Effect of Dietary Supplementation of Pigment Sources on Pigmentation of the Round Tailed Paradise Fish Macropodus chinensis and the Pale Chub Zacco platypus (사료내 색소원료 첨가가 버들붕어 Macropodus chinensis 및 피라미 Zacco platypus 표피의 색택에 미치는 영향)

  • This study investigated the effect of supplement of the pigment source in diets on body color for round tailed paradise fish Macropodus chinensis and the pale chub Zacco platypus. Fish (the round tailed paradise fish: 1.6 g/ fish, pale chub: 2.4 g/fish) were fed diets containing various pigment sources (Porphyra powder, Spirulina powder. yeast astaxanthin and paprika extract) twice a day for 6 weeks. Survival of all treatments was above 80%. Weight gain and total carotenoid contents of round tailed paradise fish fed the diet containing Porphyra powder was higher than that of fish fed the other diets. The apparent redness of skin and fin in round tailed paradise fish fed the diets containing paprika extract and yeast astaxanthin was better than that of fish fed the diets containing Porphyra powder, Spirulina powder and the control diet. Weight gain and total carotenoid contents of pale chub fed the diet containing paprika extract were higher than that of fish fed the containing Porphyra powder, yeast astaxanthin, Spirulina powder and the control diet. The apparent redness of skin and fin in pale chub fed the diets containing parika extract and yeast astaxanthin was better than that of fish fed the diets containing Porphyra powder, Spirulina and the control diet. The results of this study suggest that dietary supplementation with paprika extract and yeast astaxanthin can improve redness of skin and fin of round tailed paradise fish and pale chub.

Changes on Initial Growth and Physiological Characteristics of Larix kaempferi and Betula costata Seedlings under Elevated Temperature (온도 증가에 따른 일본잎갈나무와 거제수나무 유묘의 초기 생장과 생리 특성의 변화)

  • Larix kaempferi and Betula costata seedlings were grown under an elevated temperature ($27^{\circ}C$) for four weeks to understand initial changes on physiological characteristics caused by temperature rising in connection with global warming. At the end of the treatment, growth performance, leaf pigment content, antioxidative enzyme activities and malondialdehyde (MDA) content were measured and analyzed. Relative growth rates of the height of two tree species grown under elevated temperature ($27^{\circ}C$) were lower than those of control ($24^{\circ}C$) and dry weights of leaves, stems and roots were also reduced at higher temperature. Particularly, the root growth reduction of two tree species increased markedly at $27^{\circ}C$ over the study period, which increased the ratio of shoot to root. Under higher temperature, leaf pigment contents decreased, whereas anti-oxidative enzyme activities such as ascorbate peroxidase (APX) and catalase (CAT) increased as compared with the control. But MDA content was not affected by elevated temperature. In conclusion, the elevated temperature leads to root growth reduction, restriction of nutrient uptake from soil and the reduction of leaf pigment contents, which can inhibit the aboveground growth. In addition, higher temperature might act as a stress factor that causes growth reduction through the increase of energy consumption during a growth period.

  • This study was performed to evaluate the hexavalent chromium content in flexography inks and organic pigments used as colorants for the ink. The digestion of sample was carried out under alkaline condition, and the content of soluble hexavalent chromium extracted from samples was determined by UV-visible colorimetric analysis method after alkaline digestion. Duplicate sampling system to obtain two digested samples, was applied in this study. To determine the hexavalent chromium content in the flexography ink and organic pigment colorimeteric analysis was employed. Because the organic pigment is the main ingredient of flexography ink it is important to remove the errors associated with the colorant of the ink in colorimetric determination. The duplicate sampling system allowed us to correct the errors associated with the colorimeteric measurement. The additional filtration was found an essential step to exclude colorimeteric error derived from the various precipitates. The soluble hexavalent chromium content in flexography inks was generally less than 5 ppm. Yellow, violet and some magenta colors showed higher soluble hexavalent chromium contents. The content of hexavalent chromium in organic pigments was greater than flexography inks, and yellow, violet and some magenta pigments contained greater amount of hexavalent chromium, which indicated that the hexavalent chromium in inks derives from the colorants. The soluble hexavalent chromium content in linerboards were below 1 ppm, and no hexavalent chromium was detected in UKP. Results suggested that flexography ink is the main factor to cause hexavalent chromium in linerboard and organic pigments.

Production and Characteristics of Hongkuk-ju using Monascus anka (Monascus anka를 이용한 홍국주의 제조 및 특성)

  • To reproduce the brewing process of Hongkuk-ju and to identify the functional properties of it, Hongkuk-ju was brewed using different additions of Hongkuk (100%, 90%, 70%, 50%) and Nuruk (0%, 10%, 30%, 50%). The quality elements, including pH, total acidity, reducing sugar content, alcohol content and pigments (yellow, red, monacolin K and citrinin), were measured. The pH values of Hongkuk-ju showed a slight difference (pH 4.08~4.58) right after the $1^{st}$ stage mash; further, the pH on all groups (H1, H2, H3 and H4) in the terminal of the $2^{nd}$ stage mash (9 days in fermentation) were similar, ranging approximately at pH 3.70. The total acidity change did not show a difference directly the $1^{st}$ stage mash (nearby 0.2 %); however, it began to show a slight difference at the terminal of the $2^{nd}$ stage mash between the range of 0.69~0.76%. The residual reducing sugar of the content was decreased with the increased Nuruk content. The alcohol concentrations of the treatment brew with Nuruk ranging from 12.3% to 13.7% were higher than Hongkuk on its own. The yellow and red pigment contents of Hongkuk-ju ranged from 7.2~8.8 O.D. units (yellow pigment) and from 4.4~5.1 O.D. units (red pigment). The production of monacolin K and citrinin was the highest (9.48 mg/kg and 10.14 mg/kg) when the treatment solely brewed Hongkuk. The concentration of Nuruk and the preparation of the seed mash from it were critical factors compared to the treatment of rice in brewing Hongkuk-ju.

Stability of the Anthocyanin Pigment Extracted from Aronia (Aronia melancocarpa) (아로니아(Aronia melancocarpa) 유래 안토시아닌 색소의 안정성)

  • The purpose of this study was to investigate the effects of pH, temperature, light, sugars, and organic acids on the stability of anthocyanin pigments extracted from aronia (Aroniamelanocarpa). Pigments from aronia were most stable at pH 3.0, with maximal absorbance at 514 nm. High temperatures and prolonged heating substantially reduced anthocyanin contents. Anthocyanin levels were as low as approximately 21.70 and 79.63% of normal levels after 300 min of heating at 30 and $100^{\circ}C$, respectively. All of the sugars tested decreased the abundance of aronia pigments, with the levels highest in the presence of maltose, and decreasing progressively in the presence of galactose, sucrose, glucose, and fructose. Among the organic acids tested, citric acid was the most effective in stabilizing the aronia pigment, followed by tartaric acid, malic acid, formic acid, and ascorbic acid. These results provide useful reference data for efforts to use pigments from aronia in processed foods or for developing natural food colorants.

Case study of Hexavalent Chromium and Silica Exposure Assessment and Respiratory Fit-test for Paint Manufacturing Worker (페인트 제조 작업자의 6가 크롬 및 실리카 노출평가와 호흡보호구 밀착도 검사 사례)

  • Objective: Paint manufacturing industry workers are exposed to various lung cancer carcinogenic substances including hexavalent chromium and crystalline silica. Studies have been conducted on lung cancer in Paint manufacturing industry workers and the concentration of hexavalent chromium in paint industry; however, the concentration of crystalline silica and hexavalent chromium and cases of lung cancer in a single Paint factory has never been reported in Korea. Methods: To determine whether the cancer was related to his work environment, we assessed the level of exposure to carcinogens during pouring and mixing talc and pigment. In addition, a mask fit test was performed for the worker. Results: Analysis of talc and silica bulk powder materials showed that crystalline silica (quartz) was 5% in talc and 100% in silica. The green and yellow pigments contained 87% and 92% of lead chromate, respectively. Our quantitative analysis of pigment powder samples showed that the hexavalent chromium contents quantified in the green and yellow pigment samples were 87% and 92%, respectively. In order to estimate his exposure level of hexavalent chromium, we measured a personal exposure level of hexavalent chromium for a worker in accordance with the National Institute for Occupational Safety and Health #7605 method. The results showed that the worker was exposed to the high level of hexavalent chromium (0.033 mg m-3). In addition, the talc powder also contained 5% quartz, and the worker's exposure level to respirable quartz exceeded OEL. As a result of the respiratory protection fit test for workers, the overall Fit Factor was '15' when wearing a second-grade mask and '25' when wearing a first-grade mask, significantly lower than the US Occupational Safety and Health Agency (OSHA) pass value of "100". Conclusion: Workers who pouring and mixing powder materials such as talc or colored pigments in paint manufacturing company may be exposed to high concentrations of carcinogenic substances. These findings indicate that it is necessary to local ventilation system inspection, safety and health education for employers and workers, and periodically monitoring and manage the working environment.

Effect of Fe2+/Fe3+ Molar Ratio on the Synthesized Magnetic Black Pigment by Hydrothermal Method (수열합성법으로 제조한 흑색 자성안료의 Fe2+/Fe3+ 몰비에 따른 특성)

  • $Fe_3O_4$ was prepared on the $TiO_2-coated$ natural mica substrate. The natural mica has an average particle size of $22{\mu}m$. The substrate was coated on $TiO_2$ thin films using hydrothermal synthesis at pH 1.5-2.5 at $75^{\circ}C$. The Fe precursor solution was prepared by mixing $FeSO_4$ (for $Fe^{2+}$ ion) and $FeCl_3$ (for $Fe^{3+}$ ions) with different molar ratios such as 1/2, 1/1, 2/1, 3/0, and $Fe_3O_4$ only. X-ray diffraction analysis shows that the crystal structure depends on the $FeCl_3-to-FeSO_4$ molar ratio. $Fe_3O_4$ crystal phase could be obtained at higher $FeSO_4$ contents.

Effects of Macrophytes on Biological Treatment of Processed-Leachate from Sanitary Landfill Sites

  • Three macrophytes species, that are, Phragmites australis, Zizania latifolia and Typha angustifolia were grown in mono culture in order to compare growth (relative biomass increase rate), variation of photosynthetic pigment (total chlorophyll, Chl a, Chl b and Chl a/Chl b) and effectiveness of nutriment removal in 15%o NaCl-salt solution or processed-leachate (salinity 19.6%o) from sanitary landfill sites. The relative biomass increase rate of p. australis was significantly higher than Z. latifolia and T. angustifolia. In the case of processed-leachate treatment, the relative biomass increase rates of above-part, rhizome and root of P. australiswere 178 %, 148 % and 157 %, respectively. Also, in 15%o NaCl-salt solution treatment, the relative biomass increase rates of P. australis increased as follows; 161 % (above-part), 183 % (rhizome) and 112 % (root). Total chlorophyll contents increased significantly in the leaves of P. australis and Z. latifolia grown in 15%o NaCl-salt solution and processed-leachate. Among three macrophytes, P. australis was evaluated as most effective macrophyte for the biological retreatment of processed-leachate from sanitary landfill sites.

Growth regulation of cow1 rice mutant seedlings by blue light

  • We assessed whether the cow1 mutant defects are associated with growth of Tos17 and T-DNA insertional rice in blue light (BL). Growth of oscow1 mutants which encoded a member of the YUCCA protein family was retarded in BL. Root to shoot ratios of the mutants were reduced about 2 times lower in the absence of NAA and about 2.5 times lower in the presence of NAA; the shoot growth was not significantly changed by NAA addition. Photosynthetic activity of the mutants was however inhibited in high light. Pigment analysis showed significant difference between wild-type (Chl a:b = 3.02) and mutants (3.84). Carotenoid contents of the mutants were also decreased considerably, implying the involvement of cow1 in pigment formation. These findings lead us to suggest that the growth retardation of oscow1 mutant plants by BL results from the difference of photosynthetic activity in part.

Anti-Melanogenic Activity of Undecylprodigiosin, a Red Pigment Isolated from a Marine Streptomyces sp. SNA-077

  • Bioassay and HPLC-UV guided fractionations of the crude extract of marine-derived Streptomyces sp. SNA-077 have led to the isolation of a red pigment, undecylprodigiosin (1). The chemical structure of undecylprodigiosin (1) was revealed by the interpretation of NMR and mass spectroscopic (MS) data. Further, anti-melanogenic effects of undecylprodigiosin (1) were investigated. First, the melanin contents of undecylprodigiosin (1)-treated B16 cells were evaluated. Furthermore, undecylprodigiosin (1) significantly inhibited the key enzymes involved in melanogenesis, including tyrosinase, tyrosinase related protein-1 (TYRP-1), and dopachrome tautomerase (DCT). The mRNA and protein expression levels of Microphthalmia-associated transcriptian factor (MiTF), a critical transcription factor for tyrosinase gene expression, were also suppressed by undecylprodigiosin (1) treatment in B16 analyses. Collectively, our results suggest for the first time that undecylprodigiosin (1), a potent component isolated from an extract of marine Streptomyces sp. SNA-077, critically exerts the anti-melanogenic ability for melanin synthesis.
