee', 'White Christmas', 'Eldorado', 'Origin', 'Red Lion', 'elstar', 'Crypsy' were cultured on the MS medium supplemented with various concentrations of 2,4-D, NAA, BA and TDZ. Among the treatments, NAA were more effective for the shoot regeneration and bulblet formation than other treatment. Addition of 0.5 ㎎/L NAA was effective for bulblet induction from explant Shoot regeneration was most effective on the medium with 1.0㎎/L NAA and 2.0 ㎎/L TDZ. The addition of 1.0-2.0㎎/L TDZ induced numerous shoots per explant but strongly inhibited root development when compared to 1.0-2.0㎎/L BA. When bulb scale segments of 'Star Van Holland' was incubated, bulblet formation was the most effective on MS medium with 0.5㎎/L NAA.

Taurine Content in Korean Foods of Plant Origin (한국인 상용 식물성 식품의 타우린함량 조사)

  • Taurine content in Korean foods of plant origin was determined for 118 commonly used food items including cereals, potatoes, pulses, nuts, seeds, vegetables and fruits. Taurine concentration in food sample was analyzed using an automated amino acid analyzer(Biochrom 20, Pharmacia LKB) based on ion-exchange chromatography. Taurine was frequently detected in plant kingdom in much lower concentrations(1/100~1/1000) than those found in marine lifes and mammals. Glutinous rice, glutinous millent and sorghum did not contain taurine, while 0.7~3.9mg taurine/100g dry wt were detected in rice, barley and their products. Potatoes and sweet potatoes contained 0.3~1.2mg taurine/100g wet wt, and seasame seeds, perilla seeds, almonds, walnuts and gigko nuts contained 0.7~3.0mg taurine/100g wt. Taurine concentration was undetectable in most of the pulses, and in a large number of vegetables. Garlic bulbs, eggplants, green peppers, lotus roots, and cabbages have a relatively high level of taurine(around 1mg taurine/100g wet wt) among vegetables. Taurine was absent or found in very low levels(<1mg taurine/100g wet wt) in most of the commonly used fruits.

Direct somatic embryogenesis, plant regeneration and genetic transformation of Panax ginseng

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

Pharmacognostical Studies on the "DangYak" (당약의 생약학적 연구)

  • A Korean folk medicine, "DangYak (當藥)" has been used as a bitter stomachic in Korea. With regard to the botanical origin of this crude drug, it has been considered to be from Swertia species (Gentianaceae), but there was no pharmacognostical confirmation on it. In this study, the morphological and anatomical characteristics of three Swertia species growing in Korea, Swertia japonica Makino, S. pseudochinensis Hara and S. tetrapetala Pall. were compared to clarify the botanical origin of "DangYak". As a result, it was determined that the commercial "DangYak" from Korean traditional market was proved to be the whole plant body mixture of Swertia japonica and Swertia pseudochinensis.

The Study on the Origin of Soybean Cultivation (대두재배(大豆栽培)의 기원(起源)에 관한 고찰(考察))

  • According to the literature, soybean cultivation originated from Wang-Gong (B.C. 685-643) of China who brought it from northeast Asia, for the first time. FUKUDA, of Japan divided soybean into three species-the wild, the cultivated and the intermediate. From the result of that study, he concluded that the soybean originated in northeast Asia. But Wang Kum Rung of China insisted that soybean originated in Hwa-Nam, because the soybean is a shortday plant and the agricultural history of Hwa-Nam, south of China, is older than that of northeast Asia. However, agriculture in northeast Asia had been already begun about B.C. 4000-6000 and the origin of culture cannot be decided only by photosensitivity. It has been proved that soybeans found in Korea were same as the ones of B.C. 2000. The soybeans of northeast Asia meet the conditions of the probable place of origin of cultivated crops established by Vabilov. Accordingly it is concluded that soybean has been originated from northeast Asia.
