Plant growth-promoting rhizobacteria (PGPR) are naturally occurring bacteria that intensively colonize plant roots and are crucial in promoting the crop growth. These beneficial microorganisms have garnered considerable attention as potential bio-inoculants for sustainable agriculture. PGPR directly interacts with plants by providing essential nutrients through nitrogen fixation and phosphate solubilization and accelerating the accessibility of other trace elements such as Cu, Zn, and Fe. Additionally, they produce plant growth-promoting phytohormones, such as indole acetic acids (IAA), indole butyric acids (IBA), gibberellins, and cytokinins.PGPR interacts with plants indirectly by protecting them from diseases and infections by producing antibiotics, siderophores, hydrogen cyanide, and fungal cell wall-degrading enzymes such as glucanases, chitinases, and proteases. Furthermore, PGPR protects plants against abiotic stresses such as drought and salinity by producing 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase and modulating plant stress markers. Bacteria belonging to genera such as Bacillus, Pseudomonas, Burkholderia, Pantoa, and Enterobacter exhibit multiple plant growth-promoting traits, that can enhance plant growth directly, indirectly, or through synergetic effects. This comprehensive review emphasizes how PGPR influences plant growth promotion and presents promising prospects for its application in sustainable agriculture.
Three hundred and seventy-four rhizobacteria were isolated from the rhizosphere soil (RS) or rhizoplane (RP) of Echinochloa crus-galli, Carex leiorhyncha, Commelina communis, Persicaria lapathifolia, Carex kobomugi, and Equisetum arvense, grown in contaminated soil with petroleum and heavy metals. The isolates were screened for plant growth-promoting trait (PGPT), including indole acetic acid (IAA) productivity, 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, and siderophore(s) synthesis ability. IAA production was detected in 86 isolates (23.0%), ACC deaminase activity in 168 isolates (44.9%), and siderophore(s) synthesis in 213 isolates (57.0%). Among the rhizobacteria showing PGPT, 162 isolates had multiple traits showing more than two types of PGPT. The PGPT-possesing rhizobacteria were more abundant in the RP (82%) samples than the RS (75%). There was a negative correlation (-0.656, p<0.05) between the IAA producers and the ACC deaminase producers. Clustering analysis by principal component analysis showed that RP was the most important factor influencing the ecological distribution and physiological characterization of PGPT-possesing rhizobacteria.
Salinity has been a threat to agriculture in some parts of the world; and recently, the threat has grown. Plant growth-promoting rhizobacteria (PGPR) may benefit plant growth, either by improving plant nutrition or producing plant growth hormones. The effects of rhizobacterial strains to attenuate the salinity stress on the germination of Chinese cabbage seeds were tested using four different concentrations of NaCl (50, 100, 150, and 200 mM). Also, PGPR strains were tested to enhance the early germination of Chinese cabbage seeds under normal conditions. Azotobacter chroococcum performed best with enhancing the radicle length of 4.0, 1.2, and 1.0 times at treatments of 50, 100, and 150 mM of NaCl, respectively. Additionally, significant differences were found in plumule length, A. chroococcum and Lactobacillus sp. showed remarkable activation either in normal or under stress conditions. Co-inoculation by three rhizobacterial strains (LAPmix) indicated synergistic effect to enhance the early germination of the seeds. The results of this study are promising for application of rhizobacterial strains that possess plant growth promoting traits to enhance the plant tolerance against salinity.

The purpose of this study was to investigate the molecular diversity of rhizobacteria associated with ginseng of varying age levels and their plant benefiting attributes. A total of 143 different isolates belonging to 15 different bacterial genera were recovered. Although variation was found in the rhizobacterial community due to age of the plant, majority of bacteria belong to Firmicutes (58%). In which, Bacillus was found to be the predominant genus irrespective of age of the ginseng. To assess the plant benefiting attributes, 30 representative isolates were selected. The results indicated that some of the isolates could exhibit multiple plant growth promoting traits like secretion of cell wall degrading enzymes, production of indole-3-acetic acid, synthesis of siderophores, solubilization of phosphates and soil pathogens inhibition. It can be suggested that strains of B. subtilis, B. amyloliquefaciens, B. velezensis, and B. licheniformis were positive for all the above traits, which have potential to be used as plant growth promoting inoculants to improve ginseng crop in the future.
This study assessed the possible role of different traits in selected plant growth-promoting rhizobacteria (PGPR) for improving wheat growth and yield under natural conditions. Rhizobacteria exhibiting 1-aminocyclopropane-1-carboxylate (ACC)-deaminase activity were isolated and screened for their growth-promoting activity in wheat under axenic conditions. Five isolates belonging to Pseudomonas and one Burkholderia caryophylli isolate that showed promising performances under axenic conditions were selected and characterized for in vitro ACC-deaminase activity, chitinase activity, auxin production, P solubilization, and root colonization. These isolates were then used as inocula for wheat cultivated under natural conditions in pot and/or field trials. Significant increases in root elongation, root weight, tillers per pot, 1,000-grain weight, and grain and straw yields were observed in response to inoculation with PGPR in the pot trials. Inoculation with these PGPR was also effective under field conditions and increased the wheat growth and yield significantly. However, the efficacy of the strains was inconsistent under the axenic, pot, and field conditions. Pseudomonas fluorescens ($ACC_{50}$), which exhibited a relatively high in vitro ACC-deaminase activity, chitinase activity, auxin production, and P solubilization and more intensive root colonization, was the most efficient isolate under the field conditions. Therefore, these results demonstrated that ACC-deaminase activity is an efficient parameter for the selection of promising PGPR under axenic conditions. However, additional traits of PGPR, including auxin production, chitinase activity, P solubilization, and root colonization, are also important for selecting PGPR as biofertilizers.
Plant growth-promoting (PGP) bacteria can be used as bioresources to enhance phytoremediation through their PGP traits and pollutant removal capacity. In this study, 49 rhizobacteria were primarily isolated from the rhizosphere of tall fescue grown in diesel- and heavy metal-contaminated soil. Their biosurfactant production, phosphate (P) solubilization, and siderophore production were qualitatively and quantitatively evaluated to identify superior PGP bacteria. The optimal conditions for the growth of PGP bacteria and the stability of their PGP traits were a temperature of 35℃, a pH of 7, and 2 days of cultivation time. Four superior PGP bacteria (Pseudomonas sp. NL3, Bacillus sp. NL6, Bacillus sp. LBY14, and Priestia sp. TSY6) were finally selected. Pseudomonas sp. NL3 exhibited superior biosurfactant production and P solubilization. Bacillus sp. NL6 showed the highest P solubilization and superior production of biosurfactants and siderophores. Bacillus sp. LBY14 offered the best siderophore production and impressive P solubilization. Priestia sp. TSY6 had superior capacity for all three PGP traits. Through their secretion of beneficial PGP metabolites, the four bacteria isolated in this study have the potential for use in the phytoremediation of contaminated soil.

Rhizosphere microorganisms possessing phytase activity are considered important for rendering phytate-phosphorus (P) available to plants. In the present study, the Citrobacter braakii phytase gene (appA) was overexpressed in rhizobacteria possessing plant growth promoting (PGP) traits, for increasing their potential as bioinoculants. AppA was cloned under the lac promoter in the broadhost-range expression vector pBBR1MCS-2. Transformation of the recombinant construct pCBappA resulted in high constitutive phytase activity in all of the eight rhizobacterial strains belonging to genera Pantoea, Citrobacter, Enterobacter, Pseudomonas (two strains), Rhizobium (two strains), and Ensifer that were studied. Transgenic rhizobacterial strains were found to display varying levels of phytase activity, ranging from 10-folds to 538-folds higher than the corresponding control strains. The transgenic derivative of Pseudomonas fluorescens CHA0, a well-characterized plant growth promoting rhizobacterium, showed the highest expression of phytase (~8 U/mg) activity in crude extracts. Although all transformants showed high phytase activity, rhizobacteria having the ability to secrete organic acid showed significantly higher release of P from Ca-phytate in buffered minimal media. AppA overexpressing rhizobacteria showed increased P content, and dry weight (shoot) or shoot/ root ratio of mung bean (Vigna radiata) plants, to different extents, when grown in semisolid agar (SSA) medium containing Na-phytate or Ca-phytate as the P sources. This is the first report of the overexpression of phytase in rhizobacterial strains and its exploitation for plant growth enhancement.
Plant growth promoting rhizobacteria and endophytic bacteria were isolated from different varieties of turmeric (Curcuma longa L.) from South India. Totally 50 strains representing, 30 PGPR and 20 endophytic bacteria were identified based on biochemical assays and 16S rDNA sequence analysis. The isolates were screened for antagonistic activity against Pythium aphanidermatum (Edson) Fitzp., and Rhizoctonia solani Kuhn., causing rhizome rot and leaf blight diseases in turmeric, by dual culture and liquid culture assays. Results revealed that only five isolates of PGPR and four endophytic bacteria showed more than 70% suppression of test pathogens in both assays. The SEM studies of interaction zone showed significant ultrastructural changes of the hyphae like shriveling, breakage and desication of the pathogens by PGPR B. cereus (RBacDOB-S24) and endophyte P. aeruginosa (BacDOB-E19). Selected isolates showed multiple Plant growth promoting traits. The rhizome bacterization followed by soil application of B. cereus (RBacDOB-S24) showed lowest Percent Disease Incidence (PDI) of rhizome rot and leaf blight, 16.4% and 15.5% respectively. Similarly, P. aeruginosa (BacDOB-E19) recorded PDI of rhizome rot (17.5%) and leaf blight (17.7%). The treatment of these promising isolates exhibited significant increase in plant height and fresh rhizome yield/plant in comparison with untreated control under greenhouse condition. Thereby, these isolates can be exploited as a potential biocontrol agent for suppressing rhizome rot and leaf blight diseases in turmeric.

Drought limits rice production under upland condition. This study quantified the effect of rhizobacteria inoculation on rice root system developmental response to drought and its role in maintaining high soil water use, and dry matter production under drought using NSIC Rc192 (rainfed lowland rice variety). The source of inoculant was Streptomyces mutabilis, a recently isolated rhizobacteria containing plant growth promoting compounds such as ACC deaminase, indole-3-acetic acid and phosphatase (Cruz et al., 2014, 2015). In the first experiment, pre-germination inoculation of seeds with S. mutabilis significantly increased the shoot and root (radicle) length as well as root hair lengths, relative to the non-inoculated control. In the second experiment, rice plants inoculated with S. mutabilis and grown in rootbox with soil generally had greater total root length under drought regardless of the timing of inoculations, relative to the non-inoculated control. Consequently, improved root system development contributed to the increase in soil water uptake under drought and thus, dry matter production. Among inoculation treatments, one-time inoculation of S. mutabilis either at pre-germination or pre-drought stress at 14 days after sowing (DAS), had significantly greater shoot dry matter production than three-time inoculation at pre-germination, at thinning (3 DAS) and at pre-drought (14 DAS). This study demonstrated the effectiveness of rhizobacteria (S. mutabilis) containing growth promoting compounds for enhancing drought dehydration avoidance root traits and improving the growth of rice plants under drought condition.
A total of 31 Acinetobacter isolates were obtained from the rhizosphere of Pennisetum glaucum and evaluated for their plant-growth-promoting traits. Two isolates, namely Acinetobacter sp. PUCM1007 and A. baumannii PUCM1029, produced indole acetic acid (10-13 ${\mu}g$/ml). A total of 26 and 27 isolates solubilized phosphates and zinc oxide, respectively. Among the mineral-solubilizing strains, A. calcoaceticus PUCM1006 solubilized phosphate most efficiently (84 mg/ml), whereas zinc oxide was solubilized by A. calcoaceticus PUCM1025 at the highest solubilization efficiency of 918%. All the Acinetobacter isolates, except PUCM1010, produced siderophores. The highest siderophore production (85.0 siderophore units) was exhibited by A. calcoaceticus PUCM1016. Strains PUCM1001 and PUCM1019 (both A. calcoaceticus) and PUCM1022 (Acinetobacter sp.) produced both hydroxamate-and catechol-type siderophores, whereas all the other strains only produced catechol-type siderophores. In vitro inhibition of Fusarium oxysporum under iron-limited conditions was demonstrated by the siderophore-producing Acinetobacter strains, where PUCM1018 was the most potent inhibitor of the fungal phytopathogen. Acinetobacter sp. PUCM1022 significantly enhanced the shoot height, root length, and root dry weights of pearl millet seedlings in pot experiments when compared with controls, underscoring the plant-growth-promoting potential of these isolates.
