Effect of dark incubation in germination of indirect date palm somatic embryos and conversion into plantlets

  • All studies on date palm somatic embryogenesis have focused on germination in the presence of light while neglecting germination in darkness, which mimics the germination process of zygotic embryos within seeds. To improve the date palm micropropagation protocol, we investigated the effects of light and darkness incubation on the germination of indirect date palm somatic embryos and their subsequent conversion into plantlets. Darkness incubation emerged as a pivotal factor in the germination of indirect date palm somatic embryos and their successful conversion into plantlets. Darkness incubation significantly decreased the time required for the conversion of indirect somatic embryos into plantlets, halving the duration from 24 weeks to only 12 weeks. The micropropagation protocol was modified, consolidating the previous two distinct stages of germination and elongation under light incubation into a single stage under darkness incubation. These findings modified the protocol and significantly reduced the overall duration of the date palm micropropagation protocol.

  • One of major research issues in cellular manufacturing is studing factors that are fovorable or detrimental to the conversion of traditional functional layouts to GT cellular layouts. Among many factors, this paper explored plausible relationships between the scope of plant conversion and the manufacturing cell design processes and outcomes. The cell design practices of 28 U. S. plants were surveyed through a mail questionaire. While most relationships were not statistically significant, some interesting findings and insights could be drawn. With this researhc, we can better understand a part of relationships between the company's conversion strategy and the cell design strategies and outcomes.

  • Effect of Ca2+ on the conversion of 1-aminocycloprophane-1-carboxylic acid (ACC) to ethylene was studied with 2.5 day-old mung bean hypocotyl segments. The conversion of ACC in these tissues was inhibited by plasmolysis and sulfosuccinimidyl (hydroxyphenyl) propionate (sulfo-SHPP). The ACC induced ethylene production in HC (high calcium)-tissue grown on the Ca2+ added medium was greater than that in N (normal)-tissue. HC-tissue had a lower inhibition rate of ACC conversion by EGTA and Ca2+ -channel blockers than N-tissue. The rates of the ACC conversion by both kinds of tissues were stimulated by the Ca2+ ionophore A23187. From these results, we suggests a mechanism for the stimulative effect of Ca2+ on the conversion of ACC to ethylene as follows; in some tissues where ACC conversion is linked with plasma membrane, Ca2+ may be transported from apoplast through Ca2+ -channel into the cytoplasm ad stimulate ACC-oxidase activity.

  • Ocean thermal energy conversion (OTEC) is an effective method of power generation, which has a small impact on the environment and can be utilized semi-permanently. This paper describes a dynamic model for a pilot OTEC plant built by the Institute of Ocean Energy, Saga University, Japan. This plant is based on Uehara cycle, in which binary mixtures of ammonia and water is used as the working fluid. Some simulation results attained by this model and the analysis of the results are presented. The developed computer simulation can be used to actual practice effectively, such as stable control in a steady operation, optimal determination of the plant specifications for a higher thermal efficiency and evaluation of the economic prospects and off-line training for the operators of OTEC plant.

Re-evaluation on conversion system of BactoScan and BactoCount for raw milk in South Korea (국내 원유 세균수 검사장비 박토스캔 및 박토카운트의 보정식 재평가)

  • The total bacteria count is significantly important factor for hygienic quality in raw milk. BactoScan FMTM and BactoCount IBCTM are the automated instruments for the determination of the total bacterial count in raw milk. They have been used after calibration by standard plate count (SPC) method in South Korea since 2000. It is necessary to re-evaluate for total bacterial counter according to the improvement of milk quality and the change of milk quality grade. Therefore, this study was evaluated the conversion mode of the two machines by SPC method. We collected 921 bulk-tank milk samples throughout the concentration range of 1,000~1,000,000 CFU/mL from June 2020 to October 2021. As a result, when compared by the SPC value, there was a slight difference in total bacterial count in BactoScan below 10,000 CFU/mL and above 200,000 CFU/mL and in BactoCount above 100,000 CFU/mL, respectively. Therefore, the conversion factor for BactoScan and the conversion equation for BactoCount were newly adjusted based on SPC value, and then the correlation coefficients (R2) was 0.85 or higher. In addition, the correlation (R2) between BactoScan and BactoCount was 0.91, which means the results were high positive correlation. These results are expected to contribute to improving the accuracy of the automated instruments for determining of total bacterial count in raw milk.

  • This paper describes the methodology of calculating the internal dose conversion coefficient in order to assess the radiological impact on non-human species. This paper also presents the internal dose conversion coefficients of 25 radionuclides ($^3H,\;^7Be,\;^{14}C,\;^{40}K,\;^{51}Cr,\;^{54}Mn,\;^{59}Fe,\;^{58}Co,\;^{60}Co,\;^{65}Zn,\;^{90}Sr,\;^{95}Nb,\;^{99}Tc,\;^{106}Ru,\;^{129}I,\;^{131}I,\;^{136}Cs,\;^{137}Cs,\;^{140}Ba,\;^{140}La,\;^{144}Ce,\;^{238}U,\;^{239}Pu,\;^{240}Pu$) for domestic seven reference animals (roe deer, rat, frog, snake, Chinese minnow, bee, and earthworm) and one reference plant (pine tree). The uniform isotropic model was applied in order to calculate the internal dose conversion coefficients. The calculated internal dose conversion coefficient (${\mu}Gyd^{-1}$ per $Bqkg^{-1}$) ranged from $10^{-6}$ to $10^{-2}$ according to the type of radionuclides and organisms studied. It turns out that the internal does conversion coefficient was higher for alpha radionuclides, such as $^{238}U,\;^{239}Pu$, and $^{240}Pu$, and for large organisms, such as roe deer and pine tree. The internal dose conversion coefficients of $^{239}U,\;^{240}Pu,\;^{238}U,\;^{14}C,\;^3H$, and $^{99}Tc$ were independent of the organism.

Conversion of organic residue from solid-state anaerobic digestion of livestock waste to produce the solid fuel through hydrothermal carbonization

  • The solid-state anaerobic digestion (SS-AD) has promoted the development and application for biogas production from biomass which operate a high solid content feedstock, as higher than 15% of total solids. However, the digested byproduct of SS-AD can be used as a fertilizer or as solid fuel, but it has serious problems: high moisture content and poor dewaterability. The organic residue from SS-AD has to be improved to address these problems and to make it a useful alternative energy source. Hydrothermal carbonization was investigated for conversion of the organic residue from the SS-AD of livestock waste to solid fuels. The effects of hydrothermal carbonization were evaluated by varying the reaction temperatures within the range of $180-240^{\circ}C$. Hydrothermal carbonization increased the calorific value through the reduction of the hydrogen and oxygen contents of the solid fuel, in addition to its drying performance. Therefore, after the hydrothermal carbonization, the H/C and O/C atomic ratios decreased through the chemical conversion. Thermogravimatric analysis provided the changed combustion characteristics due to the improvement of the fuel properties. As a result, the hydrothermal carbonization process can be said to be an advantageous technology in terms of improving the properties of organic waste as a solid-recovered fuel product.

  • A electric power conversion system in a pumped storage power plant is important equipment for converting electric motor kinetic energy into electric power. A electric power conversion monitoring system consists of high voltage thyristor firing equipment, fault detection module, data gathering module, real time data processing equipment and man machine interface system. This paper describes electric power conversion system overview, the developed SFC monitoring system configuration including system characteristics, and successful application result to San-Cheong pumped storage power plant.

  • In this paper, the similarity criterion and dimensionless conversion method combined with the elasticity condition and Hooke's law are used to derive the functional relationship of the maximum effective value of the vibration velocity between the prototype pump and the model pump. The seawater circulation pump of a nuclear power plant is used as the prototype pump, and the model pump is obtained by performance conversion and choosing the appropriate scale, and the vibration state of the model pump under different flow rates is measured and analyzed. The vibration data of the model pump through the function relationship to find out the vibration parameters of the prototype model pump, and compare with the vibration data of the seawater circulation pump in reality. It can be seen that with the increase of flow rate, the maximum effective value of the vibration velocity of both model and prototype decreases and then increases, and the relative error is small, the maximum value is 7.7757%. Therefore, it can be considered that the functional relationship of model pump converted to prototype pump derived in this paper can be used to analyze the vibration of the actual seawater circulation pump of coastal nuclear power plant.
