This study was performed to establish a production system for in situ and ex situ conservation of Elsholtzia minima Nakai, an endemic plant grown in Jeju Island. Moreover, this study aimed to identify root-growth characteristics according to the use of pre-treatment agents and seedling growth effect according to fertilizer application. The mean temperature was similar in greenhouse and vinyl-moist chamber, but air humidity was higher in vinyl-moist chamber than in greenhouse. After stem planting of Elsholtzia minima Nakai, initial root growth was observed after 10 days in greenhouse and after 7 days in vinyl-moist chamber. Root growth rate was more rapid in vinyl-moist chamber. Moreover, survival rate, root growth rate and root number was slightly higher in vinyl-moist chamber than in greenhouse, indicating that vinyl-moist chamber is more effective in plant growth. When pre-treatment agents were used to remove root growth-inhibiting substances, a higher root growth rate of more than 95% was found in pre-treatment groups, excluding the group treated with AgNO3 at 77.5%. Thus, Elsholtzia minima Nakai is thought to have less root growth inhibitors. In the analysis of nitrogen application rate and Osmocote application by seedling container, a difference was found in survival rate and growth according to application rate and container conditions. When Osmocote, a slow release fertilizer, was applied to the soil surface around each culture container, survival rate and the growth of aerial and root parts were most favorable. Thus, Osmocote fertilizer is thought to be desirable for seedling propagation of Elsholtzia minima Nakai.
To study the effect of global warming on the growth of plants and plant populations throughout their life cycle under a field-like condition, we constructed a Temperature Gradient Chamber (TGC) in Tsukuba, Japan. The chamber had slender shape : 30 m long. 3 m wide, and 2.5 m high. That satisfactory performance was confirmed by a test throughout all seasons in 1998: the projected global warming condition in the near future was simulated. That is, independent of a great daily or seasonal change in ambient meteorological conditions, air temperatures at the air outlet were warmed 5$^{\circ}C$ higher than those at the ambient (the annual mean was 14.3$^{\circ}C$) with precision of ${\pm}$0.2$^{\circ}C$ (the annual means were 19.2$^{\circ}C$) with a rising rate of approximately 1$^{\circ}C$ every 5 m. This chamber will enable us to study the effects of global warming on growth of plants and plant populations because their abilities to control air temperature are excellent. TGC is expected that it would be utilized for studying the effect of global warming on plant growth under natural weather conditions.
This study was conducted to analyze the variations of air temperature, relative humidity and pressure in a low pressure chamber for plant growth. The low pressure chamber was composed of an acrylic cylinder, a stainless plate, a mass flow controller, an elastomer pressure controller, a read-out-box, a vacuum pump, and sensors of air temperature, relative humidity, and pressure. The pressure leakage in the low pressure chamber was greatly affected by the material and connection method of tubes. The leakage rate in the low pressure chamber with the welding of the stainless tubes and a plate decreased by $0.21kPa{\cdot}h^{-1}$, whereas the leakage in the low pressure chamber with teflon tube and rubber O-ring was given by $1.03kPa{\cdot}h^{-1}$. Pressure in the low pressure chamber was sensitively fluctuated by the air temperature inside the chamber. An elastomer pressure controller was installed to keep the pressure in the low pressure chamber at a setting value. However, inside relative humidity at dark period increased to saturation level.. Two levels (25 and 50kPa) of pressure and two levels (500 and 1,000sccm) of mass flow rate were provided to investigate the effect of low pressure and mass flow rate on relative humidity inside the chamber. It was concluded that low setting value of pressure and high mass flow rate of mixed gas were the effective methods to control the pressure and to suppress the excessive rise of relative humidity inside the chamber.

To test the quality change of seedlings of three domestic medicinal plants raised in plug trays, a short term storage experiment was conducted. Seedlings were kept in growth chambers for two weeks at 4 or $8^{\circ}C$ temperature combined with 0 or $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. Quality of glasshouse-raised seedlings was assessed after two weeks of cold storage in the growth chamber and one week of acclimation in the greenhouse. After two weeks of storage in the growth chamber of Perilla frutescens var. acuta Kudo, plant height was the greatest in the treatment $8^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. Internode length of P. frutescens var. acuta Kudo was the greatest in the treatment of $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. After one week of acclimatization in a glasshouse, the growth and development, such as plant height, internode length and leaf size, were greater in the $8^{\circ}C$ combined with $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD than in the other treatments. After two weeks of storage in the growth chamber of Sophora tonkinensis, plant height increased more in the treatment of $4^{\circ}C$ than $8^{\circ}C$. After one week of acclimatization in a glasshouse, number of leaves did not change in the treatment of $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD, but it increased in the other treatments. Leaf width increased more under the dark than light condition. Leaf length did not observably change in any treatments. After two weeks of storage in the growth chamber, plant height of Angelica gigas Nakai was the greatest in the treatment of $8^{\circ}C$. Number of leaves was the greatest in the treatment of $8^{\circ}C$. Leaf growth was greater under dark than light condition. These results suggested that optimal storage environment was $8^{\circ}C$ combined with $5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD for P. frutescens var. acuta Kudo, and $4^{\circ}C$ combined with $0{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD for S. tonkinensis and A. gigas Nakai. Hence, proper combination of temperature and PPFD were necessary for better storage, and acclimatization and growth, thereafter, of the plug seedlings of theses plant species.
Studies were carried out to standardize and develop a suitable macro-propagation technology for large scale production of superior clonal stock through stem cuttings in Commiphora wightii Arnott (Bhandari), a data deficient medicinal plant of arid region. For the purpose, three experiments were conducted. The first experiment was tried to elucidate the impact of various cutting diameters (0.50-0.75 cm, 0.75-1.00 cm, 1.00-1.50 cm, and >1.50 cm) in combination with varying growing conditions (sunlight, shade house and mist chamber) on shoot sprouting and rooting without using exogenous plant growth regulators. Cutting diameter (size 0.75-1.00 cm) in mist chamber has shown maximum sprouting (90.00%) and rooting (73.33%), primary root (6.67) and secondary root (16.67) followed by 1.00-1.51 cm in mist chamber. Minimum sprouting (40.00%), rooting (33.33%), number of shoot (1.33), primary root (1.00) and number of secondary root (1.00) was recorded in cutting diameter (size >1.50 cm) in sunlight. Second experiment was performed to find out optimum growth regulator concentration of rooting hormone (100, 200, 500 and 1000 ppm) of Indole-3-acetic acid (IAA) and Indole-3-butyric Acid (IBA) on adventitious root formation on cuttings diameter (size 0.25-0.50 cm) in comparison to control. Maximum rooting percentage (93.33%) was recorded in 200 ppm followed by 500 ppm (86.66%) of IBA as compared to control, which showed only 60 per cent sprouting. Third experiment was performed with newly formed juvenile micro-cuttings treated with varying concentrations of IAA and IBA. The juvenile cuttings (size 6-10 cm, basal dia <0.25 cm) were selected as micro-cuttings. The cuttings treated with IBA (500 ppm) showed 64.30% rooting as compared to other treatments. Results of above experiments indicate that cuttings (size 0.75-1.00 cm dia) may be developed in mist chamber for better performance. While using heavier cuttings, no growth promoting hormones is required however; growth regulator 200 ppm concentration of IBA rooting hormone was observed optimum for promoting macro-propagation in stem cuttings of lower diameter class (0.25-0.50 cm).
This study was carried out to investigate optimum wind velocity for growth of Pleurotus ostreatus. In the chamber experiments, the divergence of pileus was $63^{\circ}$ in 0.3m/s, and it showed the tendency that the more the air velocity was increased, the more the divergence became small. And the ratio of commercial yields was 96% in the air velocity of 0.3m/s. In the cultivation room experiments, the divergence of pileus was $64^{\circ}$ in 0.3m/s, and it was the tendency that the more the air velocity was fast, the more the divergence became larger. And the commercial yields was similar to the trend of the chamber experiment as 94.8% in 0.3m/s, and the quality characteristics of the fruit-body showed the uniform quality in comparison with other treatment because the standard deviation of the size of the pileus, etc. was the lowest in 0.3m/s like the chamber experiment.

Plants are grown under constitutive changing of environmental conditions and response to external conditions at both protein and transcription level. The effects of heat on plant growth are broad and influence the yield directly. Heat stresses could be classified depend on intensity and duration. Fundamental changes of growth condition by climate change maybe or maybe not classified as a stress on plant growth. The effects of a short and unanticipated impact of elevated heat on plant could be different with those of under longer extension of ambient heat. To examine differently expressed gene sets by ambient heat stress of soybean, we grow the soybean in normal condition for three weeks. After that, soybean plants move to growth chamber. The temperature of growth chamber increase up to $9^{\circ}C$ for four days. We have extracted mRNA and micro RNA every 24 hours and carried RNA sequence analysis. We found major metabolic pathways affected by ambient heat stress. Mainly carbon metabolism, translation machinery and amino acid synthesis are affected. We discussed the expression patterns of genes of heat sensing and hormone responses.

This is the fundamental research on restoring damaged vegetation areas in the vicinity of DMZ using local native plant species. This research is aimed at identifying effective plant growth regulators (PGR) for seed germination and seedling growth of mountain mulberry, and developing effective methods for managing its germination and growth. Mountain mulberry seeds were collected from the regions in the DMZ vicinity, and tests with seven treatments using four PGRs including $GA_3$, kinetin, ABA and NAA, were conducted. The germination rate was calculated in two different ways of both in a growth chamber and in a greenhouse after seven days observation, and the growth characters such as leaf width/length, seedling width/length and fresh/dry weight, have been surveyed in a greenhouse for three months. Although in the growth chamber the highest germination rate was shown at a group with the kinetin treatment, it was not significantly different to that of the control group. Groups with ABA or NAA presented relatively low germination rates. As for the greenhouse test, the germination rates of all groups ranged 20~30% without significant difference each other, the reason of which might be due to low absorption by the effect of a soil drench method used in this study. The entire growth characters with the treatments of $2.15mg{\cdot}L^{-1}$ of kinetin and $10mg{\cdot}L^{-1}$ of $GA_3$ were significantly different to the control. NAA treatment only showed better growth of seedling width compared to the control. Consequently, the most effective PGRs for the germination and growth of mountain mulberry near the DMZ was kinetin. Further research on examining the most effective concentration of them was needed.
This experiment was conducted to investigate the effect of transfer date to a growth chamber and low temperature on growth and flowering of Jeffersonia dubia for free control of flowering period. According to transfer date, any plant did not sprout when plants were put in a growth chamber from Aug. 20 to Dec. 20. However, all the plants which were put in a growth chamber on Jan. 20 or Feb. 20 sprouted and resulted in flowering. The effect of beginning time and period of low temperature treatment were also observed. At the beginning time of Aug. 20, sprouting and flowering occurred as plant received more than 60 days of low temperature treatment. However, almost of al l the plants sprouted and flowered by only 30 days of low temperature treatment when the low temperature was given after Sep. 20. 60 days of low temperature given again after about three month growing period after dormancy breaking, resulted in resprouting of all plants and 70% of them flowered in this experiment.
