Construction of a Genetic Linkage Map in Radish(Raphanus sativus L.) Using RAPD Markers (RAPD 마커를 이용한 무의 유전자지도 작성)

  • Genetic map and molecular marker have a great importance in improving and facilitating crop breeding program as well as in genome analysis and map-based cloning of genes representing desirable characters. This study aimed at developing RAPD markers and constructing a genetic linkage map using 82 BC$_1$F$_1$individuals originated from the cross between '835' and B$_2$in radish (Raphanus sativus L.). One of the parents for genetic linkage map construction, '835'(P$_1$) of egg type is susceptible to Fusarium wilt and have medium resistance to virus infection and the other parent, B$_2$(P$_2$) of round type, is susceptible to Fusarium wilt and virus, Screening of 394 RAPD primers in BC$_1$F$_1$) population resulted in selecting 128 polymorphic markers which displayed 1:1 segregation pattern. Two markers failed to display 1:1 segregation and showed the segregation ratio skewed to maternal genotype. Selected markers were categorized into 14 linkage group based on LOD score represented by MAPMAKER/EXP program. Five groups composed of single marker among them were excluded from the linkage map, and consequently, the remaining groups are well matched with the number of radish chromosome (n=9). The linkage map constructed with 128 markers covers 1,688.3 cM and the average distance between markers was 13.8 cM. For developing STS marker, we determined the partial nucleotide sequence of OPE10 marker at both ends and designed a oligonucleotide primer pair based on this sequence. STS PCR using the primer pair displayed a single, clear band of which segregation is perfectly matched with that of OPE10 marker. This implies that RAPD markers could readily convert into clear and reliable STS markers.

Distributional Characteristics and Evaluation of the Population Sustainability, Factors Related to Vulnerability for a Polygonatum stenophyllum Maxim. (층층둥굴레(Polygonatum stenophyllum Maxim.)의 분포특성과 개체군의 위협요인 및 지속가능성 평가)

  • Plants interact with various biotic and abiotic environmental factors. It requires much information to understand the traits of a plant species. A shortage of information would restrict the assessment, especially in the evaluation of what kind of factors influence a plant species to face extinction. Polygonatum stenophyllum Maxim. is one of the northern plants of which Korea is the southern distribution edge. The Korean Ministry of Environment had designated it to be the endangered species until December 2015. Although it is comparatively widespread, and a large population has recently been reported, it is assessed to be vulnerable due to the low population genetic diversity. This study evaluated the current distribution of Polygonatum stenophyllum Maxim. We investigated the vegetational environment, population structures, phenology, soil environment, and self-incompatibility based on the results. Lastly, we evaluated the current threats observed in the habitats. The habitats tended to be located in the areas where the masses at the edge of the stream accumulated except for those that were located on slopes of some mountainous areas. Most of them showed a stable population structure and had re-established or recruited seedlings. Polygonatum stenophyllum Maxim. had the difference in time when the shoots appeared above the ground depending on the depth of the rhizome located in the underground. In particular, the seedlings and juveniles had their rhizome located shallow in the soil. Visits by pollinator insects and success in pollination were crucial factors for bearing of fruits by Polygonatum stenophyllum Maxim. The threats observed in the habitat of Polygonatum stenophyllum Maxim. included the expansion of cultivated land, construction of new buildings, and construction of river banks and roads. Despite such observed risk factors, it is not likely that there would be rapid population reduction or extinction because of its widespread distribution with the total population of more than 2.7 million individuals and the new populations established by the re-colonization.

Wind and Flooding Damages of Rice Plants in Korea (한국의 도작과 풍수해)

  • The Korean peninsular having the complexity of the photography and variability of climate is located within passing area of a lots of typhoon occurring from the southern islands of Philippines. So, there are various patterns of wind and flooding damages in paddy field occuring by the strong wind and the heavy rain concentrated during the summer season of rice growing period in Korea. The wind damages to rice plants in Korea were mainly caused by saline wind, dry wind and strong wind when typhoon occurred. The saline wind damage having symptom of white head or dried leaves occurred by 1.1 to 17.2 mg of salt per dry weight stuck on the plant which was located at 2. 5km away from seashore of southern coastal area during the period(from 27th to 29th, August, 1986) of typhoon &Vera& accompanying 62-96% of relative humidity, more than 6 m per second of wind velocity and 22.5 to 26.4$^{\circ}C$ of air temperature without rain. Most of the typhoons accompanying 4.0 to 8. 5m per second of wind and low humidity (lesp an 60%) with high temperature in the east coastal area and southen area of Korea. were changed to dry and hot wind by the foehn phenomenon. The dry wind damages with the symptom of the white head or the discolored brownish grain occurred at the rice heading stage. The strong wind caused the severe damages such as the broken leaves, cut-leaves and dried leaves before heading stage, lodging and shattering of grain at ripening stage mechanically during typhoon. To reduce the wind damages to rice plant, cultivation of resistant varieties to wind damages such as Sangpoongbyeo and Cheongcheongbyeo and the escape of heading stage during period of typhoon by accelerating of heading within 15th, August are effective. Though the flood disasters to rice plant such as earring away of field, burying of field, submerging and lodging damage are getting low by the construction of dam for multiple purpose and river bank, they are occasionally occurred by the regional heavy rain and water filled out in bank around the river. Paddy field were submerged for 2 to 4 days when typhoon and heavy rain occurred about the end of August. At this time, the rice plants that was in younger growing stage in the late transplanting field of southern area of Korea had the severe damages. Although panicles of rice plant which was in the meiotic growing stage and heading stage were died when flooded, they had 66% of yield compensating ability by the upper tilling panicle produced from tiller with dead panicle in ordinary transplanting paddy field. It is effective for reduction of flooding damages to cultivate the resistant variety to flooding having the resistance to bacterial leaf blight, lodging and small brown planthopper simultaneously. Especially, Tongil type rice varieties are relatively resistant to flooding, compared to Japonica rice varieties. Tongil type rice varieties had high survivals, low elongation ability of leaf sheath and blade, high recovering ability by the high root activity and photosynthesis and high yield compensating ability by the upper tillering panicle when flooded. To minimize the flooding and wind damage to rice plants in future, following research have to be carried out; 1. Data analysis by telemetering and computerization of climate, actual conditions and growing diagnosis of crops damaged by disasters. 2. Development of tolerant varieties to poor natural conditions related to flooding and wind damages. 3. Improvement of the reasonable cropping system by introduction of other crops compensating the loss of the damaged rice. 4. Increament of utilization of rice plant which was damaged.

A prognosis discovering lethal-related genes in plants for target identification and inhibitor design (식물 치사관련 유전자를 이용하는 신규 제초제 작용점 탐색 및 조절물질 개발동향)

  • New technologies will have a large impact on the discovery of new herbicide site of action. Genomics, combinatorial chemistry, and bioinformatics help take advantage of serendipity through tile sequencing of huge numbers of genes or the synthesis of large numbers of chemical compounds. There are approximately $10^{30}\;to\;10^{50}$ possible molecules in molecular space of which only a fraction have been synthesized. Combining this potential with having access to 50,000 plant genes in the future elevates tile probability of discovering flew herbicidal site of actions. If 0.1, 1.0 or 10% of total genes in a typical plant are valid for herbicide target, a plant with 50,000 genes would provide about 50, 500, and 5,000 targets, respectively. However, only 11 herbicide targets have been identified and commercialized. The successful design of novel herbicides depends on careful consideration of a number of factors including target enzyme selections and validations, inhibitor designs, and the metabolic fates. Biochemical information can be used to identify enzymes which produce lethal phenotypes. The identification of a lethal target site is an important step to this approach. An examination of the characteristics of known targets provides of crucial insight as to the definition of a lethal target. Recently, antisense RNA suppression of an enzyme translation has been used to determine the genes required for toxicity and offers a strategy for identifying lethal target sites. After the identification of a lethal target, detailed knowledge such as the enzyme kinetics and the protein structure may be used to design potent inhibitors. Various types of inhibitors may be designed for a given enzyme. Strategies for the selection of new enzyme targets giving the desired physiological response upon partial inhibition include identification of chemical leads, lethal mutants and the use of antisense technology. Enzyme inhibitors having agrochemical utility can be categorized into six major groups: ground-state analogues, group specific reagents, affinity labels, suicide substrates, reaction intermediate analogues, and extraneous site inhibitors. In this review, examples of each category, and their advantages and disadvantages, will be discussed. The target identification and construction of a potent inhibitor, in itself, may not lead to develop an effective herbicide. The desired in vivo activity, uptake and translocation, and metabolism of the inhibitor should be studied in detail to assess the full potential of the target. Strategies for delivery of the compound to the target enzyme and avoidance of premature detoxification may include a proherbicidal approach, especially when inhibitors are highly charged or when selective detoxification or activation can be exploited. Utilization of differences in detoxification or activation between weeds and crops may lead to enhance selectivity. Without a full appreciation of each of these facets of herbicide design, the chances for success with the target or enzyme-driven approach are reduced.

Base Study for Improvement of School Environmental Education with the Education Indigenous Plants - In the case of Mapo-Gu Elementary School in Seoul - (자생식물 교육을 통한 학교 환경교육 개선에 관한 기초연구 - 서울시 마포구 초등학교를 중심으로 -)

  • Due to the urbanization, concentrated population, and limited land exploitation in the modern society, the environment surrounding that we live in is getting polluted more and more, and it has become hard even to let urban children experience the nature. This research was conducted to help people recognize the importance of our natural resources through the environmental education of elementary school and to use school's practical open-space for the Indigenous Plants education. The results of this study are as follows : First, the status of a plant utilization in our institutional education : There were 362 species totally of 124 species of Trees, 156 species of Herbs, 63 species of Crops, and 19 species of Hydrophytes which appear in the elementary school text book. Of all, the most frequently appearing species of tree were the Malus pumila var. dulcissima, Pinus densijlora, Citrus unshiu, Diospyros kaki. Second, the effect of plant education using the land around schools : The result of research on the open-space of the 19 elementary schools located in Mapo-gu showed that most of the species planted are the Juniperus chinensisrose, Hibiscus syriacus. Pelargonium inquinans in the order of size, and the plants appearing in text book were grown in the botanical garden organized in 7 schools. Especially most of the Indigenous Plants were being planted in botanical garden, and Pinus densijlora, Abeliophyllum distichum, Polygonatum var. plurijlorum, Liriope platyphylla and so on. Last, the result of this research on recognition of Environment, Planting education and Indigenous plants : It showed that educational necessity of students and teachers about environment and Indigenous Plants was more than 80%. The management of botanical garden was conducted by some teachers and managers. The results of this study suggested that we needed the reconstruction of curriculum, the efficient application of plant education for effectiveness of using school environment and monitoring continually and construction information sources for the better environment education in the elementary schools.

A Study on the Power Supply and Demand Policy to Minimize Social Cost in Competitive Market (경쟁시장 하에서 사회적 비용을 고려한 전력수급정책 방향에 관한 연구)

  • In this paper, the resource adequacy as well as the optimum fuel mix is obtained by the following procedures. First, the regulation body, the government agency, determine the reliability index as well as the optimum portfolio of the fuel mix during the planning horizon. Here, the resources with the characteristics of public goods such as demand-side management, renewable resources are assigned in advance. Also, the optimum portfolio is determined by reflecting the economics, environmental characteristics, public acceptance, regional supply and demand, etc. Second, the government announces the required amount of each fuel-type new resources during the planning horizon and the market participants bid to the government based on their own estimated fixed cost. Here, the government announces the winners of the each auction by plant type and the guaranteed fixed cost is determined by the marginal auction price by plant type. Third, the energy market is run and the surplus of each plant except their cost (guaranteed fixed cost and operating cost) is withdrew by the regulatory body. Here, to induce the generators to reduce their operating cost some incentives for each generator is given based on their performance. The performance is determined by the mechanism of the performance-based regulation (PBR). Here the free-riding performance should be subtracted to guarantee the transparent competition. Although the suggested mechanism looks like very regulated one, it provides two mechanism of the competition. That is, one is in the resource construction auction and the other is in the energy spot market. Also the advantages of the proposed method are it guarantee the proper resource adequacy as well as the desired fuel mix. However, this mechanism should be sustained during the transient period of the deregulation only. Therefore, generation resource planning procedure and market mechanisms are suggested to minimize possible stranded costs.

A Study on Hydromorphology and Vegetation Features Depending on Typology of Natural Streams in Korea (국내 자연하천의 유형별 물리적 구조 및 식생 특성 연구)

  • The purpose of this study is to identify the type and characteristics of the domestic natural streams in order to establish a basis for stream restoration and evaluation. To this end, 95 domestic natural stream areas, which have various natural environments, were selected except for the province of island and then the characteristics of natural environment, hydromorpholoy, plant and vegetation were investigated and analyzed in each stream area. As a result, 95 stream areas were classified into total 24 types according to 3 criteria such as stream size (4 types), altitude (3 types), bed material (5 types). Depending on altitude class that is the environmental factor showing the highest correlation with each stream types, the emergence of vegetation and plant, 24 stream types were reclassified into 3 types such as lowland (altitude less than 200m), mountain (altitude from 200m to 500m), highland (altitude more than 500m), and hydromorpholoy, plant and vegetation characteristics of each stream type were compared. First, when compared to the mountain and highland streams, the typical features of lowland streams were as follows: Stream size was large but bed material size was small and there were many valley forms where flood plane were developed well. In addition, the more large stream size was, the more cross-section width variability, bars and sinuosity were in good conditions. In lowland stream, representative vegetation community was Salix koreensis community. On the other hand, when compared to the lowland streams, the typical features of mountain and highland streams were as follows: Stream size was small but bed material was coarse-grained and its size was large. Mountain and highland streams valley form where flood plane was not developed well was narrow, and sinuosity and bars development were weak. Representative vegetation communities of mountain streams were Quercus serrata -, Quercus variabilis -, Styrax japonica community and representative vegetation communities of highland streams were Pinus densiflora -, Quercus mongolica -, Fraxinus rhynchophylla community.

Hydrogeochemical Characteristics and Contamination of Dissolved Major ions and Heavy Metals in Waters and Sediments from the Tancheon River (탄천의 하상퇴적물과 하천수내 주요 용존 이온과 중금속의 수리지구화학적 특성과 오염)

  • In order to investigate the hydrogeochemical characteristics and contamination of dissolved major ions and heavy metals in the Tancheon River, river water and sediment samples were collected at 18 locations, along a distance of 69 km, between Yongin-si in Kyunggi-do and Samsung-dong in Seoul on October in 2000 and April in 2001. After appropriate sample preparation, waters were analyzed for the dissolved constituents and sediments. The pH values of river waters were in the range of 7.0 to 9.3 and could be plotted in the area of surface environment. The level of $Ca^{2+}$, , CI-, sol-, N0$_{3}$ and HC0$_{3}$ in the Tancheon River were higher than those in world average river water. Most of dissolved constituents in the river waters increased toward downstream from upstream. In particular, high concentrations of Zn2+, Na$_{+}$, CI$^{-}$, SO$_{4}^{2-}$ and N03- were found near densely residential areas and the Sungnam waste water treatment plant. The relative ion enrichment was caused by the inflow of local domestic and industrial sewages. Also, Ca2+ and HC03- concentrations were enriched in the middle of the Tancheon River due to the dissolution of cements. This indicates that the apartment complexes were built on a large scale in the upriver since these ten years and large amounts of construction materials such as cements were flowed into the Tancheon River. Concentrations of heavy metals (Mn, Cd, Cu, Pb, Zn) in sediments from the Tancheon River exceeded the lower limit of tolerence level in bottom sediment established by the Ontario Ministry of the Environment (OME) of Canada. In particular, these metals were highly elevated in sediment (TSM-12) collected from near the Sungnam waste water treatment plant. Heavy metals were higher enriched in sediments collected from dry period rather than wet period.

Planting Method of Buffer Green Space in the Reclaimed Seaside Areas, Rokko Island, Kobe, Japan (일본 고베시(신호시(神戶市)) 로코(육갑(六甲))아일랜드 임해매립지의 완충녹지 식재기법 연구)

  • This study was carried out to suggest the basic data of planting method for construction of buffer green space based on the land use in case of reclaimed land by analyzing land structure, planting concept, and planting structure in buffer green space, Rokko Island, Kobe, Japan. Rokko Island(total area: 580ha) is divided into port and logistics industry area and urban area by constructing the box type large-scale buffer green space. The land structure of buffer green space were biased mounding type, parallel mounding type, and complex mounding type. The width of buffer green space was 50meters in case of northern area, from 28 to 32meters in case of eastern area, and 37.5meters in case of western area, and the slope of that was from 18 to 25 degrees and the height of that was from 2 to 15meters. There were applied landscape and buffer planting concept on the sea side area of northern buffer green space, on the other hand landscape and shade planting concept on the Inner city side area of that. According to the result of planting structure analysis of northern buffer green space, the main woody species were those of deciduous-evergreen species grow in warm-temperate forest zone such as Quercus glauca, Cinnamomum camphora, Machilus thunbergii, Elaeagnus maritima. The results of maximum number of species and planting density by $100mm^2$ was that 9 species 22 individuals in canopy layer, 9 species 15 individuals in understory layer, 3 species 67 individuals in shrub layer, and 14 species 104 individuals in total. The plant coverage of northern buffer green space based on the ecological planting method was from 69 to 139% in case of canopy layer, from 26 to 38% in case of understory layer, from 6 to 7% in case of shrub layer, and from 101 to 184% in total. Index of plant crown volume of northern buffer green space based on the ecological planting method was from 1.40 to $3.12m^3/m^2$ in case of canopy layer, from 0.43 to $0.55m^3/m^2$ in case of understory layer, $0.06m^3/m^2$ in case of shrub layer, and from 1.89 to $3.73m^3/m^2$ in total.

A study on ecological reprocessing and creation of biotope by reuse of treated waste water and nonpoint pollution source of stream (하수처리수와 하천 비점오염원을 이용한 생태적 재처리 효과와 생물 서식처 창출 방안 - 왕포천 생태적수질정화비오톱(Sustainable Structured wetland Biotop) 시스템 사례를 중심으로 -)

  • The Wangpo stream located in the Buyeo-gun was a small stream with both low water quality and quantity due to the cultivated land and settlement area through the stream. In order to restore ecosystem, the Sustainable Structured wetland Biotop system was applied to treat 1,500 to 7,000 ㎥/d amount of effluent water pumped from the Buyeo sewage treatment plant facility as well as inflowed from nonpoint pollution source of Wangpo stream. As a result of continuous monitoring for 2 years from 2016 to 2018 after completing restorative construction, the average BOD5 as an index of organic pollution was 7.3 mg/L and the average effluent concentration became 2.1 mg/L, showing an improvement by 71.2%. The average inflow concentration of T-N was 7.953 mg/L and the average outflow concentration was 3.379 mg/L, showing 57.5% of improvement. The average inflow concentration of T-P was 0.177 mg/L and the average outflow concentration was 0.052 mg/L, showing about 70.7% improvement. The results of ecological monitoring after creating biotope by reuse of treated waste water and nonpoint pollution source of the Wangpo Stream are as follows. The plant taxa founded in water SSB(Sustainable Structured wetland Biotop) system of the Wangpo Stream was total 41 species in 21 families, showing a higher proportion of naturally introduced plant than that of artificially planted species. In case of other terrestrial animals, both amphibian and reptile group were confirmed as 3 species in 6 families, avian group was 25 species of 15 families, and mammal group observed 5 species in 5 families, respectively. All species have been created and enhanced through purified water inhabited in the SSB(Sustainable Structured wetland Biotop) system as a treatment wetland, eventually migrating to the Wangpo Stream.
