식물 유용 방선균 2종의 배양 및 포자생성 최적화 조건 탐색 (Optimization of Culture and Sporulation for Two Plant Beneficial Streptomyces Strains)

  • 관행적 화학농약의 식물병 관리 및 치료의 한계적인 효과는 유익한 세균이나 미생물을 이용한 식물병 제어 방법 개발의 필요성을 나타낸다. 지속 가능한 농업은 특히 다양한 항생물질과 이차 대사 생성물을 생산하는 방선균의 농업적 중요성을 시사하며, Streptomyces globiosporus SP6C4균주와 Streptomyces sp. S8은 강력한 항균 작용을 가지고 있으며, 지속가능한 농업에서 작물 생장을 개선하기 위한 우수한 균주로 인정되고 있다. 본 연구에서는 다양한 질소원과 탄소원을 활용하여 대상 미생물 Streptomyces 두 균주의 생장을 촉진시키는 방법을 조사하였다. L-글루타민과 L-시스테인이 첨가된 조건에서 S. globiosporus SP6C4와 Streptomyces sp. S8 균주의 포자 생성 능력이 증가하였으며, 각 균주의 생장이 촉진되었다. 이러한 결과는 유용 미생물의 대량배양 기술의 범위 확장과 농업미생물의 실용화에 기여할 것으로 생각된다.

Fermentation Process Development of Recombinant Hansenula polymorpha for Gamma-Linolenic Acid Production

  • Development of the strain and the fermentation process of Hansenula polymorpha was implemented for the production of ${\gamma}$-linolenic acid ($GLA,\;C18:3{\Delta}^{6,9,12}$), an n-6 polyunsaturated fatty acid (PUFA) that has been reported to possess a number of health benefits. The mutated ${\Delta}^6$-desaturase (S213A) gene of Mucor rouxii was expressed in H. polymorpha under the control of the methanol oxidase (MOX) promoter. Without the utilization of methanol, a high-cell-density culture of the yeast recombinant carrying the ${\Delta}^6$-desaturase gene was then achieved by fed-batch fermentation under glycerol-limited conditions. As a result, high levels of the ${\Delta}^6$-desaturated products, octadecadienoic acid ($C18:2{\Delta}^{6,9}$), GLA, and stearidonic acid ($C18:4{\Delta}^{6,9,12,15}$), were accumulated under the derepression conditions. The GLA production was also optimized by adjusting the specific growth rate. The results show that the specific growth rate affected both the lipid content and the fatty acid composition of the GLA-producing recombinant. Among the various specific growth rates tested, the highest GLA concentration of 697 mg/l was obtained in the culture with a specific growth rate of 0.08 /h. Interestingly, the fatty acid profile of the yeast recombinant bearing the Mucor ${\Delta}^6$-desaturase gene was similar to that of blackcurrant oil, with both containing similar proportions of n-3 and n-6 essential fatty acids.

  • 메탄발효폐액(醱酵廢液)의 비효(肥效)와 그 시용량(施用量)을 구명(究明)하기 위하여 청예(靑刈)옥수수(수원(水原) 19호(號))를 공시(供試)하여 폐액(廢液)을 시비(施肥)로 시용(施用)한 결과(結果)는 다음과 같다. 가. 폐액시용(廢液施用)으로 수분공급(水分供給)이 원활해져 발아(發芽)에 효과(效果)가 있었다. 나. 옥수수의 수량(收量)(생초중(生草重), 건초중(乾草重))은 NPK+폐액(廢液)12톤구(區)>NPK+폐액(廢液) 8톤구(區)>NPK+폐액(廢液) 4톤구(區)>NPK+물 4톤구(區)>NPK구(區) 순(順)으로 증수(增收) 하였으며 생육상황(生育狀況)도 같은 경향(傾向)이었다. 다. 옥수수의 식물체(植物體) 및 토양중(土壤中)의 질소(窒素) 및 인산함량(燐酸含量)은 수량(收量)과 같은 경향(傾向)이었으며 가리함량(加里含量)은 일정(一定)한 경향(傾向)이 없었다. 라. 메탄폐액(廢液)은 비료(肥料) 및 관개용수(灌漑用水)로 활용(活用)이 가능(可能)할 것으로 판단(判斷)되었다.

Effect of Lactic Fermentation and Spray Drying Process on Bioactive Compounds from Ngoc Linh Ginseng Callus and Lactobacillus plantarum Viability

  • Ngoc Linh ginseng is one of the most valuable endemic medicinal herbs in Vietnam. In this study, Ngoc Linh ginseng callus was fermented by Lactobacillus plantarum ATCC 8014 (at 6, 7, and 8 log CFU/ml) to evaluate the extraction efficiency of bioactive compounds. The post-fermentation solution was spray-dried using maltodextrin with or without Stevia rebaudiana (3% and 6% v/v) as the wall material. Bioactive compounds such as polyphenols, polysaccharides, and total saponins, and L. plantarum viability during fermentation and after spray-drying, as well as under simulated gastric digestion, were evaluated in this study. The results showed that probiotic density had a significant effect on bioactive compounds, and L. plantarum at 8 log CFU/ml showed the best results with a short fermentation time compared to other tests. The total content of polyphenols, polysaccharides, and saponins reached 5.16 ± 0.18 mg GAE/g sample, 277.2 ± 6.12 mg Glu/g sample, and 4.17 ± 0.15 mg/g sample, respectively after 20 h of fermentation at the initial density of L. plantarum (8 log CFU/ml). Although there was no difference in the particle structure of the preparation, the microencapsulation efficiency of the bioactive compound in the samples containing S. rebaudiana was higher than that with only maltodextrin. The study also indicated that adding S. rebaudiana improved the viability of L. plantarum in gastric digestion. These results showed that S. rebaudiana, a component stimulating probiotic growth, combined with maltodextrin as a co-prebiotic, improved the survival rate of L. plantarum in simulated gastric digestion.

Protozoa population and carbohydrate fermentation in sheep fed diet with different plant additives

  • Objective: The aim of the study was to compare the effect of two plant additives, rich in polyphenolic compounds, supplemented to sheep diets on microorganisms and carbohydrate fermentation in rumen. Methods: In the experiment, 6 ewes of the Polish Mountain breed were fitted with ruminal cannulas. Sheep were divided into three feeding groups. The study was performed in a cross-over design of two animals in each group, with three experimental periods (n = 6 per each group). The animals were fed a control diet (CON) or additionally received 3 g of dry and milled lingonberry leaves (VVI) or oak bark (QUE). Additionally, plant material was analyzed for tannins concentration. Results: Regardless of sampling time, QUE diet increased the number of total protozoa, as well as Entodinium spp., Diplodinium spp. and Isotrichidae family, while decreased bacterial mass. In turn, a reduced number of Diplodinium spp. and increased Ophryoscolex spp. population were noted in VVI fed sheep. During whole sampling time (0, 2, 4, and 8 h), the number of protozoa in ruminal fluid of QUE sheep was gradually reduced as opposed to animals receiving CON and VVI diet, where rapid shifts in the protozoa number were observed. Moreover, supplementing sheep with QUE diet increased molar proportions of butyrate and isoacids in ruminal fluid. Unfortunately, none of the tested additives affected gas production. Conclusion: The addition of VVI or QUE in a small dose to sheep diets differently affected rumen microorganisms and fermentation parameters, probably because of various contribution of catechins in tested plant materials. However, it is stated that QUE diet seems to create more favorable conditions for growth and development of ciliates. Nonetheless, the results of the present study showed that VVI and QUE additives could serve as potential natural modulators of microorganism populations and, consequently, carbohydrate digestion in ruminants.

산업폐기물(産業廢棄物)의 비료화(肥料化)에 관(關)한 연구(硏究) II. 수도(水稻)에 대(對)한 주정오니(酒精汚泥) 비효시험(肥效試驗) (Use of Industrial Wastes as Sources of Organic Fertilizer II. Effect of Activated Sludge from Alcohol Fermentation on Rice)

  • 비료자원(肥料資源)으로서 유망시(有望視)되는 주정오이(酒精汚泥)의 비효(肥效) 및 시용적량(施用適量)을 구명(究明)하기 위(爲)하여 수도(水稻)에 시용(視用)한 결과(結果)는 다음과 같다. 주정오이(酒精汚泥)의 무기화(無機化)는 퇴비(堆肥)에 비(比)하여 비교적(比較的) 빨랐으며 수도생육량(水稻生育量) 및 식물체중(植物體中) N함량(含量)은 오이시용량(汚泥施用量)이 증가(增加)되었으나 수량(收量)은 1.600kg/10a부터 급격(急激)히 감소(減少)되어 수도(水稻)에서 오이(汚泥)의 적정시용량(適正施用量)은 800kg/10a로 추정(推定)되었다. 따라서 주정오이(酒精汚泥)는 유기질비료자원(有機質肥料資源) 및 토양개량제(土壤改良劑)로 활용(活用)이 가능(可能)할 것으로 판단(判斷)되었다.

Dihydrodaidzein production from soybean hypocotyl extract by human intestinal bacterium MRG-1

  • Phytoestrogenic S-equol production in human gut exclusively depends on the biotransformation of daidzein to dihydrodaidzein (DHD). With a growing demand for the DHD enriched biomaterials, the commercial soybean hypocotyl extract (SHE) was chosen as a substrate for the microbial DHD production by human gut bacterium MRG-1, anaerobic DHD producer. To optimize the production of DHD, anaerobic fermentation conditions, including sterilization time, growth stage of inoculum, and growth media, were investigated. Maximum DHD production (1.2 g/L) was achieved after 48 h incubation when 1% (w/v) of SHE in the 20-min-sterilized Gifu Anaeboic Medium media was inoculated with OD600 0.3-0.4 of MRG-1. This is the first report that crude soy biomaterial, instead of pure compounds, such as daidzin and daidzein, is utilized for the production of the DHD enriched biomaterial.

표고버섯 재배폐골목의 유기질 비료화 (Manufacturing of Organic Composts from Shiitake Bedlogs)

  • 본 연구는 표고버섯 재배폐골목 및 지조를 이용한 유기질비료 제조를 위한 적정 조건을 찾고, 제조한 유기질비료를 사용하여 식물의 생육 및 토양온도에 미치는 영향을 검토하기 위하여 실시하였다. 시멘트블록조 발효상을 사용하여 제조한 톱밥과 발효첨가제를 적량 첨가하여 퇴적후, 1 개월마다 뒤집기하면서 발효부숙시켜 표고버섯 재배폐골목 및 신선지조로부터 양질의 유기질비료를 제조할 수 있었다. 적정 제조조건으로서는 목질계 재료 1톤에 대하여 요소비료 12kg, 석회 25kg, 계분 40kg 및 적당량의 발효첨가제가 필요하였으며, 함수율은 55% 였다. 유식물 파종실험에서 유기질비료의 혼합량이 많아질수록 무우의 생장이 우수한 것으로 나타났으며, 유기질비료의 혼합율이 증가함에 따라 토양의 온도가 다소 높아지는 경향을 나타냈다. 유기질비료는 아침 외기의 온도가 내려가면, 토양의 온도를 높게 유지시키며, 외기의 온도가 높은 시간대는 오히려 토양의 온도를 낮추는 온도완충효과를 주었는데, 이러한 효과는 유기질비료의 혼합비가 높아질수록 더욱 커지는 것으로 나타났다.

유기성 폐기물 장기시용 후 토양에서 무 (Raphanus sativus cv. sodamaltari)의 중금속 흡수 (Uptake of Heavy Metals by Radish (Raphanus sativus cv. sodamaltari) from the Soils after Long-Term Application of Organic Wastes)

  • This study was carried out to understand the long-term effects of organic waste treatments on the fate of heavy metals in soils originated from the organic wastes and consequent uptake of heavy metals by plant, together with examination of changes in soil properties and plant growth performance. In this study, the soils treated with three different organic wastes (municipal sewage sludge, alcohol fermentation processing sludge, pig manure compost) at three different rates (12.5, 25.0, 50.0 ton $ha^{-1}yr^{-1}$) for 7 years (1994 - 2000) were used. To see the long-term effect, plant growth study and soil examination were conducted twice in 2000 and 2010, respectively. There was no additional treatments of organic wastes for 10 years after the organic waste treatment for 7 years. Compared to plant growth examination conducted in 2000 using radish (Raphanus sativus cv. sodamaltari), it appeared that height, root length and diameter, fresh weight of radish grown in 2010 decreased in the plots treated with municipal sewage sludge and alcohol fermentation processing sludge and that the extent of decrease was higher with increase of sludge application rates. On the other hand, pig compost treatment increased plant height, root length and diameter, fresh weight with increasing application rates. Cu and Pb concentrations in radish root and leaves increased in 2010 compared to those in 2000 while Ni concentrations in root and leaves decreased. Zn concentration was increased only in the soils treated with pig manure compost. Multiple regression analysis among heavy metal species fractions in soils, soil pH, and metal concentrations in radish root and leaves indicated that the metal uptake by radish was governed mainly by the soil pH and subsequent increase of available heavy metal fractions in soils with organic waste treatments.

사료 발효원으로 느타리버섯 종균이 흰점박이꽃무지 생육에 미치는 영향 (Role of oyster mushroom as an alternative fermentation source in the growth of white-spotted flower chafer(Protaetia brevitarsis seulensis))

  • 흰점박이꽃무지(Protaetia brevitarsis seulensis) 유충의 대체사료인 한약재 부산물 발효원인 유용미생물(Effective microorganism, EM)과 느타리버섯 종균(Oyster mushroom fungi, OM)을 비교하였다. 각 발효된 사료의 영양성분을 비교한 결과, 조회분을 제외한 조단백, 조지방, 조섬유 함량이 OM 발효사료에서 높게 나타났다. 각 발효된 사료로 사육된 흰점박이꽃무지 유충의 영양성분을 비교한 결과 차이가 관찰되지 않았다. 각 발효사료별 흰점박이꽃무지 유충의 생체중을 주별 비교 분석한 결과, 3주차 관찰시기부터 EM과 OM을 이용한 사료에서 흰점박이꽃무지 유충 평균중량이 유의하게 높았다. 유충 사육 시 생존율은 발효사료의 경우 동일하게 96.7%이나, 비발효사료의 경우 9.8%로 매우 낮았다. 본 실험결과, 흰점박이꽃무지의 생육에 먹이 원의 발효는 꼭 필요했으며, OM은 EM을 대체할 수 있는 흰점박이꽃무지 대체사료의 발효원으로 더 안정적이었다.
