Didymella gigantis sp. nov. Causing Leaf Spot in Korean Angelica

  • During a disease survey in October 2019, leaf spot symptoms with a yellow halo were observed on Korean angelica (Anglica gigas) plants grown in fields in Pyeongchang, Gangwon Province, Korea. Incidence of diseased leaves of the plants in the investigated fields ranged from 10% to 60%. Morphological and cultural characteristics of two single-spore isolates from the leaf lesions indicated that they belonged to the genus Didymella. Molecular phylogenetic analyses using combined sequences of LSU, ITS, TUB2, and RPB2 regions showed distinct clustering of the isolates from other Didymella species. In addition, the morphological and cultural characteristics of the isolates were somewhat different from those of closely related Didymella spp. Therefore, the novelty of the isolates was proved based on the investigations. Pathogenicity of the novel Didymella species isolates was confirmed on leaves of Korean angelica plants via artificial inoculation. This study reveals that Didymella gigantis sp. nov. causes leaf spot in Korean angelica.

Trends in energy intake among Korean adults, 1998-2015: Results from the Korea National Health and Nutrition Examination Survey

  • BACKGROUND/OBJECTIVES: Assessing changes in energy intake and dietary sources is important to understand trends in the prevalence of obesity. Thus, we examined trends in energy intake and its nutrient and food sources in Korean adults from 1998 through 2015. SUBJECTS/METHODS: This study included 70,769 subjects aged ${\geq}19$ years who completed a nutrition survey. Subject data were obtained from the 1998, 2001, 2005, 2007-2009, 2010-2012, and 2013-2015 Korea National Health and Nutrition Examination Surveys. Dietary intake was assessed by a 1-day 24-hour recall method. RESULTS: In men, the daily energy intake significantly increased from 2,196 kcal in 1998 to 2,489 kcal in 2013-2015 (P for trend < 0.0001). However, the daily energy intake among women did not change significantly over the same period (P for trend = 0.5772). The percentages of energy intake from animal foods (e.g., meat and milk) and beverages increased during the study period in both men and women. However, the percentage of energy intake from plant foods decreased due to a marked decrease in the intake of white rice. Changes in food sources of energy intake led to changes in the nutrient sources of energy intake; for example, the increase of energy intake from fat and decrease of energy intake from carbohydrate. CONCLUSIONS: This study suggests that since 1998, energy intake has increased among Korean adult men, but not among women. However, the composition of food and nutrient sources of energy intake has changed in both men and women. Energy intake and its nutrient and food sources should continue to be monitored regularly in the Korean adult population.

Brown Rot of Apricot and Mume Caused by Phomopsis vexans

  • Brown rot symptoms severely occurred on fruits of apricot and mume grown in Changnyeong, Suwon and Yeongi in Korea during a disease survey in June 2002. A total of 32 isolates of Phomopsis sp. was obtained from the fruit rot symptoms. All isolates were identified as Phomopsis vexans based on their morphological and cultural characteristics. Four isolates of the fungus were tested for pathogenicity to fruits of apricot and mume by artificial inoculation. All isolates induced brown rot symptoms on the fruits of apricot and mume by wound inoculation. Unwounded inoculation also induced symptoms on mume fruits but not on apricot fruits. The symptoms on the fruits induced by artificial inoculation were similar to those observed in the orchard. The pathogenicity tests revealed that mume was more susceptible to the pathogen than apricot. This is the first report of brown rot of apricot and mume caused by P. vexans.

Testing for Detection of Xanthomonas campestris pv. campestris in Crucifer Seeds and Seed Disinfection (십자화과 채소종자의 검은빛썩음병 감염검정 및 종자소독)

  • Total 29 commercial crucifer seed lots were tested for seed transmission of Xanthomonas campestris pv. campestris by seed washing liquid plating assay. One imported cabbage seed lot was found to carry Xanthomonas campestris pv. campestris. Several methods and chemicals for eradication of Xanthomonas campestris pv. campestris in and on the cabbage seed were tested for effectiveness. Soaking cabbage seed in $3\3%$ hydrogen peroxide solution for 30 minutes effectively eradicated Xanthomonas campestris pv. campestris in a naturally infested seed lot. In a field survey, black rot turned out to be an important disease in cabbage in Korea.

  • A total of 65 isolates of Penicillium were isolated from decayed post-harvest fruits of apple pear citrus and grape. The Penicillium species isolated from the apple were idnetified as P. aurantiogriseum and P. expansum those from the pear were P. crustosum and P. expansum and those from the grape were P. aurantiogriseum and P. expansum, From decayed citrus fruits. P. digitatum and P. italicum were isolated. Citrinin and patulin from these species in the YES(yeast extract sucrose) broth were extracted with ethyl acetate and purified by thin-layer chromatography(TLC) and high performance liquid chromatography(HPLC) Among 51 isolates of Penicillium from apple pear and grape 7 isolates produced citrinin 13 isolates produces patulin and 12 isolates produced citrinin and patulin also. All 14 isolates of Penicillium from citrus produced only patulin.

Distribution of Bacterial Angular Leaf Spot of Strawberry and Characterization of Xanthomonas fragariae Strains from Korea (한국의 딸기세균모무늬병 발생분포 및 딸기세균모무늬병균 특성조사)

  • Nationwide survey for angular leaf spot (ALS) of strawberry caused by Xanthomonas fragariae, a quarantine disease in Korea, was performed in November 2012. In the survey, ALS was observed in eighty eight farmers' fields of Sukok, Jinju and Okjong, Hadong in Gyeongnam Province, and one field in Namwon of Jeollabuk Province. The infected field of Namwon closed immediately after the disease diagnosed ALS. In detailed survey of Sukok and Okjong areas during February 2012 to January 2015, ALS occurrence decreased from 45% farmer's fields on December 2012 to 5% on January 2015, and from 38% on November 2013 to 5% on January 2015, respectively. Phenotypic characteristics of the Korean strains were similar to those of the type strain of X. fragariae. A multilocus sequence analysis of Korean strains of X. fragariae was conducted using four genes; dnaK, fyuA, gyrB, and rpoD. All the Korean strains had the same sequences of the four genes. The concatenated sequences of the Korean strains shared 100% with that of the type strain of X. fragariae. All strawberry cultivars tested were susceptible to the strains of X. fragariae two weeks after inoculation. The inoculated sites were necrosis and expanded, which were rated 4 based on evaluation of inoculation site.

Ecological Characteristics of Pine Wilt Disease affected areas in Andong city, Korea (안동시 소나무재선충병 피해지에 대한 생태학적 특성)

  • A field survey was carried out targeting 59 pine wilt disease control areas distributed in Andong, and the ecological characteristics of the areas affected by pine wilt disease were analyzed using vegetation information, ecological information, and land use information. Vegetation characteristics of Pine wilt disease affected forest area showed a 3-layer vegetation structure, high percentage of accidental occurrence species (37%) and secondary vegetation species (59.6%), appearing 12 taxa naturalized plants and 3 taxa ecosystem disturbance organisms designated by the Ministry of Environment. Ecological information of Pine wilt affected area showed frequent occurrence of water stress in south and west slopes, low lying grounds in mountains, and in well-drained soils. Also, surrounding the area has been used as roads, tombs, and cultivation where intensive human activities were the cause of disturbance and stress to the pine forest. It was analyzed that the pine forest in Andong city suffered extensive damage due to the onset of pine wilt disease while the pine trees were weakened due to continuous human activities. Conclusively, the spread and onset of pine wilt disease are worsened by artificial factors than natural environmental conditions.

Investigation on the Management Status of Pear and Apple Orchards Where Fire Blight Disease Was Partially Controlled in Korea (국내 과수화상병을 부분 방제한 배와 사과 과원의 관리 현황 조사)

  • Recently, the domestic plant disease control policy for fire blight has been implemented partial control in addition to burial control. In this study, an on-site management survey was conducted targeting orchards that implemented partial disease control from 2019 to 2020 in order to find efficient implementation methods for partial disease control. As a result of an investigation into 22 pear and apple orchards in Cheonan and Chungju, 7 orchards were buried. The upper part of the cut infected plants was burned at 16 orchards and covered with plastic vinyl after lime treatment at 6 orchards. The lower stumps of cut infected plants were burned at 7 orchards and covered with plastic vinyl after lime treatment at 15 orchards. There were two orchards where suckers appeared on the stumps even though covers were applied. There was no infection by Erwinia amylovora in the suckers. The conservation condition of lime treatment was good, but warning signs were absent at 6 orchards. Most orchards treated the stumps and surrounding areas with glyphosate-isopropylamine herbicide. The effect of partial control was judged to be safe.

Diaporthe phaseolorum var. caulivora, a Causal Agent for Both Stem Canker and Seed Decay on Soybean

  • Northern stem canker caused by $Diaporthe$ $phaseolorum$ var. $caulivora$ ($Dpc$) has become a serious disease in soybean. The objectives of this study were to survey the existence of $Dpc$ on soybean in Korea, and to examine the potential pathogenicity of $Dpc$ in seed decay. One such isolate, SSLP-4, isolated from a field-grown plant of the Korean soybean cultivar Danbaekkong, was identified as $Dpc$, based on its morphological and molecular characteristics by sequences of internal transcribed spacer (ITS), translation elongation factor (TEF) 1-${\alpha}$ and ${\beta}$-tubulin regions, as well as pathogenic analyses. Moreover, morphological and molecular analyses revealed that isolate SSLP-4 was nearly identical to $Dpc$ strains from the United States. Pathogenicity tests on hypocotyls of soybean seedlings and detached leaves resulted in typical symptoms of soybean northern stem canker and inoculation on plants at R5-R7 stage caused seed decay. All results suggest that the $Dpc$ strain SSLP-4 can cause both stem canker and seed decay on soybean. Thus, the SSLP-4 isolate has the potential to contribute greatly to understanding of host plant resistance mechanisms, both at vegetative and reproductive growth stages in soybean.

Occurrence of Major Diseases in Vegetable Growing under the Furnihsed Condition in Southern Part of Korea (남부 시설원예지대의 주요병 발생생태에 관한 연구)

  • The study was conducted to obtain the basic information on the diseases epidemics of vegetable crops grown in Namji, Jinju, Gimhae and Suncheon under the extremely varied and specified conditions, plastic film house. The disease survey was conducted from the end of April, 1951 to April 1982. Leaf mold and late blight were serious foliar disease in tomato during the seedling stage, especially when the infected seeds were sown. The diseases increased rapidly 35 days after seeding. In both continuous cultivation of cucumber and rotation with upland crops, incidence of Fusarium wilt was severe while incidence of the disease was negligible in cultivations after paddy rice or grafting on pumpkin. Downy mildew of cucumber was severe in Jinju and Suncheon area, however, it was not so serious in Namji area where the growing season of cucumber was unfavorable for the maximum disease incidence. Cucumber mosaic virus disease was prevalent in the areas surveyed and the disease incidence was increased rapidly after June. Powdery mildew prevailed at the early stage of cucumber growth after transplanting in Namji area. Root rot and wilting caused by Phytophthora capsici was as the most destructive disease in pepper grown under the vinyl house, especially in Namji and Jinju area where the pepper has been cultivated intensively. The Phytophthora attacked most parts of young plants during the winter time and then induced crown rot on the adults plants. Cultivation of pepper in vinyl house was almost impossible because of the Phytophora disease by the end of June. Virus diseases to tomato plants were prevalent throughout the surveyed area and the damage was also severe. In Jinju and Gimhae area leaf mold and late blight showed high infection rate in tomato during the harvest time.
