When whole cells are subjected to pyrolysis gas chromatography/mass spectrometry (Py-GC/MS) analysis, it provides biochemical profiles containing overlapping signals of the majority of compounds. To determine marker compounds that discriminate embryogenic calluses from nonembryogenic calluses, samples of embryogenic and nonembryogenic calluses of five higher plant species were subjected to Py-GC/MS. Genetic programming of Py-GC/MS data was able to discriminate embryogenic calluses from nonembryogenic calluses. The content ratio of 5-meyhyl-2-furancarboxaldehyde and 5-(hydroxymethyl)-2-furancarboxaldehyde was greater in nonembryogenic calluses than in embryogenic calluses. However, the content ratio of phenol, p-cresol, and $^1H-indole$ in embryogenic calluses was 1.2 to 2.4 times greater than the ratio in nonembryogenic calluses. These pyrolysates seem to be derived from the components of the cell walls, which suggests that differences in cell wall components or changes in the architecture of the cell wall playa crucial role in determining the embryogenic competence of calluses.
From the single cell cultures of haploid tobacco (Nicotiana tabacum L. cv. NC 2326) glyphosate-resistant plants were regenerated. After treated with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and then inoculated onto the LS medium supplemented with 1 mM glyphosate, the single cells survived formed colonies and calluses in 65 and 95 days after culture, respectively, and then whole plants were regenerated in 0.1 mM glyphosate-containing medium from the selected calluses. There was no difference in fresh weight and shikimate content between the selected and normal haploid calluses. When sprayed with 0.1 mM glyphosate, the shikimate contents in the regenerated and normal plants were 0.659 and 20.816 mol/g fr. wt., while that in other normal plants which were not sprayed was 0.921. In addition, the calluses induced from the regenerated plants grew without showing any retardation when treated with glyphosate. These results indicate that the secelcted calluses and regenerated plants are resistant to glyphosate.
Mature seeds of Artemisia annua L. were placed onto Murashige and Skoog's (MS) medium supplemented with $4.52\;{\mu}M$ 2,4-dichlorophenoxyacetic acid (2,4-D). After 6 weeks of culture, off-white, compact calluses were formed on the plumule of seedlings at a frequency of 5.9%. Calluses were subcultured on the same medium. After an additional 2 weeks of subculture, calluses produced a few somatic embryos at a frequency of 28.8%. Upon transfer to MS basal medium, calluses producing a few somatic embryos gave rise to numerous somatic embryos, which subsequently developed into plantlets. Plantlets were successfully transplanted to potting soil and grown to maturity in a greenhouse.
The highest degree of callus formation was obtained from the shoot tips of Dianthus superbus when cultured on the MS medium supplemented with 2.0 mg/L NAA and 0.5 mg/L BAP. Embryogenic calluses were obtained from the seperated friable calluses on MS medium containing 2.0 mg/L 2,4-D after 7-8 wk of culture. For plant regeneration, embryogenic calluses were selected and cultured on te proliferation medium. After 3 wk, somatic embryos appeared on MSK medium (0.5 mg/L NAA, 2.0 mg/L kinetin) and N6 medium (2.0 mg/L kinetin, 0.1 mg/LNAA, 0.1 mg/L 2,4-D and 2.0 g/L casein hydrolysate). When these somatic embryos were kept under continuous illumination, shoots were successfully regenerated on the both media. The shoots were rooted on MS medium supplemented with 2.0 mg/L NAA.
Young haploid leaf derived from the anthers of tobacco plant was cultuerd and plantlets of various ploidies were obtained. When the leaf was put on the medium supplemented with kinetin as growth regulator, plantlets developed directly from the leaf, and the plants coming out in early stage of culture were all haploid. Plants developing in later stage were mostly haploids with some exception of diploid and aneuploid. Leaves were also cultured on the callus-inducing media supplemented with 2,4-D and kinetiion, and the calluses were sub-cultured for six months. Plants developed from these calluses were mostly aneuploids of various chromosome numbers. In view of the fact that the plants directly developed from the leaf were all haploid, the tissue of the original leaf explant was assumed to be uniform as far as chromosome number was concerned. On the other hand, it seemed that the occurrence of various ploidies in the plants derived from the calluses of same origin was the result of the influence of in vitro culture. Apical meristem tissues and various multicellular bodies were formed in the epidermal and inner mesophyll tissues as well as in the sub-epidermal cells.
Single cells obtained from suspension culture of mature embryo-derived callus in wheat(Triticum aestivum L. cv Jang Kwang) were cultured to regenrated into the plantlet. Cell clusters and embryogenic calluses were efficiently developed from when the single cells clutured on the MS medium supplemented with 10${\mu}{\textrm}{m}$ 2,4-D. Upon transfer to hormone-free MS medium containing 10 mg/I AgNO3, embryogenic calluses gave rise to shoots, probably through somatic embryogenesis.
Off-white, friable embryogenic calluses were formed on the internal integument of mature seeds of yuzu (Citrus junos) cultured on Murashige and Skoog's basal medium at a frequency of 1.2%. Embryogenic calluses were proliferated when cultured on medium with 1 mg/L 2,4-D. Upon transfer to medium with 0.1 mg/L kinetin, embryogenic calluses produced numerous somatic embryos. Embryogenic suspension cultures were established by placing embryogenic calluses into liquid medium with 1 mg/L 2,4-D. When plated onto medium with 0.5 mg/L ABA, embryogenic cells developed into somatic embryos at a high frequency, and then regenerated into plantlets. Plantlets were successfully transplanted to potting soil and grown in a greenhouse.
Rugosa rose (Rosa rugosa) is cultivated as a garden flower and an important genetic resource for the breeding of roses (R. hybrida). This study describes culture conditions for high frequency plant regeneration from zygotic embryo explants via somatic embryogenesis in rugosa rose. Mature zygotic embryo, cotyledon, and radicle explants formed embryogenic calluses at frequencies of 38, 6.7, and 8.8% when cultured on half-strength Murashige and Skoog medium (${\frac{1}{2}}MS$) supplemented with 2.26, 9.05, and $9.05{\mu}M$ 2,4-dichlorophenoxyacetic acid, respectively. Embryogenic calluses produced numerous somatic embryos, which then developed into plantlets on ${\frac{1}{2}}MS$ without growth regulators. Regenerated plantlets were grown to whole plants in a growth chamber.
The gesneriaceous perennial plant Titanotrichum oldhamii has beautiful foliage and attractive bright yellow flowers. However, breeding of T. oldhamii by conventional sexual hybridization may be difficult because sexual reproduction of this species is very rare. In the present study, plant regeneration systems via both direct and indirect formation of adventitious shoots from leaf explants were established as the first step toward breeding T. oldhamii by using biotechnological techniques. Adventitious shoots were formed efficiently on medium containing $0.1mg\;l^{-1}$ benzyladenine. Histological observation showed that shoot formation on this medium occurred directly from leaf epidermal cells without callus formation. On the other hand, leaf explants formed calluses on medium containing $0.1mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid. The calluses could be maintained by monthly subculturing to fresh medium of the same composition. When the calluses were transferred to plant growth regulator-free medium, they formed adventitious shoots. Directly and indirectly formed shoots rooted well on medium containing $0.1mg\;l^{-1}$ indole-3-butyric acid. Plantlets thus obtained were successfully acclimatized and grew vigorously in the greenhouse. Flow cytometry analysis indicated that no variation in the ploidy level was observed in plants regenerated via direct shoot formation, whereas chromosome doubling occurred in several plants regenerated via indirect shoot formation. Regenerated plants with the same ploidy level as the mother plants showed almost the same phenotype as the mother plants, whereas chromosome-doubled plants showed apparent morphological alterations: they had small and crispate flowers, and round and deep green leaves.
Efficient plant regeneration system was established through the culture of rice (Oryza sativa L.) microspores. Microspores released by anther shedding culture developed into proembryos and calluses in B5 medium within two weeks of culture. The optimal hormone and carbon sources were dependent on the genotypes used. Microspore's viability decreased rapidly in culture time, therefore less than 3% of the total microspores were viable at the 9th day when the first microspore division was observed. Of two types of microspores (pollen dimorphism) observed in culture, only the P-grain, larger microspores than normal one was able to divide. Using 4',6-diamidino-2-phenylindole (DAPI) fluorescence staining, it was confirmed that the symmetrical division of uninucleate microspore was the first step leading to continuous microspore development. Microspore-derived proembryos and calluses were regenerated to plants in N6 medium containing 1 mg/L NAA and 5 mg/L kinetin, and 78.3% of the regenerated plants were haploids.
