Antioxidant Activity, Total Polyphenol Content, and Total Flavonoid Content of Boehmeria nivea var. tenacissima (Gaudich.) Miq. Collected from Six Regions (채집지역에 따른 섬모시풀(Boehmeria nivea var. tenacissima (Gaudich.) Miq.)의 항산화활성, 총 폴리페놀 함량, 총 플라보노이드 함량 차이)

  • This study measured the antioxidant activity and phytochemical content of 192 Boehmeria nivea var. tenacissima (Gaudich.) Miq. collected from six regions in order to identify the possibility of its industrial application. Two antioxidant activity assay (DPPH radical scavenging activity and ABTS radical scavenging activity) and two phytochemical content assay (total polyphenol content (TPC) and total flavonoid content (TFC)) of 192 samples were analyzed. The results showed that the DPPH radical scavenging activity of 192 samples was ranged from 0.02 to 1.35 mgAAE/g, and among them, the samples collected from Goheung (0.53 ± 0.37) and Ulleungdo (0.52 ± 0.28) showed the highest activity. The ABTS radical scavenging activity was showed the ranged from 0.07 to 1.81 mgAAE/g, and the samples collected at Ulleungdo (0.47 ± 0.36) showed the highest activity. The total polyphenol content of 192 samples was 18.6 to 234.8 ugGAE/g, and the samples collected at Mokpo (93.4 ± 34.7) and Jindo (90.4 ± 24.5) showed the highest content. The total flavonoid content was 0.10 to 1.22 mgQE/g and the samples collected at Ulleungdo (0.49 ± 0.31) showed the highest content. In the correlation analysis, there was no significant relationship between the environmental conditions and the antioxidant activity. The total polyphenol content showed a positive correlation with daily temperature difference, and negative correlation with average wind speed and average humidity, and total flavonoid content showed negative correlations with the average temperature, maximum temperature, and minimum temperature. This result can be used as a basic data establish the cultivation conditions of B. nivea var. tenacissima (Gaudich.) Miq. as a functional raw material to increase the usefulness of B. nivea var. tenacissima (Gaudich.) Miq..

Anti-inflammatory Effects, Skin Wound Healing, and Stability of Bluish-purple Color Extracted from Platycodon grandiflorus (Jacq.) A.DC. Flower Extract (도라지꽃 추출물의 항염증, 피부재생 효과 및 색소 안정성 연구)

  • Platycodon grandiflorus (P. grandiflorus) flower is a perennial plant belonging to the family Campanulaceae and has many excellent pharmacological effects, so it has been used as a medicinal ingredient since ancient times. In addition, anthocyanin is a purple or blue natural pigment contained in plant flowers and fruits, and is known as a powerful antioxidant. The purpose of this study was to confirm the dermatological functionality of P. grandiflorus flower extract and the value of the bluish anthocyanin contained in flowers as a cosmetic material as a natural pigment. Firstly, 50% ethanol and 80% ethanol were added to the P. grandiflorus flower and extracted under reflux for 4 h at 25, 60, and 80 ℃, and the pH of each treatment group was similar. Based on the anthocyanin content and chromaticity (E*ab), 50% ethanol 60 ℃ extraction conditions showing the color development most similar to the natural color of the P. grandifloras flower were selected, and a sample was prepared by concentrating and lyophilizing. The analysis results showed that the total phenol, total flavonoid, and total anthocyanin contents were in the ranges of 23 ㎍/mL, 16 ㎍/mL, and 0.17 ㎍/mL, respectively. The P. grandiflorus flower extract suppressed the production of nitric oxide (NO) and interleukin-6 (IL-6) in lipopolysaccharide (LPS) induced RAW264.7 cells. Furthermore, the P. grandiflorus flower extract showed wound healing effects through the promotion of skin cell migration in TNF-α stimulated human keratinocytes. The stability of anthocyanin and extract color was studied during a storage period of 50 days at various temperatures (4 ℃, 25 ℃, and 45 ℃). Color values (L, a, and b) of the P. grandiflorus flower extract changed over 50 days, whereas the bluish-purple color of the extract was stabilized using 5% maltodextrin. These results suggest that P. grandiflorus flower extract may be useful as a natural cosmetic pigment.

Evaluation of the Effects of Hangover-Releasing Agent Containing Vinegar Extract in Common Buckwheat and Tartary Buckwheat on Alcohol Metabolism and Hangover Improvement (일반메밀과 쓴메밀의 식초 추출물의 알코올 대사 및 숙취개선 효능 평가)

  • The aim of this study was to explore the effects of vinegar extract from seed of common buckwheat (Fagopyrum esculentum Moench) and seed of tartary buckwheat (F. tataricum Gaertner) on acute ethanol-induced hangover in Sprague-Dawley rats. Vinegar extract from buckwheat is rich choline, quercetin and its glycoside, rutin known as flavonoid antioxidants. The test extract containing buckwheat was proven to alleviate hangovers through a significant reduction in the concentration of alcohol and acetaldehyde in the context of an alcohol-induced hangover model. Hepatic alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities were significantly higher in buckwheat vinegar-treated rats than in ethanol-treated rats. Moreover, tartary buckwheat vinegar upregulated antioxidant enzyme such as superoxide dismutase and Catalase activities in liver tissues. These results suggest that buckwheat vinegar extract could alleviate ethanol-induced hangover symptoms by elevating activities related to hepatic ethanol-metabolizing enzymes against ethanol induced metabolites, and in particular, tartary buckwheat should be further developed to be a novel anti-hangover material.

  • The viability loss or death of weed seeds buried in soil can be induced by infrared irradiation which has good penetration in moist soil. By using this principle of pre-emergence soil-treatment, the study was carried out to obtain basic information needed to develop the effective weed control method for the production of less polluted agricultural products. An apparatus for irradiating infrared was constructed by using ceramic material with high emissivity. The LPG was used as fuel for producing infrared by heating ceramic material. The soil heated in this study was loam with four levels of moisture contents (0.6, 5.7, 10.7, 15.1 % wb). The temperature distribution was measured at various soil depths when soil with different moisture content was irradiated with infrared for three different times (30, 60, 90 sec). The soil depths with duration time of minimum 3 minutes over $80^{\circ}C$, temperature inducing viability loss of weed seeds, were investigated. When the moisture content of soil was 0.6 and 5.7 % wb, the soil depths which can induce viability loss of weed seeds was greatly increased with increasing irradiation time. However, any depths of soil tested in this study was not reached to the temperature of $80^{\circ}C$ when 30 seconds of irradiation time was applied on soil with moisture content of 10.7 or 15.1 % wb. Generally, the soil depth needed for viability loss of weed seeds was decreased with increasing moisture content of soil. Also, longer irradiation time was required to induce viability loss of weed seeds with increasing moisture content of soil.

  • The experiment was carried out to investigate the chemical properties of cement kiln dusts, abundantly produced from cement industry as a byproduct, and their effectiveness on rice yield. The field experiment was conducted on the acid paddy soil developed on basalt at Dongsong-Myon, Chulwon-Kun, Gangwon-Do. Two kinds of cement kiln dusts were used ; By Pass (BP) collected from the suspension preheater and Electric Precipitate (EP) from the cottrell electric precipitator. The levels of cement kiln dust applied were 100kg/10a, 200kg/10a and 300kg/10a, and the recommended variety "Nong Back" was adopted for this experiment. The results obtained are summarized as follows ; 1. The component of cement kiln dusts seems to be quite suitable for liming material. BP has 55% alkalinity, 41.7% of soluble calcium, 9.8% of soluble magnesium and 4.5% of water soluble silicate, while EP has 53.5% alkalinity, 41.7% soluble calcium, 8.3% soluble magnesium and 1% water soluble silicate. 2. The relative effectiveness of cement kiln dust in the soil will be superior due to very fine particle size. EP pass through completely 270 mesh screen, and 95% of BP pass through 150 mesh screen, 68% passing 270 mesh. 3. BP application at the rate of 100kg/10a increased 21% of rice yield as compared with control and EP 15%. It was observed that the affected yield components were increased panicle number per hill, grain number per panicle and 1,000 grains weight. 4. The application of optimum amount (100kg/10a) of cement kiln dusts accelerated the uptake of nutrients by rice plant and increased rice yield. However, the excess amounts (200kg/10a, 300kg/10a) of cement kiln dusts retarded the uptake of nutrients from soil.

  • The chemical components of Citrus junos produced in Korea were divided into two parts; common and special components respectively. In the former the relation between the physiological effects of the plant and its ripening process was observed periodically while the latter was analyzed the ripening fruits for their effective utilization as food. The results are summarized as follows: 1. The analytical result of seasonal change showed that the rind ratio was higher than the flesh ratio and on a regional basis, the rind ratio was higher in the islands than on land areas. 2. In the experiment the moisture was increased until the third period, but afterwards it was made constant. While the content of crude fat, cellulose, ash, total acid and soluble non-nitrogen material were decreased until the third period and the cotent of cellulose and total acid were continuousely redused until the last period. In con trast with the above the content of reducing sugars was increased but the content of crude fat, cellulose, ash, crude protein and soluble non-nitrogen material were increased until last period. 3. The content of vitamin C was richer in the rind than in the flesh, in the Korean species than in the Japanese. 4. Free sugars; xylose, fructose, glucose were richer in the rind than in the flesh. 5. The content of volatile organic acids was richer in the rind than in the flesh. Among them, volatile acids, acetic acid, formic and n-valeric acid were found in the rind and formic acid, acetic acid and propionic acid were deteceed in the flesh. 6. The total content of non-volatile acids was richer in the flesh than in the rind. In the kind of non-volatile acids, citric acid,glutaric acid, malic acid, tartaric acid, oxalic acid, malonic acid, succinic acid and an unknown acid were found in the rind and citric acid, malic acid, succinic acid, oxalic acid, glutaric acid and malonic acid in the flesh. 7. Three kinds of aromatic components: D-limonene, ${\alpha}-pinene$, p-cymene and seven other kinds of unknown aromatic components were detected in neutral essential oils. Among them, D-limonene seemed to be main aromatic component in the fruits. 8. From the above results it is confirmed that both rind and flesh of the ripened fruit could be utilized for food effectively, and unripened fruits are suitable for producing citric acid, ripened fruits are also useful for producing juice.

Structural Safety Test and Analysis of Type IP-2 Transport Packages with Bolted Lid Type and Thick Steel Plate for Radioactive Waste Drums in a NPP (원자력발전소의 방사성폐기물 드럼 운반을 위한 볼트체결방식의 두꺼운 철판을 이용한 IP-2형 운반용기의 구조 안전성 해석 및 시험)

  • If a type IP-2 transport package were to be subjected to a free drop test and a penetration test under the normal conditions of transport, it should prevent a loss or dispersal of the radioactive contents and a more than 20% increase in the maximum radiation level at any external surface of the package. In this paper, we suggested the analytic method to evaluate the structural safety of a type IP-2 transport package using a thick steel plate for a structure part and a bolt for tying a bolt. Using an analysis a loss or dispersal of the radioactive contents and a loss of shielding integrity were confirmed for two kinds of type IP-2 transport packages to transport radioactive waste drums from a waste facility to a temporary storage site in a nuclear power plant. Under the free drop condition the maximum average stress at the bolts and the maximum opening displacement of a lid were compared with the tensile stress of a bolt and the steps in a lid, which were made to avoid a streaming radiation in the shielding path, to evaluate a loss or dispersal of radioactive waste contents. Also a loss of shielding integrity was evaluated using the maximum decrease in a shielding thickness. To verify the impact dynamic analysis for free drop test condition and evaluate experimentally the safety of two kinds of type IP-2 transport packages, free drop tests were conducted with various drop directions. For the tests we examined the failure of bolts and the deformation of flange to evaluate a loss or dispersal of radioactive material and measured the shielding thickness using a ultrasonic thickness gauge to assess a loss of shielding integrity. The strains and accelerations acquired from tests were compared with those by analyses to verify the impact dynamic analysis. The analytic results were larger than the those of test so that the analysis showed the conservative results. Finally, we evaluated the safety of the type IP-2 transport package under the stacking test condition using a finite element analysis. Under the stacking test condition, the maximum Tresca stress of the shielding material was 1/3 of the yielding stress. Two kinds of a type IP-2 transport package were safe for the free drop test condition and the stacking test condition.

Analysis of the Spent Fuel Cooling Time for a Deep Geological Disposal (심지층 처분을 일한 사용후핵연료 냉각기간 분석)

  • The purpose of the HLW deep geological disposal is to isolate and to delay the radioactive material release to human beings and the environment for a long time so that the toxicity does not affect to the environment. The main requirements for the HLW repository design is to keep the buffer temperature below $100\;^{\circ}C$ in order to maintain its integrity. So the cooling time of spent fuels discharged from the nuclear power plant is the key consideration factors for efficiency and economic feasibility of the repository. The disposal tunnel/disposal hole spacing, the disposal area and thermal capacity required for the deep geological repository layout which satisfies the temperature requirement of the disposal system is analyzed to set the optimized spent fuels cooling time. To do this, based on the reference disposal concept, thermal stability analyses of the disposal system have been performed and the derived results have been compared by setting the spent fuels cooling time and the disposal tunnel/disposal hole spacing in various ways. From these results, desirable spent fuels cooling time in view of disposal area is derived. The results shows that the time reaching the maximum temperature within the design limit of the temperature in the disposal site is likely shortened as the cooling time of spent fuels becomes short. Also it seems that the temperature-rising and-dropping patterns in the disposal site are of smoothly varying form as the cooling time of spent fuels becomes long. In addition, it is revealed that a desirable cooling time of spent fuels is approximately 40-50 years when spent fuels are supposedly disposed in the deep geological disposal site with its structural scale under consideration in this study.

Antioxidative and Cytoprotective Effects of Annona muricata (Graviola) Extract for HDF Cell Damage Induced by Hydrogen Peroxide (H2O2에 의해 유도된 HDF 세포 손상에 대한 그라비올라 추출물의 항산화 및 세포 보호 효과)

  • As interest in functionality and environmentally friendly cosmetics is growing in recent years, materials that use safe and effective plant extracts have been developed. Therefore, this study also attempted to check the possibility of the graviola extract, which is known to have various efficacy mainly as a health functional material as a functional cosmetic material. In order to find out the antioxidant activity of graviola, we measured total polyphenol, total flavonoid content and DPPH radical scavenging activity and measured the ROS activity inhibition effect and cytoprotective effect on oxidative stress by treating HDF with hydrogen peroxide cells at an appropriate concentration after checking cytotoxicity in HDF cells. Based on the results of this experiment, the graviola extract was found to contain as high as 26.6 mg(CA)/100g, 14.3 mg(Q)/100g of total polyphenol and flavonoid, which are the antioxidant indexes and to have the high radical scavenging activity. The cell survival rate of the HDF cells was measured, and as a result, no significant cytotoxicity was observed at all concentrations and the experiment was carried out at a concentration of $100{\mu}g/mL$ afterwards. Inhibition of ROS activity in HDF cells induced by hydrogen peroxide was measured and the concentration-dependent inhibition of ROS activity was found and the cell protection effect of graviola was measured after hydrogen peroxide was treated for 4, 24 and 48 hours. As a result, the cell protection effect as high as 89.92% was confirmed at a $25{\mu}g/mL$ concentration up to 24 hours. As these results show that the graviola extract has excellent antioxidant activity, almost no toxicity to HDF cells, an effective activity inhibitory effect on active oxygen generated by hydrogen peroxide and excellent cytoprotective effect, the possibility as various functional materials with antioxidant and cytoprotective effects was confirmed.

A Study on the Solubilizing and Emulsifying Action of Tocopheryl Acetate using Plant Surfactant (식물성계면활성제를 사용한 토코페릴아세테이트의 가용화와 유화력에 관한 연구)

  • This study is a study on solubilization and emulsifying power of tocopheryl acetate using vegetable surfactants. High purity polyglyceryl-10 isostearate and polyglyceryl-10 oleate were mixed to synthesize a vegetable surfactant with excellent solubilizing power and emulsifying power. The mixed raw material was named Solubil EWG-1100. The appearance of this raw material was a pale yellowish paste with a specific smell, specific gravity of 1.12, and acid value of 0.085. The HLB value of this surfactant was calculated by the Griffin's equation with an average value of 15.17. The behavior of this surfactant to solubilize tocopheryl acetate was mechanically verified. The performance of solubilization was evaluated by a method of visual evaluation and was measured by a transmittance rate at 650 nm using a UV spectrophotometer. As a result, in the formulation using 3% ethanol as a co-solvent, the concentration of surfactant was required to solubilize tocopheryl acetate was required about 5 times of natural surfactant. In the formulation without ethanol as a co-solvent, the concentration of surfactant was required to solubilize tocopheryl acetate required about 7 times of natural surfactant. In addition, the concentration of surfactant required to make an emulsifivation 10 % of tocopheryl acetate was 1 wt% of Solubil EWG-1100, and the emulsified particle size was 3.5 mm in cream formula. In order to obtain stable and fine emulsified particles, it was found that as the concentration of tocopheryl acetate increased, the concentration of Solubil EWG-1100 also was to increase. As a result of testing the solubilizing power of the surfactant according to the pH various change, it showed stable solubilizing power in the acidic region of pH=3.2, the neutral region of pH=7.0, and the alkaline region of pH=11.8. As application, based on these results, it is expected that it can be widely applied to the cosmetics field that develops skin care prescriptions, sensitive skin products, and heavy dry skin products.
