Effects of corn particle size on energy and nutrient digestibility in diets fed to young pigs and adult sows

  • Objective: This research was carried out to investigate the effects of corn particle size on the apparent total tract digestibility (ATTD) of energy and nutrients fed to pigs at four different growth stages and therefore to provide basis for better application of corn in pig feeds. Methods: Eighteen weanling piglets, 18 growing barrows, 24 gestating sows and 24 lactating sows were used in this study. Within each stage, pigs were allotted to 1 of 3 or 4 corn-soybean meal diets which were formulated with different corn particle size in a completely randomized design with 6 replicate pigs per diet. Each stage lasted for 19 days, including 7 days for cages adaptation, 7 days for adaptation to diets and followed by 5 days for total collection of feces and urine. Results: For nursery and growing stages, the results showed that digestible energy content and ATTD of gross energy (GE), dry matter (DM), neutral detergent fiber (NDF), and acid detergent fiber (ADF) was increased (p<0.05) as the corn particle size reduced. Meanwhile, the metabolizable energy content and ATTD of crude protein (CP) tended to increase. For gestating sows, no differences were found in the ATTD of nutrients among dietary treatments. As for lactating sows, there were linear and quadratic increases (p<0.05) in the ATTD of DM, GE, NDF as the corn being finer milled. Quadratic response in ATTD of ADF and CP (p<0.05) were observed as sows fed with four different diets. Conclusion: Reducing corn particle size can increase digestibility of nutrients fed to young pigs and lactating sows. No effects were observed in present experiment when gestating sows were fed with different particle sized corn.

비육돈에 미생물제제 급여시 분뇨 특성에 미치는 효과 (The Effects of Feeding Feed Additives Containing Microorganisms on Characteristics of Excreta in Finishing Pigs)

  • 본시험은 비육돈사료에 미생물제제를 사료에 미생물제제 A 및 B 0.1 미생물제제 C를 0.2% 혼합 급여할 경우 사료섭취량 및 돈분의 오염물질 배설농도에 미치는 영향을 분석하기 위하여 4처리$\times$반복당 5두로서 총 20두를 공시하여 실시하였는데 그 결과를 요약하면 다음과 같다. 1. 비육돈의 일일 평균사료섭취량은 대조구 3.15 kg/일.두였고 미생물A, B, C구는 각각 3.14kg/일/두, 3.31, 3.42로 미생물제제 C구에서 일일 사료섭취량이 가장 높게 조사되었으며(p<0.05), 2. 일일평균 음수량은 사료섭취량이 높았던 미생물 C구에서 3.95kg/일/두로 가장 높게 조사되었다(p<0.05). 3. 미생물제제 처리구별로 분뇨배설량은 사료섭취량이 높았던 미생물제제 C구에서 가장 많이 배설되는 것으로 조사되었으며(p<0.05), 돈뇨의 배설량도 미생물제제 C구에서 2.23kg/일/두에서 높았다(p<0.05). 4. 돈분뇨의 수분 함량은 및 비료성분인 T-N, $P_{2}O_{5}$, $K_{2}O$ 성분도 처리 간에 큰 차이를 보이지 않았다(p<0.05). 5. 돈분뇨의 평균 BOD 농도는 돈분의 경우 미생물제제 B, C제제 급여구가 유의적으로 높게 조사되었다(p<0.05). 그리고 돈뇨의 BOD의 경우에는 대조구에서 $8,657.5mg/{\ell}$로 가장 높은 것으로 조사되었다(p<0.05). 6. COD 농도는 대조구에서 가장 높게 조사되었으며(p<0.05). 돈뇨의 경우에는 미생물제제 A급여구에서 평균 $9,545mg/{\ell}$로 가장 높았다(p<0.05). 7. SS 농도는 미생물제제 B급여구에서 가장 높게 조사되었으며(p<0.05), 돈분뇨중의 T-N 농도는 처리구간에 유의적인 차이가 나타나지 않았다(p<0.05). 그리고 T-P 농도의 경우에는 미생물제제 C급여구에서 유의적인 차이가 나는 것으로 조사되었다(p<0.05). 이상의 결과를 요약해보면 비육돈에 미생물제제 혼합급여시 사료섭취량과 음수량을 증가시키는데 효과가 있는 것으로 조사되었으나, 비료성분 배설량에는 큰 차이를 보이지 않는 것으로 조사되었으나, BOD 등 오염물질농도의 경우에는 미생물제제 A급여구에서 가장 낮게 조사되어 비육돈사료에 미생물제제 급여시 오염물질 저감효과가 있는 것으로 조사되었다.

퇴비 부숙과정중 뒤집기 횟수에 따른 퇴적 깊이별 이화학성 및 미생물상 변화 (Changes of Physico-chemical Properties and Microflora of Pig Manure due to Composting with Turning Times and Depth)

  • 양질의 퇴적식 퇴비생선을 위한 일환으로 돈분과 분쇄왕겨를 동일비율로 혼화 퇴적 후 뒤집기 횟수를 달리하여 퇴적깊이별로 70일동안 부숙시키면서 이화학성 및 미생물상을 조사하였다. 수분함량과 pH는 부숙화가 진행됨에 따라 뒤집기 횟수가 많을수록 감소하였으나 퇴적 깊이가 깊을수록 높은 값을 보였다. C/N율과 $NH_4-N$함량은 부숙화가 진행됨에 따라 감소한 반면 $NO_3-N$함량은 증가하였고 뒤집기 회수에 따라 이들의 값은 각기 다르게 나타났다. 깊이별로는 $NH_4-N$$NO_3-N$함량이 대부분 표층일수록 높았고 그 결과 $NH_3$가스 발생도 표층에서 높았으며 부숙화가 진행됨에 따라 점차 감소되었다. 퇴비화 70일째에 있어서 미생물상의 분포중 총 호기성 세균은 $10^7{\sim}10^9CFU\;g^{-1}$로 뒤집기 횟수가 증가할수록 표층에서 높은 편이었으며 사상균은 표층으로부터 60cm 부위까지는 $10^2{\sim}10^4CFU\;g^{-1}$ 유지되었으나 부숙화가 진행됨에 따라 내부에는 거의 생존하지 못하였다. 셀루로스 분해균과 호열성균은 각각 $10^6{\sim}10^8CFU\;g^{-1}$, $10^6{\sim}10^9CFU\;g^{-1}$ 으로써 2회 뒤집기의 30cm이상인 심층일수록 높은 경향이었다. 이상과 같이 깊이별 미생물상 분포의 경향은 뒤집기 횟수가 증가함에 따라 층위별 밀도차이를 줄일수 있어 부숙촉진을 위해서 2~3회 정도 뒤집기를 해야 하나 그 이상을 상회할 경우 수분부족으로 오히려 부숙이 지연될 우려가 있다고 판단된다.

설사 증상의 돼지 분변에서 분리된 용혈성 대장균의 항생제 내성과 독소의 인체로부터 분리된 균주로의 전이 (Transfer of Genes for Antimicrobial Resistance and Toxin of Hemolytic Escherichia coli Isolated from Feces of Pig Suffering Diarrhea to Human Isolates)

  • 1997년과 1998년 사이에 설사 증상을 보이는 돼지의 분변으로부터 총56균주의 Escherichia coli를 분리하여 이중 용혈성을 나타내는 38균주의 항생제 내성과 독소 생산능을 확인하였다. 항생제 한천 희석법으로 최소억제농도(minimal inhibitory concentration)를 측정한 결과 36균주$(94.7\%)$가 tetracycline에 대해 내성을 나타내었고, 27주$(71.0\%)$는 ampicillin에 내성, 26균주$(68.4\%)$는 chloramphenicol에 내성, 그리고 21균주$(55.2\%)$는 tri-methoprim에 내성을 나타내었으나, aztreonam, amikacin, norfloxacin에 내성을 나타낸 균주는 발견되지 않았다. 이중 4가지 이상의 항생제에 대해 내성을 가지는 다제내성(multiple drug resistance, MDR)을 보인 균주는 총 38 균주 중 21균주$(55.3\%)$였다. 또한 이중 디스크 시험법(Double Disk Synergy Test)을 수행한 결과 extended spectrum $\beta-lactamase$를 생산하는 균주는 없는 것으로 나타났다. 이들 중 가장 많은 수의 균주가 내열성 독소$(ST,89.5\%)$를 생산하였고, 다음으로 베로 독소(VT와 VTe, 각각$47.4\%$)와 이열성 독소$(LT,31.6\%)$를 생산하는 것으로 나타났다. 이 중에서 8균주$(21.0\%)$는 57만을 생산하였던 반면에 12균주$(31.6\%)$는 LT와 ST를 동시에 생산하였고, 13균주$(34.2\%)$는 ST, VT, VTe를 동시에 생산하였으며, 5균주$(13.2\%)$는 VT와 VTe를 동시에 생산하는 것으로 나타났다. 그러나 네 가지 독소를 동시에 생산하는 균주는 없었다. 또한 이 균주들은 매우 다양한 혈청형(serotype)을 가지고 있음이 확인되었다. 사람에 직접적인 유해성을 가지고 있는 지 확인하기 위해 사람 방광 유래의 T-24세포와 장내 표피 유래의 Caco-2세포에 대한 부착능을 시험하였을 때, 16균주$(42.1\%)$가 T-24방광 세포에, 그리고 17균주$(44.7\%)$가 Caco-2장세포에 대해 강한 부착능을 나타내었다. 특히 11균주$(28.9\%)$는 두 세포 모두에 강한 부착능을 가지고 있었다. Filter mating method를 수행하여 이들 균주들의 독소 생산 유전자와 항생제 내성 유전자가 사람에서 분리된 균주로 전달되는 것을 확인할 수 있었다. 본 실험의 결과는 설사 중상을 나타내는 돼지로부터 분리된 용혈성 E. coli의 독성과 세포 부착능력, 그리고 항생제 내성간의 상호 연관성을 보여주지 않았으나 동물 분리 세균의 항생제 내성과 독소 생산 능력이 유전자 전달을 통해서 뿐만 아니라 세균의 직접 접촉에 의해서도 인체로 전달될 수 있는 것을 보여주는 것이다.

Effect of inclusion level and adaptation duration on digestible energy and nutrient digestibility in palm kernel meal fed to growing-finishing pigs

  • Objective: An experiment was conducted to evaluate effects of inclusion level of palm kernel meal (PKM) and adaptation duration on the digestible energy (DE) and apparent total tract digestibility (ATTD) of chemical constituents in diets fed to growing-finishing pigs. Methods: Thirty crossbred barrows ($Duroc{\times}Landrace{\times}Large\;White$) with an average initial body weight of $85.0{\pm}2.1kg$ were fed 5 diets in a completely randomized design. The diets included a corn-soybean meal basal diet and 4 additional diets in which corn and soybean meal were partly replaced by 10%, 20%, 30%, or 40% PKM. After 7 d of adaptation to the experimental diets, feces were collected from d 8 to 12, d 15 to 19, d 22 to 26, and d 29 to 33, respectively. Results: The DE and ATTD of gross energy (GE), dry matter (DM), ash, organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and crude protein (CP) in diets decreased linearly as the dietary PKM increased within each adaptation duration (p<0.01). Diet containing 19.5% PKM had less DE value and ATTD of all detected items compared with other diets when fed to pigs for 14 days (p<0.05). The ATTD of CP in PKM calculated by 19.5% and 39.0% linearly increased as adaptation duration prolonged from 7 to 28 days (p<0 .01). Conclusion: Inclusion level of PKM and adaptation duration had an interactive effect on DE and the ATTD of GE, DM, OM, and CP (p<0.01 or 0.05) but ash, NDF, and ADF in diet (p>0.05). Considering a stable determination, 21 days of adaptation to a diet containing 19.5% PKM is needed in pigs and a longer adaptation time is recommended as dietary PKM increases.

산성 Cellulase를 분비하는 Bacillus amyloliquefaciens ATC6의 분리 (Isolation of Bacillus amyloliquefaciens ATC6 Producing Acidic Cellulase)

  • 사료곡물을 효율적으로 이용하기 위해, 산성 cellulase를 분비하는 ATC6 균주를 선발하였다. 선발된 ATC6 균주의 형태, 당 이용성 및 16S rDNA 서열분석 등을 통해 동정한 결과 Bacillus amyloliquefaciens에 속하는 것으로 나타났으므로, B. amyloliquefaciens ATC6로 명명하였다. B. amyloliquefaciens ATC6가 분비하는 cellulase 효소의 활성은 대수생장기 중반부터 급격히 증가하였고, 균의 생장이 정체기에 이르면 효소의 활성도 더 이상 증가하지 않는 것으로 나타났다. 효소의 최적 pH를 조사한 결과, pH 4.5에서 최대의 효소 활성을 나타내었으며, pH 4.0에서도 최고 활성의 80% 이상을 유지하였다. 또한 pH 4.0~pH 8.0까지 상대적으로 넓은 범위에서 최대 활성의 60% 이상을 유지하였다. Cellulase 활성의 최적 온도는 $55^{\circ}C$ 이었으며, $75^{\circ}C$의 고온에서도 최대 활성의 50% 정도의 활성을 나타내었다. 효소의 열 안정성을 조사한 결과, $45^{\circ}C$$55^{\circ}C$에서는 2시간 후에도 효소의 활성이 안정하게 유지되었으나, $65^{\circ}C$에서는 시간의 경과와 함께 효소 활성이 급격히 감소하는 것으로 나타났다.

Effects of exogenous phytase and xylanase, individually or in combination, and pelleting on nutrient digestibility, available energy content of wheat and performance of growing pigs fed wheat-based diets

  • Objective: Two experiments were conducted to determine the effects of adding exogenous phytase and xylanase, individually or in combination, as well as pelleting on nutrient digestibility, available energy content of wheat and the performance of growing pigs fed wheat-based diets. Methods: In Experiment 1, forty-eight barrows with an initial body weight of $35.9{\pm}0.6kg$ were randomly assigned to a $2{\times}4$ factorial experiment with the main effects being feed form (pellet vs meal) and enzyme supplementation (none, 10,000 U/kg phytase, 4,000 U/kg xylanase or 10,000 U/kg phytase plus 4,000 U/kg xylanase). The basal diet contained 97.8% wheat. Pigs were placed in metabolic cages for a 7-d adaptation period followed by a 5-d total collection of feces and urine. Nutrient digestibility and available energy content were determined. Experiment 2 was conducted to evaluate the effects of pelleting and enzymes on performance of wheat for growing pigs. In this experiment, 180 growing pigs ($35.2{\pm}9.0kg\;BW$) were allocated to 1 of 6 treatments according to a $2{\times}3$ factorial treatment arrangement with the main effects being feed form (meal vs pellet) and enzyme supplementation (0, 2,500 or 5,000 U/kg xylanase). Results: In Experiment 1, there were no interactions between feed form and enzyme supplementation. Pelleting reduced the digestibility of acid detergent fiber (ADF) by 6.4 percentage units (p<0.01), increased the digestibility of energy by 0.6 percentage units (p<0.05), and tended to improve the digestibility of crude protein by 0.5 percentage units (p = 0.07) compared with diets in mash form. The addition of phytase improved the digestibility of phosphorus (p<0.01) and calcium (p<0.01) by 6.9 and 7.6 percentage units respectively compared with control group. Adding xylanase tended to increase the digestibility of crude protein by 1.0 percentage units (p = 0.09) and increased the digestibility of neutral detergent fiber (NDF) (p<0.01) compared with control group. Supplementation of the xylanase-phytase combination improved the digestibility of phosphorus (p<0.01) but impaired NDF digestibility (p<0.05) compared with adding xylanase alone. In Experiment 2, adding xylanase increased average daily gain (p<0.01) and linearly improved the feed:gain ratio (p<0.01) compared with control group. Conclusion: Pelleting improved energy digestibility but decreased ADF digestibility. Adding xylanase increased crude protein digestibility and pig performance. Phytase increased the apparent total tract digestibility of phosphorus and calcium. The combination of phytase-xylanase supplementation impaired the effects of xylanase on NDF digestibility.

Effects of Dietary Energy Concentration and Lysine on the Digestible Energy Ratio for Apparent Amino Acid Digestibility in Finishing Barrows

  • This experiment was performed to investigate the effects of two energy levels and four lysine:digestible energy (DE) ratios on the apparent digestibility of nutrients in finishing pigs. The experiment was conducted using a $2{\times}4$ randomized complete block (RCB) design with three replicates. Twenty-four cross-bred finishing barrows ((Landrace${\times}$Yorkshire)${\times}$Duroc) with an average body weight of $64.2{\pm}0.69kg$ were assigned to one of eight treatments. Each barrow was placed in an individual metabolism crate and dietary treatment and water was provided ad libitum. Diets were designed to contain lysine:ME ratios of 1.5, 1.8, 2.1 and 2.4 g/Mcal at 3.35 and 3.6 Mcal/kg of diet in a $4{\times}2$ factorial arrangement. Dry matter (DM), ash, Ca and P digestibility were not affected by energy density or lysine:DE ratios. Crude fat digestibility increased as the energy density increased from 3.35 to 3.6 Mcal of DE/kg. Increasing the lysine:DE ratio also increased crude protein digestibility. There were no interactions between energy density and lysine:DE ratio in terms of nutrient digestibility. Nitrogen excretion via feces was not affected by energy density and lysine:DE ratio, while nitrogen excretion via urine was significantly affected by energy density and lysine:DE ratio. The apparent digestibility of all amino acids except for isoluecine, arginine and aspartic acid as well as average values of essential amino (EAA), non-essential amino acids (NEAA) and total amino acid digestibility (p>0.05) were not affected by energy density. The apparent digestibility of all amino acids except for leucine, proline, alanine and tyrosine, NEAA and total amino acid digestibility were significantly affected by lysine: DE ratio (p<0.05). Interactive effects of energy and lysine:DE ratio also significantly affected amino acid digestibility except for isoleucine, alanine, cystine, leucine, phenylalanine, glutamine and proline (p<0.05). In conclusion, these results suggest that maintaining the appropriate lysine:DE ratio becomes more important as the energy density of the diet increases. Consequently, increasing the lysine:DE ratio can result in increased crude protein digestibility and urinary nitrogen excretion, although apparent protein digestibility and nitrogen excretion were not affected by energy density Furthermore, increasing the lysine:DE ratio also increased the apparent digestibility of essential amino acids, except for leucine, regardless of energy density. The optimum lysine:DE ratio for maximum essential amino acid digestibility of the $64.2{\pm}0.69kg$ pig is approximately 2.4 g of lysine/Mcal of DE.

Moderate tetrabasic zinc chloride supplementation improves growth performance and reduces diarrhea incidence in weaned pigs

  • Objective: Two experiments were conducted to evaluate tetrabasic zinc chloride (TBZC) on the health of weaned pigs, and to determine the optimal supplemental concentrations and whether dietary TBZC could replace the pharmacological concentrations of dietary zinc oxide (ZnO) to improve growth performance and decrease Zn excretion in weaned pigs. Methods: In Exp. 1, 180 weaned pigs (8.92±1.05 kg body weight [BW]) were randomly assigned to 1 of 5 treatments, including the basal diet containing 125 mg/kg zinc sulfate (ZnSO4), and the basal diet with 1,200, 1,800, 2,400, or 3,000 mg/kg TBZC supplementation. In Exp. 2, 240 weaned pigs (7.66±1.09 kg BW) were randomly assigned to 1 of 5 treatments, including a negative control diet without Zn supplementation, a positive control diet (2,250 mg/kg ZnO), and 3 experimental diets with different concentrations of TBZC supplementation (1,000, 1,250, and 1,500 mg/kg). Results: In Exp. 1, the average daily gain (ADG), feed efficiency (G:F) and diarrhea incidence responded quadratically (p<0.01) as the TBZC supplemental concentrations increased, and pigs fed 1,200 and 1,800 mg/kg TBZC showed the best growth performance. Moreover, 1,800 mg/kg TBZC supplementation showed the greatest (p<0.01) total antioxidant capacity and glutathione peroxidase activities in liver of pigs. Histopathological examination revealed lesions in heart, liver, lung and kidney, and mild or severe histological lesions mainly occurred with the supplementation of 2,400 and 3,000 mg/kg TBZC. In Exp. 2, 1,000 and 1,250 mg/kg TBZC supplementation in diets significantly (p<0.01) increased ADG and G:F of weaned pigs, reduced Zn excretion in feces, and had no effect on diarrhea-reducing compared to 2,250 mg/kg ZnO supplementation. Conclusion: The TBZC is a potential alternative to ZnO. The recommended concentration of TBZC in weaned pig diets is 1,000 to 1,250 mg/kg.

경주에서 분리된 탄저균에 대한 연구 (Investigation on Bacillus anthracis isolated from Kyong-Ju)

  • The present study was conducted to investigate results of B. anthracis isolated from Anthrax in the Kyong-Ju of Feb. 12. 1994. 1. In biochemical feature, B. anthracis was a gram-positive rod, non-motility, sporulation, capsulation. It was positive in gelatinase, starch hydrolysis, glucose. But negative in urease, arabinose, mannitol, xylose. 2. B. anthracis grew well on B4 Br A TSA after incubation for 24 hours. The organisim grew well on BA, Br. A, NA, TSA after incubation for 72 hours. The media grew well on Br A instead of BA. 3. On 5% blood agar by laboratory animal, ${\beta}$ -hemolysis was produced from 36 hours to 48 hours incubation. There was perfect ${\beta}$-hemolysis after incubation for 48 hours. On the other side ${\beta}$-hemolysis was begun on 5% goat blood agar after incubation for 60 hours. 4. In the test of antimicrobial susceptibility, B. anthracis was very sensitive to AM, CF, TE, ENR, GM, AN, DFX, S, P, TYLO, N, KM, C, E, Lins+Sp, NN, CC, CFP, CB were sensitive one by one. B. anthracis was no-sensitive to L, XNL, TIA, CL, SXT 5. B. anthracis had never sensitivity to direct inoculation of rat and chicken, after subcutanous inj. It was very sensitive to mouse and goat, hamster, guinea pig, rabbit had a sensibility one by one. 6. The dead laboratory animal which had been inoculated with B. anthracis preserved at $37^{\circ}C$ incubation, B. anthracis didn't cultivate on non-dissected animal after 80 hours but cultivate on dissected animal after 360 hours. 7. The rapidly death could cause high concentration, died from 420 after S. C. 8.
