Objectives : The objective of present study was to investigate the vasorelaxant effects of 20 Korean native plants on isolated rat thoracic aorta precontracted with phenylephrine (PE). Methods : Dried 20 plant materials were extracted 3 times with water, ethanol, or methanol for 3h in the reflux apparatus at 70 ± 5℃. Male SD rats were anesthetized by ether inhalation, and their aorta rings were isolated and placed in 10 ㎖ Krebs Henseleit (KH) buffer. While using an isolated organ-chamber technique, the aorta rings were maintained by bubbling with a gas mixture of 95% O2-5% CO2 at 37℃. Changes in isometric tension of aorta rings were recorded via isometric transducers connected to a Powerlab Data Acquisition System. Results : Among the 20 native plants, Chrysanthemum indicum L. flower, Nelumbo nucifera Gaertn. rhizome, Schisandra chinensis (Turcz.) Baill. fruit, Anthriscus sylvestris (L.) Hoffm. root, Corydalis turtschaninovii Besser tuber, Corydalis decumbens (Thunb.) Pers. tuber, and Dolichos lablab L. seed showed significant vasorelaxant effect on the contraction of aorta rings induced by PE. In contrast, Mertensia maritima subsp. asiatica Takeda whole plant, Ajuga decumbens Thunb. whole plant, Trigonotis peduncularis (Trevis.) Benth. ex Baker & S.Moore whole plant, Dioscorea quinquelobate Thunb. rhizome, Allium microdictyon Prokh aerial part, Momordica charantia L. fruit, Carthamus tinctorius L. flower, and Clematis terniflora DC. root constricted more the aorta rings precontracted by PE Conclusion : These results suggest that the possibility as useful herbal resources for the development of functional foods or medicines for hypertension treatment.
This study was conducted to investigate the influence of hydrophilic polymer (HP) mixture ratio (Control, 1.0%, 2.5%, 5.0%, and 10.0%) on growth of eggplant (Solanum melongena) for lower maintenance urban agriculture via green roofs. Although it was not statistically significant (p > 0.05), substrate temperature was decreased as hydrophilic polymer mixture ratio were increased. High substrate water content (95%) was found consistently in growing media under elevated hydrophilic polymer mixture ratio at over 5% during the entire growing period. Substrate electronic conductivity was increased as hydrophilic polymer mixture ratio were increased. Growth index was decreased as hydrophilic polymer mixture ratio was increased. It was reduced about 1/3 and 1/5 compared to that of Control in HP5.0 and HP10.0 treatment plants, respectively. Number of leaves, leaf length, and leaf width were decreased in following order: Control> HP1.0> HP2.5> HP5.0> HP10.0 treatments. There numbers were significantly lower in HP5.0 and HP10.0 treatment plants. Dry weight of shoot and root were decreased as hydrophilic polymer mixture ratio was increased. They were reduced by 1/4 compared to those of Control treatment plants. In addition, visual value was decreased as hydrophilic polymer mixture ratio was increased. Plants grown in HP1.0, HP2.5, and HP5.0 treatments all survived. However, plants grown in the HP10.0 treatment had the lowest survival rate (56%) after 3 months of growing. These results indicate that the advantage of adding hydrophilic polymer to green roof growing media may greater during dry periods. However, the proper mixture proportion of hydrophilic polymer should be determined according to different characteristics of growing media and plant species.
타타리메밀의 생력재배기술 확립을 목표로 파종, 제초, 수확방법의 개선을 도모하였다. 산파(손뿌림)는 종자량이 10a당 6 kg 정도 소요되었으며, 줄뿌림파종기는 4.5 kg, 복토직파기는 3.2 kg정도 소요되었다. 복도직파기를 이용한 기계파종의 경우 종실 수량이 주당 3.4 g으로 산파 시 종실수량 주당 2.4 g에 비하여 월등한 수량성을 보였다. 이것을 10a당 수량으로 환산하면 복토직파기 파종에서 113 kg으로 산파 80 kg에 비하여 우수하였다. 라쏘만 처리한 경우에 비하여 혼용과 조합 체계 처리에서 방제가가 90% 이상으로 높았으며 수량은 혼용(라쏘+그라목손)의 체계 처리 시 가장 높았다.
기내에서 성장한 배추의 자엽, 하배축과 계대배양한 지 2~3주 되는 어린잎이 원형질체 분리재료로 사용되었다. 배추의 원형질체 분리에 가장 적합한 효소의 조합은 0.4M mannitol 을 삼투조절제로 한 1% Cellulysin과 0.5% Macerozyme의 효소조합이 배추 원형질체 분리에 가장 적합한 것으로 나타났으며, 잎조직을 27$\pm$1$^{\circ}C$에서 30rpm의 속도로 12~16시간 동안 효소와 반응시켰을 때 7.6$\times$$10^{5}$ protoplast/g의 가장 많은 원형질체를 얻었다. 세포분열을 유기시키기 위하여 K8p 배지에 5 mg/L 2,4-D와 2 mg/L에 첨가하였을 때 배양 7~10일에 자엽과 하배축에서 분리한 원형질체에서 세포군들이 형성되었다. 분열한 세포가 8~10 세포크기 단계에 이르면 0.2% agarose 반고체 배지에 고정시켜 배양하였다. 얻어진 calli들을 100가지 다양한 재분화 배지에 옮겼으나 식물체의 재분화는= 이뤄지지 않았지만, 간혹 캘러스에서 뿌리가 분화되는 것은 관찰할 수 있었다.
간편하고 효과적인 정유성분 분석을 위해 SPME fiber를 선별하고 산초와 초피의 개체 정유성분 분석에 적합한 SPME fiber를 찾고자 실험한 결과는 다음과 같다. 단일물질별 흡착율은 PDMS, PDMS/DVB, CAR/PDMS 가 우수했다. 10종의 단일물질 표준품 혼합액 흡착 실험에서 직접주입 했을때와 가장 유사한 결과를 보인 것은 PDMS, PDMS/DVB fiber이였다. 최종 선발된 PDMS와 PDMS/DVB를 이용하여 24종의 단일물질 표준품 흔합액 흡착과 직접주입시를 비교한 결과 이들 간에 차이가 없었다. 또한 GC주입구의 split ratio별 성분의 검출에서도 차이가 없었다. 흡착시간이 길어질수록 분자량이 큰 성분의 흡착율이 높아지는 경향을 보였지만, 직접주입시와 비교해 볼 때 적절한 흡착시간은 30-40분이였다. SDE 추출물을 이용한 산초와 초피나무 개체 정유성분 분석에는 산초와 초피 모두 PDMS/DVB가 흡착시간 30-40분에서 분자량이 큰 sesquiterpene류까지 흡착하는데 유리하였다.
This research was conducted to get the basic materials necessary to obtain the somatic hybrid plant between Solanum sisymbriifolium and other Solanum species (S. integrifolium and S. toxicarium). Regarding the formation of colony from the protoplast in S. sisymbriifolium, S. integrifolium and the fused protoplast mixture; for the S. sisymbriifolium, a colony was observed in F medium(Kao medium containing $5.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ 2,4-D and $1.0mg{\cdot}L^{-1}$ BA); and for the S. integrifolium, in G medium (a half strength MS medium containing 0.03 M sucrose, 0.4 M mannitol, $1.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ kinetin) respectively. In mixed cultured protoplast after electriofusion treatment, the cell division and colony formation were observed in both media F and G. For the shoot and root formation rate, there was no difference between the parent of each breed and mixed protoplast regardless of the medium. In the fused protoplast mixture of S. sisymbriifolium and S. toxicarium, a colony formation was also observed in both media F and H(a half strength MS medium containing 0.03 M sucrose, 0.4 M mannitol, $1.0mg{\cdot}L^{-1}\;NAA,\;1.0mg{\cdot}L^{-1}$ kinetin); and there was no difference in the shoot and root formation rate between the parent and the mixed protoplast.

In order to utilize the tuber of Jerusalem artichoke (Jerusalem tuberosus L.) which has been considered one of the most economical resources for bioenergy, the tubers of the plant were processed into silage in 11 lab-silo with three levels of mixture of concentrates i.e. 10, 20, 30%. The silage was stored for 20 to 40 days before analyses. The quality of silage was compared with those of the tube-only silage without concentrates. Aproximate composition, pH, ammonium nitrogen, total nitrogen, and organic acids were analyzed. The results obtained are summarized ; 1. With the mixture of concentrates, the water content of the silage was able to be adjusted to 70 percent and NDF significantly increased (P<0.05) while ADF significantly decreased. 2. The content of water-soluble carbohydrate (WSC) was significantly decreased in silages of the mixture of concentrate and the tuber (P<0.05), while total nitrogen and ammonium nitrogen increased. 3. Tube-only silage gave satisfaction in fermentation, particularly organic-acid fermentation. As the storage period extended, Flieg's score of the silage was improved. Conclusively, the tuber of Jerusalem artichoke found to satisfy the requirements of good silage and to be recommended as a good resource for all-inane silage provided the tuber was ensilaged with 20 to 30 percent concentrate.
Cactus (Opuntia ficus-indica var saboten Makino) is a tropical or subtropical plant, which is widely used as folk medicine for burned wound, edema and indigestion. We previously found that the ethanol extract of cactus stem showed anti-inflanunatory action. This investigation was designed to isolate the active fraction of anti-inflanimatory action from cactus stem by solvent extraction and colunm chromatography. Carrageenan-induced paw edema in rats and acetic acid-induced writhing test in mice were used as animal models to search anti-inflammatory and analgesic activities. respectively. The ethanol extract of cactus stem was consecutively extracted with hexane, ethyl acetate, and n-butanol. The hexane fraction was the most effective in carrageenan-induced paw edema, and then was separated in colunm chromatography of silica gel by the elution with hexane/ethyl acetate mixture. The most effective fraction 1 was separated in a second colunm chromatography by eluting with hexane/diethyl ether mixture. The most effective fraction 1-5 was obtained, and separated in a third column chromatography by eluting with hexane/chloroform mixture. It produced the most effective fraction 1-5-1. Moreover, fraction 1-5-1 showed an inhibitory effect on acetic acid-induced writhing in the doses of 30mg/kg and 60mg/kg,p.o.,indicating that it also contained analgesic activity.
This study aimed to evaluate the anti-atherosclerotic and anti-hypertensive effects of six different plant extracts using a N(G)-nitro-L-arginine-methyl ester (L-NAME)-induced rat model of hypertension. All extracts were administered orally for six weeks. At the end of the study period blood pressure, blood flow, aortic histopathology, and hepatic endothelial nitric oxide synthase (eNOS) expression were measured. Subsequently, we also measured the levels of intracellular reactive oxygen species, nitric oxide (NO), and anti-inflammatory cytokines in vitro. Based on these screening results, we selected extracts of Cinnamomum cassia (C. cassia) and Salvia miltiorrhiza (S. miltiorrhiza) for further evaluation. C. cassia and S. miltiorrhiza extracts ameliorated hypertension and atherosclerosis in L-NAME-treated rats in a dose-dependent manner. In addition, a mixture of C. cassia and S. miltiorrhiza had an additive effect to reduce blood pressure, increase blood flow, and normalize aortic tissue. This mixture demonstrated anti-oxidative and anti-inflammatory activities in vitro. In conclusion, although further analysis of the therapeutic mechanism is required, the anti-hypertensive and anti-atherosclerotic effects of this mixture are likely mediated by increased eNOS expression, and its anti-oxidative and anti-inflammatory activities.
